| 1 | MicroRNA-125b Induces Cancer Cell Apoptosis Through Suppression of Bcl-2 Expression显示文摘MicroRNAs(miRNAs) are small,noncoding RNAs which can often act as an oncogene or a tumor suppressor.Several miRNAs are associated with the development of hepatocellular carcinoma(HCC).We demonstrated that miR-125b significantly suppresses HCC cell proliferation and promotes apoptosis by inhibiting the gene expression of the anti-apoptotic protein,Bcl-2.Bioinformatic analysis indicated that the 3'UTR of Bcl-2 has binding sites for miR-125b.Luciferase reporter assay confirmed the ability of miR-125b to dramatically suppress Bcl-2 transcription,suggesting that Bcl-2 is a target gene for miR-125b.We concluded that miR-125b acts as a tumor suppressor in hepatic tumor development by targeting Bcl-2 and inducing cancer cell apoptosis. | Aihua Zhao Quan Zeng Xiaoyan Xie Junnian Zhou Wen Yue Yali Li Xuetao Pei | 2012 | Journal of Genetics and Genomics2012,39,1: | 30 |
| 2 | Weldability, microstructure and mechanical properties of laser-welded selective laser melted 304 stainless steel joints显示文摘Laser welding is a promising process for joining small components produced by selective laser melting (SLM) to fabricate the large-scale and complex-shaped parts. In the work, the morphology, microstructure, microhardness, tensile properties and corrosion resistance of the laser welded stress-relieved SLMed 304 stainless steel joints are investigated, as the different sections of stress-relieved SLMed 304 stainless steel are joined. Results show that the SLMed 304 stainless steel plates have a good laser weldability. The microstructure of laser-welded joints consists of the cellular dendrites in austenite matrix within columnar grains, exhibiting a coarser dendrite structure, lower microhardness (~220HV) and tensile properties (tensile strength of ~750 MPa, and area reduction of ~27.6%), but superior corrosion resistance to those of SLMed plates. The dendrite arm spacing of the joints varies from ~3.7μm in center zone, to ~5.0 (xm in fusion zone, to ~2.5 in epitaxial zone. The SLMed anisotropy shows a negligible effect on the microstructure and performance of the laser-welded joints. | 3 | Performance of a completely autotrophic nitrogen removal over nitrite process for treating wastewater with different substrates at ambient temperature显示文摘The stability and parameters of a bio-ceramic filter for completely autotrophic nitrogen removal were investigated.The completely autotrophic nitrogen removal over nitrite(CANON) reactor was fed with different concentrations of ammonia(400,300,and 200mg N/L) but constant influent ammonia load.The results showed that the CANON system can achieve good treatment performance at ambient temperature(15-23°C).The average removal rate and removal loading of NH4+-N and TN was 83.90%,1.26kg N/(m3·day),and 70.14%,1.09kg N/(m3·day),respectively.Among the influencing factors like pH,dissolved oxygen and alkalinity,it was indicated that the pH was the key parameter of the performance of the CANON system.Observing the variation of pH would contribute to better control of the CANON system in an intuitive and fast way.Denaturing gradient gel electrophoresis analysis of microorganisms further revealed that there were some significant changes in the community structure of ammonium oxidizing bacteria,which had low diversity in different stages,while the species of anaerobic ammonium oxidizing(anammox) bacteria were fewer and the community composition was relatively stable.These observations showed that anaerobic ammonia oxidation was more stable than the aerobic ammonia oxidation,which could explain that why the CANON system maintained a good removal efficiency under the changing substrate conditions. | Xiaoyan Chang Dong Li Yuhai Liang Zhuo Yang Shaoming Cui Tao Liu Huiping Zeng Jie Zhang | 2013 | Journal of Environmental Sciences2013,25,4: | 12 |
| 4 | Comparison of the short-term and long-term outcomes of laparoscopic hysterectomies and of abdominal hysterectomies: a case study of 4,895 patients in the Guangxi Zhuang Autonomous Region, China显示文摘Background: To evaluate the short-term and long-term outcomes after laparoscopic hysterectomy(LH) compared with abdominal hysterectomy(AH) in case of benign gynecological disease. Methods: A multi-center cohort retrospective comparative study of population among 4,895 hysterectomies(3,539 LH vs.1,356 AH) between 2007 and 2013 was involved. Operative time(OT), estimated blood loss(EBL), intra-operative and post-operative complications, passing flatus; days with indwelling catheter, questionnaires covering pelvic floor functions and sexual functions were assessed.Results: The EBL(174.1±157.4 vs. 263.1±183.2 cc, LH and AH groups, respectively), passing flatus(38.7±14.1 vs. 48.1±13.2 hours), days with indwelling catheter(1.5±0.6 vs. 2.2±0.8 days), use of analgesics(6.5% vs. 73.1%), intra-operative complication rate(2.4% vs. 4.1%), post-operative complication rate(2.3% vs. 5.7%), post-operative constipation(12.1% vs. 24.6%), mild and serious stress urinary incontinence(SUI) post-operative(P<0.001; P=0.014), and proportion of Female Sexual Functioning Index(FSFI) total score <26.55 post-operative(P<0.001) of the LH group were significantly less than those of AH group. There wereno significant differences in OT(106.5±34.5 vs. 106.2±40.3 min) between the two groups.Conclusions: LH is a safe and efficient operation for improving patients' long-term quality of life(Qo L), and LH is a cost-effectiveness procedure for treating benign gynecological disease. | 5 | The chromosome-level Hemerocallis citrina Borani genome provides new insights into the rutin biosynthesis and the lack of colchicine显示文摘Hemerocallis citrina Borani(huang hua cai in Chinese)is an important horticultural crop whose flower buds are widely consumed as a delicious vegetable in Asia.Here we assembled a high-quality reference genome of H.citrina using single-molecule sequencing and Hi-C technologies.The genome assembly was 3.77 Gb and consisted of 3183 contigs with a contig N50 of 2.09 Mb,which were further clustered into 11 pseudochromosomes.A larger portion(3.25 Gb or 86.20%)was annotated as a repetitive content and 54,295 protein-coding genes were annotated in the genome.Genome evolution analysis showed that H.citrina experienced a recent whole-genome duplication(WGD)event at~15.73 million years ago(Mya),which was the main factor leading to many multiple copies of orthologous genes.We used this reference genome to predict 20 genes involved in the rutin biosynthesis pathway.Moreover,our metabolomics data revealed neither colchicine nor its precursors in H.citrina,challenging the long-standing belief that this alkaloid causes poisoning by the plant.The results of our disruptive research are further substantiated by our genomic finding that H.citrina does not contain any genes involved in colchicine biosynthesis.The high-quality genome lays a solid foundation for genetic research and molecular breeding of H.citrina. | Zhixing Qing Jinghong Liu Xinxin Yi Xiubin Liu Guoan Hu Jia Lao Wei He Zihui Yang Xiaoyan Zou Mengshan Sun Peng Huang Jianguo Zeng | 2021 | Horticulture Research2021,8,1: | 6 |

| 8 | Highly efficient and genotype-independent barley gene editing based on anther culture显示文摘Recalcitrance to tissue culture and genetic transformation is the major bottleneck for gene manipulation in crops.In barley,immature embryos of Golden Promise have typically been used as explants for transformation.However,the genotype dependence of this approach limits the genetic modification of commercial varieties.Here,we developed an anther culture-based system that permits the effective creation of transgenic and gene-edited plants from commercial barley varieties.The protocol was tested in Golden Promise and four Australian varieties,which differed in phenology,callus induction,and green plant regeneration responses.Agrobacterium-mediated transformation was performed on microspore-derived callus to target the HvPDS gene,and T0 albinos with targeted mutations were successfully obtained from commercial varieties.Further editing of three targets was achieved with an average mutation rate of 53%in the five varieties.In 51 analyzed T0 individuals,Cas9 induced a large proportion(69%)of single-base indels and two-base deletions in the target sites,with variable mutation rates among targets and varieties.Both ontarget and off-target activities were detected in T1 progenies.Compared with immature embryo protocols,this genotype-independent platformcan deliver a high editing efficiency and more regenerant plants within a similar time frame.It shows promise for functional genomics and the application of CRISPR technologies for the precise improvement of commercial varieties. | Yong Han Sue Broughton Li Liu Xiao-Qi Zhang Jianbin Zeng Xiaoyan He Chengdao Li | 2021 | Plant Communications2021,2,2: | 3 |
| 9 | Intestinal dysbiosis exacerbates the pathogenesis of psoriasis-like phenotype through changes in fatty acid metabolism显示文摘The intestinal microbiota has been associated with host immunity as well as psoriasis;however,the mechanism of intestinal microbiota regulating psoriasis needs to be demonstrated systematically.Here,we sought to examine its role and mechanism of action in the pathogenesis of psoriasis.We found that the severity of psoriasis-like skin phenotype was accompanied by changes in the composition of the intestinal microbiota.We performed co-housing and fecal microbial transplantation(FMT)experiments using the K14-VEGF transgenic mouse model of psoriasis and demonstrated that the transfer of intestinal microbiota from mice with severe psoriasis-like skin phenotype exacerbated psoriasiform skin inflammation in mice with mild symptoms,including increasing the infiltration and differentiation of Th17,and increased the abundance of Prevotella,while decreasing that of Parabacteroides distasonis,in the colon.These alterations affected fatty acid metabolism,increasing the abundance of oleic and stearic acids.Meanwhile,gentamicin treatment significantly reduced the abundance of Prevotella and alleviated the psoriasis-like symptoms in both K14-VEGF mice and imiquimod(IMQ)-induced psoriasis-like mice.Indeed,administration of oleic and stearic acids exacerbated psoriasis-like symptoms and increased Th17 and monocyte-derived dendritic cell infiltration in the skin lesion areas in vivo,as well as increased the secretion of IL-23 by stimulating DCs in vitro.At last,we found that,treatment of PDE-4 inhibitor alleviated psoriasis-like phenotype of K14-VEGF mice accompanied by the recovery of intestinal microbiota,including the decrease of Prevotella and increase of Parabacteroides distasonis.Overall,our findings reveal that the intestinal microbiota modulates host metabolism and psoriasis-like skin inflammation in mice,suggesting a new target for the clinical diagnosis and treatment of psoriasis. | Qixiang Zhao Jiadong Yu Hong Zhou Xiaoyan Wang Chen Zhang Jing Hu Yawen Hu Huaping Zheng Fanlian Zeng Chengcheng Yue Linna Gu Zhen Wang Fulei Zhao Pei Zhou Haozhou Zhang Nongyu Huang Wenling Wu Yifan Zhou Jiong Li | 2023 | Signal Transduction and Targeted Therapy2023,8,2: | 2 |

Firstly, the convolution results of several convolution kernels are combined by this method, and then the results after combination are made pooling operation, three sorts of CNN models(we named TBCNN, MCT-CNN and MMCT-CNN respectively) are constructed and then corresponding algorithmic thought are detailed on this basis. Secondly, relevant experiments and analyses are respectively designed to show the effects of three key parameters(convolution kernel, combination kernel number and word embedding) on three kinds of CNN models and to further demonstrate the effect of the models proposed.
