| 1 | Parameterizing soil organic carbon's impacts on soil porosity and thermal parameters for Eastern Tibet grasslands显示文摘This study investigates the stratification of soil thermal properties induced by soil organic carbon (SOC) and its impacts on the parameterization of the thermal properties. Soil parameters were measured for alpine grassland stations and North China flux stations, with a total of 34 stations and 77 soil profiles. Measured data indicate that the topsoils of alpine grasslands contain high SOC contents than underlying soil layers, which leads to higher soil porosity values and lower thermal conductivity and bulk density values in the topsoils. However, this stratification is not evident at the lowland stations due to low SOC contents. Evaluations against measured data show that three thermal conductivity schemes used in land surface models severely overestimate the values for soils with high SOC content (i.e. topsoils of alpine grassland), but they are better for soils with low SOC content. A new parameterization is then developed to take the impacts of SOC into account. | 5 | Development of a daily soil moisture product for the period of2002–2011 in Chinese mainland显示文摘Soil moisture is an essential climate variable(ECV) concerned widely. Due to its high spatial variability, it is costly to measure soil moisture at tens of kilometers scale. In this study, a ten-year(2002–2011) daily soil moisture dataset at 0.25° spatial resolution for Chinese mainland was produced through assimilating the Advanced Microwave Scanning Radiometer for Earth Observing System(AMSR-E) brightness temperature(TB) data into a land surface model(LSM). The obtained soil moisture data was evaluated against soil moisture-measuring networks deployed in two wet areas and one dry area of the Tibetan Plateau.The results show that for the wet areas the accuracy of the soil moisture product obtained from the assimilation is considerably higher than that of both AMSR-E official soil moisture products and land surface simulation results, and for the dry area their accuracy is comparable to each other. The spatial pattern of the soil moisture from the new product is consistent with that of soil porosity from an independent survey-based dataset, further confirming the credibility of the new product. According to this product, the transition regions in China show stronger seasonal variation of soil moisture than dry and wet regions, and drier regions have stronger inter-annual variability of soil moisture than wetter regions, particularly during transitional seasons(spring and autumn). | 6 | Self-adjusted elastic action and its CBM pool-forming effect of the high rank coal reservoir显示文摘The coal reservoir is a kind of geologic body with considerable elasticity, and its response to stress is more sensitive than the routine ones. In the pool-forming process of the coalbed methane, (CBM) the action of many external geodynamic factors in the coal reservoir is represented by the ability of CBM’s diffusion and migration in the coal reservoir. Therefore, the occurrence of the natural fissures in the coal reservoir becomes a key that determines whether the CBM pool can be formed or not. Based on the principle, the authors have designed a comprehensive method of physical modeling to study the elasticity of the coal matrix block with different coal ranks in the light of the solid to fluid coupling, have established the model of the volume deformation (self-adjusted elastic effect) of the coal reservoir under the condition of the effective stress-adsorption/desorption, and have obtained a correct understanding of the law of the elastic deformation. | 9 | Analysis of Commonly and Specifically Dysregulated Pathways in Three Women Cancers显示文摘Breast, ovarian and endometrial cancer are three most prevalent gynaecological malignancies. Identifying their common and specific biomarkers is significant for cancer prediction and therapy in females. We propose a method to identify dysregulated pathways in cancer through scoring pathways based on the molecular interaction data and genomic data. Commonly and specifically dysregulated pathways are analyzed across the above three female cancers, which have not been studied as a whole to the best of our knowledge. Our results demonstrate that all the three cancers have close relationships with Type II diabetes and cell cycle-related biology processes. Breast cancer is specifically related to immune system while ovarian cancer and endometrial cancer are associated with blood vascular-related systems such as renin-angiotensin system and coagulation system. | 10 | Detection of Porcine Epidemic Diarrhea Virus in Guangxi Province from 2011 to 2014 and Sequence Analysis of Its M Gene显示文摘Detection of pigs epidemic diarrhea virus( PEDV) was conducted on 331 piglets diarrhea fecal samples collected in Nanning,Yulin and other 12 areas of Guangxi Province from January of 2011 to April of 2014 by the method of reverse transcription-polymerase chain reaction( RT-PCR). The results showed that the positive samples of PEDV were 210 and the positive rate was 63. 44%. The clone and sequencing of M gene was carried out on 25 positive samples. PEDV reference strains were selected from Gene Bank to conduct the sequence homology alignment analysis and the phylogenetic tree of M gene. The M gene homology and amino acid sequence identity between 25 isolated strains and 51 reference strains were 96. 0%- 99. 6% and 94. 3%- 99. 6%,respectively. The genetic variation analysis of M gene showed that the genetic relationship of PEDV prevalent strains in Guangxi Province from 2013 to 2014 was close to that of the prevalent strains in Beijing,Anhui,Wuhan,Hebei and Guangdong from 2010 to 2013,and which were far from that of the Chinese early isolates CH / S( Gen Bank number: JN547228),vaccine strain CV777( Gen Bank number: AF353511) and Attenuated DR13( Gen Bank number: JQ023162). | 12 | Homocysteine aggravates cerebral hemorrhage in rats: Correlation with oxygen-free radical production and cell apoptosis in tissue surrounding hematoma显示文摘BACKGROUND:Previous studies have demonstrated that homocysteine is an independent risk factor for ischemic stroke,as determined by detection of apoptosis and oxygen-free radical scavengers following cerebral ischemia.However,the mechanisms of homocysteine remain unclear.Several reports have addressed the effects of homocysteine on ischemic stroke.OBJECTIVE:To analyze the effects of homocysteine on apoptosis,intracellular superoxide dismutase(SOD) activity,and malondialdehyde content in tissue surrounding hematoma in rats with cerebral hemorrhage,and to determine the action pathway of malondialdehyde following cerebral hemorrhage.DESIGN,TIME AND SETTING:The randomized,controlled,animal experiment was performed at the Laboratory of Molecular Biology,Hospital Affiliated to Luzhou Medical College,China from April 2007 to April 2008.MATERIALS:In situ apoptosis detection kit(Roche,Mannheim,Germany),SOD detection kit and malondialdehyde detection kit(Nanjing Jiancheng Bioengineering Institute,China),and homocysteine(Sigma,St Louis,MO,USA) were used in the present study.METHODS:A total of 75 Sprague Dawley rats were equally and randomly assigned to sham surgery,model,and homocysteine groups.Autologous blood was infused into the caudate putamen of rats to establish models of cerebral hemorrhage in model and homocysteine groups.Homocysteine was injected directly into the brain through the skull at the hematoma hemisphere at 30 minutes after model induction in the homocysteine group.MAIN OUTCOME MEASURES:At 6,12,24,and 72 hours,as well as 1 week,post-surgery,neurological deficits were observed in each group.Brain water content was measured using the dry-wet weight method.Cell apoptosis in tissue surrounding the hematoma was detected utilizing terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end labeling(TUNEL).SOD activity and malondialdehyde content in tissue surrounding the hematoma were respectively measured using the xanthine oxidase and thiobarbituric acid methods.RESULTS:Neurological function was similar between model and homocysteine groups following cerebral hemorrhage(P > 0.05).Brain water content was increased at 12 hours post-surgery,peaked at 3 days,and remained unchanged at 7 days in the model group.Brain edema was not significantly aggravated following homocysteine intervention(P > 0.05),but SOD activity significantly decreased and malondialdehyde content significantly increased(P < 0.05).The number of apoptotic cells increased in rats with cerebral hemorrhage at 12 hours(P < 0.05),and numbers peaked at 72 hours following model establishment(P < 0.05).The time of peak value was identical between model and homocysteine groups.Brain water content was negatively associated with SOD activity(rmodel group =-0.448,P < 0.05;rhomocysteine group =-0.612,P < 0.05),but was positively associated with malondialdehyde content(rmodel group = 0.542,P < 0.05;rhomocysteine group = 0.684,P < 0.05) in brain tissues surrounding the hematoma following surgery in model and homocysteine groups.CONCLUSION:Homocysteine aggravates neurological dysfunction and brain edema in rats with cerebral hemorrhage.The mechanisms of action are likely associated with production of oxygen-free radical and cellular apoptosis following cerebral hemorrhage. | Yingying Liu Xinjun Li Chun Wang Chun Chen Ping Liu Qin Feng Duanxiu Liao Xiaogang Li | 2010 | Neural Regeneration Research2010,5,13: | 3 |

| 14 | Synergistic effects of autophagy/mitophagy inhibitors and magnolol promote apoptosis and antitumor efficacy显示文摘Mitochondria as a signaling platform play crucial roles in deciding cell fate.Many classic anticancer agents are known to trigger cell death through induction of mitochondrial damage.Mitophagy,one selective autophagy,is the key mitochondrial quality control that effectively removes damaged mitochondria.However,the precise roles of mitophagy in tumorigenesis and anticancer agent treatment remain largely unclear.Here,we examined the functional implication of mitophagy in the anticancer properties of magnolol,a natural product isolated from herbal Magnolia officinalis.First,we found that magnolol induces mitochondrial depolarization,causes excessive mitochondrial fragmentation,and increases mitochondrial reactive oxygen species(mtROS).Second,magnolol induces PTEN-induced putative kinase protein 1(PINK1)-Parkin-mediated mitophagy through regulating two positive feedforward amplification loops.Third,magnolol triggers cancer cell death and inhibits neuroblastoma tumor growth via the intrinsic apoptosis pathway.Moreover,magnolol prolongs the survival time of tumor-bearing mice.Finally,inhibition of mitophagy by PINK1/Parkin knockdown or using inhibitors targeting different autophagy/mitophagy stages significantly promotes magnolol-induced cell death and enhances magnolol's anticancer efficacy,both in vitro and in vivo.Altogether,our study demonstrates that magnolol can induce autophagy/mitophagy and apoptosis,whereas blockage of autophagy/mitophagy remarkably enhances the anticancer efficacy of magnolol,suggesting that targeting mitophagy may be a promising strategy to overcome chemoresistance and improve anticancer therapy. | Yancheng Tang Liming Wang Tao Yi Jun Xu Jigang Wang Jiang-Jiang Qin Qilei Chen Ka-Man Yip Yihang Pan Peng Hong Yingying Lu Han-Ming Shen Hu-Biao Chen | 2021 | Acta Pharmaceutica Sinica B2021,11,12: | 2 |

| 19 | Study on the effects of the different polar group of EPA-enriched phospholipids on the proliferation and apoptosis in 95D cells显示文摘EPA-enriched phosphatidylcholine(EPA-PC)and EPA-enriched phosphatidylethanolamine(EPA-PE)are newly identified marine phospholipids.The polar group of phospholipids is known to influence EPA-phospholipid activity.However,the differences in anti-tumor effects between EPA-PC and EPA-PE have not been reported.In this study,we evaluated the effects of two forms of EPA on the proliferation and apoptosis in the lung-cancer cell line 95D as well as possible molecular mechanisms.Our results showed that EPA-PC effectively inhibited proliferative activity and promoted apoptosis of 95D cells in a dose-dependent manner,while EPA-PE had no effect on cell proliferation,although it slightly promoted apoptosis.Western blot results showed that EPA-PC and EPA-PE upregulated the expression of PPARy,RXRoc,and PTEN,and downregulated the PI3K/AKT signaling pathway.Furthermore,EPA-PC and EPA-PE induced the expression of the pro-apoptotic gene,Bax,and reduced the expression of the anti-apoptotic gene,Bcl-xl.Additionally,EPA-PC and EPA-PE promoted the release of cytochrome c and activated the apoptotic enzyme-cleaved caspase-3.These data suggest that the anti-tumor effect of EPA-phospholipids may be exerted via a PPARy-related mechanism.EPA-PC was more efficacious as compared to EPA-PE,which might be due to the different polar groups of phospholipids. | Yao Guo Qin Zhao Yingying Tian Yuanyuan Liu Ziyi Yan Changhu Xue Jingfeng Wang | 2021 | Marine Life Science & Technology2021,3,4: | 1 |
| 20 | Brain organoid-on-chip system to study the effects of breast cancer derived exosomes on the neurodevelopment of brain显示文摘Early human brain development can be affected by multiple prenatal factors that involve chemical exposures in utero,maternal health characteristics such as psychiatric disorders,and cancer.Breast cancer is one of the most common cancers worldwide arising pregnancy.However,it is not clear whether the breast cancer might influence the brain development of fetus.Exosomes secreted by breast cancer cells play a critical role in mediating intercellular communication and interplay between different organs.In this work,we engineered human induced pluripotent stem cells(hiPSCs)-derived brain organoids in an array of micropillar chip and probed the influences of breast cancer cell(MCF-7)derived-exosomes on the early neurodevelopment of brain.The formed brain organoids can recapitulate essential features of embryonic human brain at early stages,in terms of neurogenesis,forebrain regionalization,and cortical organization.Treatment with breast cancer cell derived-exosomes,brain organoids exhibited enhanced expression of stemness-related marker OCT4 and forebrain marker PAX6.RNA-seq analysis reflected several activated signaling pathways associated with breast cancer,medulloblastoma and neurogenesis in brain organoids induced by tumor-derived exosomes.These results suggested that breast cancer cell-derived exosomes might lead to the impaired neurodevelopment in the brain organoids and the carcinogenesis of brain organoids.It potentially implies the fetus of pregnant women with breast cancer has the risk of impaired neurodevelopmental disorder after birth. | Kangli Cui Wenwen Chen Rongkai Cao Yingying Xie Peng Wang Yunsong Wu Yaqing Wang Jianhua Qin | 2022 | Cell Regeneration2022,11,1: | 1 |
