| 3 | Aligned fibrin/functionalized self-assembling peptide interpenetrating nanofiber hydrogel presenting multi-cues promotes peripheral nerve functional recovery显示文摘Nerve guidance conduits with hollow lumen fail to regenerate critical-sized peripheral nerve defects(15 mm in rats and 25 mm in humans),which can be improved by a beneficial intraluminal microenvironment.However,individual cues provided by intraluminal filling materials are inadequate to eliminate the functional gap between regenerated nerves and normal nerves.Herein,an aligned fibrin/functionalized self-assembling peptide(AFG/fSAP)interpenetrating nanofiber hydrogel that exerting synergistic topographical and biochemical cues for peripheral nerve regeneration is constructed via electrospinning and molecular self-assembly.The hydrogel possesses an aligned structure,high water content,appropriate mechanical properties and suitable biodegradation capabilities for nerve repair,which enhances the alignment and neurotrophin secretion of primary Schwann cells(SCs)in vitro,and successfully bridges a 15-mm sciatic nerve gap in rats in vivo.The rats transplanted with the AFG/fSAP hydrogel exhibit satisfactory morphological and functional recovery in myelinated nerve fibers and innervated muscles.The motor function recovery facilitated by the AFG/fSAP hydrogel is comparable with that of autografts.Moreover,the AFG/fSAP hydrogel upregulates the regeneration-associated gene expression and activates the PI3K/Akt and MAPK signaling pathways in the regenerated nerve.Altogether,the AFG/fSAP hydrogel represents a promising approach for peripheral nerve repair through an integration of structural guidance and biochemical stimulation. | Shuhui Yang Jinjin Zhu Changfeng Lu Yi Chai Zheng Cao Jiaju Lu Zhe Zhang He Zhao Yin-Yuan Huang Shenglian Yao Xiangdong Kong Peixun Zhang Xiumei Wang | 2022 | Bioactive Materials2022,7,2: | 3 |
| 4 | Changes in Vegetation and Assessment of Meteorological Conditions in Ecologically Fragile Karst Areas显示文摘Meteorological conditions have an important impact on changes of vegetation in ecologically fragile karst areas.This study aims to explore a method for quantitative evaluation of these meteorological conditions. We analyzed the changing trend of vegetation during 2000–2018 and the correlations between vegetation changes and various meteorological factors in karst rocky areas of Guangxi Zhuang Autonomous Region, China. Key meteorological factors in vegetation areas with varying degrees of improvement were selected and evaluated at seasonal timescale. A quantitative evaluation model of comprehensive influences of meteorological factors on vegetation was built by using the partial least-square regression(PLS). About 91.45% of the vegetation tended to be improved, while only the rest 8.55% showed a trend of degradation from 2000 to 2018. Areas with evident vegetation improvement were mainly distributed in the middle and northeast, and those with obvious vegetation degradation were scattered. Meteorological factors affecting vegetation were significantly different among the four seasons. Overall, high air humidity, small temperature difference in spring and autumn, and low daily minimum temperature and air pressure were favorable conditions. Low temperature in winter as well as high temperature in summer and autumn were unfavorable conditions. The Climate Vegetation Index(CVI) model was established by PLS using the maximum, minimum, and average temperatures;vapor pressure;rainfall;and air pressure as key meteorological factors. The Enhanced Vegetation Index(EVI) was well fitted by the CVI model, with the average coefficient of determination(r2) and root mean square error(RMSE) of 0.856 and 0.042, respectively. Finally, an assessment model of comprehensive meteorological conditions was built based on the interannual differences in CVI. | 5 | HER3 intracellular domains play a crucial role in HER3/HER2 dimerization and activation of downstream signaling pathways显示文摘Dimerization among the EGFR family of tyrosine kinase receptors leads to allosteric activation of the kinase domains of the partners.Unlike other members in the family,the kinase domain of HER3 lacks key amino acid residues for catalytic activity.As a result,HER3 is suggested to serve as an allosteric activator of other EGFR family members which include EGFR,HER2 and HER4.To study the role of intracellular domains in HER3 dimerization and activation of downstream signaling pathways,we constructed HER3/HER2 chimeric receptors by replacing the HER3 kinase domain(HER3-2-3)or both the kinase domain and the C-terminal tail(HER3-2-2)with the HER2 counterparts and expressed the chimeric receptors in Chinese hamster ovary(CHO)cells.While over expression of the intact human HER3 transformed CHO cells with oncogenic properties such as AKT/ERK activation and increased proliferation and migration,CHO cells expressing the HER3-2-3 chimeric receptor showed significantly reduced HER3/HER2 dimerization and decreased phosphorylation of both AKT and ERK1/2 in the presence of neuregulin-1(NRG-1).In contrast,CHO cells expressing the HER3-2-2 chimeric receptor resulted in a total loss of downstream AKT activation in response to NRG-1,but maintained partial activation of ERK1/2.The results demonstrate that the intracellular domains play a crucial role in HER3’s function as an allosteric activator and its role in downstream signaling. | Byung-Kwon Choi Xiumei Cai Bin Yuan Zhao Huang Xuejun Fan Hui Deng Ningyan Zhang Zhiqiang An | 2012 | Protein & Cell2012,3,10: | 3 |
