1Connexin 43 hemichannels protect bone loss during estrogen deficiency显示文摘Estrogen deficiency in postmenopausal women is a major cause of bone loss,resulting in osteopenia,osteoporosis,and a high risk for bone fracture.Connexin 43 (Cx43) hemichannels (HCs) in osteocytes play an important role in osteocyte viability,bone formation,and remodeling.We showed here that estrogen deficiency reduced Cx43 expression and HC function.To determine if functional HCs protect osteocytes and bone loss during estrogen deficiency,we adopted an ovariectomy model in wild-type (WT) and two transgenic Cx43 mice:R76W (dominant-negative mutant inhibiting only gap junction channels) and Cx43 Δ130–136 (dominant-negative mutant compromising both gap junction channels and HCs).The bone mineral density (BMD),bone structure,and histomorphometric changes of cortical and trabecular bones after ovariectomy were investigated.Our results showed that the Δ130–136 transgenic cohort had greatly decreased vertebral trabecular bone mass compared to WT and R76W mice,associated with a significant increase in the number of apoptotic osteocyte and empty lacunae.Moreover,osteoclast surfaces in trabecular and cortical bones were increased after ovariectomy in the R76W and WT mice,respectively,but not in Δ130–136 mice.These data demonstrate that impairment of Cx43 HCs in osteocytes accelerates vertebral trabecular bone loss and increase in osteocyte apoptosis,and further suggest that Cx43 HCs in osteocytes protect trabecular bone against catabolic effects due to estrogen deficiency.Liang Ma Rui Hua Yi Tian Hongyun Cheng Roberto Jose Fajardo Joseph J. 2Reversible and irreversible heat generation of NCA/Si–C pouch cell during electrochemical energy-storage process显示文摘To meet the requirements of electronic vehicles(EVs) and hybrid electric vehicles(HEVs),the high energy density Li Ni_(0.8) Co_(0.15) Al_(0.05) O_2(NCA) cathode and Si–C anode have attracted more attention.Here we report the thermal behaviors of NCA/Si–C pouch cell during the charge/discharge processes at different current densities.The total heat generations are derived from the surface temperature change during electrochemical Li+insertion/extraction in adiabatic surrounding.The reversible heat is determined by the entropic coefficients,which are related with open-circuit voltage at different temperatures; while the irreversible heat is determined by the internal resistance,which can be obtained via V–I characteristic,electrochemical impedance spectroscopy and hybrid pulse power characterization(HPPC).During the electrochemical process,the reversible heat contributes less than 10% to total heat generation; and the heat generated in charge process is less than that in discharge process.The results of thermal behaviors analyses are conducive to understanding the safety management and paving the way for building a reliable thermal model of high energy density lithium ion battery.Ying Bai Limin Li Yu Li Guanghai Chen Huichun Zhao Zhaohua Wang Chuan Wu Hongyun Ma Xinquan Wang Hongyue Cui Jiang Zhou 2019Journal of Energy Chemistry2019,28,2:7
3The ACTIVE study protocol:apatinib or placebo plus gefitinib as first-line treatment for patients with EGFR-mutant advanced non-small cell lung cancer(CTONG1706)显示文摘Background:Gefitinib,as the first epidermal growth factor receptor tyrosine kinase inhibitors(EGFR-TKI)approved for the treatment of advanced non-small cell lung cancer(NSCLC),has been proved to significantly improve the progression-free survival(PFS)in the first-line setting but suffers from resistance 7-10 months after treatment initia-tion.Apatinib(YN968D1),a potent vascular endothelial growth factor receptor(VEGFR)2-TKI,specifically binds to VEGFR2 and leads to anti-angiogenetic and anti-neoplastic effect.Concurrent inhibition of VEGFR and EGFR path-ways represents a rational approach to improve treatment responses and delay the onset of treatment resistance in EGFR-mutant NSCLC.This ACTIVE study aims to assess the combination of apatinib and gefitinib as a new treatment approach for EGFR-mutant NSCLC as a first-line setting.Methods:This multicenter,randomized,double-blind,placebo-controlled phase III study(NCT02824458)has been designed to assess the efficacy and safety of apatinib or placebo combined with gefitinib as a first-line treatment for patients with EGFR-mutant advanced NSCLC.A total of 310 patients with EGFR-mutation(19del or 21L858R),pathological stage IIIB to IV non-squamous NSCLC were to be enrolled.The primary endpoint is investigator assessment of PFS,and the secondary endpoints include independent radiological central(IRC)-confirmed PFS,overall survival(OS),objective response rate(ORR),disease control rate(DCR),time to progressive disease(TTPD),duration of response(DoR),quality of life(QoL)and safety.The patients are randomized in a 1:1 ratio to receive gefitinib(250 mg,p.o.q.d.)plus apatinib(500 mg,p.o.q.d.)or gefitinib plus placebo,given until disease progression or intolerable adverse events.Exploratory biomarker analysis will be performed.This study is being conducted across China and comprises of 30 participating centers.Enrollment commenced in August 2017 and finished in December 2018,most of the patients are in the follow-up period.Anticipated outcomes and significance:The present study will be the first to evaluate the efficacy and safety profile of the combination of apatinib plus gefitinib as a first-line therapy for patients with EGFR-positive advanced non-squamous NSCLC.Importantly,this trial will provide comprehensive evidence on the treatment of EGFR-TKIs combined with antiangiogenic therapy.Zhonghan Zhang Fan Luo Yang Zhang Yuxiang Ma Shaodong Hong Yunpeng Yang Wenfeng Fang Yan Huang Li Zhang Hongyun Zhao 2019Cancer Communications2019,39,1:5
4Effects of Driving Mode on the Performance of Multiple-Chamber Piezoelectric Pumps with Multiple Actuators显示文摘Due to the limited output capability of piezoelectric diaphragm pumps, the driving voltage is frequently increased to obtain the desired output. However, the excessive voltage application may lead to a large deformation in the piezoelectric ceramics, which could cause it to breakdown or become damaged. Therefore, increasing the number of chambers to obtain the desired output is proposed. Using a check-valve quintuple-chamber pump with quintuple piezoelectric actuators, the characteristics of the pump under different driving modes are investigated through experiments. By changing the number and connection mode of working actuators, pump performances in terms of flow rate and backpressure are tested at a voltage of 150 V with a frequency range of 60 Hz ?400 Hz. Experiment results indicate that the properties of the multiple-chamber pump change significantly with distinct working chambers even though the number of pumping chambers is the same. Pump performance declines as the distance between the working actuators increases. Moreover, pump performance declines dramatically when the working piezoelectric actuator closest to the outlet is involved. The maximum backpressures of the pump with triple, quadruple, and quintuple actuators are increased by 39%, 83%, and 128%, respectively, compared with the pump with double working actuators; the corresponding maximum flow rates of the pumps are simply increased by 25.9%, 49.2%, and 67.8%, respectively. 5Identification and characterization of the cellular subclones that contribute to the pathogenesis of mantle cell lymphoma显示文摘Mantle cell lymphoma(MCL)is a B-cell malignancy with poor clinical outcome and undefined pathogenesis.Development of clinically relevant cellular models for MCL research is an urgent need.Our preliminary observations lead the development of two novel hypotheses that we tested in this study:1.multicellular spheroid might be a unique growth mode of earlystage cells in MCL;2.MCL might be a polyclonal tumor.We made the following original observations that have not been reported:First,we have provided a new experiment method for enriching MCL early-stage cells and characterized the spheroid mode of growth as a unique feature of early-stage MCL cells in cell line as well as in clinical samples.Second,we have established a clinically relevant cellular model of MCL,the JeKo-1-spheroid cell line,that was highly enriched in early-stage sub-clones.JeKo-1-spheroid cells and the spheroid growing cells enriched from MCL patients exhibited comparably enhanced tumorigenic abilities and similar biological features.Third,Immunophenotypic analysis has revealed that MCL may be derived from precursor-B(pre-B),immature-B and mature-B cells,not only the mature-B cells as WHO classified in 2016.Fourth,MCL may be a polyclonal disease composed of CD19e/IgMe,CD19 e/IgMt,CD19t/IgMt three sub-clones,of which the CD19e/IgMt sub-clone might be the dominant sub-clone with the strongest tumorigenic ability.Fifth,CD19t/IgMe that differentiates MCL and normal B cells may represent a new marker for MCL early detection,minor residual disease monitoring after therapies and prognosis.Junling Tang Li Zhang Tiejun Zhou Zhiwei Sun Liangsheng Kong Li Jing Hongyun Xing Hongyan Wu Yongli Liu Shixia Zhou Jingyuan Li Mei Chen Fang Xu Jirui Tang Tao Ma Min Hu Dan Liu Jing Guo Xiaofeng Zhu Yan Chen Ting Ye Jianyu Wang Xiaoming Li H.Rosie Xing 2019Genes & Diseases2019,6,4:2

9Effects of trace calcium and strontium on microstructure and properties of Cu-Cr alloys显示文摘Cu-0.57Cr-0.01Ca and Cu-0.58Cr-0.01Sr(wt.%)alloys were fabricated and processed by thermo-mechanical treatment.Their mechanical and electrical properties and microstructure were investigated in detail and compared with those of a Cu-0.57Cr(wt.%)alloy.The results showed that the softening resistance of the Cu-Cr alloy was significantly improved by the additions of Ca and Sr elements.Compared with the Cu-Cr alloy,the deformation microstructure of the Cu-Cr-Ca and Cu-Cr-Sr alloys was more difficult to recrystallize at elevated temperatures,and the Cr precipitates in the Cu-Cr-Ca and Cu-Cr-Sr alloys were smaller in size and had an FCC structure at any given aging state.The high strengths of the Cu-Cr-Ca and Cu-Cr-Sr alloys were mainly attributed to the dislocation strengthening provided by high-density dislocations and the precipitate strengthening provided by fine Cr precipitates.First-principles calculation showed that the segregations of Ca and Sr atoms at interface between Cr precipitates and copper matrix were favorable in energetics.This segregation effectively hindered the growth of Cr precipitates and significantly enhanced the pinning effect on the motion of dislocations and subgrain boundaries,eventually leading to the improvement in the softening resistance of the Cu-Cr alloy.Muzhi Ma Zhu Xiao Xiangpeng Meng Zhou Li Shen Gong Jie Dai Hongyun Jiang Yanbin Jiang Qian Lei Haigen Wei 2022Journal of Materials Science & Technology2022,,17:1

12Impacts of Roof/Ground Mitigation Strategies on Improving the Urban Thermal Environment and Human Comfort over the Yangtze River Delta, China显示文摘The combined effects of global warming and the urban heat islands exacerbate the risk of urban heat stress. It is crucial to implement effective cooling measures in urban areas to improve the comfort of the thermal environment. In this study, the Weather Research and Forecasting Model(WRF), coupled with a single-layer Urban Canopy Model(UCM), was used to study the impact of heat mitigation strategies. In addition, a 5-km resolution land-cover dataset for China(ChinaLC), which is based on satellite remote sensing data, was adjusted and used, and 18 groups of numerical experiments were designed, to increase the albedo and vegetation fraction of roof/ground parameters. The experiments were conducted for four heatwave events that occurred in the summer of 2013 in the Yangtze River Delta urban agglomeration of China. The simulated results demonstrated that, for the single roof/ground schemes, the mitigation effects were directly proportional to the albedo and greening. Among all the experimental schemes, the superposed schemes presented better cooling effects. For the ground greening scheme, with similar net radiation flux and latent heat flux, its storage heat was lower than that of the roof greening scheme, resulting in more energy flux into the atmosphere, and its daytime cooling effect was not as good as that of the roof greening scheme. In terms of human thermal comfort(HTC), the improvement achieved by the ground greening scheme was better than any other single roof/ground schemes, because the increase in the relative humidity was small. 13Precipitation behavior and grain refinement of burnishing Al-Zn-Mg alloy显示文摘Burnishing is a unique strengthening approach to improve the strength of surface layer and remains the ductility of the interior of metallic materials. In this work, burnishing treatment was employed to improve the surface microstructure of naturally aged Al-Zn-Mg alloys after solid solution. Transmission electron microscopy, highresolution transmission electron microscopy, X-ray diffraction and nano-indentation were used to characterize the effects of the burnishing on the microstructures of surface layer and Guinier-Preston(GP) zones. It was indicated that GP zones uniformly distributed and dispersed in the matrix before burnishing, and the amount of GP zones decreased dramatically after burnishing processing. Additionally, the grains in the surficial layer were refined into nano-crystals with an average grain size of 78 nm. Burnishing treatment not only led to formation of large number of dislocation substructures in the sub-surface and near-matrix surface, but also promoted the precipitation of metastable η' phase at grain boundaries. 18Characterization of DNA polymerase δ from deep-sea hydrothermal vent shrimp Rimicaris exoculata显示文摘DNA polymerase δ(Polδ)plays a crucial and versatile role in DNA replication and DNA repair processes.Vent shrimp Rimicaris exoculata is the primary megafaunal community living in hydrothermal vents.In this study,the Polδfrom shrimp Rimicaris exoculata was cloned,expressed and characterized.The results showed that the Polδcatalytic subunit(POLD1),852 amino acids in length,shared high homology with crayfish Procambarus clarkii and shrimp Oratosquilla oratoria.The recombinant POLD1 expressed in Escherichia coli showed that the enzyme was active in a range of 20℃ to 40℃ with an optimum temperature at 25℃ and in a wide range of p H with an optimum at pH 6.0.The activities of POLD1 were significantly enhanced in the presence of Triton-X 100,Tween 20 and Mn^(2+).The K_(m)(dNTP)value of POLD1 was 4.7μmol/L.The present study would be helpful to reveal the characterization of Polδof deep-sea vent animals.Wenlin Wu Hongyun Li Tiantian Ma Xiaobo Zhang 2021Acta Oceanologica Sinica2021,40,8:0
19Using a Particle Tracking Method to Quantify Groundwater Circulation rates: a Case Study in the Ordos Plateau显示文摘Groundwater is a valuable resource in many areas, and commonly plays a key role in economic development. Therefore, groundwater circulation rates at different depths are key issues for groundwater development and management in the Ordos Plateau. In this study, a particle tracking method was used to quantify circulation rates in the Ordos plateau. The results show that groundwater mainly circulates in shallow groundwater systems up to 400m depth.
