| 2 | Targeting the PI3K/Akt signaling pathway in gastric carcinoma: A reality for personalized medicine?显示文摘Frequent activation of phosphatidylinositol-3 kinases(PI3K)/Akt/m TOR signaling pathway in gastric cancer(GC) is gaining immense popularity with identification of mutations and/or amplifications of PIK3 CA gene or loss of function of PTEN,a tumor suppressor protein,to name a few; both playing a crucial role in regulating this pathway. These aberrations result in dysregulation of this pathway eventually leading to gastric oncogenesis,hence,there is a need for targeted therapy for more effective anticancer treatment. Several inhibitors are currently in either preclinical or clinical stages for treatment of solid tumors like GC. With so many inhibitors under development,further studies on predictive biomarkers are needed to measure the specificity of any therapeutic intervention. Herein,we review the common dysregulation of PI3K/Akt/m TOR pathway in GC and the various types of single or dual pathway inhibitors under development that might have a superior role in GC treatment. | 4 | Factors associated with time to laparoscopic cholecystectomy for acute cholecystitis显示文摘AIM: To determine patient and process of care factors associated with performance of timely laparoscopic cholecystectomy for acute cholecystitis. METHODS: A retrospective medical record review of 88 consecutive patients with acute cholecystitis was conducted. Data collected included demographic data, co-morbidities, symptoms and physical findings at presentation, laboratory and radiological investigations, length of stay, complications, and admission service (medical or surgical). Patients not undergoing cholecystectomy during this hospitalization were excluded from analysis. Hierarchical generalized linear models were constructed to assess the association of pre-operative diagnostic procedures, presenting signs, and admitting service with time to surgery.RESULTS: Seventy cases met inclusion and exclusion criteria, among which 12 were admitted to the medical service and 58 to the surgical service. Mean ± SD time to surgery was 39.3 ± 43 h, with 87% of operations performed within 72 h of hospital arrival. In the adjusted models, longer time to surgery was associated with number of diagnostic studies and endoscopic retrograde cholangio-pancreatography (ERCP, P = 0.01) as well with admission to medical service without adjustment for ERCP (P < 0.05). Patients undergoing both magnetic resonance cholangiopancreatography (MRCP) and computed tomography (CT) scans experienced the longest waits for surgery. Patients admitted to the surgical versus medical service underwent surgery earlier (30.4 ± 34.9 vs 82.7 ± 55.1 h, P < 0.01), had less post-operative complications (12% vs 58%, P < 0.01), and shorter length of stay (4.3 ± 3.4 vs 8.1 ± 5.2 d, P < 0.01).CONCLUSION: Admission to the medical service and performance of numerous diagnostic procedures, ERCP, or MRCP combined with CT scan were associated with longer time to surgery. | 5 | A subfossil chironomid-total phosphorus inference model for lakes in the middle and lower reaches of the Yangtze River显示文摘The results of an investigation into the relationship between surface sediment subfossil chi- ronomid distribution and water quality are presented. Data from 30 lakes in the middle and lower reaches of the Yangtze River indicate that the nutrient gradi- ent was the major factor affecting the distribution of chironomids across these sites. Canonical corre- spondence analysis (CCA) revealed that of 12 sum- mer water environmental variables, total Phosphorus was most important, accounting for 20.1% of the variance in the chironomid data. This was significant enough to allow the development of quantitative in- ference models. A TP inference model was devel- oped using weighted averaging (WA), partial least squares (PLS) and weighted averaging partial least squares (WA-PLS). An optimal two-component WA-PLS model provided a high jack-knifed coeffi- cient of prediction for conductivity r 2jack = 0.76, with a low root mean squared error of prediction (RMSEPjack = 0.13). | 8 | INTERVENTIONAI DIAGNOSIS AND TREATMENT OF VASCULOGENEIC PULSATILE TINNITUS显示文摘Objective To retrospectively study clinical features and diagnostic imaging of vasculogeneic pulsatile tinnitus, and the feasibility and efficacy of transvascular interventional treatment for this condition. Methods Data from 82 cases of arterial or venous pulsatile tinnitus were reviewed. DSA characteristics and possible pathophysiological mechanisms of pulsatile tinnitus in these cases were studied. Diagnoses in this group included intracranial arterovenous fistula(AVF)(n=3), spontaneous skull base dural AVF(n=16), traumatic carotid-cavernous sinus fistula(n=5), subclavian artery stenosis(n=2), internal carotid artery stenosis(n=3), intracranial arterial stenosis(n=1), kinked and/or elongated vertebrobasilar artery(n=2), venous sinus diverticulum(n=2), venous sinus stenosis on the dominant drainage side(n=46) and occipital sinus stenosis(n=2).Treatments included embolization and stenting using coils, NBCA glue, Balt balloons, self-expansion stents and intracranial micro-stents via either the femoral artery or femoral vein. Results Procedures were successful in all cases with no surgery-related complications. Tinnitus disappeared within 2 days after the procedure in all cases. Follow up duration was 5-36 months. Recurrence occurred in 4 cases of arterial tinnitus within 3 months following the initial procedure, which improved after revision embolization or symptom management. There was no recurrence in venous tinnitus cases following stent plastic or stent-coiling embolization treatments. Conclusions Endovascular intervention provides a new approach to the diagnosis and treatment of intractable pulsatile tinnitus. | 9 | Multiple locus genealogies and phenotypic characters reappraise the causal agents of apple ring rot in China显示文摘Apple ring rot inflicts severe economic losses in the main apple producing areas of East Asia.The causal agent of the disease has been variously identified as Macrophoma kuwatsukai,Physalospora piricola and Botryosphaeria berengeriana f.sp.piricola,although B.dothidea is currently the most widely accepted pathogen name.The taxonomic uncertainty has delayed research that is needed to manage effectively this destructive disease.In the present study,genealogical concordance phylogenetic species recognition(GCPSR)was applied to pathogenic fungal isolates from apple and pear from several locations in China,along with several reference isolates.Phylogenetic results based on sequences of four nuclear loci(ITS,EF-1α,HIS and HSP)revealed the existence of two species within the examined isolates.One includes an ex-epitype isolate of B.dothidea and the other includes an isolate that was previously designated as B.berengeriana f.sp.piricola.Morphologically,the latter taxon presented an appressed mycelial mat on PDA whereas B.dothidea displayed columns of aerial mycelia reaching the lids,and conidia of the latter species were longer than B.dothidea.Botryosphaeria dothidea had a faster growth rate than the latter taxon under relatively high temperatures.Pathogenicity tests showed that on pear stems the latter taxon caused large-scale cankers along with blisters whereas B.dothidea was non-pathogenic,but on apple shoots the two fungi induced large and small wart-like prominences,respectively.Overall,this cryptic species demonstrated sufficient genetic variations and biological differences from B.dothidea.As a result of taxonomic study,we described here the latter taxon in a new combination,Botryosphaeria kuwatsukai and designate an epitype.Both B.kuwatsukai and B.dothidea are considered to be the main causal agents for apple ring rot in China and Japan. | Chao Xu Chunsheng Wang Liangliang Ju Rong Zhang Alan R.Biggs Eiji Tanaka Bingzhi Li Guangyu Sun | 2015 | Fungal Diversity2015,,2: | 6 |

| 12 | Garcinol sensitizes human head and neck carcinoma to cisplatin in a xenograft mouse model despite downregulation of proliferative biomarkers显示文摘OBJECTIVE Platinum compounds such as cisplatin and carboplatin are frequently used as the first-line chemotherapy for the treatment of the head and neck squamous cell carcinoma(HNSCC).In the present study,we investigated whether garcinol,apolyisoprenylated benzophenone can chemosensitize HNSCC to cisplatin.METHODS The effect of garcinol and cisplatin on HNSCC was assessed by MTT,Western blotting,real time PCR,FACS,immunohistochemistry,DNA binding assay and xenograft mouse model.RESULTS We found that garcinol inhibited the viability of a panel of diverse HNSCC cell lines,enhanced the apoptotic effect of cisplatin,suppressed constitutive as well as cisplatin-induced NF-κB activation,and downregulated the expression of various oncogenic gene products(cyclin D1,Bcl-2,survivin and VEGF).In vivo study showed that administration of garcinol alone(0.5 mg·kg-1,ip five times/week)significantly suppressed the growth of the tumor,and this effect was further increased by cisplatin.Both the markers of proliferation index(Ki-67)and microvessel density(CD31)were downregulated in tumor tissues by the combination of cisplatin and garcinol.The pharmacokinetic results of garcinol indicated that good systemic exposure was achievable after ip administration of garcinol at 0.5and 2mg·kg-1 with mean peak concentration(cmax)of 1825.4 and 6635.7nmol·L-1 in the mouse serum,respectively.CONCLUSION Overall,our results suggest that garcinol can indeed potentiate the effects of cisplatin by negative regulation of various inflammatory and proliferative biomarkers. | Feng Li Muthu K. | 18 | 3D urchin like V-doped CoP in situ grown on nickel foam as bifunctional electrocatalyst for efficient overall water-splitting显示文摘Cobalt phosphide (CoP) is considered to be a potential candidate in the field of electrocatalysis due to its low-cost, abundant resources and high electrochemical stability. However, there is a great space for further improvement of its electrocatalytic performance since its charge transfer rate and catalytic activity have not reached a satisfactory level. Herein, we design and fabricate a three dimensional urchins like V-doped CoP with different amounts of V-doping on nickel foam electrode. The V-doped CoP/NF electrode with optimized amounts of V-doping (10%) exhibits outstanding hydrogen evolution reaction (HER) performance under universal-pH conditions and preeminent oxygen evolution reaction (OER) performance in alkaline media. Notably, the assembled water-splitting cell displays a cell voltage of only 1.53 V at 10 mA·cm−2 and has excellent durability, much better than many reported related bifunctional catalysts. The experiment results and theoretical analysis revealed that vanadium atoms replace cobalt atoms in CoP lattice. Vanadium doping can not only raise the density of electronic states near the Fermi level enhancing the conductivity of the catalyst, but can also optimize the free energy of hydrogen and oxygen-containing intermediates adsorption over CoP, thus promoting its catalytic activity.
