2Whole-genome sequencing provides insights into the genetic diversity and domestication of bitter gourd(Momordica spp.)显示文摘Bitter gourd(Momordica charantia)is a popular cultivated vegetable in Asian and African countries.To reveal the characteristics of the genomic structure,evolutionary trajectory,and genetic basis underlying the domestication of bitter gourd,we performed whole-genome sequencing of the cultivar Dali-11 and the wild small-fruited line TR and resequencing of 187 bitter gourd germplasms from 16 countries.The major gene clusters(Bi clusters)for the biosynthesis of cucurbitane triterpenoids,which confer a bitter taste,are highly conserved in cucumber,melon,and watermelon.Comparative analysis among cucurbit genomes revealed that the Bi cluster involved in cucurbitane triterpenoid biosynthesis is absent in bitter gourd.Phylogenetic analysis revealed that the TR group,including 21 bitter gourd germplasms,may belong to a new species or subspecies independent from M.charantia.Furthermore,we found that the remaining 166 M.charantia germplasms are geographically differentiated,and we identified 710,412,and 290 candidate domestication genes in the South Asia,Southeast Asia,and China populations,respectively.This study provides new insights into bitter gourd genetic diversity and domestication and will facilitate the future genomics-enabled improvement of bitter gourd.Junjie Cui Yan Yang Shaobo Luo Le Wang Rukui Huang Qingfang Wen Xiaoxia Han Nansheng Miao Jiaowen Cheng Ziji Liu Changyuan Zhang Chengcheng Feng Haisheng Zhu Jianwen Su Xinjian Wan Fang Hu Yu Niu Xiaoming Zheng Yulan Yang Dai Shan Zhensheng Dong Weiming He Narinder P.S.Dhillon Kailin Hu 2020Horticulture Research2020,7,1:7
3Fine relocation, mechanism, and tectonic indications of middle-small earthquakes in the Central Tibetan Plateau显示文摘The medium-small earthquakes that occurred in the middle part of Tibetan Plateau(32°N–36°N, 90°E–93°E) from August 2016 to June 2017 were relocated using the absolute earthquake location method Hypo2000. Compared to the reports of Chinese Seismological Networks, our relocation results are more clustered on the whole, the horizontal location differences exceed 10 km, and the focal depths are concentrated in 0–8 km, which indicates that the upper crust inside the Tibetan Plateau is tectonically active. In June2017 altogether eight earthquakes above magnitude 3.0 took place; their relocated epicenters are concentrated around Gêladaindong.The relocation results of M<3.0 small earthquakes also showed obvious differences. Therefore, we used the CAP method to invert for the focal mechanisms of the M ≥3.0 earthquakes; results generally tally with the surface geological structures, indicating that the Tibetan Plateau is still under the strong compressional force from the India Plate. 4Clinical and pathological features of miR-10b and RHOC gene expression in hepatocellular carcinoma显示文摘Aberrant expression of microRNAs(miRNAs)was reported frequently in different human cancers.The major role of miRNA is targeting 30-UTR of coding gene and causing translational repression or mRNA degradation.miR-10b overexpression was reported to promote breast cancer metastasis by up-regulating RHOC expression.But its expression in hepatocellular carcinoma(HCC)remains unclear.Our study indicated that the expression of miR-10b was different in HCC and adjacent tissue samples,and reduced expression of miR-10b in HCC was related tovein invasion.High-level expression of RHOC was also related to vein invasion in HCC.But no correlation was found between miR-10b and RHOC expression.These results suggest that miR-10b and RHOC are independent predictors of HCC invasion and metastasis.Jingli Du Yulan Wang Hanjiang Fu Xiaofei Zheng Minggui Lin Mingzhou Guo Lixin Wei 2014Chinese Science Bulletin2014,59,19:4

10SARS-CoV-2 immunity and functional recovery of COVID-19 patients 1-year after infection显示文摘The long-term immunity and functional recovery after SARS-CoV-2 infection have implications in preventive measures and patient quality of life.Here we analyzed a prospective cohort of 121 recovered COVID-19 patients from Xiangyang,China at 1-year after diagnosis.Among them,chemiluminescence immunoassay-based screening showed 99%(95%CI,98–100%)seroprevalence 10–12 months after infection,comparing to 0.8%(95%CI,0.7–0.9%)in the general population.Total anti-receptor-binding domain(RBD)antibodies remained stable since discharge,while anti-RBD IgG and neutralization levels decreased over time.A predictive model estimates 17%(95%CI,11–24%)and 87%(95%CI,80–92%)participants were still 50%protected against detectable and severe re-infection of WT SARS-CoV-2,respectively,while neutralization levels against B.1.1.7 and B.1.351 variants were significantly reduced.All non-severe patients showed normal chest CT and 21%reported COVID-19-related symptoms.In contrast,53%severe patients had abnormal chest CT,decreased pulmonary function or cardiac involvement and 79%were still symptomatic.Our findings suggest long-lasting immune protection after SARS-CoV-2 infection,while also highlight the risk of immune evasive variants and long-term consequences for COVID-19 survivors.Yan Zhan Yufang Zhu Shanshan Wang Shijun Jia Yunling Gao Yingying Lu Caili Zhou Ran Liang Dingwen Sun Xiaobo Wang Zhibing Hou Qiaoqiao Hu Peng Du Hao Yu Chang Liu Miao Cui Gangling Tong Zhihua Zheng Yunsheng Xu Linyu Zhu Jin Cheng Feng Wu Yulan Zheng Peijun Liu Peng Hong 2021Signal Transduction and Targeted Therapy2021,6,11:0

12PICOTEES:a privacy-preserving online service of phenotype exploration for genetic-diagnostic variants from Chinese children cohorts显示文摘The growth in biomedical data resources has raised potential privacy concerns and risks of genetic information leakage. For instance, exome sequencing aids clinical decisions by comparing data through web services, but it requires significant trust between users and providers. To alleviate privacy concerns, the most commonly used strategy is to anonymize sensitive data. Unfortunately, studies have shown that anonymization is insufficient to protect against reidentification attacks. Recently, privacy-preserving technologies have been applied to preserve application utility while protecting the privacy of biomedical data. We present the PICOTEES framework, a privacy-preserving online service of phenotype exploration for genetic-diagnostic variants (https://birthdefectlab.cn:3000/). PICOTEES enables privacy-preserving queries of the phenotype spectrum for a single variant by utilizing trusted execution environment technology, which can protect the privacy of the user's query information, backend models, and data, as well as the final results. We demonstrate the utility and performance of PICOTEES by exploring a bioinformatics dataset. The dataset is from a cohort containing 20,909 genetic testing patients with 3,152,508 variants from the Children's Hospital of Fudan University in China, dominated by the Chinese Han population (>99.9%). 13Seasonality modeling of the distribution of Aedes albopictus in China based on climatic and environmental suitability显示文摘Background:Aedes albopictus is a highly invasive mosquito species and a major vector of numerous viral pathogens.Many recent dengue fever outbreaks in China have been caused solely by the vector.Mapping of the potential distribution ranges of Ae.albopictus is crucial for epidemic preparedness and the monitoring of vector populations for disease control.Climate is a key factor influencing the distribution of the species.Despite field studies indicating seasonal population variations,very little modeling work has been done to analyze how environmental conditions influence the seasonality of Ae.albopictus.The aim of the present study was to develop a model based on available observations,climatic and environmental data,and machine learning methods for the prediction of the potential seasonal ranges of Ae.albopictus in China.Methods:We collected comprehensive up-to-date surveillance data in China,particularly records from the northern distribution margin of Ae.albopictus.All records were assigned long-term(1970–2000)climatic data averages based on the WorldClim 2.0 data set.Machine learning regression tree models were developed using a 10-fold crossvalidation method to predict the potential seasonal(or monthly)distribution ranges of Ae.albopictus in China at high resolution based on environmental conditions.The models were assessed based on sensitivity,specificity,and accuracy,using area under curve(AUC).WorldClim 2.0 and climatic and environmental data were used to produce environmental conduciveness(probability)prediction surfaces.Predicted probabilities were generated based on the averages of the 10 models.Results:During 1998–2017,Ae.albopictus was observed at 200 out of the 242 localities surveyed.In addition,at least 15 new Ae.albopictus occurrence sites lay outside the potential ranges that have been predicted using models previously.The average accuracy was 98.4%(97.1–99.5%),and the average AUC was 99.1%(95.6–99.9%).The predicted Ae.albopictus distribution in winter(December–February)was limited to a small subtropical-tropical area of China,and Ae.albopictus was predicted to occur in northern China only during the short summer season(usually June–September).The predicted distribution areas in summer could reach northeastern China bordering Russia and the eastern part of the Qinghai-Tibet Plateau in southwestern China.Ae.albopictus could remain active in expansive areas from central to southern China in October and November.Conclusions:Climate and environmental conditions are key factors influencing the seasonal distribution of Ae.albopictus in China.The areas predicted to potentially host Ae.albopictus seasonally in the present study could reach northeastern China and the eastern slope of the Qinghai-Tibet Plateau.Our results present new evidence and suggest the expansion of systematic vector population monitoring activities and regular re-assessment of epidemic risk potential.Xueli Zheng Daibin Zhong Yulan He Guofa Zhou 2020Infectious Diseases of Poverty2020,9,1:0
