2Population Genomic Analysis and De Novo Assembly Reveal the Origin of Weedy Rice as an Evolutionary Game显示文摘Crop weediness,especially that of weedy rice(Oryza sativa f.spontanea),remains mysterious.Weedy rice possesses robust ecological adaptability;however,how this strain originated and gradually formed proprietary genetic features remains unclear?Here,we demonstrate that weedy rice at Asian high latitudes(WRAH)is phylogenetically well defined and possesses unselected genomic characteristics in many divergence regions between weedy and cultivated rice.We also identified novel quantitative trait loci underlying weedy-specific traits,and revealed that a genome block on the end of chromosome 1 is associated with rice weediness.To identify the genomic modifications underlying weedy rice evolution,we generated the first de novo assembly of a high-quality weedy rice genome(WR04-6),and conducted a comparative genomics study between WR04-6 with other rice reference genomes.Multiple lines of evidence,including the results of demographic scenario comparisons,suggest that differentiation between weedy rice and cultivated rice was initiated by genetic improvement of cultivated rice and that the essence of weediness arose through semi-domestication.A plant height model further implied that the origin of WRAH can be modeled as an evolutionary game and indicated that strategy-based selection driven by fitness shaped its genomic diversity.Jian Sun Dianrong Ma Liang Tang Minghui Zhao Guangchen Zhangh Wenjia Wang Jiayu Song Xiang Li Zimeng Liu Wenxing Zhang Quan Xu Yuncheng Zhou Jianzhong Wu Toshio Yamamoto Fei Dai Yan Lei Song Li Gang Zhou Hongkun Zheng Zhengjin Xu Wenfu Chen 2019Molecular Plant2019,12,5:9

4Sortilin promotes macrophage cholesterol accumulation and aortic atherosclerosis through lysosomal degradation of ATP-binding cassette transporter A1 protein显示文摘Sortilin is closely associated with hyperlipidemia and the risk of atherosclerosis(AS).The role of sortilin and the underlying mechanism in peripheral macrophage are not fully understood.In this study,we investigated the effect of macrophage sortilin on ATP-binding cassette transporter A1(ABCA1)expression,ABCA1-mediated cholesterol efflux,and aortic AS.Macrophage sortilin expression was upregulated by oxidized low-density lipoproteins(ox-LDLs)in both concentration-and time-dependent manners.Its expression reached the peak level when cells were incubated with 50μg/ml ox-LDL for 24 h.Overexpression of sortilin in macrophage reduced cholesterol efflux,leading to an increase in intracellular total cholesterol,free cholesterol,and cholesterol ester.Sortilin was found to bind with ABCA1 protein and suppress macrophage ABCA1 expression,resulting in a decrease in cholesterol efflux from macrophages.The inhibitory effect of sortilin in cholesterol efflux was partially reversed by treatment with chloroquine,a lysosomal inhibitor.On the contrary,the ABCA1 protein level and ABCA1-mediated cholesterol efflux is increased by sortilin short hairpin RNA transfection.The fecal and biliary cholesterol 3H-sterol from cholesterol-laden mouse peritoneal macrophage was reduced by sortilin overexpression through lentivirus vector(LV)-sortilin in low-density lipoprotein receptor knockout mice,which was prevented by co-treatment with chloroquine.Treatment with LV-sortilin reduced plasma high-density lipoprotein and increased plasma ox-LDL levels.Accordingly,aortic lipid deposition and plaque area were exacerbated,and ABCA1 expression was reduced in mice in response to infection with LV-sortilin alone.These effects of LV-sortilin were partially reversed by chloroquine.Sortilin enhances lysosomal degradation of ABCA1 protein and suppresses ABCA1-mediated cholesterol efflux from macrophages,leading to foam cell formation and AS development.Yuncheng Lv Jing Yang Anbo Gao Sha Sun Xilong Zheng Xi Chen Wei Wan Chaoke Tang Wei Xie Suyun Li Dongming Guo Tianhong Peng Guojun Zhao Liyuan Zhong 2019Acta Biochimica et Biophysica Sinica2019,51,5:4
5Distribution of δ^(34)S and δ^(18)O in SO_4^(2-) in Groundwater from the Ordos Cretaceous Groundwater Basin and Geological Implications显示文摘The Ordos Cretaceous Groundwater Basin,located in an arid-semiarid area in northwestern China,is a large-style groundwater basin.SO_4^(2-) is one of the major harmful components in groundwater.Dissolved SO_4^(2-) concentrations,andδ^(34)S-SO_4^(2-) andδ^(18)O-SO_4^(2-) in groundwater from 14 boreholes and in gypsum from aquifer were analyzed.Results show that SO_4^(2-) in shallow groundwaters originates from precipitation,sulfide oxidation,and dissolution of stratum sulphate,with a big range ofδ^(34)S values,from-10.7‰to 9.2‰,and addition of SO_4^(2-) in deep groundwater results from dissolution of stratum sulphate,with biggerδ^(34)S values,from 7.8‰to 18.5‰,compared with those in shallow groundwater.This research also indicates that three types of sulphate are present in the strata,and characterized by highδ^(34)S values and highδ^(18)O values-style,highδ^(34)S values and middleδ^(18)O valuesstyle, middleδ^(34)S values and lowδ^(18)O values-style,respectively.Theδ^(34)S-SO_4^(2-) andδ^(18)O-SO_4^(2-) in groundwater have a good perspective for application in distinguishing different groundwater systems and determining groundwater circulation and evolution in this area.YANG Yuncheng SHEN Zhaoli WEN Dongguang HOU Guangcai SHE Hongquan ZHAO Zhenhong WANG Dong LI Jingwen 2010Acta Geologica Sinica(English Edition)2010,84,2:3
6A novel secretagogin/ATF4 pathway is involved in oxidized LDL-induced endoplasmic reticulum stress and islet β-cell apoptosis显示文摘Excessive accumulation of cholesterol inβcells initiates endoplasmic reticulum(ER)stress and associated apoptosis.We have reported that excessive uptake of cholesterol by MIN6 cells decreases the expression of secretagogin(SCGN)and then attenuates insulin secretion.Here,we aimed to determine whether cholesterol-induced SCGN decrease is involved in the modulation of ER stress and apoptosis in pancreaticβcells.In this study,MIN6 cells were treated with oxidized low-density lipoprotein(ox-LDL)for 24 h,and then intracellular lipid droplets and cell apoptosis were quantified,and SCGN and ER stress markers were identified by western blot analysis.Furthermore,small interfer RNA(siRNA)-mediated SCGN knockdown and recombinant plasmid-mediated SCGN restoration experiments were performed to confirm the role of SCGN in ER stress and associated cell apoptosis.Finally,the interaction of SCGN with ATF4 was computationally predicted and then validated by a co-immunoprecipitation assay.We found that ox-LDL treatment increased the levels of ER stress markers,such as phosphorylated protein kinase-like endoplasmic reticulum kinase,phosphorylated eukaryotic initiation factor 2 alpha,activating transcription factor 4(ATF4),and transcription factor CCAAT-enhancer-binding protein homologous protein,and promoted MIN6 cell apoptosis;in addition,the expression of SCGN was downregulated.siRNA-mediated SCGN knockdown exacerbatedβ-cell ER stress by increasing ATF4 expression.Pretreatment of MIN6 cells with the recombinant SCGN partly antagonized ox-LDL-induced ER stress and apoptosis.Furthermore,a co-immunoprecipitation assay revealed an interaction between SCGN and ATF4 in MIN6 cells.Taken together,these results demonstrated that pancreaticβ-cell apoptosis induced by ox-LDL treatment can be attributed,in part,to an SCGN/ATF4-dependent ER stress response.Li Wu Yuncheng Lv Ying Lv Sunmin Xiang Zhibo Zhao Ziqing Tang Linling Ou Bin Yan Xinhua Xiao Gebo Wen Renxian Cao Jing Yang 2021Acta Biochimica et Biophysica Sinica2021,53,1:2
7Temporal-spatial distributions of road silt loadings and fugitive road dust emissions in Beijing from 2019 to 2020显示文摘Road silt loading(sL)is an important parameter in the fugitive road dust(FRD)emissions.In this study,the improved Testing Re-entrained Aerosol Kinetic Emissions from Roads(TRAKER)combined with the AP-42 method was firstly developed to quickly measure and estimate the sLs of paved roads in Beijing,China.The annual average sLs in Beijing was 0.59±0.31 g/m^(2)in 2020,and decreased by 22.4%compared with that in 2019.The seasonal variations of sLs followed the order of spring>winter>summer>autumn in the two years.The seasonal mean road sLs on the same type road in the four seasons presented a decline trend from^(2)019 to 2020,especially on the Expressway,decreasing 47.4%-72.7%.The road sLs on the different type roads in the same season followed the order of Major arterial∼Minor arterial∼Branch road>Express road,and Township road∼Country highway>Provincial highway∼National highway.The emission intensities of PM10 and PM_(2.5)from FRD in Beijing in 2020 were lower than those in 2019.The PM10 and PM_(2.5)emission intensities at the four planning areas in the two years all presented the order of the capital functional core area>the urban functional expansion area>the urban development new area>the ecological conservation and development area.The annual emissions of PM10 and PM_(2.5)from FRD in Beijing in 2020 were 74,886 ton and 18,118 ton,respectively,decreasing by∼33.3%compared with those in 2019.Haibin Wang Lihui Han Tingting Li Song Qu Yuncheng Zhao Shoubin Fan Tong Chen Haoran Cui Junfang Liu 2023Journal of Environmental Sciences2023,,10:1

10Air pollutant emissions from the asphalt industry in Beijing,China显示文摘Improving our understanding of air pollutant emissions fromthe asphalt industry is critical for the development and implementation of pollution control policies.In this study,the spatial distribution of potential maximum emissions of volatile organic compounds(VOCs)in the complete life cycle of asphaltmixtures,as well as the particulate matter(PM),asphalt fume,nonmethane hydrocarbons(NMHCs),VOCs,and benzoapyrene(BaP)emissions from typical processes(e.g.,asphalt and concrete mixing stations,asphalt heating boilers,and asphalt storage tanks)in asphalt mixing plants,were determined in Beijing in 2017.The results indicated that the potential maximum emissions of VOCs in the complete life cycle of asphalt mixtures were 18,001 ton,with a large contribution from the districts of Daxing,Changping,and Tongzhou.The total emissions of PM,asphalt fume,NMHC,VOCs,and BaP from asphalt mixing plants were 3.1,12.6,3.1,23.5,and 1.9×10^(−3)ton,respectively.The emissions of PMfromasphalt and concretemixing stations contributed themost to the total emissions.The asphalt storage tankwas the dominant emission source of VOCs,accounting for 96.1%of the total VOCs emissions in asphalt mixing plants,followed by asphalt heating boilers.The districts of Daxing,Changping,and Shunyi were the dominant regions for the emissions of PM,asphalt fume,NMHC,and BaP,while the districts of Shunyi,Tongzhou,and Changping contributed the most emissions of VOCs.Song Qu Shoubin Fan Gang Wang Wanqing He Kangli Xu Lei Nie Yuncheng Zhao Qingchun Zhu Tingting Li Guohao Li 2021Journal of Environmental Sciences2021,33,11:1
11PAID study design on the role of PKC activation in immune/inflammation-related depression:a randomised placebo-controlled trial protocol显示文摘Background Inflammation that is mediated by microglia activation plays an important role in the pathogenesis of depression.Microglia activation can lead to an increase in the levels of proinflammatory cytokines,including TNF-α,which leads to neuronal apoptosis in the specific neural circuits of some brain regions,abnormal cognition and treatment-resistant depression(TRD).Protein kinase C(PKC)is a key regulator of the microglia activation process.We assume that the abnormality in PKC might result in abnormal microglia activation,neuronal apoptosis,significant changes in emotional and cognitive neural circuits,and TRD.In the current study,we plan to target at the PKC signal pathway to improve the TRD treatment outcome.Methods and analysis This is a 12-week,ongoing,randomised,placebo-controlled trial.Patients with TRD(N=180)were recruited from Shanghai Mental Health Center,Shanghai Jiao Tong University.Healthy control volunteers(N=60)were recruited by advertisement.Patients with TRD were randomly assigned to‘escitalopram+golimumab(TNF-αinhibitor)’,‘escitalopram+calcium tablet+vitamin D(PKC activator)’or‘escitalopram+placebo’groups.We define the primary outcome as changes in the 17-item Hamilton Depression Rating Scale(HAMD-17).The secondary outcome is defined as changes in anti-inflammatory effects,cognitive function and quality of life.Discussion This study might be the first randomised,placebo-controlled trial to target at the PKC signal pathway in patients with TRD.Our study might help to propose individualised treatment strategies for depression.Trial registration number The trial protocol is registered with ClinicalTrials.gov under protocol ID 81930033 and ClinicalTrials.gov ID NCT04156425.Xiaoyun Guo Ruizhi Mao Lvchun Cui Fan Wang Rubai Zhou Yun Wang Jia Huang Yuncheng Zhu Yamin Yao Guoqing Zhao Zezhi Li Jun Chen Jinhui Wang Yiru Fang 2021General Psychiatry2021,34,2:0
