| 5 | Channel Measurement and Path Loss Modeling from 220 GHz to 330 GHz for 6G Wireless Communications显示文摘Terahertz(THz)communication has been envisioned as a key enabling technology for sixthgeneration(6G).In this paper,we present an extensive THz channel measurement campaign for 6G wireless communications from 220 GHz to 330 GHz.Furthermore,the path loss is analyzed and modeled by using two single-frequency path loss models and a multiplefrequencies path loss model.It is found that at most frequency points,the measured path loss is larger than that in the free space.But at around 310 GHz,the propagation attenuation is relatively weaker compared to that in the free space.Also,the frequency dependence of path loss is observed and the frequency exponent of the multiple-frequencies path loss model is 2.1.Moreover,the cellular performance of THz communication systems is investigated by using the obtained path loss model.Simulation results indicate that the current inter-site distance(ISD)for the indoor scenario is too small for THz communications.Furthermore,the tremendous capacity gain can be obtained by using THz bands compared to using microwave bands and millimeter wave bands.Generally,this work can give an insight into the design and optimization of THz communication systems for 6G. | Pan Tang Jianhua Zhang Haoyu Tian Zhaowei Chang Jun Men Yuxiang Zhang Lei Tian Liang Xia Qixing Wang Jingsuo He | 2021 | China Communications2021,18,5: | 7 |

| 7 | All-trans retinoic acid induces leukemia resistance to NK cell cytotoxicity by down-regulating B7-H6 expression via c-Myc signaling显示文摘Background:The interaction between activating receptor NKp30 and its major tumor ligand B7-H6 is important for NK cell-mediated tumor rejection.However,the regulation of B7-H6 by tumor therapeutics remains largely unknown.In this study,we investigated the regulation of B7-H6 by all-trans retinoic acid(atRA),a terminal differentiation inducer of tumor cells that is extensively used for clinical leukemia therapy.Methods:We investigated the role of NKp30:B7-H6 axis in NK cell-mediated tumor lysis against leukemia cells and the influence of atRA treatment on the cytotoxicity of NK cells using NK cell lines(NK92 and NKG)and leukemia cell lines(U-937 and THP-1).We evaluated the effect of atRA treatment on the expression of B7-H6 using real-time PCR,flow cytometry and western blotting.We used CRISPR/Cas9 to knockdown B7-H6 expression and siRNA to knockdown c-Myc in U-937 cells to evaluate the role of B7-H6 and c-Myc in atRA-induced tumor resistance against NK cells.Results:NK cell-mediated U-937 cell lysis was mainly dependent on NKp30/B7-H6 interaction.Blockade of B7-H6 by monoclonal antibody significantly impaired NK cytotoxicity.atRA treatment induced U-937 resistance to NK cell cytotoxicity by reducing B7-H6 expression,and showed no effect on NK cytotoxicity against B7-H6 knockdown U-937 cells.Epigenetic modifications,such as DNA methylation and histone deacetylase(HDAC),were not responsible for atRA-mediated B7-H6 down-regulation as inhibitors of these pathways could not restore B7-H6 mRNA expression.On the other hand,atRA treatment reduced c-Myc expression,which in turn inhibited the transcription of B7-H6 on leukemia cells.Conclusion:atRA treatment promotes tumor cell resistance against NK cellmediated lysis by down-regulating B7-H6 expression via the c-Myc signaling pathway,suggesting that more attention needs to be paid to the immunological adverse effects in the clinical use of atRA treatment. | Guoshuai Cao Ying Cheng Xiaodong Zheng Haiming Wei Zhigang Tian Rui Sun Haoyu Sun | 2021 | Cancer Communications2021,41,1: | 1 |

| 9 | Cell-free synthesis system-assisted pathway bottleneck diagnosis and engineering in Bacillus subtilis显示文摘Metabolic engineering is a key technology for cell factories construction by rewiring cellular resources to achieve efficient production of target chemicals.However,the existence of bottlenecks in synthetic pathway can seriously affect production efficiency,which is also one of the core issues for metabolic engineers to solve.Therefore,developing an approach for diagnosing potential metabolic bottlenecks in a faster and simpler manner is of great significance to accelerate cell factories construction.The cell-free reaction system based on cell lysates can transfer metabolic reactions from in vivo to in vitro,providing a flexible access to directly change protein and metabolite variables,thus provides a potential solution for rapid identification of bottlenecks.Here,bottleneck diagnosis of the N-acetylneuraminic acid(NeuAc)biosynthesis pathway in industrially important chassis microorganism Bacillus subtilis was performed using cell-free synthesis system.Specifically,a highly efficient B.subtilis cell-free system for NeuAc de novo synthesis was firstly constructed,which had a 305-fold NeuAc synthesis rate than that in vivo and enabled fast pathway dynamics analysis.Next,through the addition of all potential key intermediates in combination with substrate glucose respectively,it was found that insufficient phosphoenolpyruvate supply was one of the NeuAc pathway bottlenecks.Rational in vivo metabolic engineering of NeuAc-producing B.subtilis was further performed to eliminate the bottleneck.By down-regulating the expression level of pyruvate kinase throughout the growth phase or only in the stationary phase using inhibitory Nterminal coding sequences(NCSs)and growth-dependent regulatory NCSs respectively,the maximal NeuAc titer increased 2.0-fold.Our study provides a rapid method for bottleneck diagnosis,which may help to accelerate the cycle of design,build,test and learn cycle for metabolic engineering. | Rongzhen Tian Minghu Wang Jintian Shi Xiaolong Qin Haoyu Guo Xuanjie Jia Jianghua Li Long Liu Guocheng Du Jian Chen Yanfeng Liu | 2020 | Synthetic and Systems Biotechnology2020,5,3: | 1 |
| 10 | Trispecific killer engager 161519 enhances natural killer cell function and provides anti-tumor activity against CD19-positive cancers显示文摘Objective:Natural killer(NK)cells have gained considerable attention due to their potential in treating'cold tumors,'and are therefore considered as one of the new strategies for curing cancer,by using worldwide development of their new possibilities and interventions with NK cell-related therapeutic products.Methods:We constructed a trispecific killer engager(TriKE)consisting of anti-CD16,IL-15,and anti-CD19.This TriKE was designed to attract CD19^(+)tumor cells to CD16^(+)NK cells,whereas IL-15 sustained the proliferation,development,and survival of NK cells.Results:Treatment with 161519 TriKE in the presence of CD19^(+)targets upregulated expression of CD69,CD107 a,TRAIL,IFN-γ,and TNF-α in NK cells,and significantly improved the proliferation and cytotoxicity of NK cells.NK cells'armed'with 161519 TriKE showed stronger cytolysis against CD19+targets compared with that of'unarmed'NK cells.A preclinical model of B-cell lymphoma in human peripheral blood mononuclear cell-reconstituted xenograft mice showed significant inhibition of tumor growth and prolonged overall survival after treatment with 161519 TriKE,when compared with that in control mice or mice treated with 1619 BiKE.Combined use of IL-2 was a more effective treatment with 1619 BiKE,when compared with that using 161519 TriKE.Conclusions:The newly generated 161519 TriKE enhanced the proliferation,activation,cytokine secretion,and cytotoxicity of NK cells in the presence of CD19+tumor cells.The 161519 TriKE aided inhibition of tumor growth and prolonged the overall survival of murine xenografts,and could be used to treat CD19-positive cancers. | Ying Cheng Xiaodong Zheng Xuefu Wang Yongyan Chen Haiming Wei Rui Sun Zhigang Tian Haoyu Sun | 2020 | Cancer Biology & Medicine2020,17,4: | 1 |

| 14 | Nomogram for predicting the overall survival of patients after D2/R0 resection and chemoradiotherapy for stage Ⅲ gastric cancer or gastroesophageal junction cancer显示文摘Objective:To explore the independent factors associated with the overall survival(OS)after D2/R0 resection and adjuvant chemoradiotherapy(CRT)for stage Ⅲ gastric cancer(GC)and gastroesophageal junction(GEJ)cancer,and establish and validate a nomogram model to predict the OS in this patient population.Methods:A total of 569 stage Ⅲ GC or GEJ cancer patients who underwent D2/R0 resection and adjuvant CRT at our center from January 2013 to May 2018 were retrospectively enrolled in this study.The patients were randomly divided into training and validation cohorts in a 3:1 ratio.In the training cohort,univariate and multivariate analysis were conducted to identify clinical factors associated with OS,and a nomogram model was established on the basis of the independent factors.The concordance index(C-index)and the calibration curve were used to validate the prediction power of the nomogram.Results:Overall,427 and 142 patients were enrolled in the training and validation cohorts,respectively.In the univariate and multivariate analyses,age,Lauren's classification,TNM stage,and administration of adjuvant CRT were independent factors for OS to construct the nomogram.The C-index values were 0.654 and 0.613 in the training and validation cohorts,respectively.The calibration curves were identical to the actual OS.Risk stratification based on the nomogram model showed good risk stratification for OS in both groups.Conclusion:Our study included adjuvant CRT as a predictive factor and established a nomogram model for predicting the OS in patients with stage III GC or GEJ after D2/R0 resection and adjuvant therapy.This nomogram could facilitate more individualized prediction of OS in clinical practice. | Jinming Shi Wenzhe Kang Songlin Gao Wenyang Liu Yuan Tang Ning Li Qiang Zeng Huiying Ma Haoyue Li Yantao Tian Jing Jin | 2023 | Radiation Medicine and Protection2023,4,3: | 0 |

| 18 | LncRNA ZFAS1 regulates cardiomyocyte differentiation of human embryonic stem cells显示文摘Background:Cardiomyocytes derived from human embryonic stem cells(hESCs)are regulated by complex and stringent gene networks during differentiation.Long non-coding RNAs(lncRNAs)exert critical epigenetic regulatory functions in multiple differentiation processes.However,the involvement of lncRNAs in the differentiation of hESCs into cardiomyocytes has not yet been fully elucidated.Here,we identified the key roles of ZFAS1(lncRNA zinc finger antisense 1)in the differentiation of cardiomyocytes from hESCs.Methods:A model of cardiomyocyte differentiation from stem cells was established using the monolayer differentiation method,and the number of beating hESCs-derived cardiomyocytes was calculated.Gene expression was analyzed by quantitative real-time PCR(qRTPCR).Immunofluorescence assays were performed to assess the expression of cardiac troponin T(cTnT)andα-actinin protein in cardiomyocytes.Results:qRT-PCR showed that ZFAS1 expression in the mesoderm was significantly higher than that in embryonic stem cells,cardiac progenitor cells,and cardiomyocytes.Knockdown of ZFAS1 inhibited cardiomyocyte differentiation from hESCs,which was characterized by reduced expression of the cardiac-specific markers cTnT,α-actinin,myosin heavy chain 6(MYH6),and myosin heavy chain 7(MYH7).In contrast,ZFAS1 overexpression remarkably increased the percentage of spontaneously beating cardiomyocytes.In terms of the mechanism,we found that ZFAS1 is an antisense lncRNA at the 5′end of the protein-coding gene ZNFX1.Knockdown of ZFAS1 could increase the mRNA expression level of ZNFX1.Furthermore,qRT-PCR demonstrated that the silencing of ZNFX1 led to an increase in cardiac-specific markers that predicted the promotion of cardiomyocyte differentiation.Conclusion:Altogether,these data suggest that lncRNA-ZFAS1 is required for cardiac differentiation by functionally inhibiting the expression of ZNFX1,which may provide a reference for the treatment of heart disease to a certain extent. | YANG CAO YINING LIU YANG YU XIAOFEI GUO XIUXIU WANG WENYA MA HANJING LI ZHONGYU REN XINLU GAO SIJIA LI HAOYU JI HONGYANG CHEN HONG YAN YANAN TIAN XIN WANG BENZHI CAI | 2023 | BIOCELL2023,47,6: | 0 |

| 20 | Panoramic comparison between NK cells in healthy and cancerous liver through single-cell RNA sequencing显示文摘Objective:NK cells play crucial roles in the immune defense mechanisms against viral infections and transformed cells.However,the developmental progression,transcriptomic landscape,and functional subtypes of liver NK cells are not well defined.Hepatocellular carcinoma(HCC)accounts for approximately 80%of primary liver cancer worldwide,yet the biological characteristics of NK cells in the HCC environment are unclear.Therefore,we aimed to determine these cells’roles in tumorigenesis and prognosis.Methods:We compared the single-cell RNA sequencing profiles of NK cells purified from blood(n=1),healthy liver tissues(n=3),HCC tumor tissues(n=4),and peritumor liver tissues(n=1)to identify NK cell subsets.Furthermore,we performed bioinformatics analysis by using The Cancer Genome Atlas(TCGA)data to identify prognostic biomarkers simultaneously overexpressed in the blood and tumor tissues of patients with HCC.Results:Transcriptomic analysis revealed 5 NK cell subsets(L1-NK-CD56bright,L2-NK-CD56dim,L3-NK-HLA,L4-LrNK-FCGR3A,and L5-LrNK-XCL1)in the healthy liver tissues.However,the transitional L3 subset and the CXCR6+CD16+L4 subset with strong anti-tumor activity were absent in the HCC and peritumor liver tissues.Furthermore,4 common prognosis-associated genes(RHOB,TALDO1,HLA-DPA1,and TKT)were significantly overexpressed in the paired tumor tissue and blood.Conclusions:Our study revealed 5 specific subsets of NK cells in healthy human liver tissues.However,only 3 of the 5 NK cell subsets were present in HCC and peritumor tissues.The cytotoxic NK cell subsets were absent in HCC tissues.Furthermore,we identified 4 potential non-invasive prognostic biomarkers in patients with HCC. | Huan Liu Ronghua Zhao Rongrong Qin Haoyu Sun Qiang Huang Lianxin Liu Zhigang Tian Björn Nashan Cheng Sun Rui Sun | 2022 | Cancer Biology & Medicine2022,19,9: | 0 |
