| 1 | A rapid advice guideline for the diagnosis and treatment of 2019 novel coronavirus(2019-nCoV) infected pneumonia(standard version)显示文摘In December 2019, a new type viral pneumonia cases occurred in Wuhan, Hubei Province;and then named '2019 novel coronavirus(2019-nCoV)' by the World Health Organization(WHO) on 12 January 2020. For it is a never been experienced respiratory disease before and with infection ability widely and quickly, it attracted the world’s attention but without treatment and control manual. For the request from frontline clinicians and public health professionals of 2019-nCoV infected pneumonia management, an evidence-based guideline urgently needs to be developed. Therefore, we drafted this guideline according to the rapid advice guidelines methodology and general rules of WHO guideline development;we also added the first-hand management data of Zhongnan Hospital of Wuhan University. This guideline includes the guideline methodology, epidemiological characteristics, disease screening and population prevention, diagnosis, treatment and control(including traditional Chinese Medicine), nosocomial infection prevention and control, and disease nursing of the 2019-nCoV. Moreover, we also provide a whole process of a successful treatment case of the severe 2019-nCoV infected pneumonia and experience and lessons of hospital rescue for 2019-nCoV infections. | 3 | Advances in low-protein diets for swine显示文摘Recent years have witnessed the great advantages of reducing dietary crude protein(CP) with free amino acids(AA)supplementation for sustainable swine industry, including saving protein ingredients, reducing nitrogen excretion, feed costs and the risk of gut disorders without impairing growth performance compared to traditional diets. However, a tendency toward increased fatness is a matter of concern when pigs are fed low-protein(LP) diets. In response, the use of the net energy system and balanced AA for formulation of LP diets has been proposed as a solution. Moreover, the extent to which dietary CP can be reduced is complicated. Meanwhile, the requirements for the first five limiting AA(lysine, threonine, sulfur-containing AA, tryptophan, and valine) that growing-finishing pigs fed LP diets were higher than pigs fed traditional diets, because the need for nitrogen for endogenous synthesis of non-essential AA to support protein synthesis may be increased when dietary CP is lowered. | 4 | Early kidney injury during long-term adefovir dipivoxil therapy for chronic hepatitis B显示文摘AIM: To evaluate urine β2-microglobulin(β2-M), retinol-binding protein(RBP) excretion, and renal impairment with adefovir dipivoxil(ADV) for chronic hepatitis B. METHODS: We enrolled 165 patients with chronic hepatitis B infection who were treated with ADV monotherapy(n = 90) or ADV plus lamivudine combination therapy(n = 75). An additional 165 chronic hepatitis B patients treated with entecavir were recruited as controls. We detected serum creatinine, urine β2-M, and RBP levels, and estimated the glomerular filtration rate(e GFR) at the initiation of antiviral therapy and every 6 mo for a period of five years. RESULTS: Urine β2-M abnormalities were observed in patients during the first(n = 3), second(n = 7), third(n = 11), fourth(n = 16), and fifth(n = 21) year of ADV treatment. Urinary RBP abnormalities were observed in patients during the first(n = 2), second(n = 8), third(n = 12), fourth(n = 15), and fifth(n = 22) year of ADV treatment. e GFR decreased 20%-30% from baseline in 20 patients, 30%-50% in 12 patients, and > 50% in 3 patients during the five years of treatment. Further analysis indicated that decreases in e GFR of ≥ 30% relative to the baseline level correlated significantly with urine RBP and β2-M abnormalities. In contrast, both serum creatinine and e GFR remained stable in patients treated with entecavir, and only one of these patients developed a urine β2-M abnormality, and two developed urine RBP abnormalities during the five years of treatment. | 5 | Telomere and telomerase in the initial stage of immortalization of esophageal epithelial cell显示文摘AIM: To search for the biomarker of cellular immortalization,the telomere length, telomerase activity and its subunits incultured epithelial cells of human fetal esophagus in theprocess of immortalization.METHODS: The transgenic cell line of human fetalesophageal epithelium (SHEE) was established with E6 E7genes of bt man papillomavirus (HPV) type 18 in ourlaboratory. Morphological phenotype of cultured SHEE cellsfrom the 6th to 30th passages, was examined by phasecontrast microscopy, the telomere length was assayed bySouthern blot method, and the activity of telomerase wasanalyzed by telomeric repeat amplification protocol (TRAP).Expressions of subunits of telomerase, hTR and hTERT,were assessed by RT-PCR. DNA content in cell cycle wasdetected by flow cytometry. The cell apoptosis wasexamined by electron microscopy (EM) and TUNEL label.RESULTS: SHEE cells from the 6 th to 10 th passagesshowed cellular proliferation with a good differentiation.From the 12 th to the 16 th passages, many senescent andapoptotic cells appeared, and the telomere length sharplyshortened from 23 kb to 17 kb without expression of hTERTand telornerase activity. At the 20 th passage, SHEE cellsovercame the senescence and apoptosis and restored theirproliferative activity with expression of telomerase andhTERT at low levels, but the telomere length shortenedcontinuously to the lowest of 3 kb. After the 30 th passagecells proliferation was restored by increment of cells at S andG2M phase in the cell cycle end telomerase activity expressedat high levels and with maintenance of telomere length.CONCLUSION: At the early stage of SHEE cells, telomeresare shortened without expression of telomerase and hTERTcausing cellular senescence and cell death. From the 20 thto the 30 th passages, the activation of telomerase andmaintenance of telomere length show a progressive processfor immortalization of esophageal epithelial cells. | 6 | LncRNA SNHG12 ameliorates brain microvascular endothelial cell injury by targeting miR-199a显示文摘Long non-coding RNAs regulate brain microvascular endothelial cell death, the inflammatory response and angiogenesis during and after ischemia/reperfusion and oxygen-glucose deprivation/reoxygenation(OGD/R) insults. The long non-coding RNA, SNHG12, is upregulated after ischemia/reperfusion and OGD/R in microvascular endothelial cells of the mouse brain. However, its role in ischemic stroke has not been studied. We hypothesized that SNHG12 positively regulates ischemic stroke, and therefore we investigated its mechanism of action. We established an OGD/R mouse cell model to mimic ischemic stroke by exposing brain microvascular endothelial cells to OGD for 0, 2, 4, 8, 16 or 24 hours and reoxygenation for 4 hours. Quantitative real-time polymerase chain reaction showed that SNHG12 levels in brain microvascular endothelial cells increased with respect to OGD exposure time. Brain microvascular endothelial cells were transfected with pc DNA-control, pc DNA-SNHG12, si-control, or si-SNHG12. After exposure to OGD for 16 hours, these cells were then analyzed by 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide, trypan blue exclusion, western blot, and capillary-like tube formation assays. Overexpression of SNHG12 inhibited brain microvascular endothelial cell death and the inflammatory response but promoted angiogenesis after OGD/R, while SNHG12 knockdown had the opposite effects. miR-199a was identified as a target of SNHG12, and SNHG12 overexpression reversed the effect of miR-199a on brain microvascular endothelial cell death, the inflammatory response, and angiogenesis. | 8 | Biodegradable chitosan scaffolds containing microspheres as carriers for controlled transforming growth factor-β_1 delivery for cartilage tissue engineering显示文摘Background Natural articular cartilage has a limited capacity for spontaneous regeneration.Controlled release oftransforming growth factor-β_1(TGF-β_1)to cartilage defects can enhance chondrogenesis.In this study,we assessed thefeasibility of using biodegradable chitosan microspheres as carriers for controlled TGF-β_1 delivery and the effect ofreleased TGF-β_1 on the chondrogenic potential of chondrocytes.Methods Chitosan scaffolds and chitosan microspheres loaded with TGF-β_1 were prepared by the freeze-drying andthe emulsion-crosslinking method respectively.In vitro drug release kinetics,as measured by enzyme-linkedimmunosorbent assay,was monitored for 7 days.Lysozyme degradation was performed for 4 weeks to detect in vitrodegradability of the scaffolds and the microspheres.Rabbit chondrocytes were seeded on the scaffolds containingTGF-β_1 microspheres and incubated in vitro for 3 weeks.Histological examination and type Ⅱcollagenimmunohistochemical staining was performed to evaluate the effects of released TGF-β_1 on cell adhesivity,proliferationand synthesis of the extracellular matrix.Results TGF-β_1 was encapsulated into chitosan microspheres and the encapsulation efficiency of TGF-β_1 was high(90.1%).During 4 weeks of incubation in lysozyme solution for in vitro degradation,the mass of both the scaffolds andthe microspheres decreased continuously and significant morphological changes was noticed.From the releaseexperiments,it was found that TGF-β_1 could be released from the microspheres in a multiphasic fashion including aninitial burst phase,a slow linear release phase and a plateau phase.The release amount of TGF-β_1 was 37.4%,50.7%,61.3%,and 63.5% for 1,3,5,and 7 days respectively.At 21 days after cultivation,type Ⅱ collagen immunohistochemicalstaining was performed.The mean percentage of positive cells for collagen type II in control group(32.7%±10.4%)wassignificantly lower than that in the controlled TGF-β_1 release group(92.4%±4.8%,P<0.05).Both the proliferation rateand production of collagen type Ⅱ in the transforming growth factor-β_1 microsphere incorporated scaffolds weresignificantly higher than those in the scaffolds without microspheres,indicating that the activity of TGF-β_1 was retainedduring microsphere fabrication and after growth factor release.Conclusion Chitosan microspheres can serve as delivery vehicles for controlled release of TGF-β_1,and the releasedgrowth factor can augment chondrocytes proliferation and synthesis of extracellular matrix.Chitosan scaffoldsincorporated with chitosan microspheres loaded with TGF-β_1 possess a promising potential to be applied for controlledcytokine delivery and cartilage tissue engineering. | CAI Dao-zhang ZENG Chun QUAN Da-ping BU Li-si WANG Kun LU Hua-ding LI Xiao-feng | 2007 | Chinese Medical Journal2007,,3: | 20 |
| 9 | Expansion and differentiation of human hepatocyte-derived liver progenitor-like cells and their use for the study of hepatotropic pathogens显示文摘The study of pathophysiological mecha nisms in huma n liver disease has bee n constrained by the in ability to expa nd primary hepatocytes in vitro while maintaining proliferative capacity and metabolic function. We and others have previously shown that mouse mature hepatocytes can be con verted to liver progenitor-like cells in vitro with defined chemical factors. Here we describe a protocol achieving efficient conversion of human primary hepatocytes into liver progenitor-like cells (HepLPCs) through delivery of developmentally relevant cues, including NAD +-dependent deacetylase SIRT1 signaling. These HepLPCs could be expanded sign ifica ntly duri ng in vitro passage. The expa nded cells can readily be con verted back into metabolically functional hepatocytes in vitro and upon tran splantati on in vivo. Under three-dime nsional culture conditions, differentiated cells gen erated from HepLPCs regai ned the ability to support infect io n or reactivatio n of hepatitis B virus (HBV). | 10 | Extracorporeal shock-wave versus pneumatic ureteroscopic lithotripsy in treatment of lower ureteral calculi显示文摘Aim:To compae the efficacy and complications of extracorporeal shock-wave lithotripsy (SWL) and pneumatic ureteroscopic lithotripsy(URS) in the treatment of lower ureteral calculi.Methods:From August 1997 to June 1999,210 patients with calculi in the distal third of the ureter were treated with SWL and other 180 with URS.The stones were fragmented with either HB-ESWL-V lithotripter or JML-93 pneumatic lithotripter through Wolf 7.5-9.0 Fr ureteroscope.The outcome was assessed in terms of stone clearance rate,re-treatment rate and complication incidence.Results:the stone clearance rate was 78.1% with SWL and 93.3% with URS (p<0.05).SWL had a re-treatment rate of 11.9%,vs 2.2% in the URS group (P<0.05).URS caused ureteral perforation in 3.3% of patients,while it was o with SWL (P<0.05).The differences in the incidence of other complications such as infection and stricture between the two groups were insignificant.conclusion:Though the selection of these two options depends on equipments available and the expertise of the operator,we recomment URS as the optimal treatment for distal ureteral calculi. | Zeng, GQ Zhong, WD Cai, YB Dai, QS Hu, JB Wei, HA | 2002 | Asian Journal of Andrology2002,4,4: | 17 |
| 11 | Experimental study of an aircraft fuel tank inerting system显示文摘In this work, a simulated aircraft fuel tank inerting system has been successfully established based on a model tank. Experiments were conducted to investigate the influences of different operating parameters on the inerting effectiveness of the system, including flow rate of the inert gas(nitrogen-enriched air), inert gas concentration, fuel load of the tank and different inerting approaches. | 12 | Research advances in the detection of miRNA显示文摘MicroRNAs (miRNAs) are a family of endogenous, small (approximately 22 nucleotides in length), noncoding, functional RNAs. With the development of molecular biology, the research of miRNA biological function has attracted significant interest, as abnormal miRNA expression is identified to contribute to serious human diseases such as cancers. Traditional methods for miRNA detection do not meet current demands. In particular, nanomaterial-based methods, nucleic acid amplification-based methods such as rolling circle amplification (RCA), loop-mediated isothermal amplification (LAMP), strand-displacement amplification (SDA) and some enzyme-free amplifications have been employed widely for the highly sensitive detection of miRNA. MiRNA functional research and clinical diagnostics have been accelerated by these new techniques. Herein, we summarize and discuss the recent progress in the development of miRNA detection methods and new applications. | 13 | Outcomes of preoperative chemoradiotherapy followed by surgery in patients with unresectable locally advanced sigmoid colon cancer显示文摘Background: Complete resection of locally advanced sigmoid colon cancer(LASCC) is sometimes difficult. Patients with LASCC have a dismal prognosis and poor quality of life, which has encouraged the evaluation of alternative multimodality treatments. This prospective study aimed to assess the feasibility and efficacy of neoadjuvant chemora?diotherapy(neo CRT) followed by surgery as treatment of selected patients with unresectable LASCC.Methods: We studied the patients with unresectable LASCC who received neo CRT followed by surgery between October 2010 and December 2012. The neoadjuvant regimen consisted of external?beam radiotherapy to 50 Gy and capecitabine?based chemotherapy every 3 weeks. Surgery was scheduled 6–8 weeks after radiotherapy.Results: Twenty?one patients were included in this study. The median follow?up was 42 months(range, 17–57 months). All patients completed neo CRT and surgery. Resection with microscopically negative margins(R0 resection) was achieved in 20 patients(95.2%). Pathologic complete response was observed in 8 patients(38.1%). Multivisceral resection was necessary in only 7 patients(33.3%). Two patients(9.5%) experienced grade 2 postopera?tive complications. No patients died within 30 days after surgery. For 18 patients with pathologic M0(yp M0) disease, the cumulative probability of 3?year local recurrence?free survival, disease?free survival and overall survival was 100.0%, 88.9% and 100.0%, respectively. | 14 | Polysaccharide extracts of Astragalus membranaceus and Atractylodes macrocephala promote intestinal epithelial cell migration by activating the polyamine-mediated K^+ channel显示文摘Astragalus membranaceus(Radix Astragali, RA) and Atractylodes macrocephala(Rhizoma Atractylodis Macrocephalae, RAM) are often used to treat gastrointestinal diseases. In the present study, we determined the effects of polysaccharides extracts from these two herbs on IEC-6 cell migration and explored the potential underlying mechanisms. A migration model with IEC-6 cells was induced using a single-edged razor blade along the diameter of cell layers in six-well polystyrene plates. The cells were grown in control--media or media containing spermidine(5 μmol·L^(-1), SPD), alpha-difluoromethylornithine(2.5 mmol·L^(-1), DFMO), 4-Aminopyridine-(40 μ-mol·L^(-1), 4-AP), the polysaccharide extracts of RA or RAM(50, 100, or 200 mg·L^(-1)), DFMO plus SPD, or DFMO plus polysaccharide extracts of RA or RAM for 12 or 24 h. Next, cytosolic free Ca^(2+)([Ca^(2+)]cyt) was measured using laser confocal microscopy, and cellular polyamine content was quantified with HPLC. Kv1.1 mRNA expression was assessed using RT-qPCR and Kv1.1 and RhoA protein expressions were measured with Western blotting analysis. A cell migration assay was carried out using Image-Pro Plus software. In addition, GC-MS was introduced to analyze the monosaccharide composition of both polysaccharide extracts. The resutls showed that treatment with polysaccharide extracts of RA or RAM significantly increased cellular polyamine content, elevated [Ca^(2+)]cyt and accelerated migration of IEC-6 cells, compared with the controls(P < 0.01). Polysaccharide extracts not only reversed the inhibitory effects of DFMO on cellular polyamine content and [Ca^(2+)]cyt, but also restored IEC-6 cell migration to control level(P < 0.01 or < 0.05). Kv1.1 mRNA and protein expressions were increased(P < 0.05) after polysaccharide extract treatment in polyamine-deficient IEC-6 cells and RhoA protein expression was increased. Molar ratios of D-ribose, D-arabinose, L-rhamnose, D-mannose, D-glucose, and D-galactose was 1.0:14.1:0.3:19.9:181.3:6.3 in RA and 1.0:4.3:0.1:5.7:2.8:2.2 in RAM. | 15 | Pediatric reference intervals in China(PRINCE):design and rationale for a large,multicenter collaborative cross-sectional study显示文摘There is a lack of accurate pediatric reference intervals(RIs) in China, with most commonly used RIs established without consideration of the effect of age and gender. The Pediatric Reference Intervals in China(PRINCE) project aims to establish and verify pediatric RIs for 31 common laboratory measurands.The project will be a large, multicenter cross-sectional study:14,490 healthy children and adolescents aged up to 19 years will be surveyed by 10 children's hospitals and one pediatric department of a university hospital. To evaluate the feasibility and efficiency of the study methods, 602 children were surveyed in the pilot phase of the PRINCE study in April 2017: it found that some measurands were distinctly age dependent and that there were differences between values for males and females. The results of the pilot study affirmed the necessity of the PRINCE project for Chinese pediatrics. The pilot also indicated potential difficulties in the full survey, e.g., difficulties in recruiting children aged under 3 years and insufficient collection of blood samples from infants. | 18 | Exogenous application of a low concentration of melatonin enhances salt tolerance in rapeseed(Brassica napus L.) seedlings显示文摘Melatonin is a naturally occurring compound in plants. Here, we tested the effect of exogenous melatonin on rapeseed(Brassica napus L.) grown under salt stress. Application of 30 μmol L^(–1) melatonin alleviated salt-induced growth inhibition, and the shoot fresh weight, the shoot dry weight, the root fresh weight, and the root dry weight of seedlings treated with exogenous melatonin increased by 128.2, 142.9, 122.2, and 124.2%, respectively, compared to those under salt stress. In addition, several physiological parameters were evaluated. The activities of antioxidant enzymes including peroxidase(POD), catalase(CAT) and ascorbate peroxidase(APX) were enhanced by 16.5, 19.3, and 14.2% compared to their activities in plants without exogenous melatonin application under salt stress, while the H_2O_2 content was decreased by 11.2% by exogenous melatonin. Furthermore, melatonin treatment promoted solute accumulation by increasing the contents of proline(26.8%), soluble sugars(15.1%) and proteins(58.8%). The results also suggested that higher concentrations(>50 μmol L^(–1)) of melatonin could attenuate or even prevent the beneficial effects on seedling development. In conclusion, application of a low concentration of exogenous melatonin to rapeseed plants under salt stress can improve the H_2O_2-scavenging capacity by enhancing the activities of antioxidant enzymes such as POD, CAT and APX, and can also alleviate osmotic stress by promoting the accumulation of osmoregulatory substances such as soluble proteins, proline, and water soluble glucan. | 20 | Submarine Landslides on the North Continental Slope of the South China Sea显示文摘Recent and paleo-submarine landslides are widely distributed within strata in deep-water areas along continental slopes, uplifts, and carbonate platforms on the north continental margin of the South China Sea(SCS). In this paper, high-resolution 3D seismic data and multibeam data based on seismic sedimentology and geomorphology are employed to assist in identifying submarine landslides. In addition, deposition models are proposed that are based on specific geological structures and features, and which illustrate the local stress field over entire submarine landslides in deep-water areas of the SCS. The SCS is one of the largest fluvial sediment sinks in enclosed or semi-enclosed marginal seas worldwide. It therefore provides a set of preconditions for the formation of submarine landslides, including rapid sediment accumulation, formation of gas hydrates, and fluid overpressure. A new concept involving temporal and spatial analyses is tested to construct a relationship between submarine landslides and different time scale trigger mechanisms, and three mechanisms are discussed in the context of spatial scale and temporal frequency: evolution of slope gradient and overpressure, global environmental changes, and tectonic events. Submarine landslides that are triggered by tectonic events are the largest but occur less frequently, while submarine landslides triggered by the combination of slope gradient and overpressure evolution are the smallest but most frequently occurring events.
