1Prognostic value of programmed death.1, programmed death-ligand 1, programmed death-ligand 2 expression, and CD8(+) T cell density in primary tumors and metastatic lymph nodes from patients with stage T1.4N+M0 gastric adenocarcinoma显示文摘Background: Anti-programmed death-1/programmed death-ligand 1(PD-1/PD-L1) immunotherapy has been proved to be effective on gastric cancer in ongoing clinical trials. However, the value of PD-L1 in predicting responses of patients with gastric cancer to anti-PD-1/PD-L1 immunotherapy is controversial. Some studies suggested that intra-and inter-tumoral heterogeneity of PD-L1 expression might explain the controversy.This study aimed to analyze the expression of PD-L1, PD-L2, and PD-1 as well as CD8(+) T-cell density in primary tumors and lymph nodes from patients with stage T1-4 N+M0 gastric adenocarcinoma to explore the heterogeneity of PD-1 signaling pathway molecules.Methods: In primary tumors and metastatic as well as non-metastatic lymph nodes from patients with stage T1-4 N+M0 gastric adenocarcinoma, we detected PD-L1 and PD-L2 expression with immunohistochemistry. CD8(+)T-cell density in primary tumors and PD-1 expression on CD8(+)T cells were detected with immunofluorescence. Univariate analysis was used to determine the prognostic values of them. Cox proportional hazard regression model was used to identify independent risk factors that affect patients' overall survival and disease-free survival.Results: Among 119 eligible patients who had undergone surgical resection, the positive rate of PD-L1 was higher in metastatic lymph nodes than in primary tumors(45.4% vs. 38.7%, P = 0.005); the positive rate of PD-1 on CD8(+)T cells was significantly higher in primary tumors and metastatic lymph nodes than in tumor-free lymph nodes(both P < 0.001). The intensity of PD-1 expression on CD8(+) T cells in primary tumors and in metastatic lymph nodes were stronger than that in tumor-free lymph nodes from the same patient. Beside, the positive rate of PD-L2 did not show any differences between primary tumors and metastatic lymph nodes. 3Sensitivity Experiments on the Poleward Shift of Tropical Cyclones over the Western North Pacific under Warming Ocean Conditions显示文摘Recent studies found that in the context of global warming, the observed tropical cyclones(TCs) exhibit significant poleward migration trend in terms of the mean latitude where TCs reach their lifetime-maximum intensity in the western North Pacific(WNP). This poleward migration of TC tracks can be attributed to not only anthropogenic forcing(e.g., continuous increase of sea surface temperature(SST)), but also impacts of other factors(e.g., natural variability). In the present study, to eliminate the impacts of other factors and thus focus on the impact of unvaried SST on climatological WNP TC tracks, the mesoscale Weather Research and Forecasting(WRF) model is used to conduct a suite of idealized sensitivity experiments with increased SST. Comparisons among the results of these experiments show the possible changes in climatological TC track, TC track density, and types of TC track in the context of SST increase. The results demonstrate that under the warmer SST conditions, the climatological mean TC track systematically shifts poleward significantly in the WNP, which is consistent with the previous studies. Meanwhile, the ocean warming also leads to the decreased(increased) destructive potential of TCs in low(middle) latitudes, and thus northward migration of the region where TCs have the largest impact. 4A Multifunctional Anti-Proton Electrolyte for High-Rate and Super-Stable Aqueous Zn-Vanadium Oxide Battery显示文摘Large volumetric expansion of cathode hosts and sluggish transport kinetics in the cathode–electrolyte interface,as well as dendrite growth and hydrogen evolution at Zn anode side are considered as the system problems that cause the electrochemical failure of aqueous Zn-vanadium oxide battery.In this work,a multifunctional anti-proton electrolyte was proposed to synchronously solve all those issues.Theoretical and experimental studies confirm that PEG 400 additive can regulate the Zn^(2+) solvation structure and inhibit the ionization of free water molecules of the electrolyte.Then,smaller lattice expansion of vanadium oxide hosts and less associated by-product formation can be realized by using such electrolyte.Besides,such electrolyte is also beneficial to guide the uniform Zn deposition and suppress the side reaction of hydrogen evolution.Owing to the integrated synergetic modifica-tion,a high-rate and ultrastable aqueous Zn-V_(2)O_(3)/C battery can be constructed,which can remain a specific capacity of 222.8 m Ah g^(-1)after 6000 cycles at 5 A g^(-1),and 121.8 m Ah g^(-1) even after 18,000 cycles at 20 A g^(-1),respectively.Such“all-in-one”solution based on the electrolyte design provides a new strategy for developing high-performance aqueous Zn-ion battery.Yangwu Chen Dingtao Ma Kefeng Ouyang Ming Yang Sicheng Shen Yanyi Wang Hongwei Mi Lingna Sun Chuanxin He Peixin Zhang 2022Nano-Micro Letters2022,14,9:3
5Impact of Initial Storm Intensity and Size on the Simulation of Tropical Cyclone Track and Western Pacific Subtropical High Extent显示文摘Typhoon Megi, the 13 th typhoon of the 2010 typhoon season, was selected for case study by utilizing the Weather Research and Forecasting(WRF) model. Twelve sensitivity experiments with various initial tropical cyclone(TC) intensities and sizes were conducted to investigate their impacts on the simulation of typhoon track. Interaction between TC and the western Pacific subtropical high(WPSH) was also analyzed to explore the mechanism for the impact on TC track of the initial TC intensity and size. Numerical results indicate that the simulated TC size and TC track are sensitive to initial TC intensity and size. Stronger initial TC intensity and larger initial TC size often lead to larger simulated TC size and make TC turn northward earlier. Further analysis suggests that, with the increase of initial TC intensity and size, more air mass enters into the TC region, which subsequently reduces the extent of WPSH.As a result, the steering flow changes significantly and eventually causes the TC to turn northward earlier. 10Junction plakoglobin regulates and destabilizes HIF2α to inhibit tumorigenesis of renal cell carcinoma显示文摘Background:Increased hypoxia-inducible factor 2α(HIF2α)activation is a common event in clear cell renal cell carcinoma(ccRCC)progression.However,the function and underlying mechanism of HIF2α in ccRCC remains uninvestigated.We conducted this study to access the potential link between junction plakoglobin(JUP)and HIF2αin ccRCC.Methods:Affinity purification and mass spectrometry(AP-MS)screening,glutathione-s-transferase(GST)pull-down and co-immunoprecipitation(Co-IP)assays were performed to detect the interacting proteins of HIF2α.Quantitative PCR(qPCR)and Western blotting were used to detect the expression of JUP in human ccRCC samples.Luciferase reporter assays,chromatin immunoprecipitation(ChIP),cycloheximide chase assays,and ubiquitination assays were conducted to explore the regulation of JUP on the activity of HIF2α.Cell Counting Kit-8(CCK-8)assays,colony formation assays,transwell assays,and xenograft tumor assays were performed to investigate the effect of JUP knockdown or overexpression on the tumorigenicity of renal cancer cells.Results:We identified JUP as a novel HIF2α-binding partner and revealed an important role of JUP in recruiting von Hippel-Lindau(VHL)and histone deacetylases 1/2(HDAC1/2)to HIF2α to regulate its stability and transactivation.JUP knockdown promoted and overexpression suppressed the tumorigenicity of renal cell carcinoma in vitro and in vivo.Importantly,the low expression of JUP was found in clinical ccRCC samples and correlated with enhanced hypoxia scores and poor treatment outcomes.Conclusion:Taken together,these data support a role of JUP in modulating HIF2α signaling during ccRCC progression and identify JUP as a potential therapeutic target.Ke Chen Jin Zeng Yi Sun Wei Ouyang Gan Yu Hui Zhou Yangjun Zhang Weimin Yao Wei Xiao Junhui Hu Jinchun Xing Kefeng Xiao Lily Wu Zhiqiang Chen Zhangqun Ye Hua Xu 2021Cancer Communications2021,41,4:1
