1Mouse macrophage specific knockout of SIRT1 influences macrophage polarization and promotes angiotensin Ⅱ-induced abdominal aortic aneurysm formation显示文摘Abdominal aortic aneurysm(AAA) is a vascular degenerative disease. Macrophage polarization and the balance between classically activated macrophages(M1) and alternatively activated macrophages(M2)are crucial for AAA pathogenesis. The present study aims to investigate the roles of macrophage SIRT1 in AAA formation and macrophage polarization. We found that in mouse peritoneal macrophages, SIRT1 expression was decreased after M1 stimulation, but was enhanced after M2 stimulation. Results from SIRT1^(flox/flox) mice and macrophage specific SIRT1 knockout mice with treatment of angiotensin II(Ang II)for 4 weeks showed that macrophage specific deficiency of SIRT1 increased the incidence of AAA and exacerbated the severity, including more severe aneurysm types, enlarged diameter of the aneurysm and increased degradation of elastin. In mouse aortas, SIRT1 deficiency increased the proinflammatory M1 molecule inducible nitric oxide synthase(i NOS), and decreased M2 molecules such as arginase 1(Arg1) and mannose receptor(MR). Furthermore, in peritoneal macrophages, SIRT1 deficiency increased the expression of M1 inflammatory molecules, but decreased the expression of M2 molecules. Overexpression of SIRT1 had the opposite effects. 2Synergistic defensive mechanism of phytochelatins and antioxidative enzymes in Brassica chinensis L. against Cd stress显示文摘Brassica chinensis L. was chosen and exposed to different concentrations of Cd exposure to evaluate its Cd-accumulating capacity and its potential cellular defensive mechanisms. Cd accumulation in the shoots and roots of B. chinensis was up to 1348.3±461.8 and 3761.0±795.0 mg per killogram of dry weight, respectively, under 200 μmol/L of Cd exposure. Increasing Cd accumulation in the plant was accompanied by rapid accumulation of phytochelatins (PCs), and the sequestration of Cd by PCs provided a primary cellular mechanism for Cd detoxification and tolerance of B. chinensis. Furthermore, malondialdehyde formation, hydrogen peroxide content and antioxidative enzyme activities such as superoxide dismutase, catalase, guaiacol peroxidase and ascorbate peroxidase were observed in the shoots of Cd-stressed B. chinensis. Increasing enzyme activities in response to concentrations of 5 to 50 μmol/L Cd showed an efficient defense against oxidative stress, suggesting that the antioxidative system was a secondary defensive mechanism. These resulted in reduced free Cd damage and enhanced Cd accumulation and tolerance. Glutathione plays a pivotal role in these two detoxification pathways. 3Cellulose nanofiber separator for suppressing shuttle effect and Li dendrite formation in lithium-sulfur batteries显示文摘Lithium-sulfur battery(LSB) has high energy density but is limited by the polysulfides shuttle and dendrite growth during cycling. Herein, a free-standing cellulose nanofiber(CNF) separator is designed and fabricated in isopropanol/water suspension through vacuum filtration progress. CNFs with abundant polar oxygen-containing functional groups can chemically immobilize the polysulfides, and suppress the formation of the dendrites by controlling the surface morphology of the SEI on lithium metal in LSB. The isopropanol content in a suspension can fine-tune the pore structure of the membrane to achieve optimal electrochemical performance. The prepared separator displays integrated advantages of an ultrathin thickness(19 μm), lightweight(0.87 mg cm^(-2)), extremely high porosity(98.05%), and decent electrolyte affinity. As a result, the discharge capacity of the LSB with CNF separator at the first and 100 th cycle is 1.4 and 1.3 times that of PP separator, respectively. 4Forensic investigation of 23 autosomal STRs and application in Han and Mongolia ethnic groups显示文摘A forensic validation study of the Early Access HuaxiaTM Platinum Polymerase Chain Reaction (PCR) kit was completed to document the performance capabilities and limitations.The genotyping of DNA samples was consistent across a large range of template DNA concentrations,with complete profiles obtained at 0.125 ng;however,no more than 2 mm× 1.2 mm punches of samples would be recommended for direct amplification.The size precision and accuracy test revealed the genotyping ability;while consistent results were obtained when comparing the kit with other commercially available systems.In addition,the whole PCR amplification can finish within approximately 45 min,making the system suitable for fastdetection.However,only partial profiles may be obtained with challenging samples,including DNA stored on Foam-Tipped Applicators (FTA) cards or some case samples.For the forensic application in ethnic groups,a total of 282 and 229 alleles were obtained in Han and Mongolia,respectively.Since the 23 short tandem repeats were independent from each other,the cumulative power of exclusion in duos was 0.999999157188 and the cumulative power of exclusion in trios was 0.999999999859 in the Han group while the cumulative power of exclusion in duos (CPEduo) was 0.999 998 848 26 and cumulative power of exclusion in trios (CPEtrio) was 0.999 999 999 79 in the Mongolia group.And good internal consistency was found between the two investigated groups and the Sichuan Han,Hui,Tibetan and Uygur according to available reference data.Xiang Sheng Yali Wang Jiashuo Zhang Liqin Chen Yuan Lin Zhenmin Zhao Chengtao Li Suhua Zhang 2018Forensic Sciences Research2018,3,2:4

6Nontrivial coupling of light into a defect:the interplay of nonlinearity and topology显示文摘The flourishing of topological photonics in the last decade was achieved mainly due to developments in linear topological photonic structures.However,when nonlinearity is introduced,many intriguing questions arise.For example,are there universal fingerprints of the underlying topology when modes are coupled by nonlinearity,and what can happen to topological invariants during nonlinear propagation?To explore these questions,we experimentally demonstrate nonlinearity-induced coupling of light into topologically protected edge states using a photonic platform and develop a general theoretical framework for interpreting the mode-coupling dynamics in nonlinear topological systems.Performed on laser-written photonic Su-Schrieffer-Heeger lattices,our experiments show the nonlinear coupling of light into a nontrivial edge or interface defect channel that is otherwise not permissible due to topological protection.Our theory explains all the observations well.Furthermore,we introduce the concepts of inherited and emergent nonlinear topological phenomena as well as a protocol capable of revealing the interplay of nonlinearity and topology.These concepts are applicable to other nonlinear topological systems,both in higher dimensions and beyond our photonic platform.Shiqi Xia Dario Jukic Nan Wang Daria Smirnova Lev Smirnov Liqin Tang Daohong Song Alexander Szameit Daniel Leykam Jingjun Xu Zhigang Chen Hrvoje Buljan 2020Light(Science & Applications)2020,9,1:3
7A regional ionospheric TEC mapping technique over China and adjacent areas: GNSS data processing and DINEOF analysis显示文摘A technique is developed to derive two-dimensional maps of total electron content(TEC) over China and adjacent areas using Global Navigation Satellite System(GNSS) data from the Crustal Movement Observation Network of China(CMONOC) and the International Global Navigation Satellite System Service(IGS). A revised self-calibration of pseudo-range errors(SCORE) algorithm is used to derive the TEC and to determine the Differential Code Biases(DCBs) simultaneously. The accuracy and validity of this technique is verified in two ways. Firstly, the estimated TEC is compared with the results derived using DCBs from Center for Orbit Determination of Europe(CODE) under different solar activity conditions and seasons; secondly, sample TEC along the receiver-to-satellite ray paths are simulated by Ne Quick model and are reprocessed by this TEC derivation technique to make the accuracy test. Two-dimensional maps of vertical TEC of ionospheric pierce points(IPPs) are obtained accordingly with a time resolution of 30 s. The data interpolating empirical orthogonal functions(DINEOF) technique is then used to make the extrapolation for the unknown or missing data points. The optimal EOF modes for data reconstruction are specified via cross-validation method. 9Two Ultraviolet Radiation Datasets that Cover China显示文摘Ultraviolet(UV) radiation has significant effects on ecosystems, environments, and human health, as well as atmospheric processes and climate change. Two ultraviolet radiation datasets are described in this paper. One contains hourly observations of UV radiation measured at 40 Chinese Ecosystem Research Network stations from 2005 to 2015. CUV3 broadband radiometers were used to observe the UV radiation, with an accuracy of 5%, which meets the World Meteorology Organization's measurement standards. The extremum method was used to control the quality of the measured datasets. The other dataset contains daily cumulative UV radiation estimates that were calculated using an all-sky estimation model combined with a hybrid model. The reconstructed daily UV radiation data span from 1961 to 2014. The mean absolute bias error and root-mean-square error are smaller than 30% at most stations, and most of the mean bias error values are negative, which indicates underestimation of the UV radiation intensity. These datasets can improve our basic knowledge of the spatial and temporal variations in UV radiation. 17Truncated glycoprotein E of varicella-zoster virus is an ideal immunogen for Escherichia coli-based vaccine design显示文摘Varicella-zoster virus(VZV)is a highly infectious agent responsible for both varicella and herpes zoster disease.Despite high efficacy,there remain safety and accessibility concerns with the licensed vaccines.Here,we sought to produce a VZV g E immunogen using an E.coli expression system.We found that the soluble expression and yield of g E protein could be enhanced via C-terminal truncations to the protein,thereby facilitating a robust and scalable purification process for the purpose of vaccine manufacturing.The lead truncated g E(aa 31–358),hereafter referred to as tg E,was a homogenous monomer in solution and showed excellent antigenicity.Finally,we assessed and compared the immunogenicity of tg E with commercial v Oka LAV and Shingrix vaccine.We found that aluminum-adjuvanted tg E was immunogenic as compared with v Oka LAV.When adjuvanted with AS01B,a two-dose immunization of tg E showed comparable or better potency in antibody responses and cell-mediated immunity with those of the Shingrix vaccine at the same dosage,especially in terms of the proportion of IFN-γ-expressing CD4^(+)T cells.In conclusion,this method of E.coli-mediate tg E expression offers a cost-effective and scalable strategy to generate an ideal VZV g E immunogen for the development of both varicella and zoster vaccines.Tingting Chen Jie Sun Sibo Zhang Tingting Li Liqin Liu Wenhui Xue Lizhi Zhou Siting Liang Zhili Yu Qingbing Zheng Hai Yu Tong Cheng Jun Zhang Ying Gu Shaowei Li Ningshao Xia 2023Science China(Life Sciences)2023,66,4:1
