| 1 | Establishment of the diploid gynogenetic hybrid clonal line of red crucian carp × common carp显示文摘This study investigated the gynogenetic cytobiological behavior of the third gynogenetic generation (G3), which was generated from the diploid eggs produced by the second gynogenetic generation (G2) of red crucian carp × common carp, and determined the chromosomal numbers of G3, G2×scatter scale carp and G2×allotetraploid hybrids of red crucian carp × common carp. The results showed that the diploid eggs of G2 with 100 chromosomes, activated by UV-irradiated sperm from scatter scale carp and without the treatment for doubling the chromosomes, could develop into G3 with 100 chromosomes. Similar to the first and second gynogenetic generations (G1 and G2), G3 was also diploid (2n=100) and presented the hybrid traits. The triploids (3n=150) and tetraploids (4n=200) were produced by crossing G2 with scatter scale carp and crossing G2 with allotetraploids, respectively. The extrusion of the second polar body in the eggs of G2 ruled out the possibility that the retention of the second polar body led to the formation of the diploid eggs. In addition, we discussed the mechanism of the formation of the diploid eggs generated by G2. The establishment of the diploid gynogenesis clonal line (G1, G2 and G3) provided the evidence that the diploid eggs were able to develop into a new diploid hybrid clonal line by gynogenesis. | 3 | Responses of Patients with Disorders of Consciousness to Habit Stimulation:A Quantitative EEG Study显示文摘Whether habit stimulation is effective in DOC patient arousal has not been reported. In this paper, we analyzed the responses of DOC patients to habit stimulation. Nineteen DOC patients with alcohol consumption or smoking habits were recruited and 64-channel EEG signals were acquired both at the resting state and at three stimulation states. Wavelet transformation and nonlinear dynamics were used to extract the features of EEG signals and four brain lobes were selected to investigate the degree of EEG response to habit stimulation. Results showed that the highest degree of EEG response was from the callname stimulation, followed by habit and music stimulations. Significant differences in EEG wavelet energy and response coefficient were found both between habit and music stimulation, and between habit and call-name stimulation. | 4 | A new type of triploid crucian crap—red crucian carp (♀)×allotetraploid (♂)显示文摘A new type of triploid hybrids (TC) was produced by crossing red crucian carp (♀) with allotetraploid hybrids (♂). This type of triploids with no barbel was spindle-shaped and gray in color. Compared with their parents, some data of the morphological traits in the triploid hybrids were intermediate to those of their parents, and some were beyond their parents with heterozygous traits. Under the same culture conditions, the growth rate of the new type triploids was faster than their maternal red crucian carp (RC). The gonadal development of triploids was obviously slower than that of diploids, and the germ cells in them were degenerated. Compared with another type of triploids produced by Japanese crucian carp (♀) with allotetraploid hybrids (♂), the new type triploids not only kept faster growth rate and sterility, but also had no barbel that was similar to the common carp. | 5 | Efficacy of moxibustion by stimulating acupoints of Danzhong(CV 17) and Ganshu(BL 18) on hyperplasia of mammary gland in rats显示文摘OBJECTIVE: To evaluate the efficacy of moxibustion, through stimulating acupoints of Danzhong(CV 17) and Ganshu(BL 18) in rats with hyperplasia of mammary gland(HMG) which induced by estrogen and progestogen.METHODS: Thirty female Sprague-Dawley rats were randomly divided into saline control group,HMG model group, and HMG moxibustion group with 10 in each group. Saline control was the group injected by saline. HMG model were created by injection of estrogen and progestogen. Moxibustion group was also injected of estrogen and progesto-gen with moxibustion at the same time. The Changes of nipple diameter and height were measured.The rats' skin temperature was recorded by an infrared thermal camera at the nipples, mammary areas,Danzhong(CV 17) and Ganshu(BL 18). Pathological changes of mammary gland in rats were also observed under light microscope.RESULTS: The diameter and height of the nipples in model group were prominently bigger and higher than that in control group(P < 0.01). The diameter and height in moxibustion group were prominently smaller and lower than that in model group(P < 0.01), and there was no significant difference between moxibustion group and control group.Compared with control group, skin temperature of the nipples, mammary area, and acupoints Danzhong(CV 17) and Ganshu(BL 18) decreased prominently in model group(P < 0.01-0.05). Compared with model group, skin temperature of that in moxibustion group increased prominently(P <0.05).CONCLUSION: Treatment with moxibustion can effectively decrease the HMG rats' nipple diameter and height, and increase the skin temperature in HMG model rats at the nipples, mammary areas,Danzhong(CV 17) and Ganshu(BL 18). | 6 | On-line near-infrared spectroscopy optimizing and monitoring biotransformation process of γ-aminobutyric acid显示文摘Near-infrared spectroscopy(NIRS) with its fast and nondestructive advantages can be qualified for the real-time quantitative analysis.This paper demonstrates that NIRS combined with partial least squares(PLS) regression can be used as a rapid analytical method to simultaneously quantify L-glutamic acid(LGlu) and γ-aminobutyric acid(GABA) in a biotransformation process and to guide the optimization of production conditions when the merits of NIRS are combined with response surface methodology.The high performance liquid chromatography(HPLC) reference analysis was performed by the o-phthaldialdehyde pre-column derivatization.NIRS measurements of two batches of 141 samples were firstly analyzed by PLS with several spectral pre-processing methods.Compared with those of the HPLC reference analysis,the resulting determination coefficients(R^2),root mean square error of prediction(RMSEP) and residual predictive deviation(RPD) of the external validation for the L-Glu concentration were 99.5%,1.62 g/L,and11.3,respectively.For the GABA concentration,R^2,RMSEP,and RPD were 99.8%,4.00 g/L,and 16.4,respectively.This NIRS model was then used to optimize the biotransformation process through a BoxBehnken experimental design.Under the optimal conditions without p H adjustment,200 g/L L-Glu could be catalyzed by 7148 U/L glutamate decarboxylase(GAD) to GABA,reaching 99% conversion at the fifth hour.NIRS analysis provided timely information on the conversion from L-Glu to GABA.The results suggest that the NIRS model can not only be used for the routine profiling of enzymatic conversion,providing a simple and effective method of monitoring the biotransformation process of GABA,but also be considered to be an optimal tool to guide the optimization of production conditions. | Guoyu Ding Yuanyuan Hou Jiamin Peng Yunbing Shen Min Jiang Gang Bai | 2016 | Journal of Pharmaceutical Analysis2016,6,3: | 2 |

| 10 | Glycolysis and gluconeogenesis are involved of glucose metabolism adaptation during fasting and re-feeding in black carp(Mylopharyngodon piceus)显示文摘Both in nature and in aquaculture,fish may experience periods of food scarcity or hunger.The metabolic regulation of fish when nutritional state changes is a complex process that involves many factors.To study glucose metabolism adaptability during fasting and re-feeding in the black carp(Mylopharyngodon piceus),we measured changes in some biochemical indicators related to glucose metabolism.Five fish were sampled on days 0,1,3,5,and 10 of fasting(F,S1,S3,S5,and S10,respectively)and days 1,3,and 5 of re-feeding(RF1,RF3,and RF5,respectively).The serum glucose concentration decreased significantly at S1,reached the lowest point at S10,and increased significantly at RF1(P<0.05).The concentration of liver glycogen decreased significantly at S1 and reached the lowest level at S3,whereas the muscle glycogen level decreased significantly at S5 and reached the lowest value at S10(P<0.05).Both liver and muscle glycogen levels returned to the pre-fasting level at RF5(P<0.05).Regarding glycolysis,the concentrations of pyruvate kinase(PK)and hexokinase(HK)decreased significantly at S5 and increased significantly at RF5 and RF1,respectively(P<0.05).The concentrations of glucokinase(GCK)and insulin decreased significantly at S1 and increased significantly at RF1 and RF3,respectively(P<0.05).The mRNA expression levels of liver GCK and glucose transporter 2(GLUT2)decreased significantly at S1 and increased significantly at RF1 and RF5,respectively(P<0.05).As for gluconeogenesis,the concentration of glucose-6-phosphatase(G6PC)increased significantly at S1 and decreased significantly at RF1(P<0.05).The concentrations of glucagon and glucocorticoid(GC)increased significantly at S3 and significantly decreased at RF1 and RF5,respectively(P<0.05).The mRNA expression levels of liver G6PC and phosphoenolpyruvate carboxykinase(PEPCK)increased significantly at S3 and S1,and both decreased significantly at RF1(P<0.05).These results indicate that coordination between glycolysis and gluconeogenesis might be crucial for glucose homeostasis during fasting and re-feeding in the black carp. | Yafan Dai Yubang Shen Jiamin Guo Hao Yang Feng Chen Wei Zhang Wenhui Wu Xiaoyan Xu Jiale Li | 2024 | Aquaculture and Fisheries2024,9,2: | 0 |

| 13 | A core-satellite-like nanoassembly reverses a decisive tyrosine hydroxylase loss in degenerative dopaminergic neurons显示文摘In recent years,neurodegenerative diseases,such as Parkinson’s or Alzheimer’s diseases,are rapidly rising in prevalence.The main hallmark of Parkinson’s disease is the falling levels of neurotransmitter dopamine in the mid-brain with dopaminergic neurons losing.Typical therapeutic solutions,including drugs,deep brain stimulation,and cell transplantation,can only alleviate the symptoms of Parkinson’s disease.It is a tremendous challenge to reverse the function degeneration of the crucial dopaminergic neurons.Herein,we develop a core-satellite-like nanoassembly(PDA-AFn(by integrating polydopamine nanoparticles and apoferritin))to raise the expression of tyrosine hydroxylase(TH),a rate-limiting enzyme in the formation of the dopamine.Both components in the nanoassembly could cooperate with each other,not only elaborately regulate the iron homeostasis and redox microenvironment,but also utilize excessive reactive oxygen species(ROS)and iron ions in the damaged neurons to supply extra dopamine and enhance TH activity,and consequently restore the function of the degenerated neurons.Remarkably,the nanoassembly-treatment relieves the dyskinesia and dramatical increases the tyrosine hydroxylase and dopamine level in the midbrain of Parkinson’s disease model mice.It is an explicit yet inspiring advance in treatment of the neurodegeneration. | Ke Yao Jiamin Gan Di Zhao Mingding Li Xiaoquan Shen Yumin Yang Peijian Feng Qundong Shen | 2023 | Nano Research2023,16,7: | 0 |
