| 1 | Characterization of M2 antibodies in asymptomatic Chinese population显示文摘AIM: To investigate the presence of M2 antibodies specific for pdmary biliary cirrhosis (PBC) in asymptomatic Chinese and identify patients with early PBC.METHODS: Enzyme-linked immunosorbent assay (ElISA)tests for M2 antibodies to recombinant protein were performed in 5 011 subjects (age range, 26-85 years; mean age: 45.81±15.02 years) who took an annual physical examination. M2-positive subjects were further analyzed for immunoglobulin (Ig) classes and subclasses of M2 antibodies.Clinical, biochemical and immunological data were obtained for M2-positive subjects. In addition, ultrasonography (US)or endoscopic retrograde cholangio-pancreatography (ERCP)was performed to exclude any disorders other than PBC.RESULTS: M2 antibodies were detected in 8 (0.16%) of the 5 0LL subjects studied. Of the 8 subjects, 7 were female and 1 was male (age range: 40-74 years). An unexplained increase of serum alkaline phosphatase (ALP) and gamma glutamyl transpeptidase (γ-GT) values, often to striking levels,was detected in 4 M2-positive subjects, 3 of them accorded with the diagnostic criteria recommended by the American Association for the Study of Liver Diseases, even though they had no symptoms of PBC (such as fatigue, pruritus or jaundice).Liver biopsy was performed in two M2-positive subjects and the histology was compatible with PBC in both cases.CONCLUSION: Our data, while not assessing the true prevalence of asymptomatic PBC in the general population,suggest that asymptomatic PBC is much more common in China than has been supposed. | Xiao-HuaJiang Ren-QianZhong Xiao-YunFan YinHu FengAn Jian-WenSun Xian-TaoKong | 2003 | World Journal of Gastroenterology2003,9,9: | 20 |
| 2 | Construction and expression of a humanized M_2 autoantigen trimer and its application in the diagnosis of primary biliary cirrhosis显示文摘AIM: To construct and express a humanized M2 autoantigen trimer designated as BPO and to apply it in the diagnosis of primary biliary cirrhosis (PBC).
METHODS: cDNA fragments encoding M2-reactive epitopes of pyruvate dehydrogenase complex Ez (PDCE2), branched chain 2-oxo-acid dehydrogenase complex E2 (BCOADC-E2) and 2-oxo-glutarate dehydrogenase complex E2 (OGDC-E2) were amplified with PCR using total RNA extracted from human peripheral mononuclear blood cells. The fragments were cloned into the plasmid vector pQE-30 and then transferred into E. coliM15 (pREP4) for expression, which was induced by isopropylthio-β-Dgalactoside. The expressed recombinant BPO protein was demonstrated by SDS-PAGE, Western-blotting and Immunoabsorption test, its antigenic reactivity and specificity were identified with seven M2-positive sera confirmed at Euroimmun Research Center (Germany).Using the purified BPO, M2 antibodies in sera from patients with PBC and other liver related diseases were detected with ELISA.
RESULTS: The expressed BPO was observed with both antigenic reactivity and specificity of M2 autoantigens. The determination of M2 antibodies by BPO with ELISA was more sensitive than using the Euroimmun's kit with the coefficients of variation less than 10 % in both interassay and intraassay.With the newly established method, M2 antibodies were found in 100 % (20/20) of patients with PBC. Six cases of liver disease with unknown etiology and 1 patient with drug induced liver injury had detectable levels of serum M2antibodies. There were also 2 patients with autoimmune cholangitis and 1 with autoimmune hepatitis showing M2-antibody positive.

| 3 | Statistical and frequency analysis of the pressure fluctuation in a fluidized bed of non-spherical particles显示文摘In this paper,the pressure fluctuation in a fluidized bed was measured and processed via standard deviation and power spectrum analysis to investigate the dynamic behavior of the transition from the bubbling to turbulent regime.Two types[Geldart B and D) of non-spherical particles,screened from real bed materials,and their mixture were used as the bed materials.The experiments were conducted in a semiindustrial testing apparatus.The experimental results indicated that the fluidization characteristics of the non-spherical Geldart D particles differed from that of the spherical particles at gas velocities beyond the transition velocity U_c.The standard deviation of the pressure fluctuation measured in the bed increased with the gas velocity,while that measured in the plenum remained constant.Compared to the coarse particles,the fine particles exerted a stronger influence on the dynamic behavior of the fluidized bed and promoted the fluidization regime transition from bubbling toward turbulent.The power spectrum of the pressure fluctuation was calculated using the auto-regressive(AR) model;the hydrodynamics of the fluidized bed were characterized by the major frequency of the power spectrum of the pressure fluctuation.By combining the standard deviation analysis,a new method was proposed to determine the transition velocity U_k via the analysis of the change in the major frequency.The first major frequency was observed to vary within the range of 1.5 to 3 Hz. | Honghao He Xiaofeng Lu Wei Shuang Quanhai Wang Yinhu Kang Liyun Yan Xuanyu Ji Guangyu Luo Hai Liu | 2014 | Particuology2014,12,5: | 6 |

| 6 | Ozonation as an efficient pretreatment method to alleviate reverse osmosis membrane fouling caused by complexes of humic acid and calcium ion显示文摘Humic acid has been considered as one of the most significant sources in feed water causing organic fouling of reverse osmosis (RO) membranes, but the relationship between the fouling behavior of humic acid and the change of its molecular structure has not been well developed yet. In this study, the RO membrane fouling behavior of humic acid was studied systematically with ozonation as a pretreatment method to control RO membrane fouling. Furthermore, the effect of ozone on the structure ofhumic acid was also explored to reveal the mechanisms. Humic acid alone (10-90 mg/L, in deionized water) was found not to cause obvious RO membrane fouling in 45-h operation. However, the presence of Ca^2+ aggravated significantly the RO membrane fouling caused by humic acid, with significant flux reduction and denser fouling layer on RO membrane, as it was observed by scanning electron microscope (SEM) and atomic force microscope (AFM). However, after the pretreatment by ozone, the influence of Ca^2+ was almost eliminated. Further analysis revealed that the addition of Ca^2+ increased the particle size of humic acid solution significantly, while ozonation reduced the SUVA254, particle size and molecular weight of the complexes of humic acid and Ca^2+(HA-Ca^2+ complexes). According to these results and literature, the bridge effect of Ca^2+ aggregating humic acid molecules and the cleavage effect of ozone breaking HA-Ca^2+ complexes were summarized.
