| 1 | Peripheral T-lymphocyte subpopulations in different clinical stages of chronic HBV infection correlate with HBV load显示文摘AIM: To characterize the peripheral T-cell subpopulation profiles and their correlation with hepatitis B virus (HBV) replication in different clinical stages of chronic HBV infection.METHODS: A total of 422 patients with chronic HBV infection were enrolled in this study. The patients were divided into three stages: immune-tolerant stage, immune active stage, and immune-inactive carrier stage. Composition of peripheral T-cell subpopulations was determined by flow cytometry. HBV markers were detected by enzyme-linked immunosorbent assay. SerumHBV DNA load was assessed by quantitative real-time polymerase chain reaction.RESULTS: CD8+ T-cells were significantly higher in patients at the immune-tolerant stage than in patients at the immune-active and -inactive carrier stages (36.87 ± 7.58 vs 34.37 ± 9.07, 36.87 ± 7.58 vs 28.09 ± 5.64, P < 0.001). The peripheral blood in patients at the immune-tolerant and immune active stages contained more CD8+ T-cells than CD4+ T-cells (36.87 ± 7.58 vs 30.23 ± 6.35, 34.37 ± 9.07 vs 30.92 ± 7.40, P < 0.01), whereas the peripheral blood in patients at the immune-inactive carrier stage and in normal controls contained less CD8+ T-cells than CD4+ T-cells (28.09 ± 5.64 vs 36.85 ± 6.06, 24.02 ± 4.35 vs 38.94 ± 3.39, P < 0.01). ANOVA linear trend test showed that CD8+ T-cells were signif icantly increased in patients with a high viral load (39.41 ± 7.36, 33.83 ± 7.50, 31.81 ± 5.95 and 26.89 ± 5.71, P < 0.001), while CD4+ T-cells were signif icantly increased in patients with a low HBV DNA load (37.45 ± 6.14, 33.33 ± 5.61, 31.58 ± 6.99 and 27.56 ± 5.49, P < 0.001). Multiple regression analysis displayed that log copies of HBV DNA still maintained its highly signif icant coefficients for T-cell subpopulations, and was the strongest predictors for variations in CD3+, CD4+ and CD8+ cells and CD4+/CD8+ ratio after adjustment for age at HBV-infection, maternal HBV-infection status, presence of hepatitis B e antigen and HBV mutation.CONCLUSION: Differences in peripheral T-cell subpopulation profi les can be found in different clinical stages of chronic HBV infection. | 2 | Meta-analyses on prevalence of selected Parkinson’s nonmotor symptoms before and after diagnosis显示文摘Background:Nonmotor symptoms are common among patients with Parkinson’s disease(PD)and some may precede disease diagnosis.Methods:We conducted a meta-analysis on the prevalence of selected nonmotor symptoms before and after PD diagnosis,using random-effect models.We searched PubMed(1965 through October/November 2012)for the following symptoms:hyposmia,constipation,rapid eye movement sleep behavior disorder,excessive daytime sleepiness,depression,and anxiety.Eligible studies were publications in English with original data on one or more of these symptoms.Results:The search generated 2,373 non-duplicated publications and 332 met the inclusion criteria,mostly(n=320)on symptoms after PD diagnosis.For all symptoms,the prevalence was substantially higher in PD cases than in controls,each affecting over a third of the patients.Hyposmia was the most prevalent(75.5%in cases vs.19.1%in controls),followed by constipation(50%vs.17.7%),anxiety(39.9%vs.19.1%),rapid eye movement sleep behavior disorder(37.0%vs.7.0%),depression(36.6%vs.14.9%),and excessive daytime sleepiness(33.9%vs.10.5%).We observed substantial heterogeneities across studies and meta-regression analyses suggested that several factors might have contributed to this.However,the prevalence estimates were fairly robust in several sensitivity analyses.Only 20 studies had data on any symptoms prior to PD diagnosis,but still the analyses revealed higher prevalence in future PD cases than in controls.Conclusion:These symptoms are common among PD patients both before and after diagnosis.Further studies are needed to understand the natural history of nonmotor symptoms in PD and their etiological and clinical implications. | Honglei Chen Edward J Zhao Wen Zhang Yi Lu Rui Liu Xuemei Huang Anna J Ciesielski-Jones Michele A Justice Deborah S Cousins Shyamal Peddada | 2015 | Translational Neurodegeneration2015,4,1: | 18 |
| 3 | Recombinant Newcastle disease virus expressing African swine fever virus protein 72 is safe and immunogenic in mice显示文摘African swine fever(ASF) is a lethal hemorrhagic disease that affects wild and domestic swine. The etiological agent of ASF is African swine fever virus(ASFV). Since the first case was described in Kenya in 1921, the disease has spread to many other countries. No commercial vaccines are available to prevent ASF. In this study, we generated a recombinant Newcastle disease virus(r NDV) expressing ASFV protein 72(p72) by reverse genetics and evaluated its humoral and cellular immunogenicity in a mouse model. The recombinant virus, r NDV/p72, replicated well in embryonated chicken eggs and was safe to use in chicks and mice. The p72 gene in r NDV/p72 was stably maintained through ten passages. Mice immunized with r NDV/p72 developed high titers of ASFV p72 specific Ig G antibody, and had higher levels of Ig G1 than IgG2 a. Immunization also elicited T-cell proliferation and secretion of IFN-γ and IL-4. | 7 | Abnormal subchondral bone remodeling and its association with articular cartilage degradation in knees of type 2 diabetes patients显示文摘Type 2 diabetes(T2 D) is associated with systemic abnormal bone remodeling and bone loss. Meanwhile,abnormal subchondral bone remodeling induces cartilage degradation, resulting in osteoarthritis(OA).Accordingly, we investigated alterations in subchondral bone remodeling, microstructure and strength in knees from T2 D patients and their association with cartilage degradation. Tibial plateaus were collected from knee OA patients undergoing total knee arthroplasty and divided into non-diabetic(n = 70) and diabetes(n = 51) groups. Tibial plateaus were also collected from cadaver donors(n = 20) and used as controls.Subchondral bone microstructure was assessed using micro-computed tomography. Bone strength was evaluated by micro-finite-element analysis. Cartilage degradation was estimated using histology. The expression of tartrate-resistant acidic phosphatase(TRAP), osterix, and osteocalcin were calculated using immunohistochemistry. Osteoarthritis Research Society International(OARSI) scores of lateral tibial plateau did not differ between non-diabetic and diabetes groups, while higher OARSI scores on medial side were detected in diabetes group. Lower bone volume fraction and trabecular number and higher structure model index were found on both sides in diabetes group. These microstructural alterations translated into lower elastic modulus in diabetes group. Moreover, diabetes group had a larger number of TRAP^+ osteoclasts and lower number of Osterix^+ osteoprogenitors and Osteocalcin^+ osteoblasts. T2 D knees are characterized by abnormal subchondral bone remodeling and microstructural and mechanical impairments, which were associated with exacerbated cartilage degradation. | 8 | A STUDY OF STRUCTURE AND PROPERTY CHANGES OF BIODEGRADABLE POLYGLYCOLIDE AND POLY(GLYCOLIDE-co-LACTIDE)FIBERS DURING PROCESSING AND IN VITRO DEGRADATION显示文摘Structure and properties of bioabsorbable polyglycolide (PGA) and poly(glycolide-co-lactide) (PGA-co-PLA)fibers were investigated during several industrial processing stages and in vitro degradation by means of wide-angle X-ray diffraction (WAXD), dynamic mechanical analysis (DMA) and mechanical property tests. In the orientation stage, the PGA fibers were found to have higher degrees of crystallinity than corresponding PGA-co-PLA samples produced under similar conditions. In the hot-stretching and post-annealing stages, after fibers were braided, PGA samples were found to gain more crystallinity and higher Tg than PGA-co-PLA samples. The higher crystallinity in PGA fibers resulted in a slower rate of degradation. DMA results showed that a great deal of internal stress that was built during orientation and hot-stretching stages was released in the post-annealing stage for all PGA and PGA-co-PLA samples. During earlier stages ofin vitro degradation, both PGA and PGA-co-PLA samples exhibited the typical cleavage-induced crystallization mechanism. | 9 | Discovering the roles of subsurface microorganisms: Progress and future of deep biosphere investigation显示文摘The discovery of the marine 'deep biosphere'-microorganisms living deep below the seafloor-is one of the most significant and exciting discoveries since the ocean drilling program began more than 40 years ago. Study of the deep biosphere has become a research frontier and a hot spot both for geological and biological sciences. Here, we introduce the history of the discovery of the deep biosphere, and then we describe the types of environments for life below the seafloor, the energy sources for the living creatures, the diversity of organisms within the deep biosphere, and the new tools and technologies used in this research field. We will highlight several recently completed Integrated Ocean Drilling Program Expeditions, which targeted the subseafloor deep biosphere within the crust and sediments. | 12 | Research on water ice content in Cabeus crater using the data from the microwave radiometer onboard Chang’e-1 satellite显示文摘The existence, formation and content of water ice in the lunar permanently shaded region is one of the important questions for the current Moon study. On October 9, 2009, the LCROSS mission spacecraft impacted the Moon, and the initial result verified the existence of water on the Moon. But the study on formation and content of water ice is still under debate. The existence of water ice can change the dielectric constants of the lunar regolith, and a microwave radiometer is most sensitive to the dielectric parameters. Based on this, in this paper, the radiation transfer model is improved according to the simulation results in high frequency. Then the mixture dielectric constant models, including Odelevsky model, Wagner and landau-Lifshitz model, Clau-sius model, Gruggeman-Hanai model, etc., are analyzed and compared. The analyzing results indicate that the biggest difference occurs between Lichtenecker model and the improved Dobson model. The values estimated by refractive model are the second biggest in all the models. And the results from Odelevsky model, strong fluctuation model, Wagner and Landau –Lifshitz model, Clausius model and Bruggeman-Hanai model are very near to each other. Thereafter, the relation between volume water ice content and microwave brightness temperature is constructed with Odelevsky mixing dielectric model and the improved radiative transfer simulation, and the volume water ice content in Cabeus crater is retrieved with the data from microwave radiometer onboard Chang’e-1 satellite. The results present that the improved radiative transfer model is proper for the brightness temperature simulation of the one infinite regolith layer in high frequency. | 13 | Influence of Temperature and Frequency on Microwave Dielectric Properties of Lunar Regolith Simulant显示文摘The dielectric constant of the lunar regolith can directly influence the reflection coefficient and the trans-mission coefficient of the Moon′s surface, and plays an important role in the Moon research. In order to study the di-electric properties of the lunar regolith, the lunar regolith simulant was made according to the making procedure of the CAS-1 simulant made by Chinese Academy of Sciences. Then the dielectric constants of the lunar regolith simulant were measured with 85070E Aiglent Microwave Network Analyzer in the frequency ranging from 0.2 GHz to 20.0 GHz and at temperature of 25.1℃, 17.7℃, 13.1℃, 11.5℃, 9.6℃, 8.0℃, 4.1℃, -0.3℃, -4.7℃, -9.5℃, -18.7℃, -27.7℃, and -32.6℃, respectively. The Odelevsky model was employed to remove the influence of water in the air on the final effective dielectric constants. The results indicate that frequency and temperature have apparent influences on the dielectric constants of the lunar regolith simulant. The real parts of the dielectric constants increase fast over the range of 0.2 GHz to 3.0 GHz, but decrease slowly over the range of 4.0 GHz to 20.0 GHz. The opposite phenomenon occurs in the imaginary parts. The influences of the frequency and temperature on the brightness temperature were also estimated based on the radiative transfer equation. | 15 | High efficiency synthesis of HKUST-1 under mild conditions with high BET surface area and CO_2 uptake capacity显示文摘This study focuses on the development of a hydrothermal method for the rapid synthesis of good quality copper benzene-1,3,5-tricarboxylate(referred to as HKUST-1)with high yield under mild preparation conditions to address the issues associated with reported methods.Different synthesis conditions and activation methods were studied to understand their influence on the properties of HKUST-1.It was found that mixing the precursors at50°C for 3 h followed by activation via methanol refluxing led to the formation of a product with the highest BET specific surface area of 1615 m^2/g and a high yield of 84.1%.The XRD and SEM data illustrated that the product was highly crystalline.The sample was also tested on its capacity in CO_2 adsorption.The results showed strong correlation between surface area of the sample and its CO_2 uptake at 1 bar and 27°C.The HKUST-1 prepared in this study demonstrated a high CO_2 uptake capacity of 4.2 mmol/g.It is therefore concluded that this novel and efficient method can be used in the rapid preparation of HKUST-1 with high surface area and CO_2 uptake capacity. | Yipei Chen Xueliang Mu Edward Lester Tao Wu | 2018 | Progress in Natural Science:Materials International2018,28,5: | 3 |

| 18 | The influence of physiological environment on the targeting effect of aptamer-guided gold nanoparticles显示文摘Aptamer guided nano medicine shows great promise in targeted cancer therapies.However the loss of targeting capacity duri ng in vivo or clinical trials has largely hindered its popularity and there are no systematic studies to elucidate the causes.Herein,we investigated such loss of targeting capacity by examining how the physiological milieu affected targeting effect.Aptamer functionalized gold nanoparticle(AuNP)was chosen as the model and exposed to human blood serum that is used to mimic physiological milieu.Dynamic light scattering(DLS),flow cytometry and label-free liquid chromatography tan dem mass spectrometry(LC-MS/MS)were employed to determi ne variations of NPs'surface chemistry and biological identities changes after serum exposure.Results showed that the targeting ability loss was caused by protein corona blocking,replacement and enzymatic cleavage of surface aptamer targeting ligands.Noteworthy,the aggregation issue is critical for the smaller NPs.Analysis of the protein corona profile indicated the accumulation of immune-related proteins on the surface of aptamer-conjugated NPs,which could induce immune response,resulting in rapid clearanee of NPs. | Ding Ding Yinling Zhang Edward A.Sykes Long Chen Zhuo Chen Weihong Tan | 2019 | Nano Research2019,12,1: | 2 |
| 19 | Organic–inorganic hybrid perovskite scintillators for mixed field radiation detection显示文摘Sensitive and fast detection of neutrons and gamma rays is vital for homeland security,high-energy physics,and proton therapy.Fast-neutron detectors rely on light organic scintillators,andγ-ray detectors use heavy inorganic scintillators and semiconductors.Efficient mixed-field detection using a single material is highly challenging due to their contradictory requirements.Here we report hybrid perovskites(C_(8)H_(12)N)_(2)Pb(Br_(0.95)Cl_(0.05))_(4)that combine light organic cations and heavy inorganic skeletons at a molecular level to achieve unprecedented performance for mixed-field radiation detection.High neutron absorption due to a high density of hydrogen,strong radiative recombination within the highly confined[PbX_(6)]^(4-)layer,and sub-nanometer distance between absorption sites and radiative centers,enable a light yield of 41000 photons/MeV,detection pulse width of 2.97 ns and extraordinary linearity response toward both fast neutrons andγ-rays,outperforming commonly used fast-neutron scintillators.Neutron energy spectrum,time-of-flight based fast-neutron/γ-ray discrimination and neutron yield monitoring were all successfully achieved using(C_(8)H_(12)N)_(2)Pb(Br_(0.95)Cl_(0.05))_(4)detectors.We further demonstrate the monitoring of reaction kinetics and total power of a nuclear fusion reaction.We envision that molecular hybridized scintillators open a new avenue for mixed-field radiation detection and imaging. | Mengling Xia Guangda Niu Linyue Liu Runlong Gao Tong Jin Pengying Wan Weicheng Pan Xianpeng Zhang Zuoxiang Xie Sam Teale Zenghua Cai Jiajun Luo Shan Zhao Haodi Wu Shiyou Chen Zhiping Zheng Qingguo Xie Xiaoping Ouyang Edward HSargent Jiang Tang | 2022 | InfoMat2022,4,9: | 2 |
