| 2 | Inhibiting Hv1 channel in peripheral sensory neurons attenuates chronic inflammatory pain and opioid side effects显示文摘Both opioids and nonsteroidal anti-inflammatory drugs(NSAIDS)produce deleterious side effects and fail to provide sustained relief in patients with chronic inflammatory pain.Peripheral neuroinflammation(PN)is critical for initiation and development of inflammatory pain.A better understanding of molecular mechanisms underlying PN would facilitate the discovery of new analgesic targets and the development of new therapeutics.Emerging evidence suggests that peripheral sensory neurons are not only responders to painful stimuli,but are also actively engaged in inflammation and immunity,whereas the intrinsic regulatory mechanism is poorly understood.Here we report the expression of proton-selective ion channel Hv1 in peripheral sensory neurons in rodents and humans,which was previously shown as selectively expressed in microglia in mammalian central nervous system.Neuronal Hv1 was up-regulated by PN or depolarizing stimulation,which in turn aggravates inflammation and nociception.Inhibiting neuronal Hv1 genetically or by a newly discovered selective inhibitor YHV98-4 reduced intracellular alkalization and ROS production in inflammatory pain,mitigated the imbalance in downstream SHP-1-pAKT signaling,and also diminished pro-inflammatory chemokine release to alleviate nociception and morphine-induced hyperalgesia and tolerance.Thus,our data reveal neuronal Hv1 as a novel target in analgesia strategy and managing opioids-related side effects. | Qiansen Zhang Yimin Ren Yiqing Mo Peipei Guo Ping Liao Yuncheng Luo Jie Mu Zhuo Chen Yang Zhang Ya Li Linghui Yang Daqing Liao Jie Fu Juwen Shen Wei Huang Xuewen Xu Yanyan Guo Lianghe Mei Yunxia Zuo Jin Liu Huaiyu Yang Ruotian Jiang | 2022 | Cell Research2022,32,5: | 3 |
| 3 | Preparation of acellular nerve grafts with triton X-100显示文摘BACKGROUND: The source of nerve allograft enriches.We may choose expediently nerve allograft to repair injured nerve and the structure of choice nerve homlolgy or similar with the injured nerve,but the immunological rejiction limits the climical application of nerve allograft.The ideal substitute of autograft never is reaearching.OBJECTIVE: In this experiment,Triton X-100 was used to extract the Schwann cells and myelin sheaths of allograft nerve and obtain the inartiflcial and eliminated antigenicity nerve-transplanter(nerve grafts).DESIGN: Controlled experiment.SETTING: Department of Hand Surgery,the Third Affiliated Hospital of Hebei Medical University;Second Department of Orthopedics,Fourth Center Hospital of Tianjin.MATERIALS: Thirty heath New Zealand big ear white rabbit .of either sex(gender).weighing 2000-3000 g,were provided by the Center of Experimental Animal of Hebei Medical University.Triton X-100 was offered by SIGMA Company.METHODS: The experiment was carried out at the Central Laboratory of the Third Affiliated Hospital of Hebei Medical University from December 2003 to December 2004.Sixty pieces of sciatic nerves.10-mm-long nerve segment,which were taken from 30 rabbits,were incised.They were randomly divided into chemical extraction group(n=50)and control group(n=10).In the chemical extraction groups,the nerves were put into 3% Triton X-100 solution.They were treated with Triton X-100 for 12 hours,24 hours,48 hours,96 hours and 1 week.respectively.They were examined in every period.The control groups did not treated with anything. ①Respectively two segments of nerve by 2 mm length were taken from each nerve in the every periods.②The laminin immunohistochemical stained sections were performed with image acquisition and analyzed with multicolor pathological image analysis system.Measured the laminin antibody reaction part of each section and computed laminin average gray degrees of the unit area.All dates were analyzed by SPSS 10.0 software.MAIN OUTCOME MEASURES:①General observation and histological observation in two groups;②Compared with laminin average gray degrees of the unit area in each section.RESULTS:①General observation:In the control groups,fresh nerve was polish,rigidity and elasticity.After the nerves were chemical extracted,the floccules was seen at two ends and around of the nerves.The nerves being extraction presented ivory and lackluster. Its diameter and length compared reduced,tenderness and tenacity with the fresh nerve.0bserved by light microscope,Schwann cells,myelin sheaths and basement membrane distribute uniformly in control groups.After the nerves were extracted,Schwann cells and myelin sheaths disappeared. Basement membrane presented barrier array in longitudinal sections. Complex 1 displays a one dimensional (1D) chain based on the binuclear [Co2 (NPH)2 ] units and 3-bpcb ligands, which is extended into a three dimensional (3D) supramolecular framework through hydrogen bonding and - stacking interactions. In complex 2, the (3,5)-connected two dimensional (2D) layers are constructed from 1D Co-NIPH chains and bidentate-bridging 3-bpcb ligands. Complex 3 is a 2D double layer based on Co-5-H2 AIP 2D layers and 3-bpcb pillars. Complex 4 also displays a 2D network, which is constructed from the Co-5-H2AIP 1D double chains and 3-bpcd ligands. Finally, complexes 2-4 are extended into 3D supramolecular frameworks by hydrogen bonding or - stacking interactions. The substituent groups of dicarboxylates play an important role in the assembly and structures of the title complexes. | 5 | Effects of Municipal Sewage Sludge on Fixation of Cr,Ni,Cu,and Zn during Co-processing of Heavy Metal-containing Waste in Cement Kilns显示文摘Cement raw meal with MSS and different heavy metals was blended to examine the fixation ratios, chemical species, and cement crystalline phases in clinkers. The results showed that blending MSS could decrease the fixation ratio of Cr, Ni, Cu, and Zn in the produced clinker by 5% to 25%. And Cr, Cu, and Zn were mainly incorporated into clinkers as metal silicates, Ni was mainly solubilized in Mg O to form magnesium nickel oxides, and the transition phases were mainly metal aluminum oxides as indicated by X-ray diffraction. The reduction of fixation ratios was likely attributed to the presence of impure elements, such as sodium and phosphorus in MSS. In addition, high concentrations(eg, 1.7 wt%) of chlorine in MSS led to metal chloride formation that could vaporize Cu, Cr, Ni, and Zn. | 17 | ‘C-type’ closed state and gating mechanisms of K2P channels revealed by conformational changes of the TREK-1 channel显示文摘Two-pore domain potassium (K2P) channels gate primarily within the selectivity filter, termed ‘C-type’ gating. Due to the lack of structural insights into the nonconductive (closed) state, ‘C-type’ gating mechanisms remain elusive. Here, molecular dynamics (MD) simulations on TREK-1, a K2P channel, revealed that M4 helix movements induce filter closing in a novel ‘deeper-down’ structure that represents a ‘C-type’ closed state. The ‘down’ structure does not represent the closed state as previously proposed and instead acts as an intermediate state in gating. The study identified the allosteric ‘seesaw’ mechanism of M4 helix movements in modulating filter closing. Finally, guided by this recognition of K2P gating mechanisms, MD simulations revealed that gain-of-function mutations and small-molecule activators activate TREK-1 by perturbing state transitions from open to closed states.
