| 2 | Formations of groundwater hydrogeochemistry in a karst system during storm events as revealed by PCA显示文摘High-frequency samples have been collected at Jiangjia Spring, the outlet of Qingmuguan underground river system (QURS) in Chongqing in late April, 2008. The variations of hydrogeochemical compositions are found responding rapidly to storm events. Principal component analysis (PCA) of the 20 variables is employed to interpret the relationships with specific processes that control the groundwater hydrogeochemical formations. Through PCA, 84.961% of the total amount information is extracted to indicate the formations of groundwater hydrogeochemical features in QURS during storm events. The first component separates the soil erosion (i.e., increases in turbidity and concentrations of Al3+, TFe, TMn, Ba2+ and NO2?), and dilution effect (i.e., decreases in specific conductance and concentrations of HCO3?, Ca2+ and Sr2+), accounting for 41.495% of the variability in the data. The second component indicates residual fertilizers and duck’s waste from farmlands (i.e., increases in specific conductance and concentrations of Na+, NO3-, PO43-,K+and Cl-), contributing to 37.449%. The dissolution of dolomite and dolomitic limestone makes up 6.017%. | 6 | 节段性小肠移植手术中采用自体髂内血管进行血管重建显示文摘背景:本研究旨在评估自体髂内动静脉可否用于异体及自体节段性小肠移植手术的血管重建。方法:2011年1月至2019年1月间34例小肠移植患者纳入研究,其中19例为亲属活体移植,15例为自体移植。收集患者临床资料、血管重建类型及术后并发症进行分析。结果:全组男性20例,女性14例,中位年龄35岁。在34例小肠移植中,22例(64.7%,其中异体移植和自体移植各11例)采用自体髂内动脉脉进行血管重建,12例(35.3%)直接行血管吻合。血管重建耗时(21˘6)min。与直接吻合组相比,血管重建组患者手术总时长(530˘226 vs 440˘116 min,P=0.208)和冷缺血时间(159˘49 vs 125˘66 min,P=0.097)均有延长的趋势,但差异并未达到统计学意义。直接吻合组术后血栓形成发生率较血管重建组有增高的趋势(16.7%vs 0%,P=0.118)。中位随访36.9个月,无一例出现静脉狭窄或假性动脉瘤。19例异体移植患者中,4例(21.1%)发生急性排斥反应,1例(5.2%)出现慢性排斥反应。结论:本组数据显示,采用自体髂内动静脉进行血管重建可有力促进小肠移植的开展,同时有望降低术后血管并发症的风险。 | Guosheng Wu Yinglun Wu Mian Wang Wentong Zhang Chaoxu Liu Tingbo Liang | 2021 | Gastroenterology Report2021,9,4: | 2 |

| 8 | Rice FLOURY ENDOSPERM 18 encodes a pentatricopeptide repeat protein required for 5′ processing of mitochondrial nad5 messenger RNA and endosperm development显示文摘Pentatricopeptide repeat(PPR) proteins, composing one of the largest protein families in plants,are involved in RNA binding and regulation of organelle RNA metabolism at the posttranscriptional level. Although several PPR proteins have been implicated in endosperm development in rice(Oryza sativa), the molecular functions of many PPRs remain obscure. Here, we identified a rice endosperm mutant named floury endosperm 18(flo18) with pleiotropic defects in both reproductive and vegetative development.Map-based cloning and complementation tests showed that FLO18 encodes a mitochondriontargeted P-type PPR protein with 15 PPR motifs.Mitochondrial function was disrupted in the flo18 mutant, as evidenced by decreased assembly of Complex I in the mitochondrial electron transport chain and altered mitochondrial morphology. Loss of FLO18 function resulted in defective 5′-end processing of mitochondrial nad5 transcripts encoding subunit 5 of nicotinamide adenine dinucleotide hydrogenase. | 11 | A novel CD81 homolog identified in lamprey, Lampetra japonica,with roles in the immune response of lamprey VLRB^+lymphocytes显示文摘The cluster of differentiation 81 (CD81),a member of the transmembrane 4 superfamily,is primarily found to be expressed in a wide variety of cells including T and B cells of vertebrates as a critical modulator.In the present study,the open reading frame of a CD81 gene homolog (Lja-CD81) was cloned in lamprey,Lampetra japonica,which is 702 bp long and encodes a protein of 233- amino acids.Although Lja-CD81 seems to be close to CD9 molecules in their full-length sequences,Lja-CD81 possesses higher identity to vertebrates CD81 than to CD9 (including a lamprey CD9)molecules in their large extracellular loops.In addition,it also possesses a myristoylation site (Met-Gly-Val-Glu-Gly-Cys-Leu-Lys)in its N-terminal region which is identical to the N-terminal regions of CD81 molecules.These data suggest that CD9 and CD81 molecules diverged no later than the emergence of jawless vertebrates.The mRNA levels of Lja-CD81 in lymphocytes and supraneural myeloid bodies were up-regulated significantly after stimulation with mixed antigens,and a similar expressional pattern of Lja-CD81 at protein level was also confirmed. However, P-type PPRs involved in both RNA splicing and editing have rarely been reported, and their underlying mechanism remains largely unknown. Here, we report a rice floury endosperm22(flo22) mutant with delayed amyloplast development in endosperm cells. Map-based cloning and complementation tests demonstrated that FLO22 encodes a mitochondrion-localized P-type PPR protein.Mutation of FLO22 resulting in defective transsplicing of mitochondrial nad1 intron 1 and perhaps causing instability of mature transcripts affected assembly and activity of complex Ⅰ, and mitochondrial morphology and function. RNA-seq analysis showed that expression levels of many genes involved in starch and sucrose metabolism were significantly down-regulated in the flo22mutant compared with the wild type, whereas genes related to oxidative phosphorylation and the tricarboxylic acid cycle were significantly upregulated. In addition to involvement in splicing as a P-type PPR protein, we found that FLO22 interacted with DYW3, a DYW-type PPR protein, and they may function synergistically in mitochondrial RNA editing. | 18 | Molecular cloning,expression pattern and phylogenetic analysis of Guanine nucleotide exchange factor Vav2 in lamprey,Lampetra japonica显示文摘The Guanine nucleotide exchange factor Vav2(Vav2) is a member of the Vav family that serves as an important regulators for the Rho family of Ras-related GTPases. In the current study, an ortholog(Lj-Vav2) of Vav2 was identified in the lamprey(Lampetra japonica). To elucidate the phylogenetic relationship of Vav2, the metazoan genome databases were analyzed to mine the ortholog of Vav. It was found that Vav2 genes were only existed in vertebrates and Lj-Vav2 was the original one found in agnathans. The evolutionary dynamics of conserved motifs of Vav2 were explored using combined amino acid sequence as markers, and it is revealed that the Calponin homology(CH) domain, Dbl-homologous(DH) domain, Pleckstrin homology(PH) domain, Cysteine-rich(C1)domains, Src homology 3(SH3) domains and Src homology 2(SH2) domain were conserved throughout the Vav2 gene family in vertebrates during gene evolution. Relative quantitative real-time PCR analysis showed that the LjVav2 was distributed in the heart, kidney, supraneural myeloid body, liver, gill and lymphocyte-like cells. The LjVav2 was found to be expressed in these tissues, and the level of which was upregulated in lymphocyte-like cells after the animal was stimulated with LPS. These results indicated that the Lj-Vav2 might be involved in the immune response of lymphocyte-like cells in lamprey.
