1Alkali metal cation doping of metal-organic framework for enhancing carbon dioxide adsorption capacity显示文摘Metal-organic frameworks(MOFs) have attracted much attention as adsorbents for the separation of CO2 from flue gas or natural gas. Here,a typical metal-organic framework HKUST-1(also named Cu-BTC or MOF-199) was chemically reduced by doping it with alkali metals(Li, Na and K) and they were further used to investigate their CO2 adsorption capacities. The structural information, surface chemistry and thermal behavior of the prepared adsorbent samples were characterized by X-ray powder diffraction(XRD), thermo-gravimetric analysis(TGA)and nitrogen adsorption-desorption isotherm analysis. The results showed that the CO2 storage capacity of HKUST-1 doped with moderate quantities of Li+, Na+and K+, individually, was greater than that of unmodified HKUST-1. The highest CO2 adsorption uptake of 8.64mmol/g was obtained with 1K-HKUST-1, and it was ca. 11% increase in adsorption capacity at 298 K and 18 bar as compared with HKUST-1. Moreover, adsorption tests showed that HKUST-1 and 1K-HKUST-1 displayed much higher adsorption capacities of CO2 than those of N2. 2Capture of carbon dioxide from flue gas on TEPA-grafted metal-organic framework Mg_2 (dobdc)显示文摘Carbon dioxide(CO2) adsorption on a standard metal-organic framework Mg2(dobdc)(Mg/DOBDC or Mg-MOF-74) and a tetraethylenepentamine(TEPA) modified Mg2(dobdc)(TEPA-Mg/DOBDC) were investigated and compared. The structural information, surface chemistry and thermal behavior of the adsorbent samples were characterized by X-ray powder difraction(XRD), infrared spectroscopy(IR), thermogravimetric analysis(TGA) and nitrogen adsorption-desorption isotherm analysis. CO2adsorption capacity was measured by dynamic adsorption experiments with N2-CO2mixed gases at 60°C. Results showed that the CO2adsorption capacity of Mg/DOBDC was significantly improved after amine modification, with an increase from 2.67 to 6.06 mmol CO2/g adsorbent.Moreover, CO2adsorption on the TEPA-Mg/DOBDC adsorbent was promoted by water vapor, and the adsorption capacity was enhanced to 8.31 mmol CO2/g absorbent. The adsorption capacity of the TEPA-Mg/DOBDC adsorbent dropped only 3% after 5consecutive adsorption/desorption cycles. 3Quantifying 3D cell-matrix interactions during mitosis and the effect of anticancer drugs on the interactions显示文摘The mechanical force between cells and the extracellular microenvironment is crucial to many physiological processes such as cancer metastasis and stem cell differentiation. Mitosis plays an essential role in all these processes and thus an in-depth understanding of forces during mitosis gains insight into disease diagnosis and disease treatment. Here, we develop a traction force microscope method based on monolayer fluorescent beads for measuring the weak traction force (tens of Pa) of mitotic cells in three dimensions. We quantify traction forces of human ovarian granulosa (KGN) cells exerted on the extracellular matrix throughout the entire cell cycle in three dimensions. Our measurements reveal how forces vary during the cell cycle, especially during cell division. Furthermore, we study the effect of paclitaxel (PTX) and nocodazole (NDZ) on mitotic KGN cells through the measurement of traction forces. Our results show that mitotic cells with high concentrations of PTX exert a larger force than those with high concentrations of NDZ, which proved to be caused by changes in the structure and number of microtubules. These findings reveal the key functions of microtubule in generating traction forces during cell mitosis and explain how dividing cells regulate themselves in response to anti-mitosis drugs. 4A microfluidic approach for synchronous and nondestructive study of the permeability of multiple oocytes显示文摘Investigation of oocyte membrane permeability plays a crucial role in fertility preservation,reproductive medicine,and reproductive pharmacology.However,the commonly used methods have disadvantages such as high time consumption,low efficiency,and cumbersome data processing.In addition,the developmental potential of oocytes after measurement has not been fully validated in previous studies.Moreover,oocytes can only maintain their best status in vitro within a very limited time.To address these limitations,we developed a novel multichannel microfluidic chip with newly designed micropillars that provide feasible and repeatable oocyte capture.The osmotic responses of three oocytes at different or the same cryoprotectant(CPA)concentrations were measured simultaneously,which greatly improved the measurement efficiency.Importantly,the CPA concentration dependence of mouse oocyte membrane permeability was found.Moreover,a neural network algorithm was employed to improve the efficiency and accuracy of data processing.Furthermore,analysis of fertilization and embryo transfer after perfusion indicated that the microfluidic approach does not damage the developmental potential of oocytes.In brief,we report a new method based on a multichannel microfluidic chip that enables synchronous and nondestructive measurement of the permeability of multiple oocytes.Zhongrong Chen Kashan Memon Yunxia Cao Gang Zhao 2020Microsystems & Nanoengineering2020,6,1:2
5Alterations in intestinal microbiota composition coincide with impaired intestinal morphology and dysfunctional ileal immune response in growing-finishing pigs under constant chronic heat stress显示文摘Background:Previous studies had shown that short-term acute heat stress(HS)affected the host’s metabolism and intestinal microbiota independent of feed intake(FI)reduction,and long-term calorie restriction caused intestinal morphological injuries and gut microbial alterations.However,research on the effects of constant chronic HS on intestinal microbial composition and the roles of FI reduction played in is limited.This study aimed to investigate the effects of 7-day constant chronic HS on the composition of intestinal microbes in growing-finishing pigs,and its relationship with pigs’performance,intestinal morphology,and ileal immune response.Twenty-four growingfinishing pigs(Duroc×Large White×Landrace,30±1 kg body weight)were randomly assigned to three treatments(n=8),1)thermal neutral(TN)conditions(25±1℃)with ad libitum FI,2)HS conditions(35±1℃)with ad libitum FI,3)pair-fed(PF)with HS under TN conditions to discriminate the confounding effects of dissimilar FI,and the FI was the previous day’s average FI of HS.The small intestinal segments(duodenum,jejunum,and ileum)and feces were collected on d 8.Results:Results indicated that HS drastically declined(P<0.05)average daily gain(ADG)and average daily feed intake(ADFI)(about 61%)in comparison with TN,and caused hyperpyrexia,meanwhile PF caused hypothermia.Morphological observation by light and electron microscopes showed that both HS and PF treatment decreased(P<0.05)the villus and microvillus height compared with TN.Additionally,HS increased(P<0.05)protein expression of heat shock protein 70 in the duodenum,jejunum,and ileum.Furthermore,the expression of tight junction protein zonula occluden-1(ZO-1)in the duodenum and ileum,and Occludin in the ileum were enhanced(P<0.05)compared with TN and PF.Moreover,HS significantly enhanced(P<0.05)the mRNA relative expression of inflammatory cytokines(TLR-2,TLR-4,and tumor necrosis factor-α(TNF-α),IL-6,IL-8,PG1–5,β-defensin 2(pBD-2)),mucins(mucin-1 and mucin-2)and P65 protein level in the ileal mucosa tissue.Intestinal microbiota analysis by 16S rRNA sequencing showed lower(P<0.10)αdiversity in both HS and PF,and a separated cluster ofβdiversity among groups.Compared with TN,HS but not PF mainly reduced(FDR<0.05)Bacteroidetes(phylum),Bacteroidia(class)and elevated the proportions of Proteobacteria(phylum,FDR<0.05),Bacillales(order,FDR<0.05),Planococcaceae(family,FDR<0.05),Kurthia(genus,FDR<0.05),Streptococcaceae(family,FDR<0.10)and Streptococcus(genus,FDR<0.10).Notably,Lactobacillales(order)was decreased(FDR<0.05)by PF alone.Furthermore,the Spearman correlation analysis indicated that the microbes prevalent in HS were positively(P<0.05)associated with intestinal morphological injuries indicators and ileal immune response parameters,and the microbes reduced in HS were negatively(P<0.05)with the performance data.Conclusions:Intestinal morphological injuries and ileal immune response caused by constant chronic HS independent of FI showed close connections with alterations in intestinal microbiota in growing-finishing pigs.Yunxia Xiong Shuting Cao Hao Xiao Qiwen Wu Hongbo Yi Zongyong Jiang Li Wang 2022Journal of Animal Science and Biotechnology2022,13,3:2

16Nanomechanical vibration profiling of oocytes显示文摘The beginning of a mammalian life commences with a fertilized oocyte.The study of oocytes is certainly one of the most intriguing scientific questions of our time.Herein,we studied oocytes from a mechanical perspective and characterized the typical life activities of oocytes by nanomechanical vibrations.During the development of oocytes from the germinal vesicle(GV)stage to the zygotes,the GV stage oocytes induced a significant nanomechanical vibration,compared with the oocytes in meiosis I(MI)and meiosis II(MII)stages and zygotes.We analyzed the characteristics of mechanical vibrations of oocytes,including the amplitude as well as the frequency.It showed that the amplitude and frequency of nanomechanical vibrations induced by oocytes were caused by the cytoskeleton(microfilaments)and the distribution of metabolic characteristics(mitochondria)within oocytes.This work provides a new perspective for clinical quality assessment and basic research of oocytes,and can open new doors for development of life science.Yongpei Peng Junhui Zhang Weiwei Xue Wenjie Wu Yu Wang Kainan Mei Ye Chen Depeng Rao Tianhao Yan Jianye Wang Yunxia Cao Shangquan Wu Qingchuan Zhang 2023Nano Research2023,16,2:0
17Cell-autonomous effect of cardiomyocyte branched-chain amino acid catabolism in heart failure in mice显示文摘Parallel to major changes in fatty acid and glucose metabolism,defect in branched-chain amino acid(BCAA)catabolism has also been recognized as a metabolic hallmark and potential therapeutic target for heart failure.However,BCAA catabolic enzymes are ubiquitously expressed in all cell types and a systemic BCAA catabolic defect is also manifested in metabolic disorder associated with obesity and diabetes.Therefore,it remains to be determined the cell-autonomous impact of BCAA catabolic defect in cardiomyocytes in intact hearts independent from its potential global effects.In this study,we developed two mouse models.One is cardiomyocyte and temporal-specific inactivation of the E1αsubunit(BCKDHA-cKO)of the branched-chainα-ketoacid dehydrogenase(BCKDH)complex,which blocks BCAA catabolism.Another model is cardiomyocyte specific inactivation of the BCKDH kinase(BCKDK-cKO),which promotes BCAA catabolism by constitutively activating BCKDH activity in adult cardiomyocytes.Functional and molecular characterizations showed E1αinactivation in cardiomyocytes was sufficient to induce loss of cardiac function,systolic chamber dilation and pathological transcriptome reprogramming.On the other hand,inactivation of BCKDK in intact heart does not have an impact on baseline cardiac function or cardiac dysfunction under pressure overload.Our results for the first time established the cardiomyocyte cell autonomous role of BCAA catabolism in cardiac physiology.These mouse lines will serve as valuable model systems to investigate the underlying mechanisms of BCAA catabolic defect induced heart failure and to provide potential insights for BCAA targeted therapy.Jia-yu Yu Nancy Cao Christoph DRau Ro-Po Lee Jieping Yang Rachel JRoth Flach Lauren Petersen Cansheng Zhu Yea-Lyn Pak Russell AMiller Yunxia Liu Yibin Wang Zhaoping Li Haipeng Sun Chen Gao 2023Acta Pharmacologica Sinica2023,44,7:0
