1Metastatic human hepatocellular carcinoma models in nude mice and cell line with metastatic potential显示文摘Metastatic human HCC model is needed for the studies on mechanism and intervention of metastatic recurrence. By using orthotopic implantation of histologically intact tissues of 30 surgical specimens, a patient like metastatic model of human HCC in nude mice (LCI-D20)and a Iow metastatic model of human HCC in nude mice LCI-D35 ) have been established. All mice with transplanted LCI-D20 tumors exhibited extremely high metastatic ability including spontaneous metastasis to liver, lungs, lymph nodes and peritoneal seeding.Remarkable difference was also found in expression of some of the invasiveness related genes and growth factors between the LCI-D20 and LCI-D35 tumors. PAI-Iincreased gradually following tumor progression in LCID20 model, and correlated with tumor size and AFP level,Phasic expression of tissue intercellular adhesion molecule-I in this model was also observed. Using corneal micropocket model, it was demonstrated that the vascular response induced by LCI-D20 tumor was stronger than that induced by LCI-D35 tumor. Similar report on metastatic human HCC model in nude mice and human HCC cell line with metastatic potential was rarely found in the literature. 5High-yield expression of recombinant SARS coronavirus nudeocapsid protein in methylotrophic yeast Pichia pastoris显示文摘AIM: Nucleocapsid (N) protein plays an important role in reproduction and pathological reaction of severe acute respiratory syndrome (SARS) coronavirus (SCoV), the antigenicity of the protein is better than spike (S) protein.This study was to find a highly specific and antigenic recombinant SCoV nucleocapsid (rSCoVN) protein, and to provide a basis for further researches on early diagnosis of SARS.METHODS: Full length cDNA of SCoV nucleocapsid (SCoVN) protein was amplified through polymerase chain reaction (PCR) and cloned into yeast expression vector pPIC3.5K to construct plasmid of pPIC3.5K-SCoVN. The plasmid was linearized and then transformed into Pichia pastoris (P.pastoris) GS115 (His^+ Mut^+) by electroporation. His^+Mut^+ recombinant strains were identified by PCR and cultivated on MM/MD plates. The influence of different factors on biomass and rSCoVN protein production during induction phase, such as various induction media, dissolved oxygen (DO) and different final concentrations of methanol, was subsequently studied. The expression level and activation were detected by SDS-PAGE and Western-blot respectively.RESULTS: All of the recombinants were His^+Mut^+ after transformation of P.pastoris with linearized plasmids. The BMMY medium was optimal for recombinant ScoVN (rSCoVN) protein expression and growth of the recombinant strains.The final optimal concentration of methanol was 20 ml_/L,the DO had a significant effect on rSCoVN protein expression and growth of recombinant strains. The rSCoVN protein expressed in recombinant strains was about 8% of the total cell protein, 520 mg/L of rSCoVN protein was achieved,and a maximum cell A at 600 nm of 62 was achieved in shake flask culture. The rSCoVN protein had a high specificity against mouse-anti-SARS-CoVN-mAb and SARS positive sera, but had no cross-reaction with normal human serum.The biological activity of rSCoVN expressed in P.pastoris was about 4-fold higher than that expressed in Ecoliwhen the same rSCoVN protein quantity was used.CONCLUSION: Active recombinant severe acute respiratory syndrome (SARS) coronavirus nucleocapsid (rSCoVN) protein can be successfully expressed in recombinant methylotrophic yeast P.pastoris GSl15. 6Production of laccase by Coriolus versicolor and its application in decolorization of dyestuffs: (Ⅱ) Decolorization of dyes by laccase containing fermentation broth with or without self-immobilized mycelia显示文摘The capability of decolorization for commercial dyes by Coriolus versicolor fermentation broth containing laccase with or without immobilized mycelium was evaluated. With cell free fermentation broth containing laccase, high decolorization ratio was achieved for acid orange 7, but not for the other dyes concerned. The immobilized mycelium was proved to be more efficient than the cell free system. All the four dyestuffs studied were found being decolourized with certain extent by immobilized mycelium. The repeated batch decolorization was carried out with satisfactory results. The experimental data showed that the continuous decolorization of wastewater from a printing and dyeing industry was possible by using the self immobilized C. 7Validity of the Chinese Version of the Patient-Generated Subjective Global Assessment (PG-SGA) in Lung Cancer Patients显示文摘The prevalence of malnutrition is high among oncology patients in China. Although the Patient- Generated Subjective Global Assessment (PG-SGA) is widely recognized as an important nutritional assessment tool, it has not been validated for Chinese cancer patients. The purpose of this study was to conduct an analysis of nutrition screening and assessment data to validate the Chinese version of the PG-SGA in Chinese patients with lung cancer. This was an observational, cross-sectional study of 2,000 consecutive adult patients with lung cancer treated at several tertiary hospitals in China. Anthropometric and patient descriptive data were collected. The PG-SGA generated a score for the nutritional risk and a global rating for the nutritional status. The internal consistency, external consistency, and construct validity were evaluated, and a known-groups comparison and principal component analyses were conducted to evaluate the reliability and validity of the PG-SGA. The known-groups comparison demonstrated the ability of the PG-SGA to differentiate lung cancer patients of different nutritional and body mass index (BMI) groups. A confirmatory factor analysis supported the original four-factor structure of the PG-SGA. In terms of the internal and external consistency, the PG-SGA demonstrated intra-class correlation coefficients (ICC) up to 0.987. Alternative form validity was also demonstrated between Box 4 of the PG-SGA and the Karnofsky Performance Status score, with excellent external consistency (r = 0.972, P < 0.001). The construct validity was supported, and selected questionnaire dimensions were evident in the principal component analysis. Significant Spearman correlations (P < 0.001) were demonstrated. The Chinese version of the PG-SGA demonstrated significant reliability and sufficient exploratory properties to support its validity. It seems to be a valid tool that can be used to access the nutritional status of Chinese lung cancer patients. 8Validity of the Chinese Version of the Patient-Generated Subjective Global Assessment (PG-SGA) in Gastric Cancer Patients显示文摘Purpose Malnutrition is severe among gastric cancer patients in China. While the Patient-Generated Subjective Global Assessment (PG-SGA) is a widely used nutritional assessment tool, the validation for Chinese patients with gastric cancer has not been performed yet. The aim of this research is to validate the Chinese version of the PGSGA in Chinese gastric cancer patients by conducting analyses of nutrition screening and assessment data. Methods Two thousand consecutive adult patients with gastric cancer were included in this observational, cross-sectional study in several tertiary hospitals in China. A survey was made among those patients, which included several questionnaires and some anthropometric parameters. We calculated the parameters to validate the use of PG-SGA categorizing the nutritional status of gastric cancer patients. Then we performed statistical analysis to evaluate the internal consistency, external consistency, and construct validity. Results This study verified the ability of the PG-SGA to differentiate gastric cancer patients into different nutritional groups. The confirmatory factor analysis verified the original four-factor structure of the PG-SGA. The internal and external consistency was measured by a concordance analysis which showed an intraclass correlation coefficients (ICC) of 0.987. The correlations of the Box 4 score with the Karnofsky performance status score demonstrated alternative form validity with good external consistency (Pearson, r = 0.643, P < 0.001). All three anthropometric dimensions of the PG-SGA were supported by principal component analysis. Spearman correlations (P < 0.001) were significant in the majority of tests. 9Prokaryotical expression of structural and non-structural proteins of hepatitis G virus显示文摘AIM To study the epitope distribution of hepatitis G virus (HGV) and to seek for the potential recombinant antigens for the development of HGV diagnositic reagents.METHODS Fourteen clones encompassing HGV gene fragments from core to NS3 and NS5 were constructed using prokaryotic expression vector pRSET and (or)pGEX. and expressed in E. coli. Western blotting and ELISA were used to detect the immunoreactivity of these recombinant proteins.``RESULTS One clone with HGV fragment from core to El(Gl). one from E2 (G31), three from NS3 (G6, G61, G7),one from NS5B (G821) and one chimeric fragment from NS3and NS5B (G61 821) could be expressed well and showed obvious immunoreactivity by Western blotting.One clone with I-KGV framment from NS5B (G82) was also well expressed, but could not show immunoreactivity by Western blotting. No obvious expression was found in the other six clones. All the expressed recombinant proteins were in inclusion body form, except the protein G61 which could be expressed in soluble form. Further purified recombinant proteins Gl, G,31, G61, G821 and G61 821were detected in indirected ELISA as coating antigen respectively. Only recombinant Gl could still show immunoreactivity, and the other four recombinant proteins failed to react to the HGV antibody positive sera.Western blotting results indicated that the immunoactivity of these four recombinant proteins were lost during purification.``CONCLUSION Core to El, E2. NS3 and NS5 fragment of HGV contain antigenic epitopes, which could be produced in prokaryotically expressed recombinant proteins. 11An initial discussion on major controlling factors on formation of coal-formed large-medium sas fields显示文摘The so-called 'large-medium gas field' means a gas field with proven reserves equal to or larger than 100×108 m3. Up to 1997, 23 coal-formed great-medium fields are discovered in China, excluding two in Taiwan Province. Their proven reserves take 50.9% in total natural gas reserves. Therefore, the coal-formed great-medium gas field plays a decisive role in natural gas reserves and the study of their controlling factors is very important both in theory and in practice. 13Direct in vivo injection of ^(131)I-GMS and its distribution and excretion in rabbit显示文摘AIM: To explore the distribution and metabolism of 131 I-gelatin microspheres ( 131 I-GMSs) in rabbits after direct injection into rabbits’ livers.METHODS: Twenty-eight healthy New Zealand rabbits were divided into seven groups,with four rabbits per group.Each rabbit’s hepatic lobes were directly injected with 41.336 ± 5.106 MBq 131 I-GMSs.Each day after 131 I-GMSs administration,4 rabbits were randomly selected,and 250 μL of serum was collected for γ count.Hepatic and thyroid functions were tested on days 1,4,8,16,24,32,48 and 64 after 131 I-GMSs administration.Single-photon emission computed tomography (SPECT) was taken for each group on days 0,1,4,8,16,24,32,48,64 after 131 I-GMSs administration.A group of rabbits were sacrificed respectively on days 1,4,16,24,32,48,64 after 131 I-GMSs administration.Their livers were taken out for histological examination.RESULTS: After 131 I-GMSs administration,the nuclide was collected in the hepatic area with microspheres.The radiation could be detected on day 48 after 131 IGMSs administration,and radiography could be seen in thyroid areas in SPECT on days 4,8,16 and 24.One day after 131 I-GMSs administration,the liver function was damaged but recovered 4 d later.Eight days after 131 I-GMSs administration,the levels of free triiodothyronine and free thyroxin were reduced,which restored to normal levels on day 16.Histological examination showed that the microspheres were degraded to different degrees at 24,32 and 48 d after 131 I-GMSs administration.The surrounding parts of injection points were in fibrous sheathing.No microspheres were detected in histological examination on day 64 after 131 I-GMSs administration.CONCLUSION: Direct in vivo injection of 131 I-GMSs is safe in rabbits.It may be a promising method for treatment of malignant tumors.Yu Ma,Yi Wan,Li-Geng Duan,Xiao-Li Chen,The Research Unit of Hepato-Bilio-Pancreatology and Department of Hepatic Surgery,West China Hospital of Sichuan University,Chengdu 610041,Sichuan Province,China Dong-Hui Luo,Chuan-Qin Xia,College of Chemistry,Sichuan University,Chengdu 610041,Sichuan Province,China Lin Li,Department of Nuclear Medicine and The National Key Discipline of Medical Imaging and Nuclear Medicine,West China Hospital of Sichuan University,Chengdu 610041,Sichuan Province,China 2010World Journal of Gastroenterology2010,16,17:3

15Effects of Delayed Second Cochlear Implant显示文摘Objective Since Helms' successful bilateral cochlear implantation with good results in 1996, there have been increasing number of reports on bilateral cochlear implantation. Most second device have been implantated within one year after the first. Considering effects of long time auditory deprivation, it is not clear whether a delayed second cochlear implant serves to add additional benefits and how it may interact with central nervous system plasticity. Methods Three cases who received delayed second cochlear implants at People's Hospital of Peking University from 2002 to 2005 were reviewed. The interval between the first and second implants was longer than 2 years in all three patients. Sound perception, and unilateral/bilateral speech discrimination in quiet and noise were evaluated. In addition, GAP detection test was conducted in one patient. Results In one case, having both implants on provided improved performance compared to using only one implant both in quiet and noise. Presumably due to visual interference from lip-reading or short interval between second implant and testing, one patient showed no improvement from using the second implant either in quiet or noise, while the last case demonstrated additional benefits from the second implant only in quiet. In all three patients, performance in recognizing the four tones in Mandarin was superior over word recognition. Conclusions Considerable plasticity in the cerebral auditory center is preserved, despite long acoustic deprivation in some children who have received unilateral cochlear implant. Delayed second implants can result in significant improvements in some of these children. Visual interference from lip-reading may be an obstacle during retraining. 19Impacts of High-Frequency Atmospheric Forcing on Southern OceanCirculation and Antarctic Sea Ice显示文摘The relative contributions of atmospheric fluctuations on 6 h?2 d,2?8 d,and 8 d?1 month time scales to the changes in the air?sea fluxes,the SO circulation,and Antarctic sea ice are investigated.It was found that the imposed forcing variability on the three time scales creates a significant increase in wind power input,and hence an increase of about 50%,97%,and 5%of eddy kinetic energy relative to the simulation driven by monthly forcing,respectively.Also,SO circulation and the strength of the upper cell of meridional overturning circulation become strengthened.These results indicate more dominant effects of atmospheric variability on the 2?8 d time scale on the SO circulation.Meanwhile,the 6 h?2 d(2?8 d)atmospheric variability causes an increase in the total sea-ice extent,area,and volume,by about 33%,30%,and 19%(17%,20%,and 25%),respectively,relative to those in the experiment forced by monthly atmospheric variables.Such significant sea-ice increases are caused by a cooler ocean surface and stronger sea-ice transports owing to the enhanced heat losses and air-ice stresses induced by the atmospheric variability at 6 h?2 d and 2?8 d,while the effects of the variability at 8 d?1 month are rather weak.The influences of atmospheric variability found here mainly result from wind fluctuations.Our findings in this study indicate the importance of properly resolving high-frequency atmospheric variability in modeling studies.Yang WU Zhaomin WANG Chengyan LIU and Xia LIN 2020Advances in Atmospheric Sciences2020,37,5:1
