1Phytophthora sojae Effector PsAvh240 Inhibits Host Aspartic Protease Secretion to Promote Infection显示文摘Plants secrete defense molecules into the extracellular space (the apoplast) to combat attacking microbes. However, the mechanisms by which successful pathogens subvert plant apoplastic immunity remain poorly understood. In this study, we show that PsAvh240, a membrane-localized effector of the soybean pathogen Phytophthora sojae, promotes P. sojae infection in soybean hairy roots. We found that PsAvh240 interacts with the soybean-resistant aspartic protease GmAP1 in planta and suppresses the secretion of GmAP1 into the apoplast. By solving its crystal structure we revealed that PsAvh240 contain six a helices and two WY motifs. The first two a helices of PsAvh240 are responsible for its plasma membrane-localization and are required for PsAvh240's interaction with GmAP1. The second WY motifs of two PsAvh240 molecules form a handshake arrangement resulting in a handshake-like dimer. This dimerization is required for the effector's repression of GmAP1 secretion. 2Maghemite(γ-Fe_2O_3) nanoparticles enhance dissimilatory ferrihydrite reduction by Geobacter sulfurreducens: Impacts on iron mineralogical change and bacterial interactions显示文摘Microbially mediated bioreduction of iron oxyhydroxide plays an important role in the biogeochemical cycle of iron.Geobacter sulfurreducens is a representative dissimilatory ironreducing bacterium that assembles electrically conductive pili and cytochromes.The impact of supplementation withγ-Fe_2O_3 nanoparticles(NPs)(0.2 and 0.6 g)on the G.sulfurreducens-mediated reduction of ferrihydrite was investigated.In the overall performance of microbial ferrihydrite reduction mediated byγ-Fe_2O_3 NPs,stronger reduction was observed in the presence of direct contact withγ-Fe_2O_3 NPs than with indirect contact.Compared to the production of Fe(Ⅱ)derived from biotic modification with ferrihydrite alone,increases greater than 1.6-and 1.4-fold in the production of Fe(Ⅱ)were detected in the biotic modifications in which direct contact with 0.2 g and 0.6 gγ-Fe_2O_3 NPs,respectively,occurred.X-ray diffraction analysis indicated that magnetite was a unique representative iron mineral in ferrihydrite when active G.sulfurreducens cells were in direct contact withγ-Fe_2O_3 NPs.Because of the sorption of biogenic Fe(Ⅱ)ontoγ-Fe_2O_3 NPs instead of ferrihydrite,the addition ofγ-Fe_2O_3 NPs could also contribute to increased duration of ferrihydrite reduction by preventing ferrihydrite surface passivation.Additionally,electron microscopy analysis confirmed that the direct addition ofγ-Fe_2O_3 NPs stimulated the electrically conductive pili and cytochromes to stretch,facilitating long-range electron transfer between the cells and ferrihydrite.The obtained findings provide a more comprehensive understanding of the effects of iron oxide NPs on soil biogeochemistry.Zheng Chen Yaxian Zhang Qingliu Luo Liuying Wang Shurui Liu Yajuan Peng Haitao Wang Liang Shen Qingbiao Li Yuanpeng Wang 2019Journal of Environmental Sciences2019,31,4:5
3Component analysis and risk assessment of biogas slurry from biogas plants显示文摘Massive amounts of biogas slurry are produced due to the development of biogas plants.The pollution features and the risk of biogas slurry were fully evaluated in this work.Thirty-one biogas slurry samples were collected from sixteen different cities and five different raw materials biogas plants(e.g.cattle manure,swine manure,straw-manure mixture,kitchen waste and chicken manure).The chemical oxygen demand(COD),ammonia nitrogen(NH_(4)^(+)-N),anions(e.g.Cl^(-1),SO_(4)^(2-),NO_(3)^(-)and PO_(4)^(3-)),antibiotics(e.g.sulphonamides,quinolones,β2-receptor agonists,macrolides,tetracyclines and crystal violet)and heavy metals(e.g.Cu,Cd,As,Cr,Hg,Zn and Pb)contents from these biogas slurry samples were systematically investigated.On this basis,risk assessment of biogas slurry was also performed.The concentrations of COD,NH_(4)^(+)-N and PO_(4)^(3-) in biogas slurry samples with chicken manure as raw material were significantly higher than those of other raw materials.Therefore,the biogas slurry from chicken manure raw material demonstrated the most serious eutrophication threat.The antibiotic contents in biogas slurry samples from swine manure were the highest among five raw materials,mostly sulphonamides,quinolones and tetracyclines.Biogas slurry revealed particularly serious arsenic contamination and moderate potential ecological risk.The quadratic polynomial stepwise regression model can quantitatively describe the correlation among NH_(4)^(+)-N,PO_(4)^(3+) and heavy metals concentration of biogas slurry.This work demonstrated a universal potential threat from biogas slurry that can provide supporting data and theoretical basis for harmless treatment and reuse of biogas slurry.Lanting Ke Xiaobin Liu Bingqing Du Yuanpeng Wang Yanmei Zheng Qingbiao Li 2022Chinese Journal of Chemical Engineering2022,35,4:5
4Nano-sized Fe_2O_3/Fe_3O_4 facilitate anaerobic transformation of hexavalent chromium in soil–water systems显示文摘The purpose of this study is to investigate the effects of nano-sized or submicro Fe_2O_3/Fe_3O_4 on the bioreduction of hexavalent chromium(Cr(VI)) and to evaluate the effects of nano-sized Fe_2O_3/Fe_3O_4 on the microbial communities from the anaerobic flooding soil.The results indicated that the net decreases upon Cr(VI) concentration from biotic soil samples amended with nano-sized Fe_2O_3(317.1 ± 2.1 mg/L) and Fe_3O_4(324.0 ± 22.2 mg/L) within21 days,which were approximately 2-fold of Cr(VI) concentration released from blank control assays(117.1 ± 5.6 mg/L).Furthermore,the results of denaturing gradient gel electrophoresis(DGGE) and high-throughput sequencing indicated a greater variety of microbes within the microbial community in amendments with nano-sized Fe_2O_3/Fe_3O_4 than the control assays.Especially,Proteobacteria occupied a predominant status on the phylum level within the indigenous microbial communities from chromium-contaminated soils.Besides,some partial decrease of soluble Cr(VI) in abiotic nano-sized Fe_2O_3/Fe_3O_4 amendments was responsible for the adsorption of nano-sized Fe_2O_3/Fe_3O_4 to soluble Cr(VI).Hence,the presence of nano-sized Fe_2O_3/Fe_3O_4 could largely facilitate the mobilization and biotransformation of Cr(VI) from flooding soils by adsorption and bio-mediated processes.Yaxian Zhang Hua Li Libo Gong Guowen Dong Liang Shen Yuanpeng Wang Qingbiao Li 2017Journal of Environmental Sciences2017,29,7:4

8Effects of dopant content on optical and electrical properties of In_2O_3: W transparent conductive films显示文摘The In 2 O 3 : W (IWO) films with different W content were deposited on glass substrate using direct current sputtering method. The structure, surface morphology, and optical and electrical properties were investigated. Results showed that both the carrier concentration and carrier mobility were increased with the doping of W. The IWO film with the lowest resistivity of 1.0×10 3 cm, highest carrier mobilityof 43.7 cm 2 V 1 s 1 and carrier concentration of 1.4×10 20 cm 3 was obtained at the content of 2.8 wt.%. 10A stable oxygen evolution splitting electrocatalysts high entropy alloy FeCoNiMnMo in simulated seawater显示文摘Stabilizing anode for oxygen evolution(OER)in chlorine-containing electrolytes is a significant challenge.Adding corrosion inhibitors in electrolytes can alleviate this problem,but the type and dosage of cor-rosion inhibitors need a lot of exploration,and side reactions may occur to reduce current efficiency.Herein,we prepared a FeCoNiMnMo High entropy alloy(HEA)electrode for OER in simulated seawa-ter without corrosion inhibitors.It exhibits great electrocatalytic activity(overpotential(η)=237 mV at 10 mA cm^(-2))and excellent stability(200 h at 100 mA cm^(-2)).Experiments and Density Functional The-ory(DFT)calculation show that the protective layer containing K_(2)MoO_(4)is the crucial factor for chlorine resistance of FeCoNiMnMo electrode.The tightly adsorbed state of Cl-and K_(2)MoO_(4)ensures that other oxides are not corroded.And the externally added K_(2)MoO_(4)cannot protect the electrode.In addition,the high-entropy effect of HEA reduces the dissolution of the alloy.This work provides an efficient way to prepare a noble-free HEA catalyst that can stabilize oxygen evolution in corrosive electrolytes.Peng Li Yuanpeng Yao Wengen Ouyang Ze Liu Huayi Yin Dihua Wang 2023Journal of Materials Science & Technology2023,,7:2
11Bacteroides ovatus-mediated CD27^(−)MAIT cell activation is associated with obesity-related T2D progression显示文摘Type 2 diabetes(T2D)is highly associated with obesity.However,the factors that drive the transition from excessive weight gain to glucose metabolism disruption are still uncertain and seem to revolve around systemic immune disorder.Mucosal-associated invariant T(MAIT)cells,which are innate-like T cells that recognize bacterial metabolites,have been reported to be altered in obese people and to lead to metabolic dysfunction during obesity.By studying the immunophenotypes of blood MAIT cells from a cross-sectional cohort of obese participants with/without T2D,we found an elevation in CD27^(-)negative(CD27−)MAIT cells producing a high level of IL-17 under T2D obese conditions,which could be positively correlated with impaired glucose metabolism in obese people.We further explored microbial translocation caused by gut barrier dysfunction in obese people as a triggering factor of MAIT cell abnormalities.Specifically,accumulation of the bacterial strain Bacteroides ovatus in the peripheral blood drove IL-17^(-)producing CD27−MAIT cell expansion and could be associated with T2D risk in obese individuals.Overall,these results suggest that an aberrant gut microbiota–immune axis in obese people may drive or exacerbate T2D.Importantly,CD27−MAIT cell subsets and Bacteroides ovatus could represent targets for novel interventional strategies.Our findings extend current knowledge regarding the clinical relevance of body mass index(BMI)-associated variation in circulating MAIT cells to reveal the role of these cells in obesity-related T2D progression and the underlying cellular mechanisms.Yue Li Yi Yang Jin Wang Peihong Cai Mei Li Xixiang Tang Ying Tan Yuchan Wang Fan Zhang Xiaofeng Wen Qiaoxing Liang Yuanpeng Nie Tufeng Chen Xiang Peng Xuemin He Yanhua Zhu Guojun Shi Wai WCheung Lai Wei Yanming Chen Yan Lu 2022Cellular & Molecular Immunology2022,19,7:2

14RAF22,ABI1 and OST1 form a dynamic interactive network that optimizes plant growth and responses to drought stress in Arabidopsis显示文摘Plants adapt to their ever-changing environment via positive and negative signals induced by environmental stimuli.Drought stress,for instance,induces accumulation of the plant hormone abscisic acid(ABA),triggering ABA signal transduction.However,the molecular mechanisms for switching between plant growth promotion and stress response remain poorly understood.Here we report that RAF(rapidly accelerated fibrosarcoma)-LIKE MITOGEN-ACTIVATED PROTEIN KINASE KINASE KINASE 22(RAF22)in Arabidopsis tha/iana physically interacts with ABA INSENSITIVE 1(ABl1)and phosphorylates ABl1 at Ser416 residue to enhance its phosphatase activity.Interestingly,ABl1 can also enhance the activity of RAF22 through dephosphorylation,reciprocally inhibiting ABA signaling and promoting the maintenance of plant growth under normal conditions.Under drought stress,however,the ASA-activated OPEN STOMATA1(OST1)phosphorylates the Ser81 residue of RAF22 and inhibits its kinase activity,restraining its enhancement of ABl1 activity.Taken together,our study reveals that RAF22,ABl1,and OST1 form a dynamic regulatory network that plays crucial roles in optimizing plant growth and environmental adaptation under drought stress.Zhihui Sun Zhenkai Feng Yanglin Ding Yuanpeng Qi Shan Jiang Zhen Li Yu Wang Junsheng Qi Chunpeng Song Shuhua Yang Zhizhong Gong 2022Molecular Plant2022,15,7:1

18Identification and analysis of serum samples by surface-enhanced Raman spectroscopy combined with characteristic ratio method and PCA for gastric cancer detection显示文摘This study aimed to explore the application of surface-enhanced Raman scattering(SERS)in the rapid diagnosis of gastric cancer.The SERS spectra of 68 serum samples from gastric cancer patients and healthy volunteers were acquired.The characteristic ratio method(CRM)and principal component analysis(PCA)were used to differentiate gastric cancer serum from normal serum.Compared with healthy volunteers,the serum SERS intensity of gastric cancer patients was relatively high at 722 cm^(-1),while it was relatively low at 588,644,861,1008,1235,1397,1445 and 1586 cm^(-1).These results indicated that the relative content of nucleic acids in the serum of gastric cancer patients rises while the relative content of amino acids and carbohydrates decreases.In PCA,the sensitivity and specificity of discriminating gastric cancer were 94.1%and 94.1%,respectively,with the accuracy of 94.1%.Based on the intensity ratios of four characteristic peaks at 722,861,1008 and 1397 cm^(-1),CRM presented the diagnostic sensitivity and specificity of 100%and 97.4%,respectively,and the accuracy of 98.5%.Therefore,the three peak intensity ratios of I_(722)/I_(861),I_(722)/I_(1008)and I_(722)/I_(1397)can be considered as biologicalfingerprint information for gastric cancer diagnosis and can rapidly and directly reflect the physiological and pathological changes associated with gastric cancer development.This study provides an important basis and standards for the early diagnosis of gastric cancer.Liu Guo Yuanpeng Li Furong Huang Jia Dong Fucui Li Xinhao Yang Siqi Zhu Maoxun Yang 2019Journal of Innovative Optical Health Sciences2019,,2:1
