1Cancer nanotechnology: Enhancing tumor cell response to chemotherapy for hepatocellular carcinoma therapy显示文摘Hepatocellular carcinoma(HCC)is one of the deadliest cancers due to its complexities,reoccurrence after surgical resection,metastasis and heterogeneity.In addition to sorafenib and lenvatinib for the treatment of HCC approved by FDA,various strategies including transarterial chemoembolization,radiotherapy,locoregional therapy and chemotherapy have been investigated in clinics.Recently,cancer nanotechnology has got great attention for the treatment of various cancers including HCC.Both passive and active targetings are progressing at a steady rate.Herein,we describe the lessons learned from pathogenesis of HCC and the understanding of targeted and non-targeted nanoparticles used for the delivery of small molecules,monoclonal antibodies,miRNAs and peptides.Exploring current efficacy is to enhance tumor cell response of chemotherapy.It highlights the opportunities and challenges faced by nanotechnologies in contemporary hepatocellular carcinoma therapy,where personalized medicine is increasingly becoming the mainstay.Overall objective of this review is to enhance our understanding in the design and development of nanotechnology for treatment of HCC.Yongbing Sun Wen Ma Yuanyuan Yang Mengxue He Aimin Li Lei Bai Bin Yu Zhiqiang Yu 2019Asian Journal of Pharmaceutical Sciences2019,14,6:8
2Pioglitazone ameliorates neuronal damage after traumatic brain injury via the PPARg/NF-kB/IL-6 signaling pathway显示文摘Traumatic brain injury(TBI)is the major cause of high mortality and disability rates worldwide.Pioglitazone is an activator of peroxisome proliferator-activated receptor-gamma(PPARγ)that can reduce inflammation following TBI.Clinically,neuroinflammation after TBI lacks effective treatment.Although there are many studies on PPARγin TBI animals,only few could be converted into clinical,since TBI mechanisms in humans and animals are not completely consistent.The present study,provided a potential theoretical basis and therapeutic target for neuroinflammation treatment after TBI.First,we detected interleukin-6(IL-6),nitric oxide(NO)and Caspase-3 in TBI clinical specimens,confirming a presence of a high expression of inflammatory factors.Western blot(WB),quantitative real-time PCR(qRTPCR)and immunohistochemistry(IHC)were used to detect PPARγ,IL-6,and p-NF-kB to identify the mechanisms of neuroinflammation.Then,in the rat TBI model,neurobehavioral and cerebral edema levels were investigated after intervention with pioglitazone(PPARγactivator)or T0070907(PPARγinhibitor),and PPARγ,IL-6 and p-NF-kB were detected again by qRT-PCR,WB and immunofluorescence(IF).The obtained results revealed that:1)increased expression of IL-6,NO and Caspase-3 in serum and cerebrospinal fluid in patients after TBI,and decreased PPARγin brain tissue;2)pioglitazone could improve neurobehavioral and reduce brain edema in rats after TBI;3)the protective effect of pioglitazone was achieved by activating PPARγand reducing NF-kB and IL-6.The neuroprotective effect of pioglitazone on TBI was mediated through the PPARγ/NF-kB/IL-6 pathway.Yongbing Deng Xue Jiang Xiaoyan Deng Hong Chen Jie Xu Zhaosi Zhang Geli Liu Zhu Yong Chengfu Yuan Xiaochuan Sun Changdong Wang 2020Genes & Diseases2020,7,2:6

4Preparation and application of ZnO doped Pt-CeO2 nanofibers as electrocatalyst for methanol electro-oxidation显示文摘ZnO doped Pt/CeO_2 nanocomposites were prepared by electrospinning and reduction impregnation.Xray diffraction(XRD),transmission electron microscopy(TEM),energy disperse spectroscopy(EDS) and X-ray photoelectron spectroscopy(XPS) were employed to characterize the nanocomposites.It is observed that ZnO and CeO_2 form the hexagonal wurtzite phase and cubic fluorite phase in the nanocomposite,respectively,whilst Pt nanoparticles(NPs) with the number-averaged size of ca.3.1 nm are uniformly distributed on the surface of nanofibers.The mass fraction of Pt NPs in the nanocomposites is about 10 wt%.The doping of ZnO is effective to promote reactive oxygen species,surface reaction sites and the interaction between Pt and oxides.The catalytic performance of nanocomposites was evaluated by the methanol electro-oxidation.indexed with the catalytic activity,stability of catalyst.As a result,it is found that the nanocomposite exhibits much higher activity and stability for methanol oxidation than the undoped Pt/CeO_2 catalyst.Yingping Zheng Xin Zhang Zhengying Zhang Ying Li Yueming Sun Yongbing Lou Xiaojun Li Yang Lu 2018Journal of Rare Earths2018,36,9:5
5Nasal delivery of broadly neutralizing antibodies protects mice from lethal challenge with SARS-CoV-2 delta and omicron variants显示文摘Multiple new variants of severe acute respiratory syndrome coronavirus 2(SARS-Co V-2)have constantly emerged,as the delta and omicron variants,which have developed resistance to currently gained neutralizing antibodies.This highlights a critical need to discover new therapeutic agents to overcome the variants mutations.Despite the availability of vaccines against coronavirus disease 2019(COVID-19),the use of broadly neutralizing antibodies has been considered as an alternative way for the prevention or treatment of SARS-Co V-2 variants infection.Here,we show that the nasal delivery of two previously characterized broadly neutralizing antibodies(F61 and H121)protected K18-h ACE2 mice against lethal challenge with SARS-Co V-2 variants.The broadly protective efficacy of the F61 or F61/F121 cocktail antibodies was evaluated by lethal challenge with the wild strain(WIV04)and multiple variants,including beta(B.1.351),delta(B.1.617.2),and omicron(B.1.1.529)at 200or 1000 TCID_(50),and the minimum antibody administration doses(5-1.25 mg/kg body weight)were also evaluated with delta and omicron challenge.Fully prophylactic protections were found in all challenged groups with both F61 and F61/H121 combination at the administration dose of 20 mg/kg body weight,and corresponding mice lung viral RNA showed negative,with almost all alveolar septa and cavities remaining normal.Furthermore,low-dose antibody treatment induced significant prophylactic protection against lethal challenge with delta and omicron variants,whereas the F61/H121 combination showed excellent results against omicron infection.Our findings indicated the potential use of broadly neutralizing monoclonal antibodies as prophylactic and therapeutic agent for protection of current emerged SARS-Co V-2 variants infection.Jia Lu Qiangling Yin Rongjuan Pei Qiu Zhang Yuanyuan Qu Yongbing Pan Lina Sun Ding Gao Cuiqin Liang Jingwen Yang Wei Wu Jiandong Li Zongqiang Cui Zejun Wang Xinguo Li Dexin Li Shiwen Wang Kai Duan Wuxiang Guan Mifang Liang Xiaoming Yang 2022Virologica Sinica2022,37,2:4
6PEPT1-mediated prodrug strategy for oral delivery of peramivir显示文摘Peramivir was a novel and highly potent neuraminidase(NA) inhibitor for the treatment of influenza A and B. However, it exhibited a very low oral bioavailability(only 3%) due to the high polarity(log P of-1.4) and the low membrane permeability across the intestine. To utilize the PEPT1-mediated prodrug strategy to improve the oral absorption and develop the oral alternative, seven amino acid ester prodrugs and seven amino acid amide prodrugs have been synthesized. The permeability of these prodrugs across Caco-2 cells were screened. Peramivr-(CH_2)_2-l-Val and Peramivir-l-Ile were of the highest permeability in ester prodrugs and amide prodrugs, respectively, and then they were selected for further studies. Glycylsarcosine(gly-sar) uptake by Caco-2 could be inbihited by Peramivir-(CH_2)_2-l-Val and Peramivir-l-Ile in a concentration-dependent manner, and the IC 50 was 1.34 ± 0.31 m M and 1.78 ± 0.48 m M, respectively. The direct uptake of Peramivir-(CH_2)_2-l-Val and Peramivirl-Ile in MDCK-PEPT1 cells were significantly higher than in MDCK mock cells, and could be markedly inhibited by gly-sar. The uptake of Peramivir-(CH_2)_2-l-Val and Peramivir-l-Ile(0.01 to 50 m M) in MDCK-hPEPT1 cells conformed to Michaelis–Menten Equation. The oral bioavailability of peramivir was 65.3% and 37.3% after the oral administration of Peramivir-(CH_2)_2-l-Val and Peramivir-l-Ile to rats, respectively. The oral absorption and bioactivation of Peramivir-(CH_2)_2-l-Val was rapid and extensive, and no Peramivir-(CH_2)_2-l-Val was found in plasma. Because the amide bond was relatively stable, Peramivir-l-Ile could not be totally converted to the parent drug in vivo. Peramivir-(CH_2)_2-l-Val with good oral profiles and rapid bioactivation might be a promising prodrug for the further clinic development. 9PTEN and AKT/GSK-3β/CRMP-2 signaling pathway are involved in neuronal apoptosis and axonal injury in early brain injury after SAH in rats显示文摘In early brain injury(EBI)after subarachnoid hemorrhage(SAH),white matter(WM)axonal injury plays a key role in the prognosis of the disease.The purpose of this study was to investigate the effects of phosphatase and tensin homolog deleted on chromosome ten(PTEN)on axonal injury and neuronal apoptosis post-SAH in rats and to find its underlying mechanism.Adeno-associated virus was injected into the lateral ventricle to suppress or promote PTEN.Neural function post-SAH in animals was determined by the modified Garcia score,beam balance,and Rotarod test,and the blood–brain barrier disruption was assessed by the brain water content.Axonal injury post-SAH was observed by TEM and determined by IF,and neuron apoptosis was measured by TUNEL staining.The mechanism was analyzed by Western blot to detect p-PTEN/PTEN,p-AKT/AKT,p-GSK-3β/GSK-3β,p-CRMP-2/CRMP-2,axonal injury markerβ-APP and pro-and anti-apoptosis proteins,including Bax and Bcl-2,expression.We found 1.After knocking down PTEN,neuronal apoptosis and axonal injury were alleviated,and nerve function and blood–brain barrier were protected;accordingly,after overexpression of PTEN,neuronal apoptosis and axon damage were aggravated,and nerve function damage and blood–brain barrier damage were increased.2.PTEN and AKT/GSK-3β/CRMP-2 pathway were jointly involved in regulating neuronal apoptosis and WM axon injury after SAH.According to our research,PTEN was a negative factor of EBI,and together with the AKT/GSK-3β/CRMP-2 signaling pathway aggravates neuronal apoptosis and WM axon damage after SAH.Inhibition of PTEN expression may become a new target for SAH treatment.Hong Chen Chao Zhou Jianfeng Zheng Zhaosi Zhang Yongbing Deng Chongjie Cheng Zongduo Guo Gang Huo Cheng Yin Xiaochuan Sun 2022Genes & Diseases2022,9,1:1

12Pre-incubated with BSA-complexed free fatty acids alters ER stress/autophagic gene expression by carboxylated multi-walled carbon nanotube exposure in THP-1 macrophages显示文摘Recently we found that multi-walled carbon nanotube (MWCNT) exposure alters the mRNA levels of endoplasmic reticulum (ER) stress/autophagic genes, but the impact of biological molecules on this response is unclear. Herein, we compared the different actions of carboxylated MWCNTs (c-MWCNTs) pre-incubated with bovine serum albumin (BSA) or BSA-complexed free fatty acid (denoted as FFA) on macrophages derived from THP-1 monocytes (denoted as THP-1 macrophages). C-MWCNTs exhibited increased diameter and hydrodynamic size as well as decreased absolute zeta potential value after pre-incubation with BSA or FFA, which suggested a coating effect. Cytotoxicity or oxidative stress were not significantly induced after exposure to BSA-or FFA-coated c-MWCNTs. BSA-pre-incubated c-MWCNTs significantly enhanced the expression of the ER stress gene, DDIT3 and the autophagic genes, ATG5, BECN1, and PLIN2, but the mRNA levels of these genes was significantly decreased by FFA-pre-incubated c-MWCNTs. FFA-pre-incubated c-MWCNTs induced significantly higher lipid accumulation and interleukin-6 (IL-6) protein level compared with BSA-pre-incubated c-MWCNTs, which suggested that FFA-pre-incubated c-MWCNTs may more effectively induce the formation of macrophage foam cells. 14Two-dimensional ferromagnetic superlattices显示文摘Mechanically exfoliated two-dimensional ferromagnetic materials(2D FMs)possess long-range ferromagnetic order and topologically nontrivial skyrmions in few layers.However,because of the dimensionality effect,such few-layer systems usually exhibitmuch lower Curie temperature(TC)compared to their bulk counterparts.It is therefore of great interest to explore effective approaches to enhance their TC,particularly in wafer-scale for practical applications.Here,we report an interfacial proximity-induced high-TC 2D FM Fe3GeTe2(FGT)via A-type antiferromagnetic material CrSb(CS)which strongly couples to FGT.A superlattice structure of(FGT/CS)n,where n stands for the period of FGT/CS heterostructure,has been successfully produced with sharp interfaces by molecular-beam epitaxy on 2-inch wafers.By performing elemental specific X-ray magnetic circular dichroism(XMCD)measurements,we have unequivocally discovered that TC of 4-layer Fe3GeTe2 can be significantly enhanced from 140 K to 230 K because of the interfacial ferromagnetic coupling.Meanwhile,an inverse proximity effect occurs in the FGT/CS interface,driving the interfacial antiferromagnetic CrSb into a ferrimagnetic state as evidenced by double-switching behavior in hysteresis loops and the XMCD spectra.Density functional theory calculations show that the Fe-Te/Cr-Sb interface is strongly FMcoupled and doping of the spin-polarized electrons by the interfacial Cr layer gives rise to the TC enhancement of the Fe3GeTe2 films,in accordance with our XMCDmeasurements.Strikingly,by introducing rich Fe in a 4-layer FGT/CS superlattice,TC can be further enhanced to near room temperature.Our results provide a feasible approach for enhancing the magnetic order of few-layer 2D FMs in wafer-scale and render opportunities for realizing realistic ultra-thin spintronic devices.Shanshan Liu Ke Yang Wenqing Liu Enze Zhang Zihan Li Xiaoqian Zhang Zhiming Liao Wen Zhang Jiabao Sun Yunkun Yang Han Gao Ce Huang Linfeng Ai Ping Kwan Johnny Wong Andrew Thye Shen Wee Alpha TN’Diaye Simon AMorton Xufeng Kou Jin Zou Yongbing Xu Hua Wu Faxian Xiu 2020National Science Review2020,7,4:0

16Boosting oxygen evolution reactivity by modulating electronic structure and honeycomb-like architecture in Ni_(2)P/N,P-codoped carbon hybrids显示文摘Oxygen evolution reaction(OER)as the foremost stumbling block to generate cost-effective clean fuels has received extensive attention in recent years.But,it still maintains the challenge to manipulate the geometric and electronic structure during single reaction process under the same conditions.Herein,we report a simple self-template strategy to generate honeycomb-like Ni_(2)P/N,P-C hybrids with preferred electronic architecture.Experiments coupled with theoretical results revealed that the synthesized catalyst has two characteristics:firstly,the unique honeycomb-like morphology not only enables the fully utilization of catalytic active sites but also optimizes the mass/electron transportation pathway,which favor the diffusion of electrolyte to accessible active sites.Secondly,N,P-C substrate,on the one hand,largely contributes the electronic distribution near Fermi level(E_(F))thus boosting its electrical conductivity.On the other hand,the support effect result in the upshift of d-band center and electropositivity of Ni sites,which attenuates the energy barrier for the adsorption of OH~àand the formation of*OOH.In consequence,the optimized Ni_(2)P/N,P-C catalysts feature high electrocatalytic activity towards OER(a low overpotential of 252 m V to achieve10 m A cm^(-2))and 10 h long-term stability,the outstanding performance is comparable to most of transition metal catalysts.This work gives a innovative tactics for contriving original OER electrocatalysts,inspirng deeper understanding of fabricating catalysts by combining theoretical simulation and experiment design.Menglei Yuan Yu Sun Yong Yang Jingxian Zhang Sobia Dipazir Tongkun Zhao Shuwei Li Yongbing Xie He Zhao Zhanjun Liu Guangjin Zhang 2021Green Energy & Environment2021,6,6:0
17Conversion of phenol to cyclohexane in the aqueous phase over Ni/zeolite bi-functional catalysts显示文摘A series of Ni/HZSM-5 and Ni/HIM-5 bi-functional catalysts were synthesized and applied to the aqueous-phase hydrodeoxygenation(HDO)of phenol.The Ni dispersibility and particle sizes were shown to be directly related to the porosity and crystal sizes of the parent zeolites,which further influenced the catalytic performances.The large pores and small crystal sizes of the parent zeolites were beneficial for dispersing Ni and forming small Ni particles,and the corresponding Ni/zeolite catalyst exhibited a higher phenol conversion and selectivity towards hydrocarbons.Importantly,the Ni/HIM-5 bi-functional catalyst exhibited a high activity(98.3%)and high selectivity for hydrocarbons(98.8%)when heated at 220°C for 1 h and is thus a new potential catalyst for the HDO of phenolics to form hydrocarbons in the aqueous phase.Jimei Zhang Fuping Tian Junwen Chen Yanchun Shi Hongbin Cao Pengge Ning Shanshan Sun Yongbing Xie 2021Frontiers of Chemical Science and Engineering2021,15,2:0

19A facial synthesis of nitrogen-doped reduced graphene oxide quantum dot and its application in aqueous organics degradation显示文摘N-doped reduced graphene oxide quantum dots(N-rGQDs) have attracted more and more attention in efficient catalytic degradation of aqueous organic pollutants.However,the synthesis of N-rGQDs is generally a complex and high energy required process for the reduction and N-doping steps.In this study,a facile and green fabrication approach of N-rGQDs is established,based on a metal-free Fenton reaction without additional energy-input.The N structures of N-rGQDs play a significant role in the promotion of their catalytic performance.The N-rGQDs with relatively high percentage of aromatic nitrogen(NAr-rGQDs) perform excellent catalytic activities,with which the degradation efficiency of pollutant is enhanced by 25 times.Density functional theory(DFT) calculation also indicates aromatic nitrogen structures with electron-rich sites are prone to transfer electron,presenting a key role in the catalytic reaction.This metal-free Fenton process provides a green and costeffective strategy for one-step fabrication of N-rGQDs with controllable features and potential environmental catalytic applications.He Zhao Juehua Wang Zhuangjun Fan Yongbing Xie Di Zhang Shanshan Sun Yi Zhang Hongbin Cao 2022Green Energy & Environment2022,7,3:0
