| 2 | Effects of dietary protein/energy ratio on growth performance,carcass trait,meat quality,and plasma metabolites in pigs of different genotypes显示文摘Background:The protein/energy ratio is important for the production performance and utilization of available feed resources by animals.Increased protein consumption by mammals leads to elevated feed costs and increased nitrogen release into the environment.This study aimed to evaluate the effects of dietary protein/energy ratio on the growth performance,carcass traits,meat quality,and plasma metabolites of pigs of different genotypes.Methods:Bama mini-pigs and Landrace pigs were randomly assigned to two dietary treatment groups(Chinese conventional diet with low protein/energy ratio or National Research Council diet with high protein/energy ratio;n = 24 per treatment) in a 2x2 factorial arrangement.Blood and muscle samples were collected at the end of the nursery,growing,and finishing phases.Results:We observed significant interactions(P< 0.05) between breed and diet for total fat percentage,intramuscular fat(IMF) content,protein content in biceps femoris(BF) muscle,and plasma urea nitrogen(UN) concentration in the nursery phase;for average daily gain(ADG),average daily feed intake(ADFI),dry matter,IMF content in psoas major(PM) muscle,and plasma total protein and albumin concentrations in the growing phase;and for drip loss and plasma UN concentration in the finishing phase.Breed influenced(P < 0.05) growth performance,carcass traits,and meat quality,but not plasma metabolites.Throughout the trial,Landrace pigs showed significantly higher(P < 0.05) ADG,ADFI,dressing percentage,lean mass rate,and loin-eye area than did Bama mini-pigs,but significantly lower(P< 0.05)feed/gain ratio,fat percentage,backfat thickness,and IMF content.Dietary protein/energy ratio influenced the pH value,chemical composition of BF and PM muscles,and plasma activities of glutamic-pyruvic transaminase and gamma-glutamyl transpeptidase,and plasma concentration of UN.Conclusions:Compared with Landrace pigs,Bama mini-pigs showed slower growth and lower carcass performance,but had better meat quality.Moreover,unlike Landrace pigs,the dietary protein/energy ratio did not affect the growth performance of Bama mini-pigs.These results suggest that,in swine production,low dietary protein/energy ratio may be useful for reducing feed costs and minimizing the adverse effects of ammonia release into the environment. | Yingying Liu Xiangfeng Kong Guoli Jiang Bi'e Tan Jinping Deng Xiaojian Yang Fengna Li Xia Xiong Yulong Yin | 2015 | Journal of Animal Science and Biotechnology2015,6,4: | 37 |

| 6 | Effects of dietary protein restriction on muscle fiber characteristics and m TORC1 pathway in the skeletal muscle of growing-finishing pigs显示文摘Background: To investigate the effects of dietary crude protein(CP) restriction on muscle fiber characteristics and key regulators related to protein deposition in skeletal muscle, a total of 18 growing-finishing pigs(62.30 ± 0.88 kg)were allotted to 3 groups and fed with the recommended adequate protein(AP, 16 % CP) diet, moderately restricted protein(MP, 13 % CP) diet and low protein(LP, 10 % CP) diet, respectively. The skeletal muscle of different locations in pigs, including longissimus dorsi muscle(LDM), psoas major muscle(PMM) and biceps femoris muscle(BFM) were collected and analyzed.Results: Results showed that growing-finishing pigs fed the MP or AP diet improved(P < 0.01) the average daily gain and feed: gain ratio compared with those fed the LP diet, and the MP diet tended to increase(P = 0.09) the weight of LDM. Moreover, the ATP content and energy charge value were varied among muscle samples from different locations of pigs fed the reduced protein diets. We also observed that pigs fed the MP diet up-regulated(P < 0.05) muscular m RNA expression of all the selected key genes, except that myosin heavy chain(My HC) IIb,My HC IIx, while m RNA expression of ubiquitin ligases genes was not affected by dietary CP level. | 8 | Developmental changes in intercellular junctions and Kv channels in the intestine of piglets during the suckling and post-weaning periods显示文摘Background: The intestinal epithelium is an important barrier that depends on a complex mixture of proteins and these proteins comprise different intercellular junctions. The purpose of this study was to investigate the postnatal and developmental changes in morphology, intercellular junctions and voltage-gated potassium(Kv) channels in the intestine of piglets during the suckling and post-weaning periods.Results: Samples of the small intestine were obtained from 1-, 7-, 14-, and 21-d-old suckling piglets and piglets on d 1, 3, 5, and 7 after weaning at 14 d of age. The results showed that the percentage of proliferating cell nuclear antigen(PCNA)-positive cells and alkaline phosphatase(AKP) activity, as well as the abundances of E-cadherin,occludin, and Kv1.5 m RNA and claudin-1, claudin-3, and occludin protein in the jejunum were increased from d 1to d 21 during the suckling period(P < 0.05). Weaning induced decreases in the percentage of PCNA-positive cells,AKP activity and the abundances of E-cadherin, occludin and zonula occludens(ZO)-1 m RNA or protein in the jejunum on d 1, 3 and 5 post-weaning(P < 0.05). There were lower abundances of E-cadherin, occludin and ZO-1m RNA as well as claudin-1, claudin-3 and ZO-1 protein in the jejunum of weanling piglets than in 21-d-old suckling piglets(P < 0.05). The abundances of E-cadherin, occludin, ZO-1 and integrin m RNA were positively related to the percentage of PCNA-positive cells.Conclusion: Weaning at 14 d of age induced damage to the intestinal morphology and barrier. | 10 | Development and utilization of a new chemically-induced soybean library with a high mutation density显示文摘Mutagenized populations have provided important materials for introducing variation and identifying gene function in plants. In this study, an ethyl methanesulfonate(EMS)-induced soybean(Glycine max) population,consisting of 21,600 independent M_2 lines, was developed.Over 1,000 M_(4(5))families, with diverse abnormal phenotypes for seed composition, seed shape, plant morphology and maturity that are stably expressed across different environments and generations were identified. Phenotypic analysis of the population led to the identification of a yellow pigmentation mutant, gyl, that displayed significantly decreased chlorophyll(Chl) content and abnormal chloroplast development. Sequence analysis showed that gyl is allelic to Minn Gold, where a different single nucleotide polymorphism variation in the Mg-chelatase subunit gene(ChlI1a) results in golden yellow leaves. A cleaved amplified polymorphic sequence marker was developed and may be applied to marker-assisted selection for the golden yellow phenotype in soybean breeding. | 14 | Extraction and identification of the chyme proteins in the digestive tract of growing pigs显示文摘This study aimed to explore the rule of degradation of dietary proteins by identifying chyme proteins in different segments of the digestive tract of growing pigs, using proteomics techniques. Six growing pigs were fed a corn-soybean meal-based diet for 7 days. The feedstuff and chyme proteins were separately extracted and separated with SDS-PAGE. 2 D LCMS/MS combined with protein database searching identified 1,513 proteins in different segments of the gastrointestinal tract, the number of identified exogenous proteins gradually decline from the stomach to colon, with large amounts in the duodenum to the large intestine.More corn proteins than soybean proteins were identified both in the feedstuff and chyme, and these were significantly decreased after digestion in the stomach. | 16 | 4-Phenylbutyric acid accelerates rehabilitation of barrier function in IPEC-J2 cell monolayer model显示文摘As the first line of defence against pathogens and endotoxins crossing the intestine-blood barrier,the intestinal epithelial barrier plays a determinant role in pigs’health and growth.4-Phenylbutyric acid(4-PBA),an aromatic fatty acid,was reported to benefit homeostasis of endoplasmic reticulum and protein synthesis.However,whether 4-PBA affects intestinal epithelial barrier function in pigs is unknown.This study aimed to explore the effects of 4-PBA on the intestinal barrier function,using in vitro models of well-differentiated intestinal porcine epithelial cell(IPEC-J2)monolayers in the transwell plates.Cell monolayers with or without 4-PBA(1.0 mmol/L)treatment were challenged with physical scratch,deoxynivalenol(DON,2.0μg/mL,48 h),and lipopolysaccharide(LPS,5.0μg/mL,48 h),respectively.Transepithelial electrical resistance(TEER)and fluorescein isothiocyanate-dextran(FD-4)permeability were measured to indicate barrier integrity and permeability.Real-time PCR and Western blot were conducted to determine relative gene and protein expressions of tight junction proteins.As expected,physical scratch,DON,and LPS challenges decreased TEER and increased FD-4 permeability.4-PBA treatment accelerated cell mitigation and rehabilitation of the physical scratch-damaged intestinal epithelial barrier but did not alleviate DON or LPS induced barrier damage.However,once 48-h DON and LPS challenges were removed,rehabilitation of the epithelial barrier function of IPEC-J2 monolayer was accelerated by the 4-PBA treatment.Also,the relative gene and protein expressions of zonula occludens-1(ZO-1),occludin,and claudin-1 were further upregulated by the 4-PBA treatment during the barrier rehabilitation.Taken together,4-PBA accelerated the IPEC-J2 cell monolayer barrier recovering from physical scratch,DON-,and LPS-induced damage,via enhancing cell mitigation and expressions of tight junction proteins. | Qian Jiang Jie Yin Jiashun Chen Xiaokang Ma Miaomiao Wu Xilong Li Kang Yao Bi'e Tan Yulong Yin | 2021 | Animal Nutrition2021,,4: | 2 |

| 18 | Biodegradable lubricating mesoporous silica nanoparticles for osteoarthritis therapy显示文摘Osteoarthritis is characterized by lubrication failure of the articular cartilage and severe inflammation of the joint capsule.Lubricating mesoporous silica nanoparticles(MSNs)have been developed for the treatment of osteoarthritis based on enhanced lubrication and local drug delivery.However,MSNs are difficult to degrade in vivo in a short time,resulting in potential toxic effect due to bioaccumulation.In this study,biodegradable MSNs(bMSNs)were prepared through an oil–water biphase stratification method,and modified with poly(2-methacryloyloxyethyl phosphocholine)(PMPC)to synthesize lubricating drug-loaded nanoparticles(bMSNs–NH_(2)@PMPC)by photopolymerization.The in vitro degradation test demonstrated that the bMSNs and bMSNs–NH_(2)@PMPC almost degraded within 7 days.The tribiological test showed that the lubrication property of the bMSNs–NH2@PMPC was greatly improved,with a reduction of 50%in the friction coefficient(COF)compared with the bMSNs.It was attributed to hydration lubrication mechanism by which a tenacious hydration layer is formed surrounding the zwitterionic headgroups(N+(CH_(3))^(3)and PO4-)in PMPC polyelectrolyte polymer.Additionally,the bMSNs–NH_(2)@PMPC maintained excellent lubrication property under degradation and achieved sustained drug release behavior compared with the bMSNs.In summary,the biodegradable bMSNs–NH2@PMPC developed in this study with the properties of enhanced lubrication and drug delivery may be a promising approach for osteoarthritis therapy. | Li WAN Yi WANG Xiaolong TAN Yulong SUN Jing LUO Hongyu ZHANG | 2022 | Friction2022,10,1: | 2 |
| 19 | Starch supplementation improves the reproductive performance of sows in different glucose tolerance status显示文摘This study was to evaluate the effects of glucose tolerance status,maternal starch supplementation and soybean substitution in diets on the performance of dams and their offspring.Eighty-eight pregnant sows(Landrace x Large White)were selected from an initial total of 120 sows,based on blood glucose test values,and assigned to 4 experimental treatments in a 2×2 factorial design.The factors were glucose tolerance status(glucose intolerant[GIT]vs.normal glucose tolerant[NGT])or dietary treatments(corn starch diet[CS]vs.soybean substitution diet[SSI]).A higher area under the curve(AUC)for post-meal glucose was observed(P<0.05)in the GIT group than in the NGT group on d 109 of gestation.The CS group had a lower value of homeostasis model assessment-insulin resistance than the SS group(P<0.05)on d 109 of gestation.Corn starch supplementation for sows decreased the stillbirth rate(P<0.05),regardless of the sows’glucose tolerance status.The villus height of the jejunum and the villus height to crypt depth ratio of the ileum were greater in normal birth weight piglets from the CS group than from the SS group(P<0.01),and so was the activity of sucrase in the jejunum and ileum(P<0.01).Compared with the SS group,the CS group showed a reduction in pre-weaning mortality rate,an increase in the number of high-birth-weight piglets,and a decrease in the number of low-birth-weight piglets(P<0.05)under GIT status.In conclusion,sows fed CS decreased stillbirth rate and improved insulin resistance,as well as improving the intestinal morphology and digestive enzyme activities of their progeny,regardless of glucose tolerance status.Additionally,the CS group improved birth weight distribution and decreased pre-weaning mortality rate of piglets under GIT status. | Yunyu Yang Ming Deng Jianzhao Chen Xichen Zhao Kaili Xiao Wenliang He Xinggang Qiu Yanzhen Xu Yulong Yin Chengquan Tan | 2021 | Animal Nutrition2021,,4: | 2 |
