| 1 | A 2-kb Mycovirus Converts a Pathogenic Fungus into a Beneficial Endophyte for Brassica Protection and Yield Enhancement显示文摘Mycoviruses are viruses that infect fungi,and hypovirulence-associated mycoviruses have the potential to control fungal diseases.However,it is unclear how mycovirus-mediated hypovirulent strains live and survive in the field,and no mycovirus has been applied for field crop protection.In this study,we found that a previously identified small DNA mycovirus(SsHADV-1)can convert its host,Sclerotinia sclerotiorum,from a typical necrotrophic pathogen to a beneficial endophytic fungus.SsHADV-1 downregulates the expression of key pathogenicity factor genes in S.sclerotiorum during infection.When growing in rapeseed,the SsHADV-1-infected strain DT-8 significantly regulates the expression of rapeseed genes involved in defense,hormone signaling,and circadian rhythm pathways.As a result,plant growth is promoted and disease resistance is enhanced.Field experiments showed that spraying DT-8 at the early flowering stage can reduce the disease severity of rapeseed stem rot by 67.6%and improve yield by 14.9%.Moreover,we discovered that SsHADV-1 could also infect other S.sclerotiorum strains on DT-8-inoculated plants and that DT-8 could be recovered from dead plants.These findings suggest that the mycoviruses may have the ability to shape the origin of endophytism.Our discoveries suggest that mycoviruses may influence the origin of endophytism and may also offer a novel strategy for disease control in which mycovirus-infected strains are used to improve crop health and release mycoviruses into the field. | Hongxiang Zhang Jiatao Xie Yanping Fu Jiasen Cheng Zheng Qu Zhenzhen Zhao Shufen Cheng Tao Chen Bo Li Qianqian Wang Xinqiang Liu Binnian Tian David B.Collinge Daohong Jiang | 2020 | Molecular Plant2020,13,10: | 9 |
| 2 | Analysis of Dynamic Tensile Process of Fiber Reinforced Concrete by Acoustic Emission Technique显示文摘The fiber reinforced concrete has good dynamic mechanical properties. But corresponding research lacks the dynamic damage characteristics of the polypropylene fiber(fiber of low elastic modulus) and steel fiber(fiber of high elastic modulus) reinforced concrete under medium strain rate(10-6 s-1-10-4 s-1). In order to study the effect of strain rate on the damage characteristics of fiber reinforced concrete during the full curve damage process, the real time dynamic acoustic emission(AE) technique was applied to monitor the damage process of fiber reinforced concrete at three strain rates. The AE wavelet energy spectrum in ca8 frequency band and average AE peak frequency at three strain rates were analyzed. With the accumulation of damage, the AE wavelet energy spectrum in ca8 frequency band increased first and then decreased, and the average AE peak frequency increased gradually. With the increase of strain rate, the AE wavelet energy spectrum in ca8 frequency band and average AE peak frequency decreased gradually. The polypropylene fiber content has more obvious effect on the Dynamic increase factor(DIF) of the peak stress than the steel fiber content. | 3 | THE GLASS TRANSITION TEMPERATURE AND MICROSTRUCTURE OF POLYURETHANE/EPOXY RESIN INTERPENETRATING POLYMER NETWORKS NANOCOMPOSITES显示文摘Nanocomposites with various contents of organophilic montmorillonite (oMMT) have been prepared by adding oMMT to interpenetrating poly- mer networks (IPNs) of polyurethane and epoxy resin (PU/EP) which had been prepared by a sequential polymeric technique. DSC experiment indicates a novel phenomenon that the glass transition tem- perature (Tg) of the nanocomposites increases with the oMMT content up to 3 %, then decreases with further increasing oMMT content. In order to explain this phenomenon, crosslink density, hydrogen bond- ing in the hard segments, crystallization of the nanocomposites and the exfoliation degree of oMMT in the nanocomposites have been investigated by swelling method, FT-IR, XRD, SEM and TEM, re- spectively. The results indicate that the crosslink density and the hydrogen bonding index of the nanocomposites increase, but the crystallization de- gree of the nanocomposites decreases with increas- ing oMMT content. | 5 | Materials Experiment on Tiangong-2 Space Laboratory显示文摘During the China's Tiangong-2(TG-2) flight mission, the experiments of 18 kinds of material samples were conducted in space by using a Multiple Materials Processing Furnace(MMPF) mounted in the orbital module of the TG-2 space laboratory. After the experiments of 12 kinds of samples of the first and second batches were completed successfully, astronauts packed and brought them back to the ground by ShenzhouII spacecraft. By studying processing and formation on semiconductor and optoelectronics materials, metal alloys and metastable materials, functional single-crystal, micro-and nano-composite materials encapsulated in sample ampoules both in space and on Earth, we expect to explore some physical and chemical processes and mechanism of the materials formation that are normally obscured and therefore are difficult to study quantitatively on the ground due to the gravity-induced convection, to obtain the processing and synthesis technology for preparing high quality materials, and lead to the improvement and development of materials processing techniques on Earth, and also develop the experiment device and comprehensive ability for materials experiment in microgravity environment. | 6 | Damage Processes of Polypropylene Fiber Reinforced Mortar in Different Fiber Content Revealed by Acoustic Emission Behavior显示文摘The performances of the cement-based materials can be improved by the incorporation of polypropylene fiber, but the damage processes become more complex with different fiber contents at the same time. The acoustic emission(AE) technology can achieve the global monitoring of internal damage in materials. The evolution process of failure mode and damage degree of polypropylene fiber reinforced mortar and concrete were analyzed by measuring the AE energy, RA value, AF value and b value. It was found that the cement matrix cracked on the initial stage, the cracks further developed on the medium stage and the fibers were pulled out on the last stage. The matrix cracked with minor injury cracks, but the fiber broke with serious damage cracks. The cumulative AE energy was proportional to the polypropylene fiber reinforced concrete and mortar's ductility. | 7 | The pain-related behavior changes correlate with the bone damage in a rat model of rheumatoid arthritis显示文摘Objective: In view of the extensive bone damage in rheumatoid arthritis, we used a commonly utilized animal model to detect behavioral changes in pain-related and the bone damage during the early disease, and to explore the correlation between bone damage and pain-related behavioral changes. Methods: Arthritis were induced in Sprague-Dawley (SD) male rats by injecting complete Freund's adjuvant (CFA) into the tails. Pain-related behavior changes were studied using the Hargreaves, VonFrey, and acetone tests on the 0, 7, 14, day and 28 day after CFA injection. The rats were sacrificed according the same schedule. The bone damage of the right proximal tibia was studied by microCT scan and bone histological slices. Results: Animals developed soft tissue inflammation and polyarthritis on 7 days after CFA injection, and arthritic score proved obvious arthritis were established within the study period. Mechanical hyperalgesia and cold allodynia were present in the affected hind paw from the 7 day through the 28 day, but the heat hyperalgesia and the mechanical allodynia lasted a short time after CFA injection. Trabecular bone number (Tb.N), Tissue Mineral Content (TMC) and Bone Volume to Tissue Volume (BV/TV) in the proximal tibia by microCT scan were also reduced after induction, especial 14 days after CFA injection. The bone histological slices showed the trabecular bone and proteoglycan diminished, the bone damage severity scores became more severely on the 7 day after CFA injection. Using analysis of covariance, these changes had statistical significance compared with baseline. By linear regression analysis demonstrated mechanical hyperalgesia and cold allodynia correlated well with arthritic score, bone damage parameters and bone damage severity scores. Conclusion: Adjuvant-induced arthritis (AA) were observed after CFA injection and lasted within the later experimental period. Pain-related behavioral changes were observed in the early time of AA. Bone damage was also occurred with arthritis development. | 9 | Metabolic regulation of T cell development by Sinl-mTORC2 is mediated by pyruvate kinase M2显示文摘Glucose metabolism plays a key role in thymocyte development. The mammalian target of rapamycin complex 2 (mT0RC2) is a critical regulator of cell growth and metabolism, but its role in early thymocyte development and metabolism has not been fully studied. We show here that genetic ablation of Sinl, an essential component of mTORC2, in T lineage cells results in severely impaired thymocyte development at the CD4^- CD8^- double negative (DN) stages but not at the CD4^+ CD8^+ double positive (DP) or later stages. Notably, Sinl-deficient DN thymocytes show markedly reduced proliferation and glycolysis.Importantly, we discover that the M2 isoform of pyruvate kinase (PKM2) is a novel and crucial Sinl effector in promoting DN thymocyte development and metabolism. At the molecular level, we show that Sinl-mTORC2 controls PKM2 expression through an AKT-dependent PPAR-y nuclear translocation. | 13 | Composition and origin of lipid biomarkers in the surface sediments from the southern Challenger Deep, Mariana Trench显示文摘The surface sediments collected from the southern Mariana Trench at water depths between ca.4900 m and 7068 m were studied using lipid biomarker analyses to reveal the origin and distribution of organic matters. For all samples, an unresolved complex mixture(UCM) was present in the hydrocarbon fractions, wherein resistant component tricyclic terpanes were detected but C_(27)-C _(29) regular steranes and hopanes indicative of a higher molecular weight range of petroleum were almost absent.This biomarker distribution patterns suggested that the UCM and tricyclic terpanes may be introduced by contamination of diesel fuels or shipping activities and oil seepage elsewhere. The well-developed faults and strike-slip faults in the Mariana subduction zone may serve as passages for the petroleum hydrocarbons. In addition, the relative high contents of even n-alkanes and low Carbon Preference Indices indicated that the n-alkanes were mainly derived from bacteria or algae. For GDGTs, the predominance of GDGT-0 and crenarchaeol, together with low GDGT-0/Crenarchaeol ratios(ranging from0.86 to 1.64), suggests that the GDGTs in samples from the southern Mariana Trench were mainly derived from planktic Thaumarchaeota. However, the high GDGT-0/crenarchaeol ratio(10.5) in sample BC07 suggests that the GDGTs probably were introduced by methanogens in a more anoxic environment. Furthermore, the n-alkanes C_(19)-C_(22) and the n-fatty acids C_(20:0)-C_(22:0) were depleted in^(13)C by 3‰ compared to n-alkanes C_(16)-C_(18) and the n-fatty acids C_(14:0)-C_(18:0), respectively, which was interpreted to result from the preferential reaction of fatty acid fragments with carbon 'lighter' terminal carboxyl groups during carbon chain elongation from the precursors to products. The abundance of total alkanes, carboxylic acids, alcohols and total lipids were generally increased along the down-going seaward plate, suggesting the lateral organic matter inputs play an important role in organic matter accumulation in hadal trenches. The extremely high contents of biomarkers in sample BC11 were most likely related to trench topography and current dynamics, since the lower steepness caused by graben texture and proximity to the trench axis may result in higher sedimentation rate. | 16 | Glycolysis Recycling of Waste Polyurethane Rigid Foam Using Different Catalysts显示文摘Dramatically increasing waste polyurethane rigid foam(WPRF)draws the attention of the world.A mixture of ethylene glycol(EG)and diethylene glycol(DEG)is used as glycolysis agents.WPRF was subjected to alcoholysis using different catalysts which are titanium ethylene glycol and potassium hydroxide to obtain recycled polyol,respectively.The effect of a different catalyst on the viscosity and hydroxyl value of recycled polyol is discussed.The regenerated polyurethane(RPU)is performed using the recycled polyol.Infrared spectrum,compressive strength,apparent density,water absorption,scanning electron microscope,and thermogravimetric analysis are carried out to investigate the effect of WPRF degradation using different catalysts.The results show that titanium glycol is more efficient than potassium hydroxide in almost all conditions.The viscosity of the recycled polyol is relatively low,and the hydroxyl value meets the requirements of industrial use.When the titanium glycol titanium addition amount is 0.05%,the prepared RPU has a compressive strength of 0.24 MPa,an apparent density of 41.75 kg/m^(3),and a good foam structure.Besides,the water absorption rate of the RPU under the two catalytic systems is not much different,and the thermal stability is good.The recycled polyol can generally partially replace traditional polyols to prepare polyurethane rigid foams with good comprehensive properties. | Xiaohua Gu Hongxiang Luo Shiwei Lv Peng Chen | 2021 | Journal of Renewable Materials2021,9,7: | 2 |
| 17 | Development and validation of a UPLC–MS/MS assay for the determination of gemcitabine and its L-carnitine ester derivative in rat plasma and its application in oral pharmacokinetics显示文摘A simple and rapid UPLC–MS/MS method to simultaneously determine gemcitabine and its L-carnitine ester derivative(2’-deoxy-2’, 2’-difluoro-N-((4-amino-4-oxobutanoyl) oxy)-4-(trimethyl amm-onio) butanoate-cytidine, JDR) in rat plasma was developed and validated.The conventional plasma sample preparation method of nucleoside analogues is solidphase extraction(SPE) which is time-consuming and cost-expensive. In this study, gradient elution with small particles size solid phase was applied to effectively separate gemcitabine and JDR, and protein precipitation pretreatment was adopted to remove plasma protein and extract the analytes with high recovery(>81%). Method validation was performed as per the FDA guidelines, and the standard curves were found to be linear in the range of 5–4000 ng/ml for JDR and 4–4000 ng/ml for gemcitabine, respectively. The lower limit of quantitation(LLOQ)of gemcitabine and JDR was 4 and 5 ng/ml, respectively. The intra-day and inter-day precision and accuracy results were within the acceptable limits.
