| 4 | Small changes in the length of diselenide bond-containing linkages exert great influences on the antitumor activity of docetaxel homodimeric prodrug nanoassemblies显示文摘Homodimeric prodrug-based self-assembled nanoparticles,with carrier-free structure and ultrahigh drug loading,is drawing more and more attentions.Homodimeric prodrugs are composed of two drug molecules and a pivotal linkage.The influence of the linkages on the self-assembly,in vivo fate and antitumor activity of homodimeric prodrugs is the focus of research.Herein,three docetaxel(DTX)homodimeric prodrugs are developed using different lengths of diselenide bond-containing linkages.Interestingly,compared with the other two linkages,the longest diselenide bond-containing linkage could facilitate the self-delivery of DTX prodrugs,thus improving the stability,circulation time and tumor targeting of prodrug nanoassemblies.Besides,the extension of linkages reduces the redox-triggered drug release and cytotoxicity of prodrug nanoassemblies in tumor cells.Although the longest diselenide bond-containing prodrug nanoassemblies possessed the lowest cytotoxicity to 4T1 cells,their stable nanostructure maintained intact during circulation and achieve the maximum accumulation of DTX in tumor cells,which finally“turned the table”.Our study illustrates the crucial role of linkages in homodimeric prodrugs,and gives valuable proposal for the development of advanced nano-DDS for cancer treatment. | Lingxiao Li Shiyi Zuo Fudan Dong Tian Liu Yanlin Gao Yinxian Yang Xin Wang Jin Sun Bingjun Sun Zhonggui He | 2021 | Asian Journal of Pharmaceutical Sciences2021,16,3: | 3 |
| 5 | Temporal and spatial variations of net primary productivity and its response to groundwater of a typical oasis in the Tarim Basin, China显示文摘Net primary productivity (NPP) of the vegetation in an oasis can reflect the productivity capacity of a plant community under natural environmental conditions. Owing to the extreme arid climate conditions and scarce precipitation in the arid oasis regions, groundwater plays a key role in restricting the development of the vegetation. The Qira Oasis is located on the southern margin of the Taklimakan Desert (Tarim Basin, China) that is one of the most vulnerable regions regarding vegetation growth and water scarcity in the world. Based on remote sensing images of the Qira Oasis and daily meteorological data measured by the ground stations during the period 2006-2019, this study analyzed the temporal and spatial patterns of NPP in the oasis as well as its relation with the variation of groundwater depth using a modified Carnegie Ames Stanford Approach (CASA) model. At the spatial scale, NPP of the vegetation decreased from the interior of the Qira Oasis to the margin;at the temporal scale, NPP of the vegetation in the oasis fluctuated significantly (ranging from 29.80 to 50.07 g C/(m2•month)) but generally showed an increasing trend, with the average increase rate of 0.07 g C/(m2•month). The regions with decreasing NPP occupied 64% of the total area of the oasis. During the study period, NPP of both farmland and grassland showed an increasing trend, while that of forest showed a decreasing trend. The depth of groundwater was deep in the south of the oasis and shallow in the north, showing a gradual increasing trend from south to north. Groundwater, as one of the key factors in the surface change and evolution of the arid oasis, determines the succession direction of the vegetation in the Qira Oasis. With the increase of groundwater depth, grassland coverage and vegetation NPP decreased. During the period 2008-2015, with the recovery of groundwater level, NPP values of all types of vegetation with different coverages increased. | 6 | Resequencing of sweetpotato germplasm resources reveals key loci associated with multiple agronomic traits显示文摘Sweetpotato is an important crop that exhibits hexaploidy and high heterozygosity,which limits gene mining for important agronomic traits.Here,314 sweetpotato germplasm resources were deeply resequenced,and 4599509 SNPs and 846654 InDels were generated,among which 196124 SNPs were nonsynonymous and 9690 InDels were frameshifted.Based on the Indels,genome-wide marker primers were designed,and 3219 of 40366 primer pairs were selected to construct the core InDel marker set.The molecular ID of 104 sweetpotato samples verified the availability of these primers.The sweetpotato population structures were then assessed through multiple approaches using SNPs,and diverse approaches demonstrated that population stratification was not obvious for most Chinese germplasm resources.As many as 20 important agronomic traits were evaluated,and a genome-wide association study was conducted on these traits.A total of 19 high-confidence loci were detected in both models.These loci included several candidate genes,such as IbMYB1,IbZEP1,and IbYABBY1,which might be involved in anthocyanin metabolism,carotenoid metabolism,and leaf morphogenesis,respectively.Among them,IbZEP1 and IbYABBY1 were first reported in sweetpotato.The variants in the promoter and the expression levels of IbZEP1 were significantly correlated with f lesh color(orange or not orange)in sweetpotato.The expression levels of IbYABBY1 were also correlated with leaf shape.These results will assist in genetic and breeding studies in sweetpotato. | Shizhuo Xiao Xibin Dai Lingxiao Zhao Zhilin Zhou Lukuan Zhao Pan Xu Bingqian Gao An Zhang Donglan Zhao Rui Yuan Yao Wang Jie Wang Qinglian Li Qinghe Cao | 2023 | Horticulture Research2023,10,1: | 1 |

| 16 | Synergetic lethal energy depletion initiated by cancer cell membrane camouflaged nano-inhibitor for cancer therapy显示文摘Mitochondrial bioenergy plays a vital role in the occurrence and development of cancer.Although strategies to impede mitochondrial energy supply have been rapidly developed,the anticancer efficacy is still far from satisfactory,mainly attributed to the hybrid metabolic pathways of mitochondrial oxidative phosphorylation(OXPHOS)and glycolysis.Herein,we construct a cancer cell membrane camouflaged nano-inhibitor,mTPPa-Sy nanoparticle(NP),which co-encapsulates OXPHOS inhibitor(mitochondrial-targeting photosensitizers:TPPa)and glycolysis inhibitor(syrosingopine(Sy))for synergistically blocking the two different energy pathways.The mTPPa-Sy NPs exhibit precision tumor-targeting due to the high affinity between the biomimic membrane and the homotypic cancer cells.Under laser irradiation,the mitochondrial-targeting TPPa,which is synthesized by conjugating pyropheophorbide a(PPa)with triphenylphosphin,produces excessive reactive oxygen species(ROS)and further disrupts the OXPHOS.Interestingly,OXPHOS inhibition reduces O_(2) consumption and improves ROS production,further constructing a closed-loop OXPHOS inhibition system.Moreover,TPPa-initiated OXPHOS inhibition in combination with the Sytriggered glycolysis inhibition results in lethal energy depletion,significantly suppressing tumor growth even after a single treatment.Our findings highlight the necessity and effectiveness of synergetic lethal energy depletion,providing a prospective strategy for efficient cancer therapy. | Fudan Dong Qikun Jiang Lingxiao Li Tian Liu Shiyi Zuo Lin Gao Mengna Fang Yanlin Gao Bingjun Sun Cong Luo Zhonggui He Jin Sun | 2022 | Nano Research2022,15,4: | 0 |
