3UDP-glucose pyrophosphorylase2 (OsUgp2), a pollen-preferential gene in rice, plays a critical role in starch accumulation during pollen maturation显示文摘UDP-glucose pyrophosphorylase (UGPase) is predominantly present and plays significant role in carbohydrate metabolism in plants. Two homologous UGPase genes, OsUgp1 and OsUgp2, exist in rice genome. OsUgp1 has recently been reported to be essential for callose deposition during pollen mother cell and meiosis stages as well as for seed carbohydrate metabolism. In this study, a full-length cDNA of OsUgp2 was isolated from rice anther. Northern blot and RNA in situ hybridization indicated that the expression of OsUgp2 was preferentially in pollen and developmentally regulated. No transcripts were found in leaf, stem, lemma/palea, ripening grain and florets before the uninucleate microspore developmental stage, but a large quantity of OsUgp2 mRNA was found in pollen at the binucleate and mature stages. The immunolocalization of OsUgp2 showed a similar expression pattern to that by RNA in situ hybridization. The function of OsUgp2 was investigated by dsRNA-mediated transcriptional gene silencing. The pollen fertility of 16 independent transgenic rice plants was found between 25% and 90%, which was correlated with the amount of OsUgp2 mRNA. The results of morphological changes and starch variation during pollen development in transgenic rice showed that the abnormal feature of pollen development appeared after the uninucleate microspore stage. Starch failed to accumulate in pollen and thus led to sterile pollens. These results demonstrated that OsUgp2 is a pollen-preferential 'late gene' and plays a key role during pollen maturation, especially for starch accumulation. 4MAPPING OF THE RICE (ORYZA SATIVA L.) THERMO-SENSITIVE GENIC MALE STERILE GENE TMS5 WITH EST AND SSR MARKERS显示文摘With the cDNA suppression subtraction hybridization method, a spikelet-specific cDNA library was constructed that expressed at meiosis stage in rice. A total of 121 cDNA fragments were selected from the library and used as EST (expressed se-quence tags) markers to detect the polymorphism between Annong N, a normal fertile Indica rice line and Annong S-1, its spontaneous mutant with thermo-sensitive genic male sterility, using the RFLP (restriction fragment length polymorphism) technique. HN57, one of the EST probes, could detect poly-morphism between them. The results of segregation analysis with the F2 population developed from An-nong S-1 and Annong N indicate that HN57 co-seg- regates with the thermo-sensitive genic male-sterility controlled by tms5, the recessive gene in Annong S-1. This marker is located on the 31.2-cM region of the chromosome 2 of RGP (rice genome research pro-gram) genetic map. To further determine the location of tms5, 80 SSR (simple sequence repeat) markers around this region were developed, and 12 of them were polymorphic. 5Enhancement of Aggression Induced by Isolation Rearing is Associated with a Lack of Central Serotonin显示文摘Isolation rearing(IR) enhances aggressive behavior, and the central serotonin(5-hydroxytryptamine,5-HT) system has been linked to IR-induced aggression.However, whether the alteration of central serotonin is the cause or consequence of enhanced aggression is still unknown. In the present study, using mice deficient in central serotonin Tph2-/-and Lmx1 b-/-, we examined the association between central serotonin and aggression with or without social isolation. We demonstrated that central serotonergic neurons are critical for the enhanced aggression after IR. 5-HT depletion in wild-type mice increased aggression. On the other hand, application of 5-HT in Lmx1 b-/-mice inhibited the enhancement of aggression under social isolation conditions. Dopamine was downregulated in Lmx1 b-/-mice. Similar to 5-HT, L-DOPA decreased aggression in Lmx1 b-/-mice. 6Methylesterification of cell-wall pectin controls the diurnal flower-opening times in rice显示文摘Flowers are the core reproductive organ of plants, and flowering is essential for cross-pollination. Diurnal flower-opening time is thus a key trait influencing reproductive isolation, hybrid breeding, and thermostability in plants. However, the molecular mechanisms controlling this trait remain unknown. Here, we report that rice Diurnal Flower Opening Time 1 (DFOT1) modulates pectin methylesterase (PME) activity to regulate pectin methylesterification levels of the lodicule cell walls, which affect lodicule swelling to control diurnal flower-opening time. DFOT1 is specifically expressed in the lodicules, and its expression gradually increases with the approach to flowering but decreases with flowering. Importantly, a knockout of DFOT1 showed earlier diurnal flower opening. We demonstrate that DFOT1 interacts directly with multiple PMEs to promote their activity. Knockout of PME40 also resulted in early diurnal flower opening, whereas overexpression of PME42 delayed diurnal flower opening. Lower PME activity was observed to be associated with higher levels of pectin methylesterification and the softening of cell walls in lodicules, which contribute to the absorption of water by lodicules and cause them to swell, thus promoting early diurnal flower opening. Higher PME activity had the opposite effect. 7Robust superamphiphobic coatings with gradient and hierarchical architecture and excellent anti-flashover performances显示文摘Biomimetic superhydrophobic(SH)coatings have emerged as a promising alternative to traditional room temperature vulcanizing(RTV)silicone rubber coatings for improving the flashover strength of insulators.However,organic contamination occurs in outdoor applications and thus a superamphiphobic(SAP)surface is more desirable but not yet reported for improving flashover performance.Herein,we developed a novel anti-flashover technique by fabricating robust SAP coating with unique gradient and micro-nanoscale hierarchical architecture.The SAP coating was fabricated by sequentially spray-depositing a resin-based primer and a silica-based topcoat on substrates(i.e.,glass slides and insulators).The primer not only functions as an adhesive offering strong adhesion to the substrate but also offers a micromastoid-like structure facilitating the subsequent formation of hierarchical micro-nanostructure.The appropriate spraying pressure leads to a diffusion of the fluorocarbon-modified silica nanoparticles into the primer to form a unique gradient structure,by analogy to inserting bullets into a wood.These features render the SAP coating excellent robustness with strong abrasive resistance,excellent ultraviolet(UV)resistance,and excellent chemical and thermal stability.Pollution flashover property of the SAP coating was explored and compared with that of SH and RTV specimens,from which a novel organic-contamination model to evaluate the flashover performance was proposed.The coated SAP glass insulator demonstrated 42.9%pollution flashover voltage improvement than RTV-coated insulator.These stated unique features reveal the convincing potential of the present SAP coatings to be applied for not only outdoor transmission line insulators for antiflashover but also other fields for self-cleaning,anti-fouling,and anti-icing.Yi Xie Wei Xiong Shefiu Kareem Chuxiong Qiu Yongfei Hu Ivan PParkin Shengwu Wang Huayun Wang Junwu Chen Lee Li Zhi Chen Huajun Sun Xiujian Zhao 2022Nano Research2022,15,8:2
8Ubiquitinome Profiling Reveals the Landscape of Ubiquitination Regulation in Rice Young Panicles显示文摘Ubiquitination,an essential post-transcriptional modification(PTM),plays a vital role in nearly every biological process,including development and growth.Despite its functions in plant reproductive development,its targets in rice panicles remain unclear.In this study,we used proteome-wide profiling of lysine ubiquitination in rice(O.sativa ssp.indica)young panicles.We created the largest ubiquitinome dataset in rice to date,identifying 1638 lysine ubiquitination sites on916 unique proteins.We detected three conserved ubiquitination motifs,noting that acidic glutamic acid(E)and aspartic acid(D)were most frequently present around ubiquitinated lysine.Enrichment analysis of Gene Ontology(GO)annotations and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathways of these ubiquitinated proteins revealed that ubiquitination plays an important role in fundamental cellular processes in rice young panicles.Interestingly,enrichment analysis of protein domains indicated that ubiquitination was enriched on a variety of receptorlike kinases and cytoplasmic tyrosine and serine-threonine kinases.Furthermore,we analyzed the crosstalk between ubiquitination,acetylation,and succinylation,and constructed a potential protein interaction network within our rice ubiquitinome.Moreover,we identified ubiquitinated proteins related to pollen and grain development,indicating that ubiquitination may play a critical role in the physiological functions in young panicles.Taken together,we reported the most comprehensive lysine ubiquitinome in rice so far,and used it to reveal the functional role of lysine ubiquitination in rice young panicles.Liya Zhu Han Cheng Guoqing Peng Shuansuo Wang Zhiguo Zhang Erdong Ni Xiangdong Fu Chuxiong Zhuang Zexian Liu Hai Zhou 2020Genomics, Proteomics & Bioinformatics2020,18,3:2

10Real-Time Iterative Compensation Framework for Precision Mechatronic Motion Control Systems显示文摘With regard to precision/ultra-precision motion systems,it is important to achieve excellent tracking performance for various trajectory tracking tasks even under uncertain external disturbances.In this paper,to overcome the limitation of robustness to trajectory variations and external disturbances in offline feedforward compensation strategies such as iterative learning control(ILC),a novel real-time iterative compensation(RIC)control framework is proposed for precision motion systems without changing the inner closed-loop controller.Specifically,the RIC method can be divided into two parts,i.e.,accurate model prediction and real-time iterative compensation.An accurate prediction model considering lumped disturbances is firstly established to predict tracking errors at future sampling times.In light of predicted errors,a feedforward compensation term is developed to modify the following reference trajectory by real-time iterative calculation.Both the prediction and compen-sation processes are finished in a real-time motion control sampling period.The stability and convergence of the entire control system after real-time iterative compensation is analyzed for different conditions.Various simulation results consistently demonstrate that the proposed RIC framework possesses satisfactory dynamic regulation capability,which contributes to high tracking accuracy comparable to ILC or even better and strong robustness.Chuxiong Hu Ran Zhou Ze Wang Yu Zhu Masayoshi Tomizuka 2022IEEE/CAA Journal of Automatica Sinica2022,9,7:2

12OsWR2 recruits HDA704 to regulate the deacetylation of H4K8ac in the promoter of OsABI5 in response to drought stress显示文摘Drought stress is a major environmental factor that limits the growth, development, and yield of rice(Oryza sativa L.). Histone deacetylases(HDACs) are involved in the regulation of drought stress responses. HDA704 is an RPD3/HDA1 class HDAC that mediates the deacetylation of H4K8(lysine 8of histone H4) for drought tolerance in rice. In this study, we show that plants overexpressing HDA704(HDA704-OE) are resistant to drought stress and sensitive to abscisic acid(ABA), whereas HDA704 knockout mutant(hda704) plants displayed decreased drought tolerance and ABA sensitivity.Transcriptome analysis revealed that HDA704 regulates the expression of ABA-related genes in response to drought stress. Moreover, HDA704 was recruited by a drought-resistant transcription factor,WAX SYNTHESIS REGULATORY 2(Os WR2), and co-regulated the expression of the ABA biosynthesis genes NINE-CIS-EPOXYCAROTENOID DIOXYGENASE 3(NCED3), NCED4, and NCED5 under drought stress. HDA704 also repressed the expression of ABA-INSENSITIVE 5(Os ABI5) and DWARF AND SMALL SEED 1(Os DSS1) by regulating H4K8ac levels in the promoter regions in response to polyethylene glycol 6000 treatment. In agreement, the loss of Os ABI5 function increased resistance to dehydration stress in rice. 15Advantages and disadvantages of terracing:A comprehensive review显示文摘For thousands of years,terracing has been one of the most important systems for preventing soil erosion,conserving water,and increasing agricultural production.Despite having a long history,the wide-ranging effects and mechanisms of terracing are poorly understood owing to large-scale spatial and temporal distribution patterns and the challenges related to assessing the ecosystem services of terraced landscapes.Thus,our study aims to review comprehensively the effect of terraces by describing the mechanisms behind terraced systems.Terraces provide many ecosystem services,including the reduction of runoff and sediment by over 41.9%and 52%,respectively,and the improvement of grain yields and soil moisture content by 44.8%and 12.9%,respectively.In addition,terracing can also contribute toward the conservation of plant biodiversity on a local scale.However,as terraces age,a number of disadvantages gradually emerge,including interference with water circulation and the development of serious environmental problems caused by poorly designed or mismanaged terraces,where the average runoff and soil loss can be 1—5 times that of well-managed terraces.Although understanding the complexity and multifaceted effects of terracing is vital for terrace construction and management,the negative effects of terracing are often not considered,and existing studies have several shortcomings.Within this context,this paper aims to describe both the benefits and disadvantages of terracing,investigate the gaps in current research,as well as discuss preventive and remedial measures so as to negate the possible bad impacts of terracing.Chuxiong Deng Guangye Zhang Yaojun Liu Xiaodong Nie Zhongwu Li Junyu Liu Damei Zhu 2021International Soil and Water Conservation Research2021,9,3:1
16Thermal stability of soil organic carbon subjected to water erosion as a function of edaphic factors显示文摘The stability of soil organic carbon(SOC),as it relates to resistance to decomposition,is important for greenhouse gas emission and climate change.However,the SOC stabilization and its related influencing factors subjected to water erosion remain uncertain.The objective of the current study was to determine the SOC stability under long-term water erosion and to investigate the link between SOC stability and edaphic factors.Soil samples from eroded,depositional,and control sites in a closed watershed in subtropical China were collected.The SOC concentration,carbon functional groups,soil physicochemical properties,and thermal stability of SOC were determined.The study results showed that the primary component of the SOC functional groups was O-alkyl carbon(reached 40%),and its proportion has no significant difference among different sites.The TG-T50(the temperature at which 50%of the soil organic matter is lost)values indicated that the SOC thermal stability in the eroded topsoil was higher than that of the deposited topsoil.The carbon functional groups were lack of relation with SOC thermal stability(p>0.05).However,the TG-T_(50) was significantly positively correlated with the cation exchange capacity and silt,but remarkably negatively correlated with SOC,dissolved organic carbon,total nitrogen,sand,clay,pH,aluminum,specific surface area,and iron oxides.These results indicated that the edaphic factors,which were strongly affected by micro-environment,are the main factors affecting thermal stability of organic carbon.The SOC stabilization under the effects of long-term water erosion should be addressed from the perspective of the ecosystem.Zhongwu Li Linhui Xiao Chuxiong Deng Zaijian Yuan Chen Liang Qian Xiong Zeting Li Xiaodong Nie 2022International Journal of Sediment Research2022,37,1:0

18Genomic polymorphism in consecutive generation rice plants from seeds on board a spaceship and their relationship with space HZE particles显示文摘Dry rice seeds(Oryza sativa,subspecies indica)were sandwiched between nuclear track detectors,aboard the Chinese spaceship Shenzhou-3 for seven days.The seeds were recovered and the genomic polymorphism in 201 plants developed from these seeds was studied using random amplified polymorphism DNA analysis.When compared with plants from ground-based control seeds,the genomic polymorphic bands were amplified in 30.2%of the plants from the seeds exposed in space.The results for sequencing and SNP analyses of the polymorphic bands verified the single nucleotide variations in these plants.Genomic poly-morphisms in the consecutive generations of individual plants of the seeds from space were also discovered.Seven seeds receiving hits of HZE(high atomic number and high energy)particles from space were selected for further analyses and variable genomic polymorphisms were detected in all plants that developed from these seeds.Among them,the embryos of three seeds were hit at least once,and mutants with significant changes in agronomic traits were only found in later generations of these seeds.This result implies that the HZE particles of space are effective in inducing the changes of plant genome of inherited phenotypes.LUO Yi WANG Xujie MEI Mantong ZHUANG Chuxiong ZHOU Feng WEI Zengquan XIE Hongmei YAO Juan ZHAO Junliang 2007Frontiers in Biology2007,2,3:0
