| 1 | Molecular mechanosensors in osteocytes显示文摘Osteocytes, the most abundant and long-lived cells in bone, are the master regulators of bone remodeling. In addition to their functions in endocrine regulation and calcium and phosphate metabolism, osteocytes are the major responsive cells in force adaptation due to mechanical stimulation. Mechanically induced bone formation and adaptation, disuse-induced bone loss and skeletal fragility are mediated by osteocytes, which sense local mechanical cues and respond to these cues in both direct and indirect ways. The mechanotransduction process in osteocytes is a complex but exquisite regulatory process between cells and their environment, between neighboring cells, and between different functional mechanosensors in individual cells. Over the past two decades, great efforts have focused on finding various mechanosensors in osteocytes that transmit extracellular mechanical signals into osteocytes and regulate responsive gene expression. The osteocyte cytoskeleton, dendritic processes, Integrin-based focal adhesions, connexin-based intercellular junctions, primary cilium, ion channels, and extracellular matrix are the major mechanosensors in osteocytes reported so far with evidence from both in vitro and in vitro studies. | 2 | Formation and destruction processes of upper Sinian oil-gas pools in the Dingshan-Lintanchang structural belt, southeast Sichuan Basin, China显示文摘The lower Cambrian Niutitang Formation hydrocarbon source rocks at the Dingshan-Lintanchang structure in the southeast Sichuan Basin were of medium-good quality with two excellent hydrocarbon-generating centers developed in the periphery areas, with a possibility of forming a medium to large-sized oil-gas field. Good reservoir rocks were the upper Sinian (Dengying Formation) dolomites. The mudstone in the lower Cambrian Niutitang Formation with a good sealing capacity was the cap rock. The widely occurring bitumen in the Dengying Formation indicates that a paleo oil pool was once formed in the study area. The first stage of paleo oil pool formation was maturation of the lower Cambrian source rocks during the late Ordovician. Hydrocarbon generation from the lower Cambrian source rocks stopped due to the Devonian-Carboniferous uplifting. The lower Cambrian source rocks then restarted generation of large quantities of hydrocarbons after deposition of the middle Permian sediments. This was the second stage of the paleo oil pool formation. The oil in the paleo oil pool began to crack during the late Triassic and a paleo gas pool was formed. This paleo gas pool was destroyed during the Yanshan-Himalayan folding, uplifting and denudation. Bitumen can be widely seen in the Dengying Formation in wells and outcrops in the Sichuan Basin and its periphery areas. | 3 | Multi-Stage Basin Development and Hydrocarbon Accumulations: A Review of the Sichuan Basin at Eastern Margin of the Tibetan Plateau显示文摘Sichuan Basin is one of the uppermost petroliferous basins in China. It experienced three evolutionary phases which were marine carbonate platform(Ediacaran to Late Triassic), Indosinian-Yanshanian orogeny foreland basin(Late Triassic to Late Cretaceous) and uplift and tectonic modification(Late Cretaceous to Quaternary). The present-day tectonics of the Sichuan Basin and its periphery are characterized by three basic elements which are topography, basement type and surface structure, and two settings(plate margin and interior). Therefore, be subdivided into five units which have different structure and tectonic history. The basin contains five different sets of source rocks with thickness up to 2 500 m. These source rocks were well preserved due to the presence of Middel–Lower Triassic evaporites(>~200 m) and thick terrestrial sediments filling in the Indosinian-Yanshanian foreland basin(>3 000 m). The uplift and erosion since Late Cretaceous has significant influence on cross-strata migration and accumulation of oil and gas. | 6 | Hydrothermal Dolomite in the Upper Sinian (Upper Proterozoic) Dengying Formation, East Sichuan Basin, China显示文摘Hydrothermal Dolomite(HTD) is present in the Upper Sinian(Upper Proterozoic) Dengying Formation, east Sichuan Basin, China. The strata are comprised by primary dolomite. The HTD has various textures, including zebra dolomite, subhorizontal sheet-like cavities filled by saddle dolomite and breccias cemented by saddle dolomites as well occur as a fill of veins and fractures. Also co-occur MVT type lead-zinc ores in the study area. The δ13C and δ18O isotopes of HTD in the Upper Sinian Dengying Formation are lighter than those of the host rocks, while 87Sr/86 Sr is higher. The apparent difference in carbon, oxygen and strontium isotopes, especially the large difference in 87Sr/86 Sr isotopes ratio indicate crystallization from hot basinal and/or hydrothermal fluids. Saddle dolomite was precipitated at temperatures of 270-320°C. The diagenetic parasequences of mineral assemblage deposited in the Dengying Formation are:(1) dolomite host rock →sphalerite-galena-barite-fluorite;(2) dolomite host rock →saddle dolomite →quartz;(3) dolomite host rock → saddle dolomite→bitumen;(4) dolomite host rock → saddle dolomite → barite. The mean chemical composition of the host dolomite matrix and HTD didn't change much during hydrothermal process. The fluids forming the HTDs in the Dengying Formation were mixtures of freshwater from the unconformity at the top of Sinian, fluids from diagenetic compaction and hydrocarbon generation & expulsion from the Lower Cambrian Niutitang Formation mudstones or the Doushantuo Formation silty mudstones, and hydrothermal fluids from the basement. The hydrocarbon reservoirs associated with the HTD were mostly controlled by the basement faults and fractures and karsting processes at the unconformity separating Sinian and Cambrian strata. The hydrocarbon storage spaces of HTD included dissolved cavities and intercrystalline pores. Dissolution cavities are extensive at the top of Dengying Formation, up to about 46 m below the unconformity between Sinian and Cambrian and were generated mainly during karstification. Hydrothermal alteration enhanced the reservoir property of the Dengying Formation dolomites with 3%-5% increase in porosity. No agreement has been reached why zebra dolomite occurs only in the Upper Sinian strata, which would indicate that HTD mineralization occurred during two different periods, each of them related to major extensional tectonic event. The early one related to the Xingkai taphrogenesis(Z2-?1)and the later one to the Emei taphrogenesis(D2-T2). | 7 | Osteoarthritis:pathogenic signaling pathways and therapeutic targets显示文摘Osteoarthritis(OA)is a chronic degenerative joint disorder that leads to disability and affects more than 500 million population worldwide.OA was believed to be caused by the wearing and tearing of articular cartilage,but it is now more commonly referred to as a chronic whole-joint disorder that is initiated with biochemical and cellular alterations in the synovial joint tissues,which leads to the histological and structural changes of the joint and ends up with the whole tissue dysfunction.Currently,there is no cure for OA,partly due to a lack of comprehensive understanding of the pathological mechanism of the initiation and progression of the disease.Therefore,a better understanding of pathological signaling pathways and key molecules involved in OA pathogenesis is crucial for therapeutic target design and drug development.In this review,we first summarize the epidemiology of OA,including its prevalence,incidence and burdens,and OA risk factors.We then focus on the roles and regulation of the pathological signaling pathways,such as Wnt/β-catenin,NF-κB,focal adhesion,HIFs,TGFβ/ΒΜP and FGF signaling pathways,and key regulators AMPK,mTOR,and RUNX2 in the onset and development of OA.In addition,the roles of factors associated with OA,including MMPs,ADAMTS/ADAMs,and PRG4,are discussed in detail.Finally,we provide updates on the current clinical therapies and clinical trials of biological treatments and drugs for OA.Research advances in basic knowledge of articular cartilage biology and OA pathogenesis will have a significant impact and translational value in developing OA therapeutic strategies. | Qing Yao Xiaohao Wu Chu Tao Weiyuan Gong Mingjue Chen Minghao Qu Yiming Zhong Tailin He Sheng Chen Guozhi Xiao | 2023 | Signal Transduction and Targeted Therapy2023,8,3: | 11 |
| 8 | Multi-Stage Hydrocarbon Accumulation and Formation Pressure Evolution in Sinian Dengying FormationCambrian Longwangmiao Formation, Gaoshiti-Moxi Structure, Sichuan Basin显示文摘Sichuan Basin is a typical superimposed basin, which experienced multi-phase tectonic movements, meanwhile Sinian–Cambrian underwent complex hydrocarbon accumulation processes, causing exploration difficulties in the past 60 years. Based on the microscopic evidence of fluid inclusions, combined with basin-modelling, this paper determines stages and time of hydrocarbon accumulation, reconstructs evolution of formation pressure and dynamic processes of hydrocarbon accumulation in Sinian Dengying Formation-Cambrian Longwangmiao Formation of Gaoshiti-Moxi structure. Three stages of inclusions are detected, including a stage of yellow-yellowgreen fluorescent oil inclusions, a stage of blue fluorescent oil-gas inclusions and a stage of non-fluorescent gas inclusions, reflecting the study area has experienced a series of complex hydrocarbon accumulation processes, such as formation of paleo-oil reservoirs, cracking of crude oil, formation of paleo-gas reservoirs and adjustment to present gas reservoirs, which occurred during 219–188, 192–146 and 168–0 Ma respectively. During the period of crude oil cracking, Dengying Formation-Longwangmiao Formation showed weak overpressure to overpressure characteristics, then after adjustment of paleo-gas reservoirs to present gas reservoirs, the pressure in Dengying Formation changed into overpressure but finally reduced to normal pressure system. | 9 | Focal adhesion protein Kindlin-2 regulates bone homeostasis in mice显示文摘Our recent studies demonstrate that the focal adhesion protein Kindlin-2 is critical for chondrogenesis and early skeletal development. Here, we show that deleting Kindlin-2 from osteoblasts using the 2.3-kb mouse Col1 a1-Cre transgene minimally impacts bone mass in mice, but deleting Kindlin-2 using the 10-kb mouse Dmp1-Cre transgene, which targets osteocytes and mature osteoblasts, results in striking osteopenia in mice. Kindlin-2 loss reduces the osteoblastic population but increases the osteoclastic and adipocytic populations in the bone microenvironment. Kindlin-2 loss upregulates sclerostin in osteocytes,downregulates β-catenin in osteoblasts, and inhibits osteoblast formation and differentiation in vitro and in vivo. Upregulation ofβ-catenin in the mutant cells reverses the osteopenia induced by Kindlin-2 deficiency. Kindlin-2 loss additionally increases the expression of RANKL in osteocytes and increases osteoclast formation and bone resorption. Kindlin-2 deletion in osteocytes promotes osteoclast formation in osteocyte/bone marrow monocyte cocultures, which is significantly blocked by an anti-RANKLneutralizing antibody. | 10 | Formation and evolution of Sinian oil and gas pools in typical structures, Sichuan Basin, China显示文摘The only major breakthrough in the exploration of the Sinian of the Sichuan Basin has been the Weiyuan gas field. Taking the typical structures in the Sichuan Basin as examples, an apatite fission track simulation was applied to constrain the thermal evolutionary history of the source rocks in this study. Combined with trap formation and evolution, the formation, destruction, and accumulation history of the Sinian reservoirs were analyzed from a dynamic perspective. The Sinian reservoirs underwent several primary stages: the paleo-reservoir formation and destruction in the late Silurian, hydrocarbon recharge in the Permian-Triassic, the cracking of oil to gas and dissolved-gas in the late Triassic-Mid-Late Jurassic, and the exsolution and accumulation of dissolved-gas as a result of episodic uplift since the Late Cretaceous. The exsolution process of dissolved-gas is of great significance to the accumulation of natural gas. The formation of the Weiyuan gas field is also related to this process. | 11 | Circular RNA circStag1 promotes bone regeneration by interacting with HuR显示文摘Postmenopausal osteoporosis is a common bone metabolic disorder characterized by deterioration of the bone microarchitecture,leading to an increased risk of fractures.Recently,circular RNAs(circ RNAs)have been demonstrated to play pivotal roles in regulating bone metabolism.However,the underlying functions of circ RNAs in bone metabolism in postmenopausal osteoporosis remain obscure.Here,we report that circ Stag1 is a critical osteoporosis-related circ RNA that shows significantly downregulated expression in osteoporotic bone marrow mesenchymal stem cells(BMSCs)and clinical bone tissue samples from patients with osteoporosis.Overexpression of circ Stag1 significantly promoted the osteogenic capability of BMSCs.Mechanistically,we found that circ Stag1 interacts with human antigen R(Hu R),an RNA-binding protein,and promotes the translocation of Hu R into the cytoplasm.A high cytoplasmic level of Hu R led to the activation of the Wnt signaling pathway by stabilizing and enhancing low-density lipoprotein receptor-related protein 5/6(Lrp5/6)andβ-catenin expression,thereby stimulating the osteogenic differentiation of BMSCs.Furthermore,overexpression of circ Stag1 in vivo by circ Stag1-loaded adeno-associated virus(circ Stag1-AAV)promoted new bone formation,thereby preventing bone loss in ovariectomized rats.Collectively,we show that circ Stag1 plays a pivotal role in promoting the regeneration of bone tissue via Hu R/Wnt signaling,which may provide new strategies to prevent bone metabolic disorders such as postmenopausal osteoporosis. | Gaoyang Chen Canling Long Shang Wang Zhenmin Wang Xin Chen Wanze Tang Xiaoqin He Zhiteng Bao Baoyu Tan Jin Zhao Yongheng Xie Zhizhong Li Dazhi Yang Guozhi Xiao Songlin Peng | 2022 | Bone Research2022,10,3: | 7 |
| 12 | Effects of Fatty Acids on Low-Sulfur Diesel Lubricity:Experimental Investigation, DFT Calculation and MD Simulation显示文摘The continuous reduction in sulfur content of fuels would lead to diesel fuel with poor lubricity which could result in engine pump failure. In the present work, fatty acids were adopted as lubricity additives to low-sulfur diesel fuel. It was attempted to correlate the molecular structures of fatty acids, such as carbon chain length, degree of saturation and hydroxylation, to their lubricity enhancement, which was evaluated by the High-Frequency Reciprocating Rig (HFRR) method. The efficiency order was supported by the density functional theory (DFT) calculations and the molecular dynamics (MD) simulations. The lubricity enhancing properties of fatty acids are mainly determined by the cohesive energy of adsorbed films formed on iron surface. | 14 | Kindlin-2 inhibits Nlrp3 inflammasome activation in nucleus pulposus to maintain homeostasis of the intervertebral disc显示文摘Intervertebral disc(IVD) degeneration(IVDD) is the main cause of low back pain with major social and economic burdens;however, its underlying molecular mechanisms remain poorly defined. Here we show that the focal adhesion protein Kindlin-2 is highly expressed in the nucleus pulposus(NP), but not in the anulus fibrosus and the cartilaginous endplates, in the IVD tissues. Expression of Kindlin-2 is drastically decreased in NP cells in aged mice and severe IVDD patients. Inducible deletion of Kindlin-2 in NP cells in adult mice causes spontaneous and striking IVDD-like phenotypes in lumbar IVDs and largely accelerates progression of coccygeal IVDD in the presence of abnormal mechanical stress. Kindlin-2 loss activates Nlrp3 inflammasome and stimulates expression of IL-1β in NP cells, which in turn downregulates Kindlin-2. This vicious cycle promotes extracellular matrix(ECM) catabolism and NP cell apoptosis. Furthermore, abnormal mechanical stress reduces expression of Kindlin-2, which exacerbates Nlrp3 inflammasome activation, cell apoptosis, and ECM catabolism in NP cells caused by Kindlin-2 deficiency. In vivo blocking Nlrp3 inflammasome activation prevents IVDD progression induced by Kindlin-2 loss and abnormal mechanical stress. Of translational significance, adeno-associated virus-mediated overexpression of Kindlin-2 inhibits ECM catabolism and cell apoptosis in primary human NP cells in vitro and alleviates coccygeal IVDD progression caused by mechanical stress in rat. | 15 | Comparison of nitrate-removal efficiency and bacterial properties using PCL and PHBV polymers as a carbon source to treat aquaculture water显示文摘Nitrate(NO_(3)^(−))accumulation in recirculating aquaculture systems(RASs)with high stocking densities presents a problem for reared animals and the environment.The use of a biodegradable polymer as organic carbon for heterotrophic denitrification exhibits good performance for NO_(3)^(−)removal from wastewater.A comparison of NO_(3)^(−)–N removal efficiency and bacterial properties using polycaprolactone(PCL)and poly(3-hydroxybutyrateco-3-hydroxyvalerate)(PHBV)as carbon sources to treat aquaculture water was conducted for a 102-day period.The results indicated that the NO_(3)^(−)–N removal rates of 0.27±0.07 and 0.19±0.05 g/L per day,respectively,could be achieved with influent concentrations ranging from 81.1 to 132.75 mg/L and a flow rate of 1 L/h.The removal of NO_(3)^(−)–N versus consumed PCL(1:1 w/w)was significantly higher than that versus consumed PHBV(0.3:1 w/w)(P<0.05).The concentrations of effluent nitrite-nitrogen and total ammonium nitrogen were maintained at an acceptable level.The bacterial community structures between the two types of reactors varied significantly.Acidovorax and Denitratisoma were the top two genera of the bacterial community in the biofilm in the PCL beads with a dominance of 26.83%and 6.67%,respectively.In the PHBV beads,Acidovorax at 17.95%and Bdellovibrio at 6.37%were the top two genera.The PCL-denitrification reactor developed in this study showed better potential than the PHBV-denitrification reactor in removing NO_(3)^(−)from aquaculture water. | Guozhi Luo Zhiwei Hou Luqi Tian Hongxin Tan | 2020 | Aquaculture and Fisheries2020,5,2: | 5 |
| 16 | Comparing salinities of 0, 10 and 20 in biofloc genetically improved farmed tilapia (Oreochromis niloticus) production systems显示文摘A 150 days(150-d)experiment was carried out to investigate the production efficiency,inorganic nitrogen syndrome and bacteria community of indoor biofloc technology(BFT)systems used to rear genetically improved farmed tilapia(Oreochromis niloticus)under 0(S-0),10(S-10),and 20 salinities(S20).The start-up period for BFT was 50,60 and 80 d for S-0,S-10 and S-20 groups,respectively.At steady state,the total ammonium nitrogen(NH4þ-N)and nitrite nitrogen(NO2-N)were lower than 3.0 mg/L and 0.34 mg/L,respectively and no nitrate-nitrogen(NO3-N)accumulation was observed.The fish survival rate was above 95%for all the groups.The final fish biomass of the S-10 group(35.83±1.08 kg/m^(3))was not significantly different from the S-0(34.79±1.33 kg/m^(3))group but was significantly higher than S-20(32.6±1.04 kg/m3).The feed conversion ratio for the tilapia in S-20 was 1.46,which was higher than the ratio in S-0(1.40)and S-10(1.39)tilapia.There was no significant difference in the crude protein content of the back muscle from tilapia of the three experimental groups.No significant difference in blood parameters,except for aspartate aminotransferase and alkaline phosphatase was observed between the three groups.Evaluation of microorganisms in the three BFT systems revealed that Proteobacteria,Bacteroidetes,and Fusobacteria were the top three at the phylum level in all groups.However,a significant difference was observed at the genus level in the bacteria of the three BFTs at different salinity(P<0.05).©2017 Published by Elsevier B.V.on behalf of Shanghai Ocean University.This is an open access article。 | Guozhi Luo Wenqing Li Hongxin Tan Xiaoqing Chen | 2017 | Aquaculture and Fisheries2017,2,5: | 5 |
| 17 | CHIP regulates bone mass by targeting multiple TRAF family members in bone marrow stromal cells显示文摘Carboxyl terminus of Hsp70-interacting protein(CHIP or STUB1) is an E3 ligase and regulates the stability of several proteins which are involved in different cellular functions. Our previous studies demonstrated that Chip deficient mice display bone loss phenotype due to increased osteoclast formation through enhancing TRAF6 activity in osteoclasts. In this study we provide novel evidence about the function of CHIP. We found that osteoblast differentiation and bone formation were also decreased in Chip KO mice. In bone marrow stromal(BMS) cells derived from Chip^(-/-) mice, expression of a panel of osteoblast marker genes was significantly decreased. ALP activity and mineralized bone matrix formation were also reduced in Chip-deficient BMS cells. We also found that in addition to the regulation of TRAF6, CHIP also inhibits TNFα-induced NF-κB signaling through promoting TRAF2 and TRAF5 degradation. Specific deletion of Chip in BMS cells downregulated expression of osteoblast marker genes which could be reversed by the addition of NF-κB inhibitor. These results demonstrate that the osteopenic phenotype observed in Chip^(-/-) mice was due to the combination of increased osteoclast formation and decreased osteoblast differentiation. | 18 | Microwave strengthens decomposition of mixed rare earth concentrate: Microwave absorption characteristics显示文摘The potential application of microwave heating technology for processing the mixed rare earth concentrate was systematically investigated by analyzing the microwave absorption characteristics in this study. The complex permittivity was measured through resonant cavity perturbation method. The variations of permittivity, the loss factor, loss tangent and the penetration depth with the increasing temperature were investigated numerically. The results indicate that the permittivity increases as the temperature increases, and temperature has a pivotal effect on it. The mixed concentrate is high loss material at the temperature range from 600 to 800℃ according to theoretical analyses of loss tangent and penetration depth.
