| 3 | Inhibition of SARS-CoV-2 (previously 2019-nCoV) infection by a highly potent pan-coronavirus fusion inhibitor targeting its spike protein that harbors a high capacity to mediate membrane fusion显示文摘The recent outbreak of coronavirus disease(COVID-19)caused by SARS-CoV-2 infection in Wuhan,China has posed a serious threat to global public health.To develop specific anti-coronavirus therapeutics and prophylactics,the molecular mechanism that underlies viral infection must first be defined.Therefore,we herein established a SARS-CoV-2 spike(S)protein-mediated cell-cell fusion assay and found that SARS-CoV-2 showed a superior plasma membrane fusion capacity compared to that of SARS-CoV.We solved the X-ray crystal structure of six-helical bundle(6-HB)core of the HR1 and HR2 domains in the SARS-CoV-2 S protein S2 subunit revealing that several mutated amino acid residues in the HR1 domain may be associated with enhanced interactions with the HR2 domain.We previously developed a pan-coronavirus fusion inhibitor,EK1,which targeted the HR!domain and could inhibit infection by divergent human coronaviruses tested,including SARS-CoV and MERS-CoV.Here we generated a series of lipopeptides derived from EK1 and found that EK1C4 was the most potent fusion inhibitor against SARS-CoV-2 S protein-mediated membrane fusion and pseudovirus infection with IC50s of 1.3 and 15.8 nM,about 241-and 149-fold more potent than the original EK1 peptide,respectively.EK1C4 was also highly effective against membrane fusion and infection of other human coronavirus pseudoviruses tested,including SARS-CoV and MERS-CoV,as well as SARSr-CoVs,and potently inhibited the replication of 5 live human coronaviruses examined,including SARS-CoV-2.Intranasal application of EK1C4 before or after challenge with HCoV-OC43 protected mice from infection,suggesting that EK1C4 could be used for prevention and treatment of infection by the currently circulating SARS-CoV-2 and other emerging SARSr-CoVs. | Shuai Xia Meiqin Liu Chao Wang Wei Xu Qiaoshuai Lan Siliang Feng Feifei Qi Linlin Bao Lanying Du Shuwen Liu Chuan Qin Fei Sun Zhengli Shi Yun Zhu Shibo Jiang Lu Lu | 2020 | Cell Research2020,30,4: | 81 |
| 4 | A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. | 7 | Effects of different resuscitation fluid on severe acute pancreatitis显示文摘AIM: To compare effects of different resuscitation fluid on microcirculation, inflammation, intestinal barrier and clinical results in severe acute pancreatitis (SAP). METHODS: One hundred and twenty patients with SAP were enrolled at the Pancreatic Disease Institute between January 2007 and March 2010. The patients were randomly treated with normal saline (NS group), combination of normal saline and hydroxyethyl starch (HES) (SH group), combination of normal saline, hydroxyethyl starch and glutamine (SHG group) in resuscitation. The ratio of normal saline to HES in the SH and SHG groups was 3:1. The glutamine (20% glutamine dipeptide, 100 mL/d) was supplemented into the resuscitation liquid in the SHG group. Complications and outcomes including respiratory and abdominal infection, sepsis, abdominal hemorrhage, intra-abdominal hypertension, abdominal compartment syndrome (ACS), renal failure, acute respiratory distress syndrome (ARDS), multiple organ dysfunction syndrome (MODS), operation intervention, length of intensive care unit stay, length of hospital stay, and mortality at 60 d were compared. Moreover, blood oxygen saturation (SpO 2 ), gastric intramucosal pH value (pHi), intra-abdominal pressure (IAP), inflammation cytokines, urine lactulose/mannitol (L/M) ratio, and serum endotoxin were investigated to evaluate the inflammatory reaction and gut barrier. RESULTS: Compared to the NS group, patients in the SH and SHG groups accessed the endpoint more quickly (3.9 ± 0.23 d and 4.1 ± 0.21 d vs 5.8 ± 0.25 d, P < 0.05) with less fluid volume (67.26 ± 28.53 mL/kg/d, 61.79 ± 27.61 mL/kg per day vs 85.23 ± 21.27 mL/kg per day, P < 0.05). Compared to the NS group, incidence of renal dysfunction, ARDS, MODS and ACS in the SH and SHG groups was obviously lower. Furthermore, incidence of respiratory and abdominal infection was significantly decreased in the SH and SHG groups, while no significant difference in sepsis was seen. Moreover, less operation time was needed in the SH and SHG group than the NS group, but the difference was not significant. The mortality did not differ significantly among these groups. Blood SpO 2 and gastric mucosal pHi in the SH and SHG groups increased more quickly than in the NS group, while IAP was significantly decreased in the SH and SHG group. Moreover, the serum tumor necrosis factor-α, interleukin-8 and C-reactive protein levels in the SH and SHG groups were obviously lower than in the NS group at each time point. | 11 | Chinese expert consensus on cytoreductive surgery and hyperthermic intraperitoneal chemotherapy for peritoneal malignancies显示文摘Locoregional spread of abdominopelvic malignant tumors frequently results in peritoneal carcinomatosis(PC). The prognosis of PC patients treated by conventional systemic chemotherapy is poor, with a median survival of < 6 mo. However, over the past three decades, an integrated treatment strategy of cytoreductive surgery(CRS) + hyperthermic intraperitoneal chemotherapy(HIPEC) has been developed by the pioneering oncologists, with proved efficacy and safety in selected patients. Supported by several lines of clinical evidence from phases Ⅰ, Ⅱ and Ⅲ clinical trials, CRS + HIPEC has been regarded as the standard treatment for selected patients with PC in many established cancer centers worldwide. In China, an expert consensus on CRS + HIPEC has been reached by the leading surgical and medical oncologists, under the framework of the China Anti-Cancer Association. | 15 | Estimation of internal nutrient release in large shallow Lake Taihu, China显示文摘Based on field investigation of wave, sediment suspension and the changes in nutrient concentration of the water column in Lake Taihu, China, we proposed two release models to quantify nutrient release under static and dynamic conditions, respectively. Under static conditions, nutrient release from sediments to the overlying water mainly depends on chemical diffusion induced by concentration gradient, in which the nutrient release is controlled by the temperature, dissolved oxygen concentration in the sediment-water interface, oxidation-reduction potential and the concentration difference between porewater and overlying water. Under dynamic condition (or disturbed condition),both dissolved and particulate nutrients in sediments are released into the water column because of wind-induced sediment suspension. The amount of nutrient release under dynamic conditions is larger than that under the static condition. The release of dissolved nutrients, however, does not increase because the wind induced turbulence made oxidation of metallic elements such as Fe (ferric iron), Mn which are capable of precipitating soluble reactive phosphate (SRP). Under dynamic conditions, therefore, the release of total phosphorus (TP) increases dramatically but the release of SRP is close to those under static conditions. In sediments of Lake Taihu, high Fe content leads to a high ratio of Fe to P contents in sediments (Fe:P ratio). Under dynamic conditions, therefore, nutrient release is controlled by the intensity of disturbance, sediment consolidation and nutrient content in sediments.As for dissolved nutrients, especially SRP, the release is also controlled by the intensity of dynamic re-oxidation, Fe content in sediments and nutrient concentration gradient between porewater and overlying water. Based on these two release modes, the release flux in Lake Taihu has been estimated. In the static condition (i.e. laboratory experimental condition), total release of NH4+-N for whole lake is ca. 10,000 ton/a, and PO43--P is ca. 900 ton/a. In the dynamic condition, nutrient release following sediment suspension was estimated according to three different intensities of wind forcing which were defined as 'calm' (wind speed is less than 2 m/s), 'gentle' (wind speed is greater than 2 m/s and less than 6 m/s) and 'gust' (wind speed is greater than 6 m/s). The release rate in the condition of 'calm' was estimated in terms of the nutrient release in the laboratory experimental static condition; whereas the release rate in conditions of 'gentle' and 'gust' was estimated in terms of measurement during sediment resuspension conducted in flume experiments. With the observation of wind velocity and frequency in 2001, each type of wind forcing took the frequency of 12%, 82% and 6% for 'calm', 'gentle' and 'gust', respectively. | 16 | Mechanism and control of lake eutrophication显示文摘A review about lake naturally eutrophi- cating, the internal loading of nutrients from lake sediment as well as the mechanism of algal blooms and the control practices was made, especially the eutrophication problem of shallow lakes since sev- enty percent of fresh water lakes in China are shallow lakes. It was found that shallow lakes are apt toward eutrophication than deep lakes. Without any influ- ences of human activity, shallow lakes in the middle and lower reaches of Yangtze River are still easily eutrophicated, which may be owing to the effects of flood in this area. In shallow lakes, sediments are frequently disturbed by wind-wave and resuspended, which result in huge nutrients release to overlying water. This may be the major reason for higher in- ternal loading of nutrients in shallow lakes than in deep lakes. Algal bloom is an extreme response of lake ecosystem to the eutrophication. Appearance of algal blooms is related to physical condition of lakes, such as underwater radiation (or transparency), temperature, and hydrodynamic conditions, or related to geochemical conditions of lakes, like concentra- tions of nutrients and ratio of nitrogen to phosphorus, as well as the physiological advantage of cyanobac- teria such as vacuole for moving towards the radiant energy-rich zone and the mycosporine-like amino acids (MAAs) for resisting the harm of ultraviolet ra- diation. In shallow lakes, these advantages of cyanobacteria are favorable in the competition than in deep lakes. Also being the shallowness, it is more difficult to reduce nutrient loading and to control algae blooms in shallow lakes. For the control of eutrophi- cation, people should follow the sequence from pollution sources control, ecological restoration tocatchment management. To control the internal nu- trient release, physical, chemical, biological tech- niques, and even bionic techniques could be selected. The idea of ecological restoration for a eutrophic lake is to shift the ecosystem from phytoplankton-domi- nant state to macrophyte-dominant state. To realize the shift of ecosystem state, environmental condition improvement is the fundamental work. Nowadays, we should do more work on environmental condition improvement than on planting of macrophytes since we are lack of the knowledge about the relationship between macrophyte and lake ecosystem. Empha- sizing the macrophyte planting, therefore, has blind- ness at present. | 17 | Microbial treatment in chronic constipation显示文摘Chronic functional constipation is a kind of common intestinal disease that occurs in children, adults and elderly people. This disease not only causes great influence to physiological function, but also results in varying degrees of psychological barriers. At present, constipation treatments continue to rely on traditional methods such as purgative therapy and surgery. However, these approaches can disrupt intestinal function. Recent research between intestinal diseases and gut microbiota has gradually revealed a connection between constipation and intestinal flora disturbance, providing a theoretical basis for microbial treatment in chronic constipation. Microbial treatment mainly includes probiotic preparations such as probiotics, prebiotics, synbiotics and fecal microbiota transplantation(FMT). Due to its safety, convenience and curative effect, probiotic preparations have been widely accepted, especially gradually developed FMT with higher curative effects. Microbial treatment improves clinical symptoms, promotes the recovery of intestinal flora, and has no complications during the treatment process. | 18 | Incidence and mortality of laryngeal cancer in China,2008–2012显示文摘Objective:To analyze the incidence and mortality rates of laryngeal cancer in China from 2008 to 2012.Methods:Incident and death cases of laryngeal cancer were retrieved from the National Central Cancer Registry(NCCR)database collecting from 135 cancer registries in China during 2008–2012.The crude incidence and mortality rates of laryngeal cancer were calculated by area(urban/rural),region(eastern,middle,western),gender and age group(0,1–4,5–9,…,85+).China census in 2000 and Segi’s world population were applied for age standardized rates.Join Point(Version 4.5.0.1)model was used for time trend analysis.Results:The crude incidence rate of laryngeal cancer was 1.86/100,000 ranked the 21st in overall cancers.The age-standardized incidence rates by China population(ASIRC)and by World population(ASIRW)were1.22/100,000 and 1.23/100,000,respectively.The crude mortality of laryngeal cancer in China was 1.01/100,000and it was the 21st cause of cancer-related death in overall cancers.Both the age-standardized mortality rates by Chinese standard population(ASMRC)and by world standard population(ASMRW)were 0.63/100,000.Incidence and mortality rates of laryngeal cancer were higher in males than in females and higher in urban areas than in rural areas.Middle areas had the highest incidence and mortality rates followed by eastern and western areas.Incidence and mortality rates of laryngeal cancer retained low level before age of 40 years old but increased greatly after and peaked in age group of 75.Incidence showed significant down trends in recent 10 years by 1.27%annually[95%confidence interval(95%CI):–2.2%,–0.3%].Mortality declined in females sharply by 5.18%per year although stable in males and both sexes combined.Conclusions:Appropriate targeted prevention,early detection and treatment programs should be carried out to control the local burden of laryngeal cancer. | Yuqin Liu Qin Zhao Gaoheng Ding Yitong Zhu Wenying Li Wanqing Chen | 2018 | Chinese Journal of Cancer Research2018,30,3: | 29 |
| 19 | Fibrinogen-like protein 2 fibroleukin expression and its correlation with disease progression in murine hepatitis virus type 3-induced fulminant hepatitis and in patients with severe viral hepatitis B显示文摘AIM: To evaluate the expression of fibrinogenlike protein 2 (fgl2) and its correlation with disease progression in both mice and patients with severe viral hepatitis.METHODS: Balb/cJ or A/J mice were infected intraperitoneally (ip) with 100 PFU of murine hepatitis virus type 3 (MHV-3), liver and serum were harvested at 24, 48, and 72 h post infection for further use. Liver tissues were obtained from 23 patients with severe acute chronic (AOC) hepatitis B and 13 patients with mild chronic hepatitis B. Fourteen patients with mild chronic hepatitis B with cirrhosis and 4 liver donors served as normal controls. In addition, peripheral blood mononuclear cells (PBMC) were isolated from 30 patients (unpaired) with severe AOC hepatitis B and 10 healthy volunteers as controls. Procoagulant activity representing functional prothrombinaseactivity in PBMC and white blood cells was also assayed. A polyclonal antibody against fgl2 was used to detect the expression of both mouse and human fgl2 protein in liver samples as well as in PBMC by immunohistochemistry staining in a separate set of studies. Alanine aminotransferase (ALT) and total bilirubin (TBil) in serum were measured to assess the severity of liver injury.RESULTS: Histological changes were found in liver sections 12-24 h post MHV-3 infection in Balb/cJ mice.In association with changes in liver histology, marked elevations in serum ALT and TBil were observed. Mouse fgl2 (mfgl2) protein was detected in the endothelium of intrahepatic veins and hepatic sinusoids within the liver 24 h after MHV-3 infection. Liver tissues from the patients with severe AOC hepatitis B had classical pathological features of acute necroinflammation. Human fgl2 (hfgl2)was detected in 21 of 23 patients (91.30%)with severe AOC hepatitis B, while only 1 of 13 patients(7.69%) with mild chronic hepatitis B and cirrhosis had hfgl2 mRNA or protein expression. Twenty-eight of thirty patients (93.33%) with severe AOC hepatitis B and 1of 10 with mild chronic hepatitis B had detectable hfgl2expression in PBMC. No hfgl2 expression was found either in the liver tissue or in the PBMC from normal donors. There was a positive correlation between hfgl2expression and the severity of the liver disease as indicated by the levels of TBil. | 20 | Herbal decoctosome is a novel form of medicine显示文摘Traditionally, herbal medicine is consumed by drinking decoctions produced by boiling herbs with water. The functional components of the decoction are heat stable. Small RNAs(sRNAs) were reported as a new class of functional components in decoctions. However, the mechanisms by which sRNAs survive heat treatment of the decoction and enter cells are unclear.Previous studies showed that plant-derived exosome-like nanoparticles(ELNs), which we call botanosomes, could deliver therapeutic reagents in vivo. Here, we report that heat-stable decoctosomes(ELNs) from decoctions have more therapeutic effects than the decoctions in vitro and demonstrate therapeutic efficacy in vivo. Furthermore, sRNAs, such as HJT-sRNA-m7 and PGY-sRNA-6, in the decoctosome exhibit potent anti-fibrosis and anti-inflammatory effects, respectively. Decoctosome is comprised of lipids, chemical compounds, proteins, and s RNAs. A medical decoctosome mimic is called bencaosome. A single lipid sphinganine(d22:0) identified in the decoctosome was mixed and heated with the synthesized sRNAs to form the simplest bencaosome. This simple bencaosome structure was identified by critical micelle concentration(cmc) assay that sRNAs coassembled with sphinganine(d22:0) to form the lipid layers of vesicles. The heating process facilitates co-assembly of sRNAs and sphinganine(d22:0) until a steady state is reached. The artificially produced sphinganine-HJT-sRNA-m7 and sphinganinePGY-sRNA-6 bencaosomes could ameliorate bleomycin-induced lung fibrosis and poly(I:C)-induced lung inflammation, respectively, following oral administration in mice.
