2Yin Yang 1(YY1) synergizes with Smad7 to inhibit TGF-β signaling in the nucleus显示文摘As a prototype of the TGF-βsuperfamily cytokines,TGF-βis well known for its diverse roles in embryogenesis and adult tissue homeostasis.TGF-βevokes cellular responses by signaling mainly through cell membrane receptors and transcription factor R-Smads and Co-Smad(Smad4),while an inhibitory Smad,Smad7,acts as a critical negative regulator of TGF-βsignaling.Smad7 antagonizes TGF-βsignaling by regulating the stability or activity of the receptors or blocking the DNA binding of the functional R-Smad-Smad4 complex in the nucleus.However,the function of Smad7 in the nucleus is not fully understood.Yin Yang 1(YY1)is a ubiquitously expressed transcription factor with multiple functions.It has been reported that YY1 can inhibit Smad-dependent transcriptional responses and TGF-β/BMP-induced cell differentiation independently of its DNA binding ability.In this study,we found that Smad7 interacts with YY1 and the interaction is attenuated by TGF-βsignaling.Reporter assays and target gene expression analyses revealed that Smad7 and YY1 act in concert to inhibit TGF-β-induced transcription in the nucleus.Furthermore,Smad7 could enhance the interaction of YY1 with the histone deacetylase HDAC1.Consistently,YY1 and HDAC1 augmented the transcription repression activity of Smad7 in Gal4-luciferase reporter analysis.Therefore,our findings define a novel mechanism of Smad7 and YY1 to antagonize TGF-βsignaling.YAN XiaoHua PAN Jun XIONG WanWan CHENG MinZhang SUN YingYuan ZHANG SuPing CHEN YeGuang 2014Science China(Life Sciences)2014,57,1:10
3All-solid-state flexible asymmetric supercapacitors with high energy and power densities based on NiCo_2S_4@MnS and active carbon显示文摘Electrode material based on a novel core–shell structure consisting of NiCo_2S_4(NCS) solid fiber core and Mn S(MS) sheet shell(NCS@MS) in situ grown on carbon cloth(CC) has been successfully prepared by a simple sulfurization-assisted hydrothermal method for high performance supercapacitor. The synthesized NiCo_2S_4@Mn S/CC electrode shows high capacitance of 1908.3 F g^(-1) at a current density of 0.5 A g^(-1) which is higher than those of NiCo_2S_4 and Mn S at the same current density. A flexible all-solid-state asymmetric supercapacitor(ASC) is constructed by using NiCo_2S_4@Mn S/CC as positive electrode, active carbon/CC as negative electrode and KOH/poly(vinyl alcohol)(PVA) as electrolyte. The optimized ASC shows a maximum energy density of 23.3 Wh kg^(-1) at 1 A g^(-1), a maximum power density of about7.5 kw kg^(-1) at 10 A g^(-1) and remarkable cycling stability. After 9000 cycles, the ASC still exhibited67.8% retention rate and largely unchanged charge/discharge curves. The excellent electrochemical properties are resulted from the novel core–shell structure of the NiCo_2S_4@Mn S/CC electrode, which possesses both high surface area for Faraday redox reaction and superior kinetics of charge transport. 5Effect of electroacupuncture at Zusanli(ST 36) on dorsal root nerve signals in normal rats显示文摘OBJECTIVE: To investigate how electroacupuncture (EA) at Zusanli (ST 36) with varying intensities of stimulation affects dorsal root nerve signals in normal rats. METHODS: Adult female Wistar rats were examined after drug-induced anesthesia and isolation of the L4 dorsal root associated with the Zusanli (ST 36) acupoint, using bipolar platinum electrodes. We applied EA at Zusanli (ST 36) with a continuous wave waveform for 1 min, and recorded action potentials in the L4 spinal cord dorsal root nerve with a frequency of 5 Hz and various current intensity levels (approximately 1-4 mA). In addition, we tested EA with a frequency of 15 Hz and current intensity of 1-4 mA. The L4 spinal cord dorsal nerve bundle action potentials were measured for 1 min of EA. To analyze the discharge frequency, we used the maximum Lyapunov exponent and Lempel-Ziv (LZ) complexity. RESULTS: At a fixed frequency, with increasing intensity of EA, dorsal root nerve filament discharge frequency revealed an initial increase, followed by a decreasing trend. A stimulation intensity of 3 mA induced a significantly greater discharge frequency, compared with stimulation intensities of 2 and 4 mA. EA stimulation evoked neuroelectric signals with chaotic characteristics. Increased intensity led to an initial increase in LZ complexity which then decreased, with a stimulation strength of 3 mA inducing the highest level of LZ complexity. 8Rapid and sensitive detection of urinary KIM-1 using fully printed photonic crystal microarrays显示文摘Urinary kidney injury molecule 1(uKIM-1)serves as a reliable marker for the early diagnosis of acute kidney injury(AKI).The rapid and facile detection of changes in uKIM-1 is essential for early AKI diagnosis,ultimately improving the prognosis of patients.In this study,we developed a fully printed photonic crystal-integrated microarray with photonic crystal-enhanced fluorescence properties,which can detect uKIM-1 levels at the point-of-care.We confirmed its efficacy in the early diagnosis of AKI using clinical urine specimens.Direct quantitative detection of uKIM-1 was achieved within 10 min.The lowest limit of detection is 8.75 pg·mL^(-1) with an accuracy of 94.2%.The diagnostic efficacy was validated using 86 clinical urine samples,highlighting the high sensitivity and stability of the photonic crystal microarray.Consequently,a facile and reliable immunoassay was designed and prepared for the rapid quantitative detection of uKIM-1,which is crucial for the early identification and convenient detection of AKI in hospital or community settings.Rapid,convenient,cost-effective,and long-term monitoring of changes in uKIM-1 levels can assist clinicians in making timely adjustments to treatment regimens,preventing the transition from AKI to chronic kidney disease(CKD),improving the quality of life of patients with AKI,and reducing healthcare costs.It highlights the advantages of utilizing urine samples as a noninvasive and easily accessible medium for early detection and monitoring of kidney-related conditions.Yang Liu Xuwei He Zewei Lian Qian Guo Jimei Chi Xiaoxue Lin Liyue Zhang Zheng Liu Yingyuan Liu Meng Su Keyu Wang Qiangguo Ao Qingli Cheng 2024Nano Research2024,17,5:0

10Correlation between current intensity and effective duration in the treatment of botulinus toxin for cerebral palsy by myoelectricity locating显示文摘BACKGROUND: Researches prove that the treatment of cerebral palsy by botulinus toxin (BTX-A) can improve effect and prolong effective duration. Current intensity is the important factor during injection.OBJECTIVE: To observe the changes of motor function and muscular intensity of children with spastic cerebral palsy after injecting BTX-A and analyze correlation between current intensity and effective duration. DESIGN: Cases control trial before and after nerves block.SETTING: Capital University of Medical Sciences, China Rehabilitation Research Center.PARTICIPANTS: From June 2002 to November 2004, 14 children with spastic cerebral palsy were treated by BTX-A block. All children were hospitalized in the Children Rehabilitation Department of China Rehabilitation Research Center. The children included 9 male and 5 female, and ranged from 4 to 13 years old, and average age was (6±2) years. Muscular tension ranged from grade 1 to grade 3. The diagnosis and the tape of cerebral palsy based on standard of the Fist Nationwide Cerebral Palsy Symposium, and all children were diagnosed with electroencephalogram (EEG), CT and MRI, and permitted by their guardians.METHODS: ① Locating and calculation: To locate block points by G6805-2A electro-therapeutic apparatus (Shanghai Huayi Electronic Instrument Plant) at the least stimulating current (continuous wave; impulse frequency; 2.667-83.333 Hz; current intensity: 0-6 mA; voltage: 6 V) to touch off muscles contraction. The current intensity of each point was recorded, and the average current intensity of each patient was calculated at the same time. ② Dose of BTX-A: Basing upon the spastic degree and weight of patients, the dose was made certain: dose (IU)=(scores of Modified Ashworth Scale +2.5) ×weight (kg). The number of the block points was in all 4. The dose of injection ranged from 50 IU to 160 IU with the average of (73.6±25.8) IU. The BTX-A was made by Lanzhou Institute of Biological Products, and each bottle contains 100 IU BTX-A. Muscular tension was evaluated by Modified Ashworth Scale. The scale ranged from grade 0 to grade 4, and the scores were from 0 to 4. The higher the scores were, the higher the muscular tension was. ③ Effect: Changes of muscular tension were evaluated by modified Ashworth Scale before and after block. Motor function was evaluated by Physician Rating Scale (PRS) before and after treatment. It contained Gait pattern, Hindfoot (ankle) position (stance-floor contact), Hindfoot position (foot strike), Knee position (degree of recurvation), Crouch and Speed of gait. The scores ranged from 0 to 14. The higher the points were, the better the motor function was. ④ Effective duration: The duration was definited by the recovering of the Modified Ashworth Scale. ⑤ Statistic analysis: Firstly, the current intensity and the effect duration were analyzed by One-Sample Kolmogorov-Smirnov Test, the current intensity: Z = 0.456, P = 0.985, the effective duration: Z = 0.557, P = 0.915. Both data were normal distribution. Secondly, both data were analyzed by Linear Regression. The efficiency of the BTX-A block was analyzed by paired-samples t test.MAIN OUTCOME MEASURES: ① Changes of motor function of muscular tension; ② correlation between current intensity and effective duration. RESULTS: Fourteen children with spastic cerebral palsy were all involved in the final analysis. ① Therapeutic effect: The average score of Ashworth scale after block was lower than that before block [(1.02±0.34) points vs. (2.12±0.48) points, t = 3.644, P < 0.01]. The average score of RPS after block was higher than that before block [(9.75±2.78) points vs. (6.16±0.58) points, t =13.222, P < 0.01]. ② Relation between the current intensity and the effective duration: The current intensity was (0.1857±0.0506) mA, and the effective duration was (26.36±4.48) weeks.
