2Transcriptomic landscape regulated by the 14 types of bone morphogenetic proteins(BMPs)in lineage commitment and differentiation of mesenchymal stem cells(MSCs)显示文摘Mesenchymal stem cells(MSCs)are ubiquitously-existing multipotent progenitors that can self-renew and differentiate into multiple lineages including osteocytes,chondrocytes,adipocytes,tenocytes and myocytes.MSCs represent one of the most commonly-used adult progenitors and serve as excellent progenitor cell models for investigating lineagespecific differentiation regulated by various cellular signaling pathways,such as bone morphogenetic proteins(BMPs).As members of TGFb superfamily,BMPs play diverse and important roles in development and adult tissues.At least 14 BMPs have been identified in mammals.Different BMPs exert distinct but overlapping biological functions.Through a comprehensive analysis of 14 BMPs in MSCs,we demonstrated that BMP9 is one of the most potent BMPs in inducing osteogenic differentiation of MSCs.Nonetheless,a global mechanistic view of BMP signaling in regulating the proliferation and differentiation of MSCs remains to be fully elucidated.Here,we conducted a comprehensive transcriptomic profiling in the MSCs stimulated by 14 types of BMPs.Hierarchical clustering analysis classifies 14 BMPs into three subclusters:an osteo/chondrogenic/adipogenic cluster,a tenogenic cluster,and BMP3 cluster.We also demonstrate that six BMPs(e.g.,BMP2,BMP3,BMP4,BMP7,BMP8,and BMP9)can induce ISmads effectively,while BMP2,BMP3,BMP4,BMP7,and BMP11 up-regulate Smad-independent MAP kinase pathway.Furthermore,we show that many BMPs can upregulate the expression of the signal mediators of Wnt,Notch and PI3K/AKT/mTOR pathways.While the reported transcriptomic changes need to be further validated,our expression profiling represents the first-of-its-kind to interrogate a comprehensive transcriptomic landscape regulated by the 14 types of BMPs in MSCs.Linghuan Zhang Qing Luo Yi Shu Zongyue Zeng Bo Huang Yixiao Feng Bo Zhang Xi Wang Yan Lei Zhenyu Ye Ling Zhao Daigui Cao Lijuan Yang Xian Chen Bin Liu William Wagstaff Russell R*Reid Hue H*Luu Rex C*Haydon Michael J*Lee Jennifer Moriatis Wolf Zhou Fu Tong-Chuan He Quan Kang 2019Genes & Diseases2019,6,3:11
3Highly expressed BMP9/GDF2 in postnatal mouse liver and lungs may account for its pleiotropic effects on stem cell differentiation,angiogenesis,tumor growth and metabolism显示文摘Bone morphogenetic protein 9(BMP9)(or GDF2)was originally identified from fetal mouse liver cDNA libraries.Emerging evidence indicates BMP9 exerts diverse and pleiotropic functions during postnatal development and in maintaining tissue homeostasis.However,the expression landscape of BMP9 signaling during development and/or in adult tissues remains to be analyzed.Here,we conducted a comprehensive analysis of the expression landscape of BMP9 and its signaling mediators in postnatal mice.By analyzing mouse ENCODE transcriptome datasets we found Bmp9 was highly expressed in the liver and detectable in embryonic brain,adult lungs and adult placenta.We next conducted a comprehensive qPCR analysis of RNAs isolated from major mouse tissues/organs at various ages.We found that Bmp9 was highly expressed in the liver and lung tissues of young adult mice,but decreased in older mice.Interestingly,Bmp9 was only expressed at low to modest levels in developing bones.BMP9-associated TGFβ/BMPR type I receptor Alk1 was highly expressed in the adult lungs.Furthermore,the feedback inhibitor Smads Smad6 and Smad7 were widely expressed in mouse postnatal tissues.However,the BMP signaling antagonist noggin was highly expressed in fat and heart in the older age groups,as well as in kidney,liver and lungs in a biphasic fashion.Thus,our findings indicate that the circulating BMP9 produced in liver and lungs may account for its pleiotropic effects on postnatal tissues/organs although possible roles of BMP9 signaling in liver and lungs remain to be fully understood.Wei Liu Zhongliang Deng Zongyue Zeng Jiaming Fan Yixiao Feng Xi Wang Daigui Cao Bo Zhang Lijuan Yang Bin Liu Mikhail Pakvasa William Wagstaff Xiaoxing Wu Huaxiu Luo Jing Zhang Meng Zhang Fang He Yukun Mao Huiming Ding Yongtao Zhang Changchun Niu Rex C.Haydon Hue H.Luu Jennifer Moriatis Wolf Michael J.Lee Wei Huang Tong-Chuan He Yulong Zou 2020Genes & Diseases2020,7,2:9
4Characterization of the essential role of bone morphogenetic protein 9 (BMP9) in osteogenic differentiation of mesenchymal stem cells (MSCs) through RNA interference显示文摘Mesenchymal stem cells(MSCs)are multipotent stem cells and capable of differentiating into multiple cell types including osteoblastic,chondrogenic and adipogenic lineages.We previously identified BMP9 as one of the most potent BMPs that induce osteoblastic differentiation of MSCs although exact molecular mechanism through which BMP9 regulates osteogenic differentiation remains to be fully understood.Here,we seek to develop a recombinant adenovirus system to optimally silence mouse BMP9 and then characterize the important role of BMP9 in osteogenic differentiation of MSCs.Using two different siRNA bioinformatic prediction programs,we design five siRNAs targeting mouse BMP9(or simB9),which are expressed under the control of the converging H1 and U6 promoters in recombinant adenovirus vectors.We demonstrate that two of the five siRNAs,simB9-4 and simB9-7,exhibit the highest efficiency on silencing exogenous mouse BMP9 in MSCs.Furthermore,simB9-4 and simB9-7 act synergistically in inhibiting BMP9-induced expression of osteogenic markers,matrix mineralization and ectopic bone formation from MSCs.Thus,our findings demonstrate the important role of BMP9 in osteogenic differentiation of MSCs.The characterized simB9 siRNAs may be used as an important tool to investigate the molecular mechanism behind BMP9 osteogenic signaling.Our results also indicate that recombinant adenovirus-mediated expression of siRNAs is efficient and sustained,and thus may be used as an effective delivery vehicle of siRNA therapeutics.Shujuan Yan Ruyi Zhang Ke Wu Jing Cui Shifeng Huang Xiaojuan Ji Liping An Chengfu Yuan Cheng Gong Linghuan Zhang Wei Liu Yixiao Feng Bo Zhang Zhengyu Dai Yi Shen Xi Wang Wenping Luo Bo Liu Rex C.Haydon Michael J.Lee Russell R.Reid Jennifer Moriatis Wolf Qiong Shi Hue H.Luu Tong-Chuan He Yaguang Weng 2018Genes & Diseases2018,5,2:8
5Mesoporous silica SBA-15 functionalized with phosphonate and amino groups for uranium uptake显示文摘Mesoporous silicas have a very attractive ability of sorption and enrichment of metal ions due to their huge surface area and facile functionalization by organic ligands.In this work,phosphonate-amino bifunctionalized mesoporous silica SBA-15(PA-SBA-15) as U(VI) sorbent was fabricated through post-grafting method.The obtained mesoporous silica was characterized by SEM,XRD,NMR and nitrogen sorption/desorption experiments,which revealed the existence of ordered mesoporous structure with uniform pore diameter and large surface area.The adsorptivity of PA-SBA-15 for U(VI) from aqueous solution was investigated using batch sorption technique under different experimental conditions.The preliminary results show that the U(VI) sorption by PA-SBA-15 is very quick with equilibrium time of less than 1 h,and the U(VI) uptake is as large as 373 mg/g at pH 5.5 under 95 ℃.The sorption isotherm has been successfully modeled by the Langmuir isotherm,suggesting a monolayer homogeneous sorption of U(VI) in PA-SBA-15.The sorption is pH-dependent due to the pH-dependent charge of sorbent in the aqueous solution.The thermodynamics research shows that the sorption is a feasible and endothermic process.Based on these results,PA-SBA-15 could be a promising solid phase sorbent for highly-efficient removal of U(VI) ions from waste water and enrichment of U(VI) from a solution at a very low level.WANG XiaoLiang YUANLiYong WANG YanFei LI ZiJie LIU YaLan FENG YiXiao ZHAO YuLiang CHAI ZhiFang SHI WeiQun 2012Science China Chemistry2012,55,9:6
6Establishment and functional characterization of the reversibly immortalized mouse glomerular podocytes(imPODs)显示文摘Glomerular podocytes are highly specialized epithelial cells and play an essential role in establishing the selective permeability of the glomerular filtration barrier of kidney.Maintaining the viability and structural integrity of podocytes is critical to the clinical management of glomerular diseases,which requires a thorough understanding of podocyte cell biology.As mature podocytes lose proliferative capacity,a conditionally SV40 mutant tsA58-immortalized mouse podocyte line(designated as tsPC)was established from the Immortomouse over 20 years ago.However,the utility of the tsPC cells is hampered by the practical inconvenience of culturing these cells.In this study,we establish a user-friendly and reversibly-immortalized mouse podocyte line(designated as imPOD),on the basis of the tsPC cells by stably expressing the wildtype SV40 T-antigen,which is flanked with FRT sites.We show the imPOD cells exhibit long-term high proliferative activity,which can be effectively reversed by FLP recombinase.The imPOD cells express most podocyte-related markers,including WT-1,Nephrin,Tubulin and Vinculin,but not differentiation marker Synaptopodin.The imPOD cells do not form tumor-like masses in vivo.We further demonstrate that TGFb1 induces a podocyte injury-like response in the FLP-reverted imPOD cells by suppressing the expression of slit diaphragm-associated proteins P-Cadherin and ZO-1 and upregulating the expression of mesenchymal markers,a-SMA,Vimentin and Nestin,as well as fibrogenic factors CTGF and Col1a1.Collectively,our results strongly demonstrate that the newly engineered im-POD cells should be a valuable tool to study podocyte biology both under normal and under pathological conditions.Xinyi Yu Liqun Chen Ke Wu Shujuan Yan Ruyi Zhang Chen Zhao Zongyue Zeng Yi Shu Shifeng Huang Jiayan Lei Xiaojuan Ji Chengfu Yuan Linghuan Zhang Yixiao Feng Wei Liu Bo Huang Bo Zhang Wenping Luo Xi Wang Bo Liu Rex C.Haydon Hue H.Luu Tong-Chuan He Hua Gan 2018Genes & Diseases2018,5,2:5
7Inequality of Opportunity in China's Labor Earnings: The Gender Dimension显示文摘This paper investigates the inequality of opportunity in China's labor earnings,defined as the component of inequality determined by personal circumstances that lie beyond the control of an individual,of which gender is one,as opposed to the component determined by personal efforts.Using the Survey of Women's Social Status in China (2010),we measure the share of inequality of opportunity in the total inequality of individual labor earnings for people aged 26-55 years,and separately for six birth cohorts and for female and male subsamples.Gender is revealed as the single most important circumstance determining nationwide individual labor earnings,with one's region of residence, father's occupation,father's education,birth cohort and holding rural or urban hukou also playing significant roles.A further investigation into the roles of circumstances and personal efforts (including education level,occupation,Communist Party membership, migration and marital status)confirms that circumstances play.an alarmingly high role in shaping labor earnings distribution in China,and reveals notable gender differences that cannot be attributed to personal effort alone.These results provide the basis for recommending ways to improve gender equality of opportunity in the future.Jane Golley Yixiao Zhou Meiyan Wang 2019China & World Economy2019,27,1:5

9PSO-based Optimization for Constant-current Charging Pattern for Li-ion Battery显示文摘A particle swarm optimization algorithm to search for an optimal five-stage constant-current charge pattern is proposed.The goal is to maximize the objective function for the proposed charge pattern based on the charging capacity,time,and energy efficiency,which all share the same weight.Firstly,an equivalent circuit model is built and battery parameters are identified.Then the optimal five-stage constant-current charge pattern is searched using a particle swarm optimization algorithm.At last,comparative experiments using the constant current-constant voltage(CC-CV)method are performed.Although the charging SOC of the proposed charging pattern was 2.5%lower than that of the CC-CV strategy,the charging time and charging energy efficiency are improved by 15.6%and 0.47%respectively.In particular,the maximum temperature increase of the battery is approximately 0.8℃lower than that of the CC-CV method,which indicates that the proposed charging pattern is more secure.Yixiao Wang Yong Li Li Jiang Yuduo Huang Yijia Cao 2019Chinese Journal of Electrical Engineering2019,5,2:4
10A 70 Gbps NRZ optical link based on 850 nm band-limited VCSEL for data-center intra-connects显示文摘Short-reach optical interconnects among servers in data centers have attracted extensive studies recently. High capacity and low cost are two key problems for optical link. In this paper, we demonstrate a band-limited 850 nm vertical cavity surface emitting laser(VCSEL) based optical transmission system. The optical link realizes 70 Gb/s(65 Gb/s net rate) non-return-to-zero(NRZ) signal transmission over 11 m and20 m OM4 multimode fiber(MMF), with the help of equalization for time domain interference elimination.The utilized VCSEL has a bandwidth of only 18 GHz, meeting the principle of low cost. 11Safety and efficacy of Yupingfeng granules in children with recurrent respiratory tract infection:A randomized clinical trial显示文摘Importance:Recurrent respiratory tract infection(RRTI)is common in children.Inappropriate RRTI treatment will lead to asthma and other diseases,thereby seriously affecting the growth and physical health of children.Immune function modulation can prevent and alleviate childhood RRTI.Yupingfeng(YPF),a patented traditional Chinese medicine(TCM),has immunomodulatory effects and is widely used in China to treat children with RRTI.Objective:To evaluate the safety and efficacy of YPF monotherapy in treating children with RRTI.Methods:This multicenter,randomized,double-blind,double-simulation,noninferiority clinical trial was conducted from January 2015 to August 2017,with an 8-week treatment period and 52-week follow-up after the drug withdrawal.Children aged 2–6 years with RRTI meeting the inclusion and exclusion criteria were enrolled in 13 hospitals in China and divided randomly into three groups(2:2:1 ratio)to receive YPF,pidotimod,or placebo.The primary outcome was the proportion of RRTI returning to normal standard level during the follow-up.The secondary outcomes were reduction in the number of RRTI recurrences,effect on clinical symptoms(in accord with TCM practice),effect per symptom,and safety.The trial was registered at the Chinese Clinical Trials Registry(www.chictr.org.cn)under the unique identifier ChiCTR-IPR-15006847.Results:Three hundred and fifty-one children were enrolled and randomly assigned to 3 groups;124,125,and 61 children in the YPF,pidotimod,and placebo groups,respectively,had completed the trial.During the follow-up,the proportion of RRTI returning to normal standard level was 73.13%,67.15%,and 38.81%with YPF,pidotimod,and placebo,respectively(P<0.0001).The proportion of cases who returned to normal standard level in the YPF group was 34.32%higher than that in the placebo group.The safety profile did not significantly differ among the groups.Interpretation:YPF granules were noninferior to the active control drug pidotimod oral solution for the treatment of RRTI in children,and were superior to placebo,with a high safety profile.Baoping Xu Xinmin Li Siyuan Hu Yixiao Bao Fengmei Chen Zhimin Chen Yonggang Du Enmei Liu Yufeng Liu Qinghui Mou Baoling Su Bo Wang Jianwen Xu Guiping Xu Qiaozhi Yang Liwei Gao Xiaohui Liu Lei Li Rong Ma Kunling Shen 2022Pediatric Investigation2022,6,2:3
12Patterns and dietary intake of polychlorinated dibenzo-p-dioxins and polychlorinated dibenzofurans in food products in China显示文摘The health risk of polychlorinated dibenzo-p-dioxins and dibenzofurans(PCDD/Fs) and dioxin-like PCBs(d1-PCBs) to human being should be assessed regularly.To evaluate the contamination levels in various food products in the Chinese market and to assess the dietary exposure of the Chinese population,11 varieties of food groups totaling 534 samples including beef and mutton,chicken and duck,pork,fish and seafood,milk and dairy products were evaluated.The average concentrations of PCDD/Fs in all groups ranged from0.291 to 8.458 pg/g whole weight(w.w.).The average toxic equivalency concentrations were from 0.012 pg TEQ/g w.w.for cereal to 0.357 pg TEQ/g fat for marine oil.OCDD and2,3,7,8-TCDF were the dominant congeners in foodstuffs.The dietary estimated mean intake for the Chinese rural and urban populations were 0.555 and 0.514 pg TEQ/kg body weight/day,respectively,however,the cereal group exposure were higher to the estimate daily intake and contributed 81%for rural and 48%for urban population,followed by fish and seafood which contributed 4%and 15%to the estimate daily intake.The estimated dietary intakes were compared with the toxicological reference values and showed that both rural and urban populations were well below those values.Lingyun Wang Gangdou Ding Zhiguang Zhou Xun Liu Yixiao Wang Heidi Qunhui Xie Tuan Xu Pu Wang Bin Zhao 2017Journal of Environmental Sciences2017,29,1:3
13Level and characteristics of polychlorinated dibenzo-p-dioxins and dibenzofurans in feed and feed additives显示文摘Feed security is a prerequisite for safe animal food products.In this study,13 groups of feed and feed ingredients,totaling 2067 samples,were collected in the period of 2011 to 2014 from China.The highest mean level of polychlorinated dibenzo-p-dioxins and dibenzofurans(PCDD/Fs) was found in fish meals and shell powders,with a concentration of 60.35 ng/kg,followed by mineral origin materials.In terms of the toxicity equivalent concentration,the fish oil group showed the highest PCDD/F levels because of their bio-accumulation through the aquatic food chain,with an average concentration of 1.26 ng WHO-TEQ/kg,while the lowest level was observed in compound feed for chickens and pigs,with an average value of0.16 ng WHO-TEQ/kg.OCDD and OCDF were the predominant congeners in all groups except fish oils,in which the primary congeners were 2,3,4,7,8-PeCDF and 2,3,7,8-TCDF.For zinc chloride samples,different from other zinc-based compound samples,the main congeners were 1,2,3,4,6,7,8-HpCDF(17%),1,2,3,4,7,8,9-HpCDF(15%),1,2,3,4,7,8-HxCDF(12%) and OCDF(30%).Considering toxicity equivalency factors,the dominant congeners were 2,3,4,7,8-PeCDF,1,2,3,4,7,8-HxCDF,2,3,7,8-TCDF and 1,2,3,7,8-PeCDD,and the contribution to the total TEQwas29%,16%,14%and 12%,respectively.Overall,2.1%(43 out of 2067) of all the analyzed samples exceeded the different individual 'European Union maximum limited levels for PCDD/Fs.This study is beneficial for the determination of the status of contamination levels of feed and feed ingredients.Lingyun Wang Gangdou Ding Zhiguang Zhou Xun Liu Yixiao Wang HeidQunhui Xie Tuan Xu Pu Wang Bin Zhao 2017Journal of Environmental Sciences2017,29,1:2
14A lake ice phenology dataset for the Northern Hemisphere based on passive microwave remote sensing显示文摘Lake ice phenology(LIP)is an essential indicator of climate change and helps with understanding of the regional characteristics of climate change impacts.Ground observation records and remote sensing retrieval products of lake ice phenology are abundant for Europe,North America,and the Tibetan Plateau,but there is a lack of data for inner Eurasia.In this work,enhanced-resolution passive microwave satellite data(PMW)were used to investigate the Northern Hemisphere Lake Ice Phenology(PMW LIP).The Freeze Onset(FO),Complete Ice Cover(CIC),Melt Onset(MO),and Complete Ice Free(CIF)dates were derived for 753 lakes,including 409 lakes for which ice phenology retrievals were available for the period 1978 to 2020 and 344 lakes for which these were available for 2002 to 2020.Verification of the PMW LIP using ground records gave correlation coefficients of 0.93 and 0.84 for CIC and CIF,respectively,and the corresponding values of the RMSE were 11.84 and 10.07 days.The lake ice phenology in this dataset was significantly correlated(P<0.001)with that obtained from Moderate Resolution Imaging Spectroradiometer(MODIS)data-the average correlation coefficient was 0.90 and the average RMSE was 7.87 days.The minimum RMSE was 4.39 days for CIF.The PMW is not affected by the weather or the amount of sunlight and thus provides more reliable data about the freezing and thawing process information than MODIS observations.The PMW LIP dataset pro-vides the basic freeze-thaw data that is required for research into lake ice and the impact of climate change in the cold regions of the Northern Hemisphere.The dataset is available at http://www.doi.org/10.11922/sciencedb.j00076.00081.Xingxing Wang Yubao Qiu Yixiao Zhang Juha Lemmetyinen Bin Cheng Wenshan Liang Matti Leppäranta 2022Big Earth Data2022,6,4:2

16Revealing the correlation between structure evolution and electrochemical performance of high-voltage lithium cobalt oxide显示文摘Lithium cobalt oxide(LCO)is the dominating cathode materials for lithium-ion batteries(LIBs)deployed in consumer electronic devices for its superior volumetric energy density and electrochemical performances.The constantly increasing demands of higher energy density urge to develop high-voltage LCO via a variety of strategies.However,the corresponding modification mechanism,especially the influence of the long-and short-range structural transitions at high-voltage on electrochemical performance,is still not well understood and needs further exploration.Based on ss-NMR,in-situ X-ray diffraction,and electrochemical performance results,it is revealed that the H3 to H1-3 phase transition dictates the structural reversibility and stability of LCO,thereby determining the electrochemical performance.The introduction of La and Al ions could postpone the appearance of H1-3 phase and induce various types of local environments to alleviate the volume variation at the atomic level,leading to better reversibility of the H1-3 phase and smaller lattice strain,and significantly improved cycle performance.Such a comprehensive long-range,local,and electronic structure characterization enables an in-depth understanding of the structural evolution of LCO,providing a guiding principle for developing high-voltage LCO for high energy density LIBs.Jiajia Wan Jianping Zhu Yuxuan Xiang Guiming Zhong Xiangsi Liu Yixiao Li Kelvin H.L.Zhang Chaoyu Hong Jianming Zheng Kai Wang Yong Yang 2021Journal of Energy Chemistry2021,30,3:1

18Bioinspired therapeutic platform based on extracellular vesicles for prevention of arterial wall remodeling in hypertension显示文摘Arterial stiffness due to the vessel remodeling is closely linked to raised blood pressure,and its physiopathologic mechanism is still not fully understood.We here aimed to explore whether extracellular vesicle(EV)mediated intercellular communication between endothelium and smooth muscle cell contribute to the blood vessel remodeling under hypertension.We here revealed that the arterial endothelial cells robustly secreted EV,which in turn could be circulated and/or directly taken up by the subendothelial smooth muscle cells(SMC).Under hypertension,the EV secretion increased and the miRNA profile changed significantly mainly due to the raised mechanical force and subsequent enhanced reactive oxygen species generation.Among the miRNA cargos in the EV,miR-320d/423-5p were found increased most significantly.In vivo delivery of miR-320d/423-5p mimics via engineered EV increased their expression in arterial vessels,recapitulating the phenotype in hypertension.In contrast,therapeutic delivery of miR-320d/423-5p inhibitors via engineered EV alleviated the phenotype in spontaneous hypertension rat model.Together,we have found that the injured endothelium due to the raised mechanical force in hypertension contributes to the arterial wall remodeling via the secreted EV.Our study has not only provided novel insights on the mechanism of hypertension associated blood vessel wall remodeling,but also shed light on therapeutic intervention of hypertension associated vascular diseases.Chen Wang Changyang Xing Zhelong Li Yunnan Liu Qiaoying Li Yixiao Wang Jiao Hu Lijun Yuan Guodong Yang 2022Bioactive Materials2022,7,2:1
