1Discovery of bipinnatifid fronds of Protoblechnum contractum(Gu et Zhi)显示文摘IN Permian floras of China, a dispute over the relationship among the Protoblechnum and other similar plants such as Compsopteris, Glenopteris and Supaia has been existing for a long time, and moreover it has not been completely solved up to now. Halle placed a few simple pinnate fronds from the Shihhotze Formation in the genus Protoblechnum and named them Protoblechnum Wongii Halle 70 years ago when he was studying the fossil plants from the Late Palaeozoic strata in Shanxi, China. He did not think forked or unforked fronds as an important dividing genus condition in discussing the distinction among Protoblechnum and other relative fossils. Zalessky alSO found some specimens similar to Chinese Protoblechnum while he studied the Permian fossil plants from Russia. 2Incorporating topographic factors in nonlinear mixed-effects models for aboveground biomass of natural Simao pine in Yunnan,China显示文摘A total of 128 Simao pine trees(Pinus kesiya var. langbianensis) from three regions of Pu'er City,Yunnan Province, People's Republic of China, were destructively sampled to obtain tree aboveground biomass(AGB). Tree variables such as diameter at breast height and total height, and topographical factors such as altitude,aspect of slope, and degree of slope were recorded. We considered the region and site quality classes as the random-effects, and the topographic variables as the fixedeffects. We fitted a total of eight models as follows: leastsquares nonlinear models(BM), least-squares nonlinear models with the topographic factors(BMT), nonlinear mixed-effects models with region as single random-effects(NLME-RE), nonlinear mixed-effects models with site as single random-effects(NLME-SE), nonlinear mixed-effects models with the two-level nested region and site random-effects(TLNLME), NLME-RE with the fixed-effects of topographic factors(NLMET-RE), NLME-SE with the fixed-effects of topographic factors(NLMET-SE), and TLNLME with the fixed-effects of topographic factors(TLNLMET). The eight models were compared by modelfitting and prediction statistics. The results showed: model fitting was improved by considering random-effects of region or site, or both. The models with the fixed-effects of topographic factors had better model fitting. According to AIC and BIC, the model fitting was ranked as TLNLME[ NLMET-RE [ NLME-RE [ NLMET-SE [ TLNLMET[ NLME-SE [ BMT [ BM. The differences among these models for model prediction were small. The model prediction was ranked as TLNLME [ NLME-RE [ NLMES E [ N L M E T- R E [ N L M E T- S E [ T L N L M E T [BMT [ BM. However, all eight models had relatively high prediction precision([90 %). 3Representation of conspecific vocalizations in amygdala of awake marmosets显示文摘Human speech and animal vocalizations are important for social communication and animal survival.Neurons in the auditory pathway are responsive to a range of sounds,from elementary sound features to complex acoustic sounds.For social communication,responses to distinct patterns of vocalization are usually highly specific to an individual conspecific call,in some species.This includes the specificity of sound patterns and embedded biological information.We conducted single-unit recordings in the amygdala of awake marmosets and presented calls used in marmoset communication,calls of other species and calls from specific marmoset individuals.We found that some neurons(47/262)in the amygdala distinguished‘Phee’calls from vocalizations of other animals and other types of marmoset vocalizations.Interestingly,a subset of Phee-responsive neurons(22/47)also exhibited selectivity to one out of the three Phees from two different‘caller’marmosets.Our findings suggest that,while it has traditionally been considered the key structure in the limbic system,the amygdala also represents a critical stage of socially relevant auditory perceptual processing.Guoqiang Jia Siyi Bai Yingxu Lin Xiaohui Wang Lin Zhu Chenfei Lyu Guanglong Sun Kang An Anna Wang Roe Xinjian Li Lixia Gao 2023National Science Review2023,10,11:1

7Inhibition of PRMT5 by market drugs as a novel cancer therapeutic avenue显示文摘Market drugs,suchas Foodand Drug Administration(FDA)or European Medicines Agency(EMA)-approved drugs for specific indications provide opportunities for repurposing for newer therapeutics.This potentially saves resources invested in clinical trials that verify drug safety and tolerance in humans prior to alternative indication approval.Protein arginine methyltransferase 5(PRMT5)overexpression has been linked to promoting the tumor phenotype in several cancers,including pancreatic ductal adenocarcinoma(PDAC),colorectal cancer(CRC),and breast cancer(BC),making PRMT5 an important target for cancer therapy.Previously,we showed that PRMT5-mediated methylation of the nuclear factor(NF)-kB,partially contributes to its constitutive activation observed in cancers.In this study,we utilized an AlphaLiSA-based high-throughput screening method adapted in our lab,and identified one FDA-approved drug,Candesartan cilexetil(Can,used in hypertension treatment)and one EMA-approved drug,Cloperastine hydrochloride(Clo,used in cough treatment)that had significant PRMT5-inhibitory activity,and their anti-tumor properties were validated using cancer phenotypic assays in vitro.Furthermore,PRMT5 selective inhibition of methyltransferase activity was confirmed by reduction of both NF-kB methylation and its subsequent activation upon drug treatment.Using in silico prediction,we identified critical residues on PRMT5 targeted by these drugs that may interfere with its enzymatic activity.Finally,Clo and Can treatment have exhibited marked reduction in tumor growth in vivo.Overall,we provide basis for pursuing repurposing Clo and Can as anti-PRMT5 cancer therapies.Our study offers potential safe and fast repurposing of previously unknown PRMT5 inhibitors into clinical practice.Lakshmi Prabhu Matthew Martin Lan Chen Ozlem Demir Jiamin Jin Xiumei Huang Aishat Motolani Mengyao Sun Guanglong Jiang Harikrishna Nakshatri Melissa L.Fishel Steven Sun Ahmad Safa Rommie E.Amaro Mark R.Kelley Yunlong Liu Zhong-Yin Zhang Tao Lu 2023Genes & Diseases2023,10,1:0
