| 1 | Newborn screening with targeted sequencing:a multicenter investigation and a pilot clinical study in China显示文摘Different newborn screening(NBS) programs have been practiced in many countries since the 1960 s. It is of considerable interest whether next-generation sequencing is applicable in NBS. We have developed a panel of 465 causative genes for 596 early-onset, relatively high incidence, and potentially actionable severe inherited diseases in our Newborn Screening with Targeted Sequencing(NESTS) program to screen 11,484 babies in 8 Women and Children’s hospitals nationwide in China retrospectively. The positive rate from preliminary screening of NESTS was 7.85%(902/11,484). With 45.89%(414/902) follow-up of preliminary positive cases, the overall clinically confirmative diagnosis rate of monogenic disorders was 12.07%(50/414), estimating an average of 0.95%(7.85% × 12.07%) clinical diagnosis rate, suggesting that monogenic disorders account for a considerable proportion of birth defects. The disease/gene spectrum varied in different regions of China. NESTS was implemented in a hospital by screening 3923 newborns to evaluate its clinical application. The turn-around time of a primary report, including the sequencing period of < 7 days, was within 11 days by our automatic interpretation pipeline. | 3 | Hydrochemical Dynamic Characteristics and Evolution of Underground Brine in the Mahai Salt Lake of the Qaidam Basin Qinghai-Tibet Plateau显示文摘The mineral rock salts present in the Mahai Salt Lake of the Qaidam basin exhibit high solubilities in water. In addition, the multicomponent underground brine exhibits a high salinity and is easily precipitated. In the natural state, brine transport in the brine layer is extremely slow, and the brine is in a relatively stable chemical equilibrium state with the rock salt media. However, during mining, both the seepage and the chemical fields fluctuate significantly, thereby disrupting the equilibrium and leading to variations in the chemical composition and dynamic characteristics of the brine. Therefore, we selected underground brine from the Mahai Salt Lake, collecting a total of 183 brine samples over three stages of mining(i.e., the early stage of underground brine extraction, the initial stage of mining, and the later stage of mining). Using a range of analytical techniques, the chemical dynamics of the underground brine water and its evolution were systematically studied. | 6 | Porosity Distribution Characteristics and Geological Analysis of Brine Storage Medium in the Qaidam Basin, Western China显示文摘Underground brine is an unusual water resource that contains abundant mineral resources. It is distributed widely in the Qaidam Basin, western China, a hyperarid inland basin located in the northern Tibetan Plateau. Pores in the brine storage medium act as storage space and transmission channels of underground brine. Therefore, the porosity of brine storage medium determines its ability to store brine. In this study, Mahai Salt Lake was used as the research area as a modern saline lake located in the north area of the Qaidam Basin. A total of 100 porosity samples were collected from eight sampling points in two profiles of the research area at sampling depths of 1.30–314.78 m. The porosity distribution characteristics and influencing factors in brine storage medium were analysed according to the measured porosity data. Based on analysis of the pore structure characteristics, the brine storage medium contains intercrystalline pores, unlike conventional freshwater storage mediums. Moreover, the primary salt rock is susceptible to dissolution by lighter brine, facilitating the formation of secondary porosity. Due to the formation of secondary pores, a porosity greater than 20% remains even at buried depths greater than 100 m. Based on the geological statistical analysis, due to the geographic location, salt formation time, and depositional environment, the porosity values of Mahai Salt Lake do not exhibit a wider distribution, but also show more extreme values than a nearby salt lake. | 7 | Effects of Guangxi Hepu Pearl Hydrolysate on Proliferation Activity and Apoptosis of Human Hepatic Stellate Cells显示文摘[Objective] The paper was to observe the effects of Guangxi Hepu pearl hydrolysate on proliferation and apoptosis of human hepatic stellate cells. [Method] The inhibition rate against the proliferation of human hepatic stellate cells was determined by MTT colorimetry after co-incubation with pearl hydrolysate for 48 h. The percentage of early and late apoptotic cells was determined by flow cytometry(Annexin V-FITC/PI staining). [Result] Low, medium and high concentrations of pearl hydrolysate could inhibit the proliferation of human hepatic stellate cells, and the inhibition rates at 48 h were 12.4%, 27.4% and 37.8%, respectively. The low, medium and high concentrations of pearl hydrolysate could induce the apoptosis of cells. The percentages of total apoptotic cells(early apoptotic + late apoptotic) at 48 h were 8.97%, 16.09% and 26.98%, respec-tively. [Conclusion] Pearl hydrolysate can inhibit the proliferation of human hepatic stellate cells and induce the apoptosis of these cells, which may be the mechanism of anti-hepatic fibrosis effect of pearl hydrolysate. These effects are dose-dependent. | 15 | Robust model for tunnel squeezing using Bayesian optimized classifiers with partially missing database显示文摘Accurately predicting and estimating the squeezing and ground response to tunneling remains challenging.Moreover,tunnel-squeezing hazards are much more likely to occur in deeply buried long tunnels with complex engineering-geological environments.There-fore,a high-performance predictive model for tunnel squeezing is necessary.A superior ensemble classifier is put forward in this study,which is composed of four individual classifiers(gradient boosting classifier,extra-trees classifier,AdaBoost classifier,and Logistic regression classifier)and two optimization algorithms(Bayesian optimization(BO)and sparrow search algorithm(SSA)).The training database covers five parameters:tunnel depth(H),rock tunneling quality index(Q),tunnel diameter(D),support stiffness(K),and strength stress ratio(SSR),about which the basic information is accessible at the early design phases.However,the dataset compiled from the literature is insufficient.Thus,the ten proposed methods are used to replace the missing values.During the model training pro-cess,BO shows its strong ability to optimize seventeen hyperparameters.When applied to tune the classifiers’weights,SSA achieves a fast and efficient performance.The novel Shapley Additive Explanations–LightGBM method indicates that the K is the most important input feature,followed by SSR,Q,H,and D,respectively.The ensemble classifier is then validated using the test set and additional his-torical case projects.The validation shows that the model can achieve an accuracy of 98%(i.e.,the error rate is 2%)on the test set,higher than those achieved by previous prediction models.Moreover,the predicted probability could provide warning information for timely support measures.Finally,the application results are illustrated through tests on the tunnel sections that have not yet been excavated in the line of the Sichuan–Tibet railway project.The applied predictive tendencies and laws are in line with the practical experience.In sum-mary,the proposed model’s prediction results are reasonable,and its prediction will be more accurate as more data is collected and trained for prewarning the tunnel squeezing hazard. | Yin Bo Xing Huang Yucong Pan Yanfang Feng Penghai Deng Feng Gao Ping Liu Quansheng Liu | 2023 | Underground Space2023,,3: | 0 |

| 17 | Compressive strength prediction of sprayed concrete lining in tunnel engineering using hybrid machine learning techniques显示文摘Sprayed concrete lining is a commonly employed support measure in tunnel engineering,which plays an important role in construction safety.Compressive strength is a key performance indicator of sprayed concrete lining,and the traditional measuring method is time-consuming and laborious.This paper proposes various hybrid machine learning algorithms to accomplish the advanced prediction of compressive strength of sprayed concrete lining based on the mixture design.Two hundred and five sets of experimental data were collected from a water conveyance tunnel in northwestern China for model construction,and each set of data was made up of six basic input variables(i.e.,water,cement,mineral powder,superplasticizer,coarse aggregate,and fine aggregate)and one output variable(i.e.,compressive strength).In order to eliminate the correlation between input variables,a new composite indicator(i.e.,the water-binder ratio)was introduced to achieve dimensionality reduction.After that,four hybrid models in total were built,namely BPNN-QPSO,SVR-QPSO,ELM-QPSO,and RF-QPSO,where the hyper-parameters of BPNN,SVR,ELM,and RF were auto-tuned by QPSO.Engineering application results indicated that RF-QPSO achieved the lowest mean absolute percentage error(MAPE)of 3.47% and root mean square error(RMSE)of 1.30 and the highest determination coefficient(R^(2))of 0.93 in the four hybrid models.Moreover,RFQPSO had the shortest running time of 0.15 s,followed by SVR-QPSO(0.18 s),ELM-QPSO(1.19 s),and BPNN-QPSO(1.58 s).Compared with BPNN-QPSO,SVR-QPSO,and ELM-QPSO,RF-QPSO performed the most superior performance in terms of both prediction accuracy and running speed.Finally,the importance of input variables on the model performance was quantitatively evaluated,further enhancing the interpretability of RF-QPSO. | Xin Yin Feng Gao Jian Wu Xing Huang Yucong Pan Quansheng Liu | 2022 | Underground Space2022,7,5: | 0 |
