| 2 | Exosome-mediated transfer of miR-1290 promotes cell proliferation and invasion in gastric cancer via NKD1显示文摘Currently, exosomes rich in RNAs and proteins are regarded as vital mediators of intercellular communication. Here, we aimed to explore the effects of exosomal miR-1290 in gastric cancer (GC) and understand its mechanism of action on GC progression. We first isolated exosomes from serum samples of GC patients and healthy people and characterized them by transmission electron microscopy. Then, we examined the expression level of miR-1290 contained in the exosomes by quantitative reverse-transcription polymerase chain reaction and found that exosomal miR-1290 was overexpressed in GC patients and cell lines. Promotion of proliferation, migration, and invasiveness of GC cells was noted after they were incubated with the isolated miR-1290-rich exosomes compared with incubation with a negative control. Furthermore, we predicted that naked cuticle homolog 1 (NKD1) mRNA is a direct target of miR-1290 and confirmed their interaction by a dual luciferase reporter assay. NKD1 overexpression attenuated the stimulatory effects of miR-1290 on GC cells. Collectively, our results suggest that exosomal miR-1290 enhances GC cell proliferation and invasion by targeting NKD1 mRNA and downregulating NKD1 expression. | 4 | An Ethylene-inhibited NF-YC Transcription Factor RhNF-YC9 Regulates Petal Expansion in Rose显示文摘The speed of flower opening is closely related to their ornamental period.Ethylene functions as a negative regulator involved in the regulation of the petal expansion process.In this study,we isolated a NF-YC transcription factor gene,RhNF-YC9,fromrose petals.RhNF-YC9 expression was induced at the early stages of flower opening but was inhibited by ethylene treatment.Silencing RhNF-YC9 decreased the speed of petal expansion from stage 2 to stage 5.The expressions of 11 cell expansion-related genes involved in cell wall loosening,cell turgor modulation,and cytoskeleton remodeling were significantly down-regulated in RhNF-YC9-silenced petals.We also found that silencing RhNF-YC9 decreased the expression of gibberellin acid(GA)biosynthetic gene RhGA20ox while significantly increasing the transcripts of GA catabolic gene RhGA2ox,reducing the accumulation of GA4 and GA7.The influence of ethylene treatment on the expression of RhGA20ox and RhGA2ox showed the same trend.These results together suggested that RhNF-YC9 positively regulated the speed of petal expansion and mediated the crosstalk between ethylene and GA.Our findings revealed a new insight into the function of NF-YC transcription factors involved in ethylene-regulated petal expansion. | Changxi Chen Nisar Hussain Yaru Wang Mingtong Li Lin Liu Meizhu Qin Nan Ma Junping Gao Xiaoming Sun | 2020 | Horticultural Plant Journal2020,6,6: | 9 |

| 8 | Characteristics of Oil and Gas Accumulation in Yong'an-Meitai Area of the Fushan Depression,Beibuwan Basin,South China Sea显示文摘The Yong'an-Meitai area is the focus of the present exploration in the Fushan Depression,Beibuwan Basin,South China Sea. All oils from this area are geochemically characterized by higher Pr/Ph ratio,higher proportion of heavy molecular weight hydrocarbons,and higher proportion of C29 regular steranes,which indicate that the organic matter of source rocks might have been deposited in an oxidizing palaeoenvironment and be dominated by higher plant organic matter input. The oil from E3w2 (the second member of Weizhou Fm. of the Oligocene) has a much higher density,relatively higher Pr/nC17 and Ph/nC18 ratios,and a 'UCM-unresolved complex mixture' on gas chromatograms,which indicate that it has been slightly biodegraded. CPI and other terpane and sterane isomer ratios suggest they are all mature oils. The timing of oil charging in E3w2 and E2l1 (the first member of the Liushagang Fm. of the Eocene) determined by the homogenization temperatures of fluid inclusions and thermal evolution history are from 9-3 Ma and 8-3 Ma,respectively. Thus,the interpretation of E3w2 as a secondary reservoir is unlikely. The timing of oil charging is later than that of hydrocarbon generating and expulsion of Liushagang Fm. source rocks and trap formation,which is favorable for oil accumulation in this area. All molecular parameters that are used for tracing oil filling direction decrease with shallower burial depth,which suggests vertical oil migration. | 19 | Spermatogenic Cell-Specific Gene Mutation in Mice via CRISPR-Cas9显示文摘Tissue-specific knockout technology enables the analysis of the gene function in specific tissues in adult mammals.However,conventional strategy for producing tissue-specific knockout mice is a time- and labor-consuming process,restricting rapid study of the gene function in vivo.CRISPR-Cas9 system from bacteria is a simple and efficient gene-editing technique,which has enabled rapid generation of gene knockout lines in mouse by direct injection of CRISPR-Cas9 into zygotes.Here,we demonstrate CRISPR-Cas9-mediated spermatogenic cell-specific disruption of Scp3 gene in testes in one step.We first generated transgenic mice by pronuclear injection of a plasmid containing Hspa2 promoter driving Cas9 expression and showed Cas9 specific expression in spermatogenic cells.We then produced transgenic mice carrying Hspa2 promoter driven Cas9 and constitutive expressed sgRNA targeting Scp3 gene.Male founders were infertile due to developmental arrest of spermatogenic cells while female founders could produce progeny normally.Consistently,male progeny from female founders were infertile and females could transmit the transgenes to the next generation.Our study establishes a CRISPR-Cas9-based one-step strategy to analyze the gene function in adult tissues by a temporal-spatial pattern. | Meizhu Bai Dan Liang Yinghua Wang Qing Li Yuxuan Wu Jinsong Li | 2016 | Journal of Genetics and Genomics2016,43,5: | 1 |
