| 1 | Hi-TOM: a platform for high-throughput tracking of mutations induced by CRISPR/Cas systems显示文摘The CRISPR/Cas system has been extensively applied to make precise genetic modifications in various organisms. Despite its importance and widespread use, large-scale mutation screening remains time-consuming, labour-intensive and costly. Here, we developed Hi-TOM(available at http://www.hi-tom.net/hi-tom/), an online tool to track the mutations with precise percentage for multiple samples and multiple target sites. We also described a corresponding next-generation sequencing(NGS) library construction strategy by fixing the bridge sequences and barcoding primers. Analysis of the samples from rice, hexaploid wheat and human cells reveals that the Hi-TOM tool has high reliability and sensitivity in tracking various mutations, especially complex chimeric mutations frequently induced by genome editing. Hi-TOM does not require special design of barcode primers,cumbersome parameter configuration or additional data analysis. | 6 | Uncompromised NK cell activation is essential for virus-specific CTL activity during acute influenza virus infection显示文摘Natural killer (NK) cells are indispensable components of both the innate and adaptive immune response. However,their precise roles in the cross-talk between innate and adaptive immunity during influenza virus infection remaincontroversial. By comparing NK cell dynamics and activity under a sub-lethal dose and high dose of influenza virusinfection, we showed that influenza virus PR8 directly infected NK cells during natural infection, which wasconsistent with our previous findings obtained from an in vitro investigation of human NK cells. The impairments incytotoxicity and IFN-γ production by spleen NK cells following high-dose infection were accompanied by decreasedvirus-specific killing mediated by cytotoxic T lymphocytes (CTLs). Importantly, the weakened CTL activity could bereversed by adoptive transfer of spleen NK cells harvested from low-dose-infected mice but not healthy donors.Taken together, our data provide direct evidence supporting the contribution of NK cells to antiviral T-cellresponses. | 9 | Avian influenza virus directly infects human natural killer cells and inhibits cell activity显示文摘Natural killer(NK) cell is a key component of innate immunity and plays an important role in host defense against virus infection by directly destroying infected cells. Influenza is a respiratory disease transmitted in the early phase of virus infection. Evasion of host innate immunity including NK cells is critical for the virus to expand and establish a successful acute infection. Previously,we showed that human influenza H1N1 virus infects NK cells and induces cell apoptosis, as well as inhibits NK cell activity. In this study, we further demonstrated that avian influenza virus also directly targeted NK cells as an immunoevasion strategy. The avian virus infected human NK cells and induced cell apoptosis. In addition, avian influenza virion and HA protein inhibited NK cell cytotoxicity. This novel strategy has obvious advantages for avian influenza virus, allowing the virus sufficient time to expand and subsequent spread before the onset of the specific immune response. | 10 | Nutrient resorption strategies of three oak tree species in response to interannual climate variability显示文摘Background:Nutrient resorption is critical for plants toward balancing their nutritional requirements and adapting to environmental variabilities,which further impacts litter quality and nutrient cycling.However,the interannual variability of nutrient resorption under climate change remains unclear.Methods:We investigated the five-year nutrient resorption efficiencies(NuRE,%)of 14 elements in three deciduous oak tree species(Quercus aliena var.acuteserrata,Q.glandulifera,and Q.variabilis)in a warm-temperate forest of Central China and assessed their relationships with interannual climate and soil factors.Results:Nutrient resorption did not differ between species but varied significantly between different years.For each year,N,P,S,K,C,Mg,and Zn were preferentially resorbed in all of the oak species in contrast to Ca,Na,Mn,Ba,Al,Fe,Cu,which were to some extent discriminated.Among the 14 elements,the NuRE of C,N,P,S,Ca,and Mg was more sensitive to interannual climate variations in the three oak species.The carbon resorption efficiency was significantly increased during the driest year of the study(2014);N resorption efficiency was reduced with temperature;whereas N and P resorption efficiency initially decreased and then increased with precipitation.Moreover,the elements with higher NuREs typically had lower coefficient of variation(CV)in all three oak species.Conclusions:Different oak species exhibited analogous nutrient conservation strategies in response to annual climate variabilities,and interannual climate variations strongly impacted plant nutrient resorption.Deciduous plants may establish a tradeoff mechanism to rebalance somatic nutrients for regrowth at the end of the growing season. | Baoming Du Huawei Ji Shirong Liu Hongzhang Kang Shan Yin Chunjiang Liu | 2021 | Forest Ecosystems2021,8,4: | 4 |

Springback characteristics of unannealed and annealed TRBs were investigated by means of numerical simulation and experiments taking U-Channel as an example. TRBs were annealed by the annealing process (700 ℃, holding time 10 h), then stamping and springback processes of unannealed and annealed TRBs were simulated, and corresponding experiments were also carried out. Effects of the transition zone length, the blank thickness, the friction coeffcient and the die clearance on the springback of TRB were analyzed. The results demonstrate that the springback of TRB annealed at 700 ℃ for 10 h reduces significantly. For unannealed and annealed U-Channels, the springback of TRB U-Channel is in direct proportion to the die clearance and is in inverse proportion to the transition zone length, the blank thickness and the friction coeffcient. | 16 | The Effect of 2,5-hexanedione on Myelin Protein Zero Expression,and Its Mitigation Using Ginkgo Biloba Extract显示文摘Objective To investigate the role of myelin protein zero (P 0) in 2,5-hexanedione (2,5-HD)-induced peripheral nerve injury,and the protective effect of Ginkgo biloba extract (Egb761) on 2,5-HD-induced toxic peripheral neuropathy.Methods After 4 weeks of treatment with 2,5-HD at different doses (50,100,200,400 mg/kg) in rats,changes in the levels of P 0 in rat sciatic nerves was investigated,and the effect of Egb761 on 2,5-HD-induced toxic peripheral neuropathy was studied.Results The blood-nerve barrier (BNB) permeability of the sciatic nerve increased,and the expression of P 0 mRNA and P 0 protein decreased in a dose-dependent manner after treatment with 2,5-HD for 4 weeks.Pretreatment with Egb761 protected against BNB interruption,and inhibited P 0 mRNA and protein reduction during 2,5-HD treatment.Pretreatment with Egb761 significantly reduced loss of body weight (P<0.01) and mitigated gait abnormalities (2.85±0.22) induced by 400 mg/kg 2,5-HD (P<0.01).It also reduced the signs of neurotoxicity induced by 2,5-HD.Conclusion 2,5-HD inhibited the expression of P 0 in a dose-dependent manner,and this may be an important mechanism by which toxic peripheral neuropathy is induced by 2,5-HD.Egb761 has a protective effect against 2,5-HD-induced peripheral neurotoxicity in rats. | ZHAO Lei LIU QingJun CHEN Hong DUAN HuaWei BIN Ping LIU Qing NIU Yong DAI YuFei ZHENG YuXin | 2011 | Biomedical and Environmental Sciences2011,24,4: | 3 |
| 17 | Ambient TSP concentration and dustfall variation in Urumqi,China显示文摘Total suspended particulate(TSP, particle diameter≤100 μm) was the dominant air pollutant and significantly influenced local air quality. In this paper, we investigated the interannual and seasonal variations of TSP and dustfall of the atmosphere over Urumqi, the capital city of Xinjiang Uygur autonomous region, northwestern China, basing on environmental monitoring records and meteorological data from 1986–2012. The results showed that during the study period, annual average TSP concentration decreased from 716 to 260 μg/m3, with an average level of 422.9 μg/m3, while dustfall intensity reduced from 350.4 to 166.6 t/(km2?a), with an average level of 259 t/(km2?a). Over 50% of the annual pollution days were induced mainly by TSP. Spring and winter had relatively higher dustfall intensities, and dense traffic and residential areas had the highest dustfall intensities in the Urumqi metropolitan area. With an annual average precipitation of less than 300 mm, atmospheric particulates in Urumqi could be hardly removed through wet deposition. During spring and summertime prevailing winds from northwest and northeast could carry aeolian dust particles from sandy deserts to Urumqi. | 18 | QTL mapping of drought tolerance traits in soybean with SLAF sequencing显示文摘Drought stress is an important factor affecting soybean yield.Improving drought tolerance of soybean varieties can increase yield and yield stability when the stress occurs.Identifying QTL related to drought tolerance using molecular marker-assisted selection is able to facilitate the development of drought-tolerant soybean varieties.In this study,we used a high-yielding and drought-sensitive cultivar‘Zhonghuang 35’and a drought-tolerant cultivar‘Jindou 21’to establish F6:9 recombinant inbred lines.We constructed a highdensity genetic map using specific locus amplified fragment sequencing(SLAF-Seq)technology.The genetic map contained 8078 SLAF markers distributing across 20 soybean chromosomes with a total genetic distance of 3780.98 c M and an average genetic distance of0.59 c M between adjacent markers.Two treatments(irrigation and drought)were used in the field tests,the Additive-Inclusive Composite Interval Mapping(ICIM-ADD)was used to call QTL,and plant height and seed weight per plant were used as the indicators of drought tolerance.We identified a total of 23 QTL related to drought tolerance.Among them,seven QTL(q PH2,q PH6,q PH7,q PH17,q PH19-1,q PH19-2,and q PH19-3)on chromosomes 2,6,7,17,and 19 were related to plant height,and five QTL(q SWPP2,q SWPP6,q SWPP13,q SWPP17,and q SWPP19)on chromosomes 2,6,13,17,and 19 were related to seed weight and could be considered as the major QTL.In addition,three common QTL(q PH6/q SWPP6,q PH17/q SWPP17,and q PH19-3/q SWPP19)for both plant height and seed weight per plant were located in the same genomic regions on the same chromosomes.Three(q PH2,q PH17,and q PH19-2)and four novel QTL(q SWPP2,q SWPP13,q SWPP17,and q SWPP19)were identified for plant height and seed weight per plant,respectively.Two pairs of QTL(q PH2/q SWPP2 and q PH17/q SWPP17)were also common for both plant height and seed weight per plant.These QTL and closely linked SLAF markers could be used to accelerate breeding for drought tolerant cultivars via MAS. | Honglei Ren Jianan Han Xingrong Wang Bo Zhang Lili Yu Huawei Gao Huilong Hong Rujian Sun Yu Tian Xusheng Qi Zhangxiong Liu Xiaoxia Wu Li-Juan Qiu | 2020 | The Crop Journal2020,8,6: | 2 |
| 19 | Protective effects and potential underlying mechanisms of sodium copper chlorophyllin against ethanol-induced gastric ulcer in mice显示文摘In study,we aimed to determine the mechanisms underlying the gastroprotective effects of sodium copper chlorophyllin(SCC)against ethanol-induced gastric ulcer injury in mice.First,the gastroprotective effects of SCC against gastric ulcer induced by ethanol were assessed.Then,biochemical,histopathological,immunohistochemistry assays,and western blot analysis were conducted to determine the possible mechanisms of action underlying the effects of SCC.Compared to the effects of omeprazole(OME)in a confirmed mouse model of ethanol-induced gastric ulcer injury,treatment with various doses of SCC resulted in up-regulation of Bcl-2 and down-regulation of the pro-apoptotic protein Bax.Significant decreases in the levels of the malondialdehyde(MDA),myeloperoxidase(MPO),and NO in the gastric tissues were observed.Furthermore,inflammatory cytokine analysis revealed that SCC treatment inhibited the expressions of TNF-αand IL-6,greatly reduced the phosphorylation level of IκB,and repressed the nuclear translocation of NF-κB p65,which demonstrated that SCC inhibited the activation of the NF-κB pathway.The present findings suggest that the protective effects of SCC may be beneficial as a potential preventive and therapeutic agent for gastric ulcer through the NF-κB pathway.Taken together,SCC administration significantly decreased the levels of MPO,NO,and MDA in gastric tissue and exerted a powerful anti-inflammatory activity as demonstrated by reduction in the secretions of proinflammatory mediators such as IL-6 and TNF-α in the serum of mice exposed to ethanol. | Huawei Lv Yan Lin Peigang Liu Weiqing Liang Kemin Wei Jinbao Pu Hongjian Zhang | 2019 | Acta Biochimica et Biophysica Sinica2019,51,9: | 2 |
