1Molecular immune pathogenesis and diagnosis of COVID-19显示文摘Coronavirus disease 2019(COVID-19)is a kind of viral pneumonia which is caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2).The emergence of SARS-CoV-2 has been marked as the third introduction of a highly pathogenic coronavirus into the human population after the severe acute respiratory syndrome coronavirus(SARS-CoV)and the Middle East respiratory syndrome coro-navirus(MERS-CoV)in the twenty-first century.In this minireview,we provide a brief introduction of the general features of SARS-CoV-2 and discuss current knowledge of molecular immune pathogenesis,diagnosis and treatment of COVID-19 on the base of the present understanding of SARS-CoV and MERS-CoV infections,which may be helpful in offering novel insights and potential therapeutic targets for combating the SARS-CoV-2 infection.Xiaowei Li Manman Geng Yizhao Peng Liesu Meng Shemin Lu 2020Journal of Pharmaceutical Analysis2020,10,2:31
2Spatio-temporal dynamics of vegetation and its relationship with climate factors Mongolia, China coverage in Inner显示文摘The vegetation coverage dynamics and its relationship with climate factors on different spatial and temporal scales in Inner Mongolia during 2001-2010 were analyzed based on MODIS-NDVI data and climate data.The results indicated that vegetation coverage in Inner Mongolia showed obvious longitudinal zonality,increasing from west to east across the region with a change rate of 0.2/10°N.During 2001-2010,the mean vegetation coverage was 0.57,0.4 and 0.16 in forest,grassland and desert biome,respectively,exhibiting evident spatial heterogeneities.Totally,vegetation coverage had a slight increasing trend during the study period.Across Inner Mongolia,the area of which the vegetation coverage showed extremely significant and significant increase accounted for 11.25% and 29.13% of the area of whole region,respectively,while the area of which the vegetation coverage showed extremely significant and significant decrease accounted for 7.65% and 26.61%,respectively.On inter-annual time scale,precipitation was the dominant driving force of vegetation coverage for the whole region.On inter-monthly scale,the change of vegetation coverage was consistent with both the change of temperature and precipitation,implying that the vegetation growth within a year is more sensitive to the combined effects of water and heat rather than either single climate factor.The vegetation coverage in forest biome was mainly driven by temperature on both inter-annual and inter-monthly scales,while that in desert biome was mainly influenced by precipitation on both the two temporal scales.In grassland biome,the yearly vegetation coverage had a better correlation with precipitation,while the monthly vegetation coverage was influenced by both temperature and precipitation.In grassland biome,the impacts of precipitation on monthly vegetation coverage showed time-delay effects.MU Shaojie YANG Hongfei LI Jianlong CHEN Yizhao GANG Chengcheng ZHOU Wei JU Weimin 2013Journal of Geographical Sciences2013,23,2:28
3Grassland coverage inter-annual variation and its coupling relation with hydrothermal factors in China during 1982-2010显示文摘GIMMS(Global Inventory Modeling and Mapping Studies) NDVI(Normalised Difference Vegetation Index) from 1982 to 2006 and MODIS(Moderate Resolution Imaging Spectroradiometer) NDVI from 2001 to 2010 were blended to extract the grass coverage and analyze its spatial pattern. The response of grass coverage to climatic variations at annual and monthly time scales was analyzed. Grass coverage distribution had increased from northwest to southeast across China. During 1982–2010, the mean nationwide grass coverage was 34% but exhibited apparent spatial heterogeneity, being the highest(61.4%) in slope grasslands and the lowest(17.1%) in desert grasslands. There was a slight increase of the grass coverage with a rate of 0.17% per year. Increase in slope grasslands coverage was as high as 0.27% per year, while in the plain grasslands and meadows the grass coverage increase was the lowest(being 0.11% per year and 0.1% per year, respectively). Across China, the grass coverage with extremely significant increase(P<0.01) and significant increase(P<0.05) accounted for 46.03% and 11% of the total grassland area, respectively, while those with extremely significant and significant decrease accounted for only 4.1% and 3.24%, respectively. At the annual time scale, there are no significant correlations between grass coverage and annual mean temperature and precipitation. However, the grass coverage was somewhat affected by temperature in alpine and sub-alpine grassland, alpine and sub-alpine meadow, slope grassland and meadow, while grass coverage in desert grassland and plain grassland was more affected by precipitation. At the monthly time-scale, there are significant correlations between grass coverage with both temperature and precipitation, indicating that the grass coverage is more affected by seasonal fluctuations of hydrothermal conditions. 5Direct conversion of mouse astrocytes into neural progenitor cells and specific lineages of neurons显示文摘Background:Cell replacement therapy has been envisioned as a promising treatment for neurodegenerative diseases.Due to the ethical concerns of ESCs-derived neural progenitor cells(NPCs)and tumorigenic potential of iPSCs,reprogramming of somatic cells directly into multipotent NPCs has emerged as a preferred approach for cell transplantation.Methods:Mouse astrocytes were reprogrammed into NPCs by the overexpression of transcription factors(TFs)Foxg1,Sox2,and Brn2.The generation of subtypes of neurons was directed by the force expression of cell-type specific TFs Lhx8 or Foxa2/Lmx1a.Results:Astrocyte-derived induced NPCs(AiNPCs)share high similarities,including the expression of NPC-specific genes,DNA methylation patterns,the ability to proliferate and differentiate,with the wild type NPCs.The AiNPCs are committed to the forebrain identity and predominantly differentiated into glutamatergic and GABAergic neuronal subtypes.Interestingly,additional overexpression of TFs Lhx8 and Foxa2/Lmx1a in AiNPCs promoted cholinergic and dopaminergic neuronal differentiation,respectively.Conclusions:Our studies suggest that astrocytes can be converted into AiNPCs and lineage-committed AiNPCs can acquire differentiation potential of other lineages through forced expression of specific TFs.Understanding the impact of the TF sets on the reprogramming and differentiation into specific lineages of neurons will provide valuable strategies for astrocyte-based cell therapy in neurodegenerative diseases.Kangmu Ma Xiaobei Deng Xiaohuan Xia Zhaohuan Fan Xinrui Qi Yongxiang Wang Yuju Li Yizhao Ma Qiang Chen Hui Peng Jianqing Ding Chunhong Li Yunlong Huang Changhai Tian Jialin C.Zheng 2018Translational Neurodegeneration2018,7,1:2
6Astrocytic effect of low molecular weight heparin-superoxide dismutase conjugate in interleukin-6 overexpressing mice following local cerebral ischemia显示文摘BACKGROUND:Studies have shown that low molecular weight heparin-superoxide dismutase conjugate exhibits a remarkable neuroprotective effect. OBJECTIVE:To investigate the effect of low molecular weight heparin-superoxide dismutase conjugate on astrocytes in an interleukin-6(IL-6) overexpressing mice following local cerebral ischemia. DESIGN,TIME AND SETTING:Randomized,cytological,controlled,animal study was performed in the Department of Physiology and Neuroscience,Neurology and Biochemistry and Molecular Biology,Medical University of South Carolina from January 2005 to March 2005. MATERIALS:Nine IL-6 transgenic mice,irrespective of gender,were randomly divided into three groups:sham-operated,model,and treatment,with three mice in each group.With exception of the sham-operated group,right middle cerebral artery occlusion was induced in the mice. Expression of glial fibrillary acidic protein,an astrocyte marker,was determined by immunohistochemistry.Low molecular weight heparin-superoxide dismutase conjugate was purchased from Biochemistry and Biotechnique Institute,Shandong University. METHODS:Two minutes prior to ischemia induction,0.5 mL/kg saline or 20 000 U/kg low molecular weight heparin-superoxide dismutase conjugate were administrated via the femoral artery in the model group and treatment group,respectively.The sham-operated group underwent the same protocols,with the exception of occlusion and treatment. MAIN OUTCOME MEASURES:The number of glial fibrillary acidic protein-positive cells was quantified under light microscopy(x200). RESULTS:In the sham-operated group,there were a large number of astrocytes in the IL-6 transgenic mice.However,the cell bodies were small,and the branches were few and thin.The number of astrocytes in the model group was remarkably less than the sham-operated group. Compared to the model and sham-operated groups,the number of astrocytes significantly increased,and the cell body became larger,following treatment with low molecular weight heparin-superoxide dismutase conjugate.Astrocytes exhibited hypertrophy and hyperplasia,and the processes became longer and thicker. 13A Randomized trial comparing ticlopidine with aspirin fof the prevention of ischemic cerebral stroke显示文摘OBJECTIVE To assess the effect and cafety of t iclopidine in the prevention of ischemic cerebral stroke and to compare theeffect of low-dose aspirin with t iclopidine. BACKGROUND The effect and safety of ticlopidine irn the prevention of ischemic cerebral stroke in China has not been reported. METHODS 329 patients with TIA or mild ischemic cevebral stroke wasrandonmly assigned to ticlopidine group(165 case) or aspirin group (164 case) in this study.These patrents were randomly allocated to receive either 250mg trclopidine or 50mg aspirin daily and didnd take any other platelet antiaggregating drugs. Time of eacn follow up visit was one to two months. Follow up lasted for 6 to 18 months. RESULTS The event rate for stroke or death from any cause was 8.3% in ticlopidine group arid 14.9% in aspirin group. This repesented a risk reduction of 44.3%(95% cofidence interval, 0.29-0.94) for ticiopidine group as compared with aspirin group. The event raite for ischemic cerebral stroke or myocarction of ticlopidine group(7.0%)was lower than that cf aspirin group(14.8%)(P<0.05).A riskreduction of 52.7%(95% confidence interval,0.24-0.92) for ticlopidine group compared with aspirin group. The rate of adverse effects of ticlopidine group and aspirin group were 6.9% and 11.0% during the trial ,but this was not statistically significant(P<0.05).DISCUSSION and CONCLUSION Therapeutic efficacy for the prevention oi ischemic stroke of ticlopidine was better than that of aspirin, the rate of side effects in ticlopidine group and aspirin group are not statistically significant. 14A study on levels of neuropeptide Y, neurotensin motilin and calcitonin gene-reliated peptide in plasma in patients with cerebral infarction and dinical isignificance显示文摘Objective To explore the relationship between virulent Helicobacter pylori strains infection and cerebral infarction. Method We assessed the prevalence of infection by strains bearing the cytotoxin -associated gene-A(Cag-A),a strong virulence factor ,in 83 patients with cerebral infarction and in 71 age- and sex-matched controls with similar social background. Result Prevalence of Helicobacter pylori infection was significantly higher in patients than in controls(78.3% versus 56.3%,p<0.05),with an odds ratio of 2.8(95%CI,1.46 to 5.36) adjusted for age, sex, main stroke factors. Patients with cerebral infarction also had a higher prevalence of Cag-A-positive strains(45.8% versus 19.7%, P<0.01),with an adjusted odds ratio of 3.43(95%CI 1.5 to 7.24).Conclusion It was suggested that chronic Helicobacter pylori infection, especially Cag-A-positive strains infection is an independent risk factor for cerebral infarction.Li Yizhao Sun Lin Zhang Dongjun 2000中国临床神经科学2000,8,z1:0
15The effect of ECr on HSP70 expression and morphologic changes following transient cerebral lschemia in rats显示文摘OBJECTIVE To investigate the effect of Cudrania tricuspidata root extract (ECr) on ischemic cerebral damage and the expression of the 70KDa heat shock protein (HSP70) following transient focal ischemia. BACKGROUND The role that ECr plays in cerebral ischemia has not been studied. METHODS Healthy wistar rats were randomized to the normal control group(Group A,n=4),the sham-operated control group(Group B,n=4),the ischemia and reperfusion group (Group C,n=24),the ischemia and reperfiusion after ECr pre- administration group(Group D,n=24).The rats of Group C and Group D were subjected to transient left middle cerebral artery occlusion (MCAO)as described by Zea Longa for 1 hour. Brain sections at the level of striatum were performed for HSP70 immunohistochemistry and morphology.HSP70 positive reactions and morphologic changes were semiquantiratively analyzedl. RESLULTS In the rats of Group A and Group B, there were scarcely HSP70 immunoreactivities either in cortex or in striatal neurons. In the ischemic brain regions for Group C rats,the HSP70 was induced in morphologically intact neurons and endothelial cells at hour 6 after recirculation, increased at hour12,peaked at day I, decreased at day 3,and HSP70 expression only occured in endothelial cells at day7. In Group D rats, the HSP70 was induced in the neurons of left MCA distribution at hour 1 after reperfusion, The changes in expression of HSP70 at different time points in Group D rats was in accord whth in Group C, but the number of HSP70 positive cells in Group D increased more, and HSP70 irnmunoreactivity in the HSP70-postive cells were more intense than in Group C. Histopathological study with HE staining showed no neuron pyknosis in Group A or in Group B. While pykrotic cells were present in the ipsilateral cortex and striatal neurons of MCA territory of Group C rats beginning at 6 hours after roper fusion. The change of histopathology in Group D was lighter at every time point. The number of pyknotic neurons in left MCA distribution was less than in Group C and there was no evident cell damage at 3 days and 7 days of reperfusion in Group D rats. CONCLUSION Our study demonstrated that transient focal cerebral ischenia could induce the HSP70 expression and induce neurons pyknosis.While ECr pre-treatment before transient focal ischemia in rats could increase the expression of HSP70 and reduce neuronal injury. 16Correlation between HSP70 and c-F0S expression and apoptosis in rats undergolng transient focal cerebral ischemla and reperfusion显示文摘OBJECTIVE To compare the induction of c-Fos and HSP70 and the presence of apoptosis and the influence of Ginkgo biloba extract (EGb761) in transient focal cerebral ischemic reperfusion rats.BACKGROUND Proto ancogene activation and induction of heat shock protein (HSP) occur in response to cerebral ischemia, but the correlation between these proteins and apoptosis remains uncertain. METHODS Healthy wistar rats were randomized to the normal control group(Group A, n=4), the Sham-operated control group(Group B, n=4), the ischemia and reperfusion group(Group C, n=24), the EGb761 pre-treated ischemia and reperfusion group(Group D, n=24). The rats of Group C and Group D were subjected to transient left middle cerebral artery occlusion (MCAO) as described by Zea longa for 1 hour. RESULTS Immunohistochemical analysis revealed no c-Fos or HSP70-immunoreactivity in Group A and Group B rats. However, in Group C rats, c-Fos was expressed in ipsilateral superficial cortical layers at 1 hour after reperfusion. At 6 hours, c-Fos immunoreactivities were increased in the ipsilateral cortex and were present in the contralateral cortex, while HSP70 were induced beginning in the ipsilateral neurons of MCA distribution. At 12 hours, the expression of c-Fos reached top in superficial cortical layers. At 24 hours HSP70 immunoreactivities reached top both in ipsilateral cortex and in ipsilateral striatum. At 3 days after recirculation, HSP70 expression decreased. c-Fos expression disappeared at day 7 and HSP70 expression only occured in endothelial cells. TUNEL staining showed that there was no cell apoptosis in Gronp A or in Group B. However, in Group C, TUNEL-positive neurons were observed in the border of the penumbra-like area that surrounds the ischemic core at 6 hours following reperfusion and then the number of TUNEL-positive cells reduced gradually. The changes in expression of HSP70 and c-Fos at different time points in Group D was in accord with in Group C, but the number of positive cells and the immunoreactivities in Group D were more intense than in Group C. We found only several TUNEL-positive cells at 6 hours following reperfusion in Group D and no TUNE;L-positive cells were present at other time points.CONCLUSION The present results indicate that c-Fos was expressed from an earlier stage of reperfusion and the expression occured in bilateral cerebral cortex. In contrast, HSP70 induction began later and only occured in ipsilateral neurons of MCA distribution. 17Reprogrammed astrocytes display higher neurogenic competence, migration ability and cell death resistance than reprogrammed fibroblasts显示文摘The direct reprogramming of somatic cells into induced neural progenitor cells(iNPCs)has been envisioned as a promising approach to overcome ethical and clinical issues of pluripotent stem cell transplantation.We previously reported that astrocyte-derived induced pluripotent stem cells(iPSCs)have more tendencies for neuronal differentiation than fibroblast-derived iPSCs.However,the differences of neurogenic potential between astrocytederived iNPCs(AiNPCs)and iNPCs from non-neural origins,such as fibroblast-derived iNPCs(FiNPCs),and the underlying mechanisms remain unclear.Our results suggested that AiNPCs exhibited higher differentiation efficiency,mobility and survival capacities,compared to FiNPCs.The whole transcriptome analysis revealed higher activities of TGFβsignaling in AiNPCs,versus FiNPCs,following a similar trend between astrocytes and fibroblasts.The higher neurogenic competence,migration ability,and cell death resistance of AiNPCs could be abrogated using TGFβ signaling inhibitor LY2157299.Hence,our study demonstrates the difference between iNPCs generated from neural and non-neural cells,together with the underlying mechanisms,which,provides valuable information for donor cell selection in the reprogramming approach.Xiaohuan Xia Chunhong Li Yi Wang Xiaobei Deng Yizhao Ma Lu Ding Jialin Zheng 2020Translational Neurodegeneration2020,9,1:0
18Active site identification and engineering during the dynamic evolution of copper-based catalysts for electrocatalytic CO_(2) reduction显示文摘To date,copper-based catalysts are one of the most prominent catalysts that can electrochemically reduce CO_(2)towards highvalue fuels or chemicals,such as ethylene,ethanol,and acetic acid.However,the chemically active feature of Cu-based catalysts hinders the understanding of the intrinsic catalytic active sites during the initial and the operative processes of electrochemical CO_(2)reduction(CO_(2)RR).The identification and engineering of active sites during the dynamic evolution of catalysts are thereby vital to further improve the activity,selectivity,and durability of Cu-based catalysts for high-performance CO_(2)RR.In this regard,four triggers for the dynamic evolution of catalysts were introduced in detail.Afterward,three typical active-site theories during the dynamic reconstruction of catalysts were discussed.In addition,the strategies in catalyst design were summarized according to the latest reports of Cu-based catalysts for CO_(2)RR,including the tuning of electronic structure,controlling of the external potential,and regulation of local catalytic environment.Finally,the conclusions and perspectives were provided to inspire more investigations and studies on the intrinsic active sites during the dynamic evolution of catalysts,which could promote the optimization of the catalyst system to further improve the performance of CO_(2)RR.Bangwei Deng Xueyang Zhao Yizhao Li Ming Huang Shihan Zhang Fan Dong 2023Science China Chemistry2023,66,1:0
19Evaluation of the Shallow Gas Hydrate Production Based on the Radial Drilling-Heat Injection-Back Fill Method显示文摘It has been evidenced that shallow gas hydrate resources are abundant in deep oceans worldwide.Their geological back-ground,occurrence,and other characteristics differ significantly from deep-seated hydrates.Because of the high risk of well construction and low production efficiency,they are difficult to be recovered by using conventional oil production methods.As a result,this paper proposes an alternative design based on a combination of radial drilling,heat injection,and backfilling methods.Multi-branch holes are used to penetrate shallow gas hydrate reservoirs to expand the depressurization area,and heat injection is utilized as a supplement to improve gas production.Geotechnical information collected from an investigation site close to the offshore production well in the South China Sea is used to assess the essential components of this plan,including well construction stability and gas production behavior.It demonstrates that the hydraulic fracturing of the 60mbsf overburden layer can be prevented by regulating the drilling fluid densities.However,the traditional well structure is unstable,and the suction anchor is advised for better mechanical performance.The gas produc-tion rate can be significantly increased by combining hot water injection and depressurization methods.Additionally,the suitable produc-tion equipment already in use is discussed.CHEN Qiang WAN Yizhao WU Nengyou SUN Jianye WANG Jian LIU Changling LI Yanlong LI Chengfeng HU Gaowei 2024Journal of Ocean University of China2024,23,1:0
20Behavior and respiration responses of the earthworm Eisenia fetida to soil arsenite pollution显示文摘Soil arsenic(As)pollution from mining and industrial sources is a serious issue in China.Earthworms are considered ecosystem engineers and contribute to soil fertility development and maintenance of soil physico-chemical properties.In this study,earthworms were exposed to soils with different sodium arsenite concentrations(0,5,20,and 80 mg As kg^(-1))for 60 d to investigate the changes in soil properties and the responses of the earthworms(e.g.,burrowing activity and respiration).Earthworm burrowing activity decreased with increasing arsenite concentrations,and earthworm respiration was significantly lower in soils with 20 and 80 mg As kg^(-1) compared to 0 mg As kg^(-1).Changes in soil properties were also observed after incubation of As-amended soil with earthworms.Specifically,soil pH decreased,while soil electrical conductivity and contents of soil NH3--N,Olsen-P,and available K increased.Our results suggest that arsenite negatively impacts the metabolic activity of earthworms,leading to reduced burrowing activity,which in turn modifies the effects of earthworms on soil fertility and remediation.Yali WANG Yizhao WU Jo CAVANAGH Xiuhong WANG Jiangping QIU Yinsheng LI 2021Pedosphere2021,31,3:0
