2Numerical Simulation of 2D Supersonic Magnetohydrodynamic Channel and Study on Hall Effect显示文摘In this research effort,numerical simulation of two-dimensional magnetohydrodynamic(MHD) channel is performed and Hall effect is studied.The computational model consists of the Navier-Stokes(N-S) equations coupled with electrical-magnetic source terms,Maxwell equations and the generalized Ohm's law.Boundary conditions for the electrical potential equation considering Hall effect are derived.To start with,the MHD channel with single-pair electrodes is studied and flow of the electric current is in accordance with physical principle.Then the MHD channel with five-pair electrodes is numerically simulated.The results show that the electrical current concentrates on the downstream of the anode and the upstream of the cathode due to Hall effect,and the flow field becomes asymmetrical.At the current value of the magnetic interaction parameter,the electrical-magnetic force affects the flow remarkably,decreasing the outlet Mach number and increasing the outlet pressure;what's more,the flow structure in the channel becomes extremely complex.Performances of MHD channels with continual electrodes and segmented electrodes are compared.The results show that performance of the MHD channel with segmented electrodes is better than that with continual electrodes with the increase of Hall parameter.ZHENG Xiaomei LU Haoyu XU Dajun CAI Guobiao 2011Chinese Journal of Aeronautics2011,24,2:3
3Progress and prospects of horizontal well fracturing technology for shale oil and gas reservoirs显示文摘By systematically summarizing horizontal well fracturing technology abroad for shale oil and gas reservoirs since the “13th Five-Year Plan”, this article elaborates new horizontal well fracturing features in 3D development of stacked shale reservoirs, small well spacing and dense well pattern, horizontal well re-fracturing, fracturing parameters optimization and cost control. In light of requirements on horizontal well fracturing technology in China, we have summarized the technological progress in simulation of multi-fracture propagation, horizontal well frac-design, electric-drive fracturing equipment, soluble tools and low-cost downhole materials and factory-like operation. 5Occurrence, Prevention and Control of Cryphalus eriobotryae, a New Devastating Pest of Loquat显示文摘A devastating trunk beetle has been found in Baisha loquat in Dongshan Town, Wuzhong District, Suzhou City in recent years. It is iden-tified as a new insect species, Cryphalus eriobotryae, belonging to Scolytinae, Curculionidae, Coleoptera. So far, loquat is known as the unique host of C. eriobotryae, and trees of 20-30 years old or 3-4 years old are all attacked by the insect. Holes drilled by adults with the diameter of about0.8 mm and sores(ate by adults) with the diameter of 1 mm are found on bark and trunk of damaged trees. Three-dimensional channels in 3-4 layers are formed in cortex, which are filled with insect dung. The phloem turns black-brown in late stage, and loses transport function, resulting in plant death and a great impact on loquat production and industrial development. During the occurrence period of C. eriobotryae from April to November,the damaged trunks and branches should be sprayed with 48% chlorpyrifos EC or 5% imidacloprid EC 100 times dilution once every 10 d for con-tinuous 2-3 times, and various states of the insect could be killed. 6Highly stretchable,soft and sticky PDMS elastomer by solvothermal polymerization process显示文摘Siloxane rubber shows attractive properties of high stability,elasticity and transparency.Besides,the regulation of its properties renders it widely used in many application fields.However,most of the reported performance improvement methods of siloxane rubber focus on the change of chemical composition of siloxane rubber,including the design of molecular chain and the introduction of other compounds,etc.Such a strategy is still faced with many limitations in practical application.In this work,on the premise of not changing the chemical composition of siloxane rubber,we propose a facile solvothermal polymerization process to change the structure of cross-linking networks,so as to obtain the siloxane rubber with controllable mechanical properties.Compared to the normal curing method,we realized polydimethylsiloxane elastomer(PDMS)with maximum elongation of more than 3,000%(>10 times of normally cured one)and tensile modulus lower than 0.15 MPa(<1/10 of normally cured one).In addition to superior stretchability,it gains extra high softness,stickiness and sensitive response to organic solvents.Based on our solvothermal cured PDMS,its applications in oil collection and organic solvent sensor have been demonstrated.It is expected that this method can be readily utilized widely and shows great application potentials.Jin Huang Yuchun Cai Chengyuan Xue Jin Ge Haoyu Zhao Shu-Hong Yu 2021Nano Research2021,14,10:1

8Single-cell analysis reveals the origins and intrahepatic development of liver-resident IFN-γ-producing γδ T cells显示文摘γδ T cells are heterogeneous lymphocytes located in various tissues.However,a systematic and comprehensive understanding of the origins of γδ T cell heterogeneity and the extrathymic developmental pathway associated with liver γδ T cells remain largely unsolved.In this study,we performed single-cell RNA sequencing(scRNA-seq)to comprehensively catalog the heterogeneity of γδ T cells derived from murine liver and thymus samples.We revealed the developmental trajectory of γδ T cells and found that the liver contains γδ T cell precursors(pre-γδ T cells).The developmental potential of hepatic γδ T precursor cells was confirmed through in vitro coculture experiments and in vivo adoptive transfer experiments.The adoptive transfer of hematopoietic progenitor Lin^(-)Sca-1^(+)Mac-1^(+)(LSM)cells from fetal or adult liver samples to sublethally irradiated recipients resulted in the differentiation of liver LSM cells into pre-γδ T cells and interferon-gamma^(+)(IFN-γ^(+))but not interleukin-17a^(+)(IL-17a^(+))γδ T cells in the liver.Importantly,thymectomized mouse models showed that IFN-γ-producing γδ T cells could originate from liver LSM cells in a thymus-independent manner.These results suggested that liver hematopoietic progenitor LSM cells were able to differentiate into pre-γδ T cells and functionally mature γδ T cells,which implied that these cells are involved in a distinct developmental pathway independent of thymus-derived γδ T cells.Yuan Hu Keke Fang Yanan Wang Nan Lu Haoyu Sun Cai Zhang 2021Cellular & Molecular Immunology2021,18,4:1

10Calcium overload and reactive oxygen species accumulation induced by selenium deficiency promote autophagy in swine small intestine显示文摘Selenium(Se)deficiency can seriously affect the small intestine of swine,and cause diarrhea in swine.However,the specific mechanism of Se deficiency-induced swine diarrhea has rarely been reported.Here,to explore the damage of Se deficiency on the calcium homeostasis and autophagy mechanism of swine,in vivo and in vitro models of swine intestinal Se deficiency were established.Twenty-four pure line castrated male Yorkshire pigs(45 d old,12.50±1.32 kg,12 full-sibling pairs)were divided into 2 equal groups and fed Se-deficient diet(0.007 mg Se/kg)as the Se-deficiency group,or fed Se-adequate diet(0.3 mg Se/kg)as the control group for 16 weeks.The intestinal porcine enterocyte cell line(IPEC-J2)was divided into 2 groups,and cultured by Se-deficient medium as the Se-deficient group,or cultured by normal medium as the control group.Morphological observations showed that compared with the control group,intestinal cells in the Se-deficiency group were significantly damaged,and autophagosomes increased.Autophagy staining and cytoplasmic calcium staining results showed that in the Sedeficiency group,autophagy increased and calcium homeostasis was destroyed.According to the reactive oxygen species(ROS)staining results,the percentage of ROS in the Se-deficiency group was higher than that in the control group in the in vitro model.Compared with the control group,the protein and mRNA expressions of autophagy-calcium-related genes including Beclin 1,microtubule-associated proteins 1 A(LC3-1),microtubule-associated proteins 1 B(LC3-2),autophagy-related protein 5(ATG5),autophagy-related protein 12(ATG12),autophagy-related protein 16(ATG16),mammalian target of rapamycin(mTOR),calmodulin-dependent protein kinase kinaseβ(CAMKK-β),adenosine 5’-monophosphate-activated protein kinase(AMPK),sarco(endo)plasmic reticulum Ca2+-ATPase(SERCA),and calpain in the Se-deficiency group were significantly increased which was consistent in vivo and in vitro(P<0.05).Altogether,our results indicated that Se deficiency could destroy the calcium homeostasis of the swine small intestine to trigger cell autophagy and oxidative stress,which was helpful to explain the mechanism of Se deficiency-induced diarrhea in swine.Yingying Zheng Haoyue Guan Jie Yang Jingzeng Cai Qi Liu Ziwei Zhang 2021Animal Nutrition2021,,4:0
11Realizing the Long Lifespan of Molybdenum Trioxide in Aqueous Aluminum Ion Batteries Through Potential Regulation显示文摘MoO_(3) is one of the most promising anode materials for aqueous aluminum batteries due to its high theoretical capacity and suitable aluminum insertion/de-insertion potential.However,the inferior cycling stability limits its further application,and the failure mechanism is still unclear.In this article,we provide a straightforward potential regulation technique to manage phase evolution during the charge/discharge process,which ultimately results in a markedly enhanced MoO_(3) electrode cycling stability.The failure mechanism study reveals that the excessive oxidation of the electrode during charge/discharge generates the H_(0.34)MoO_(3) phase,which has high solubility and is the primary cause of MoO_(3) deactivation.Although the dissolved Mo species will be deposited onto the electrode sheet again,the deposition is not electrochemically active and cannot contribute to the capacitance.Controlling the cutoff potential prevented the production of H_(0.34)MoO_(3),resulting in excellent cycling performance(80.1% capacity retention after 4000 cycles).The as-assembled α-MoO_(3)//MnO_(2) full battery exhibits high discharge plateaus(1.4 and 0.9 V),large specific capacity(200 mAhg^(-1) at 2 Ag^(-1)),and ultra-high coulombic efficiency(99%).The research presented here may contribute to the development of highly stable electrode materials for aqueous batteries.Haodong Fan Xuejin Li Jie Zhou Xiaoning Wang Xiuli Gao Haoyu Hu Li Zhou Tonghui Cai Yongpeng Cui Pengyun Liu Qingzhong Xue Zifeng Yan Wei Xing 2023Renewables2023,1,4:0

13Sodium butyrate alleviates deoxynivalenol-induced hepatic cholesterol metabolic dysfunction via RORγ-mediated histone acetylation modification in weaning piglets显示文摘Background:Cholesterol is an essential component of lipid rafts in cell plasma membrane,which exerts a hepatoprotective role against mycotoxin exposure in pigs,and cholesterol metabolism is vulnerable to epigenetic histone acetylation.Therefore,our present study aimed to investigate whether a histone deacetylase inhibitor(sodium butyrate [NaBu]) could protect the porcine liver from deoxynivalenol(DON) exposure by modulating cholesterol metabolism.Herein,we randomly divided 28 pigs into four groups,which were fed an uncontaminated basal diet,contaminated diet(4 mg DON/kg),uncontaminated diet supplemented with 0.2% NaBu or 4 mg/kg DON contaminated diet(4 mg DON/kg) supplemented with 0.2% NaBu for 28 d.Results:We found that the serum alanine transaminase(ALT),aspartate transaminase(AST),and alkaline phosphatase(ALP) were all increased in pigs exposed to DON,indicative of significant liver injury.Furthermore,the cholesterol content in the serum of DON-exposed pigs was significantly reduced,compared to the healthy Vehicle group.Transcriptome analysis of porcine liver tissues revealed that the cholesterol homeostasis pathway was highly enriched due to DON exposure.In which we validated by qRT-PCR and western blotting that the cholesterol program was markedly activated.Importantly,NaBu effectively restored parameters associated with liver injury,along with the cholesterol content and the expression of key genes involved in the cholesterol biosynthesis pathway.Mechanistically,we performed a ChIP-seq analysis of H3K27ac and showed that NaBu strongly diminished DON-increased H3K27ac genome-wide enrichment.We further validated that the elevated H3K27ac and H3K9ac occupancies on cholesterol biosynthesis genes were both decreased by NaBu,as determined by ChIP-qPCR analysis.Notably,nuclear receptor RORγ,a novel regulator of cholesterol biosynthesis,was found in the hyperacetylated regions.Again,a remarkable increase of RORγ at both mRNA and protein levels in DON-exposed porcine livers was drastically reduced by NaBu.Consistent with RORγ expression,NaBu also hindered RORγ transcriptional binding enrichments on these activated cholesterol biosynthesis genes like HMGCR,SQLE,and DHCR24.Furthermore,we conducted an in vitro luciferase reporter assay to verify that porcine RORγ directly bonds to the promoters of the above target genes.Conclusions:Collectively,our results demonstrate the utility of the natural product Na Bu as a potential anti-mycotoxin nutritional strategy for regulating cholesterol metabolism via RORγ-mediated histone acetylation modification.Qiufang Zong Huan Qu Yahui Zhao Haoyu Liu Shenglong Wu Shuai Wang Wenbin Bao Demin Cai 2023Journal of Animal Science and Biotechnology2023,14,2:0
14Low-cost biodegradable lead sequestration film for perovskite solar cells显示文摘Despite the high efficiency that has been achieved for the perovskite solar cells(PSCs),the hazardous lead leakage from the perovskite absorber layer is one of the crucial barriers still hindering its penetration into the commercial market for a large-scale installation.Herein,we report a novel low-cost and biodegradable lead sequestration layer with high compatibility for up-scalable encapsulation of PSCs.Through a precisely designed cross-linking reaction of chemical agents,the as-made biodegradable chitosan composite film shows enhanced mechanical strength,chemical stability,and lead adsorption capacity.The designed encapsulation strategy reduces over 99.99% lead leakage to <2 ppb under varied simulations of weather conditions(hail,rain,or flood),which meet the safe level of drinking water set by the US Environmental Protection Agency(EPA).Moreover,the PSC efficiency is improved from 21.91% to22.82% due to the improved light absorption from the printed biodegradable lead absorption film.Finally,we present a prototype process of accumulation and recycling of lead compounds in PSCs derbies via the biodegradation process.Based on the low-cost biodegradable lead sequestration film,this environmental-friendly encapsulation strategy could address the lead leakage issue for further commercialization of PSCs.Yiming Xiong Haoyu Cai Wang Yue Wenjian Shen Xuehao Zhu Juan Zhao Fuzhi Huang Yi-Bing Cheng Jie Zhong 2023Journal of Energy Chemistry2023,,9:0
15Hot-melt sub-and outercoating combined with enteric aqueous coating to improve the stability of aspirin tablets显示文摘Aspirin is apt to hydrolyze. In order to improve its stability, a new method has been developed involving the application of hot-melt sub-and outercoating combined with enteric aqueous coating. The main aim was to investigate the influence of these factors on the stability of ASA and understand how they work. Satisfactory storage stability were obtained when the aspirin tablet core coated with Eudragit L30D55 film was combined with glycerin monostearate(GMS) as an outercoat. Hygroscopicity testing indicated that the moisture penetrating into the tablet may result in a significant change in the physical properties of the coating film observed by scanning electron microscopy. Investigation of the compatibility between the drug and film excipients shows that the talc and methacrylic acid had a significant catalytic effect on ASA. A hypothesis was proposed that the hydrolysis of ASA enteric coated tablets(ASA-ECT) was mostly concentrated in the internal film and the interfaces between the film and tablet core. In conclusion, hot-melt coating technology is an alternative to subcoating or outercoating. 16LncRNA ZFAS1 regulates cardiomyocyte differentiation of human embryonic stem cells显示文摘Background:Cardiomyocytes derived from human embryonic stem cells(hESCs)are regulated by complex and stringent gene networks during differentiation.Long non-coding RNAs(lncRNAs)exert critical epigenetic regulatory functions in multiple differentiation processes.However,the involvement of lncRNAs in the differentiation of hESCs into cardiomyocytes has not yet been fully elucidated.Here,we identified the key roles of ZFAS1(lncRNA zinc finger antisense 1)in the differentiation of cardiomyocytes from hESCs.Methods:A model of cardiomyocyte differentiation from stem cells was established using the monolayer differentiation method,and the number of beating hESCs-derived cardiomyocytes was calculated.Gene expression was analyzed by quantitative real-time PCR(qRTPCR).Immunofluorescence assays were performed to assess the expression of cardiac troponin T(cTnT)andα-actinin protein in cardiomyocytes.Results:qRT-PCR showed that ZFAS1 expression in the mesoderm was significantly higher than that in embryonic stem cells,cardiac progenitor cells,and cardiomyocytes.Knockdown of ZFAS1 inhibited cardiomyocyte differentiation from hESCs,which was characterized by reduced expression of the cardiac-specific markers cTnT,α-actinin,myosin heavy chain 6(MYH6),and myosin heavy chain 7(MYH7).In contrast,ZFAS1 overexpression remarkably increased the percentage of spontaneously beating cardiomyocytes.In terms of the mechanism,we found that ZFAS1 is an antisense lncRNA at the 5′end of the protein-coding gene ZNFX1.Knockdown of ZFAS1 could increase the mRNA expression level of ZNFX1.Furthermore,qRT-PCR demonstrated that the silencing of ZNFX1 led to an increase in cardiac-specific markers that predicted the promotion of cardiomyocyte differentiation.Conclusion:Altogether,these data suggest that lncRNA-ZFAS1 is required for cardiac differentiation by functionally inhibiting the expression of ZNFX1,which may provide a reference for the treatment of heart disease to a certain extent.YANG CAO YINING LIU YANG YU XIAOFEI GUO XIUXIU WANG WENYA MA HANJING LI ZHONGYU REN XINLU GAO SIJIA LI HAOYU JI HONGYANG CHEN HONG YAN YANAN TIAN XIN WANG BENZHI CAI 2023BIOCELL2023,47,6:0
17Time-restricted feeding downregulates cholesterol biosynthesis program via RORγ-mediated chromatin modification in porcine liver organoids显示文摘Background: Time-restricted feeding(TRF) is a dieting strategy based on nutrients availability and diurnal rhythm,shown to improve lipid metabolism efficiency. We have demonstrated previously that retinoic acid-related(RAR)orphan receptor(ROR) γ is the primary transcription factor controlling cholesterol(CHO) biosynthesis program of animals. However, the functional role of RORγ in liver physiology of pigs in response to TRF has not been determined, largely due to the lack of functional models and molecular tools. In the present study, we established porcine liver organoids and subjected them to restricted nutrients supply for 10-h during the light portion of the day.Results: Our results showed that TRF regimen did not alter hepatocyte physiology, including unchanged cell viability, caspase 3/7 enzyme activity and the gene signature of cell proliferation in porcine liver organoids,compared to the control group(P > 0.05). Furthermore, we found that TRF downregulated the hepatic CHO biosynthesis program at both mRNA and protein levels, along with the reduced cellular CHO content in porcine liver organoids(P < 0.05). Using unbiased bioinformatic analysis of a previous ChIP-seq data and ChIP-qPCR validation, we revealed RORγ as the predominant transcription factor that responded to TRF, amongst the 12 targeted nuclear receptors(NRs)(P < 0.05). This was likely through RORγ direct binding to the MVK gene(encoding mevalonate kinase). Finally, we showed that RORγ agonists and overexpression enhanced the enrichment of cofactor p300, histone marks H3 K27 ac and H3K4me1/2, as well as RNA polymerase II(Pol-II) at the locus of MVK, in TRF-porcine liver organoids, compared to TRF-vector control(P < 0.05).Conclusions: Our findings demonstrate that TRF triggers the RORγ-mediated chromatin remodeling at the locus of CHO biosynthesis genes in porcine liver organoids and further improves lipid metabolism.Kexin Zhang Hao Li Zimeng Xin Yanwei Li Xiaolong Wang Yun Hu Haoyu Liu Demin Cai 2021Journal of Animal Science and Biotechnology2021,12,1:0
