| 1 | Dihydromyricetin inhibits migration and invasion of hepatoma cells through regulation of MMP-9 expression显示文摘AIM:To investigate the effects of dihydromyricetin(DHM)on the migration and invasion of human hepatic cancer cells.METHODS:The hepatoma cell lines SK-Hep-1 and MHCC97L were used in this study.The cells were cultured in RPIM-1640 medium supplemented with 10%fetal bovine serum at 37℃in a humidified 5%CO2incubator.DHM was dissolved in dimethyl sulfoxide and diluted to various concentrations in medium before applying to cells.MTT assays were performed to measure the viability of the cells after DHM treatment.Wound healing and Boyden transwell assays were used to assess cancer cell motility.The invasive capacity of cancer cells was measured using Matrigel-coated transwell chambers.Matrix metalloproteinase(MMP)-2/9 activity was examined by fluorescence analysis.Western blot was carried out to analyze the expression of MMP-2,MMP-9,p-38,JNK,ERK1/2 and PKC-δproteins.All data were analyzed by Student’s t tests in GraphPad prism 5.0software and are presented as mean±SD.RESULTS:DHM was found to strongly inhibit the migration of the hepatoma cell lines SK-Hep-1(without DHM,24 h:120±8μmol/L vs 100μmol/L DHM,24h:65±10μmol/L,P<0.001)and MHCC97L(without DHM,24 h:126±7μmol/L vs 100μmol/L DHM,24h:74±6μmol/L,P<0.001).The invasive capacity of the cells was reduced by DHM treatment(SK-Hep-1cells without DHM,24 h:67±4μmol/L vs 100μmol/L DHM,24 h:9±3μmol/L,P<0.001;MHCC97L cells without DHM,24 h:117±8μmol/L vs 100μmol/L DHM,24 h:45±2μmol/L,P<0.001).MMP2/9 activity was also inhibited by DHM exposure(SK-Hep-1 cells without DHM,24 h:600±26μmol/L vs 100μmol/L DHM,24 h:100±6μmol/L,P<0.001;MHCC97L cells without DHM,24 h:504±32μmol/L vs 100μmol/L DHM 24 h:156±10μmol/L,P<0.001).Western blot analysis showed that DHM decreased the expression level of MMP-9 but had little effect on MMP-2.Further investigation indicated that DHM markedly reduced the phosphorylation levels of p38,ERK1/2 and JNK in a concentration-dependent manner but had no impact on the total protein levels.In addition,PKC-δprotein,a key protein in the regulation of MMP family protein expression,was up-regulated with DHM treatment.CONCLUSION:These findings demonstrate that DHM inhibits the migration and invasion of hepatoma cells and may serve as a potential candidate agent for the prevention of HCC metastasis. | Qing-Yu Zhang Ran Li Guo-Fang Zeng Bin Liu Jie Liu Yang Shu Zhong-Kao Liu Zhi-Dong Qiu Dong-Jun Wang Hui-Lai Miao Ming-Yi Li Run-Zhi Zhu | 2014 | World Journal of Gastroenterology2014,20,29: | 23 |
| 2 | Sand-dust storms in China: temporal-spatial distribution and tracks of source lands显示文摘Sand-dust storm is a special natural disaster that frequently occurs in deserts and their surrounding areas. With the data published on Surface Meteorological Monthly Bulletin and Surface Chart during 1971-1996, the temporal-spatial distribution and annual variation of sand-dust storms are analyzed on the basis of the case study of atmospheric processes. Furthermore, the tracks and source areas of sand-dust storms are determined with the aid of GIS. The results show that except some parts of Qinghai Province and Inner Mongolia as well as Beijing, sand-dust storms decrease apparently in time and space in recent decades in China. Sand-dust storms occur most frequently in spring, especially in April. According to their source areas, sand-dust storms are classified into two types, i.e., the inner-source and outer-source sand-dust storms. Most of the outer-source sand-dust storms move along the north and west tracks. The north-track outer-source sand-dust storms always intrude into China across the Sino-Mongolian border from Hami, a city in the eastern part of Xinjiang, to Xilin Gol, a league in Inner Mongolia, while the west-track ones intrude into China from both southern and northern Xinjiang. | 4 | Estimation of annual actual evapotranspiration from nonsaturated land surfaces with conventional meteorological data显示文摘Views diverge greatly on the relationship between actual evapotranspiration (AE) and potential evapotranspiration (PE). Penman showed that AE is determined in intensity by PE and changes proportionally as a function of PE. In contrast, Bouchet indicated that AE deter-mines PE and varies inversely with PE. Based on nearly 30 years data from 432 weather stations and 512 hydrological stations in China, the two different theories, Penman抯 assumption and Bouchet抯 complementary relationship between AE and PE, were tested on nine river basins. With data integration technique, the complementary relationship between AE and PE was dis-played entirely. A general model to estimate the actual evapotranspiration from nonsaturated surfaces by routine meteorological observations has been established on the basis of thorough analysis of the concept of PE. | 5 | Classification of yearly extreme precipitation events and associated flood risk in the Yangtze-Huaihe River Valley显示文摘Fifty cases of regional yearly extreme precipitation events(RYEPEs)were identified over the Yangtze-Huaihe River Valley(YHRV)during 1979–2016 applying the statistical percentile method.There were five types of RYEPEs,namely Yangtze Meiyu(YM-RYEPE),Huaihe Meiyu(HM-RYEPE),southwest-northeast-oriented Meiyu(SWNE-RYEPE)and typhoon I and II(TC-RYEPE)types of RYEPEs.Potential vorticity diagnosis showed that propagation trajectories of the RYEPEs along the Western Pacific Subtropical High and its steering flow were concentrated over the southern YHRV.As a result,the strongest and most frequently RYEPEs events,about 16–21 cases with average rainfall above 100 mm,occurred in the southern YHRV,particularly in the Nanjing metropolitan area.There have been 14 cases of flood-inducing RYEPEs since 1979,with the submerged area exceeding 120 km^2as simulated by the Flood Area hydraulic model,comprising six HM-RYEPEs,five YM-RYEPEs,two TC-RYEPEs,and one SWNE-RYEPE.The combination of evolving RYEPEs and rapid expansion of urban agglomeration is most likely to change the flood risk distribution over the Nanjing metropolitan area in the future.In the RCP6.0(RCP8.5)scenario,the built-up area increases at a rate of about 10.41 km^2(10 yr)^(-1)(24.67 km^2(10 yr)^(-1))from 2010 to 2100,and the area of high flood risk correspondingly increases from 3.86 km^2(3.86 km^2)to 9.00 km^2(13.51 km^2).Areas of high flood risk are mainly located at Chishan Lake in Jurong,Lukou International Airport in Nanjing,Dongshan in Jiangning District,Lishui District and other low-lying areas.The accurate simulation of flood scenarios can help reduce losses due to torrential flooding and improve early warnings,evacuation planning and risk analysis.More attention should be paid to the projected high flood risk because of the concentrated population,industrial zones and social wealth throughout the Nanjing metropolitan area. | Zhiqing XIE Yin DU Yan ZENG Qian MIAO | 2018 | Science China Earth Sciences2018,61,9: | 10 |

| 9 | Effect of temperature on anoxic metabolism of nitrites to nitrous oxide by polyphosphate accumulating organisms显示文摘Temperature is an important physical factor, which strongly influences biomass and metabolic activity. In this study, the effects of temperature on the anoxic metabolism of nitrite(NO-2) to nitrous oxide(N2O) by polyphosphate accumulating organisms, and the process of the accumulation of N2O(during nitrite reduction), which acts as an electron acceptor, were investigated using 91% ± 4% Candidatus Accumulibacter phosphatis sludge. The results showed that N2O is accumulated when Accumulibacter first utilize nitrite instead of oxygen as the sole electron acceptor during the denitrifying phosphorus removal process. Properties such as nitrite reduction rate, phosphorus uptake rate, N2O reduction rate, and polyhydroxyalkanoate degradation rate were all influenced by temperature variation(over the range from 10 to 30°C reaching maximum values at 25°C). The reduction rate of N2O by N2O reductase was more sensitive to temperature when N2O was utilized as the sole electron acceptor instead of NO2, and the N2O reduction rates, ranging from 0.48 to 3.53 N2O-N/(hr·g VSS), increased to 1.45 to 8.60 mg N2O-N/(hr·g VSS). The kinetics processes for temperature variation of 10 to 30°C were(θ1 = 1.140–1.216 and θ2= 1.139–1.167). In the range of 10°C to 30°C, almost all of the anoxic stoichiometry was sensitive to temperature changes. | 11 | Inactivation, reactivation and regrowth of indigenous bacteria in reclaimed water after chlorine disinfection of a municipal wastewater treatment plant显示文摘Disinfection of reclaimed water prior to reuse is important to prevent the transmission of pathogens. Chlorine is a widely utilized disinfectant and as such is a leading contender for disinfection of reclaimed water. To understand the risks of chlorination resulting from the potential selection of pathogenic bacteria, the inactivation, reactivation and regrowth rates of indigenous bacteria were investigated in reclaimed water after chlorine disinfection. Inactivation of total coliforms, Enterococcus and Salmonella showed linear correlations, with constants of 0.1384, 0.1624 and 0.057 L/(mg·min) and R 2 of 0.7617, 0.8316 and 0.845, respectively. However, inactivation of total viable cells by measurement of metabolic activity typically showed a linear correlation at lower chlorine dose (0-22 (mg·min)/L), and a trailing region with chlorine dose increasing from 22 to 69 (mg·min)/L. Reactivation and regrowth of bacteria were most likely to occur after exposure to lower chlorine doses, and extents of reactivation decreased gradually with increasing chlorine dose. In contrast to total coliforms and Enterococcus, Salmonella had a high level of regrowth and reactivation, and still had 2% regrowth even after chlorination of 69 (mg·min)/L and 24 hr storage. The bacterial compositions were also significantly altered by chlorination and storage of reclaimed water, and the ratio of Salmonella was significantly increased from 0.001% to 0.045% after chlorination of 69 (mg·min)/L and 24 hr storage. These trends indicated that chlorination contributes to the selection of chlorine-resistant pathogenic bacteria, and regrowth of pathogenic bacteria after chlorination in reclaimed water with a long retention time could threaten public health security during wastewater reuse. | Dan Li Siyu Zeng April Z. | 12 | A draft sequence of the rice (Oryza sativa ssp. indica) genome显示文摘The sequence of the rice genome holds fundamental information for its biology, including physiology, genetics, development, and evolution, as well as information on many beneficial phenotypes of economic significance. Using a 'whole genome shotgun' approach, we have pro-duced a draft rice genome sequence of Oryza sativa ssp. in-dica, the major crop rice subspecies in China and many other regions of Asia. The draft genome sequence is constructed from over 4.3 million successful sequencing traces with an accumulative total length of 2214.9 Mb. The initial assembly of the non-redundant sequences reached 409.76 Mb in length, based on 3.30 million successful sequencing traces with a total length of 1797.4 Mb from an indica variant cultivar 93-11, giving an estimated coverage of 95.29% of the rice genome with an average base accuracy of higher than 99%. | 13 | A LAMP-assay-based specific microbiota analysis reveals community dynamics and potential interactions of 13 major soybean root pathogens显示文摘Soybean root diseases are associated with numerous fungal and oomycete pathogens;however,the community dynamics and interactions of these pathogens are largely unknown.We performed 13 loop-mediated isothermal amplification(LAMP)assays that targeted specific soybean root pathogens,and traditional isolation assays.A total of 159 samples were collected from three locations in the Huang-Huai-Hai region of China at three soybean growth stages(30,60,and 90 days after planting)in 2016.In LAMP results,we found that pathogen communities differed slightly among locations,but changed dramatically between soybean growth stages.Phytophthora sojae,Rhizoctonia solani,and Fusarium oxysporum were most frequently detected at the early stage,whereas Phomopsis longicolla,Fusarium equiseti,and Fusarium virguliforme were most common in the later stages.Most samples(86%)contained two to six pathogen species.Interestingly,the less detectable species tended to exist in the samples containing more detected species,and some pathogens preferentially co-occurred in diseased tissue,including P.sojae–R.solani–F.oxysporum and F.virguliforme–Calonectria ilicicola,implying potential interactions during infection.The LAMP detection results were confirmed by traditional isolation methods.The isolated strains exhibited different virulence to soybean,further implying a beneficial interaction among some pathogens. | YE Wen-wu ZENG Dan-dan XU Miao YANG Jin MA Jia-xin WANG Yuan-chao ZHENG Xiao-bo | 2020 | Journal of Integrative Agriculture2020,19,8: | 6 |
| 14 | Jixuecao(Herba Centellae Asiaticae) alleviates mesangial cell proliferation in IgA nephropathy by inducing mitofusin 2 expression显示文摘OBJECTIVE:To investigate the effect of mitofusin 2 (Mfn2) and its downstream signaling pathway on glomerular mesangial cells (GMCs) proliferation in IgA nephropathy (IgAN),as well as the mechanism of action of Jixuecao (Herba Centellae Asiaticae,HCA) in the treatment of IgAN.METHODS:Adenovirus-mediated Mfn2 gene transfection and Mfn2 expression were analyzed by real-time polymerase chain reaction (PCR) and Western blotting.IgA1 induced the proliferation of GMCs,which were then treated with HCA.Cell proliferation was detected with cell counting kit-8 (CCK-8),and Mfn2 expression was analyzed by real-time PCR and western blotting.An IgAN animal model was also established and treated with HCA.GMCs proliferation was detected by hematoxylin-eosin staining,mitochondrial structure was analyzed by electron microscopy,mitochondrial function was determined by the Clark oxygen electrode method,and the expression of Mfn2,Phospho-extracellular regulated protein kinases1/2 (P-ERK1/2),Cyclin-dependent kinase 2 (CDK2),Phospho-p27 (p-p27),and cyclin A was analyzed by Western blotting.RESULTS:In vitro,HCA inhibited GMCs in a concentration-dependent manner in association with the upregulation of Mfn2 expression.The overexpression of Mfn2 inhibited IgA1-induced GMCs proliferation and elevated the effect of HCA.In vivo,treatment with HCA could alleviate albuminuria and creatinine and GMCs proliferation.These effects were related to the upregulation of Mfn2,p-p27 and inhibition of p-ERK1/2,CDK2,and cyclinA.Mitochondrial swelling,vacuolar degeneration,and reduction of respiratory control rate were identified in IgAN,but HCA could improve the mitochondrial structure and function.CONCLUSION:HCA inhibited GMCs proliferation via the upregulation Mfn2 and the inhibition of Ras-Raf-ERK/MAPK.We revealed that changes of mitochondrial structure and function are associated with IgAN,but that HCA can improve these mitochondrial features. | Chen Hongyu Du Yuanyuan Li Yayu Zeng Jiali Miao Jianxia Jiang Xue | 2019 | Journal of Traditional Chinese Medicine2019,39,3: | 6 |

While, the effects of laser soldering technology on electric connector solder joints need to be estimated completely, especially on power consumption. The combined method based on numerical simulation and the Accelerated temperature experiment(ATE) was developed to analyze the power consumption of USB 2.0 electric connector in this paper. The ATE contains thermal cycle and thermal shock tests, and the four-electrode method is used to obtain the conductivity of solder joints. Numerical modeling and analysis was used to quantify the power consumption and optimize the geometry of solder joints, because the electric experimental measurements of power consumption during ATE are time-costing and often intractable. | 17 | Polymorphism of rs1044925 in the acyl-CoA:cholesterol acyltransferase-1 gene and serum lipid levels in the Guangxi Bai Ku Yao and Han populations显示文摘Background The association of rs1044925 polymorphism in the acyl- CoA:cholesterol acyltransferase-1(ACAT-1) gene and serum lipid profiles is not well known in different ethnic groups.Bai Ku Yao is a special subgroup of the Yao minority in China.The present study was carried out to clarify the association of rs1044925 polymorphism in the ACAT-1 gene and several environmental factors with serum lipid levels in the Guangxi Bai Ku Yao and Han populations. Methods A total of 626 subjects of Bai Ku Yao and 624 participants of Han Chinese were randomly selected from our previous stratified randomized cluster samples.Genotyping of rs 1044925 polymorphism in the ACAT-1 gene was performed by polymerase chain reaction and restriction fragment length polymorphism combined with gel electrophoresis,and then confirmed by direct sequencing.Results The levels of serum total cholesterol(TC),high-density lipoprotein cholesterol (HDL-C),apolipoprotein(Apo) AI and ApoB were lower in Bai Ku Yao than in Han(P<0.01 for all).The frequency of A and C alleles was 79.0%and 21.0%in Bai Ku Yao,and 87.3%and 12.7%in Han(P<0.001);respectively.The frequency of AA,AC and CC genotypes was 63.2%,31.4% and 5.2%in Bai Ku Yao,and 75.6%,23.2%and 1.1%in Han(P<0.001);respectively.The levels of TC,LDL-C and ApoB in Bai Ku Yao but not in Han were different between the AA and AC/CC genotypes in females but not in males (P<0.05 for all).The C allele carriers had lower serum TC, LDL-C and ApoB levels as compared with the C allele non-carriers. It covers three major physiological and developmental states: growth, starvation, and conjugation, and can be accessed through a user-friendly web interface. The gene expression profiles and candidate co-expressed genes for each gene can be retrieved using Gene ID or Gene description searches. Descriptions of standardized methods of sample preparation and the opportunity to add new Tetrahymena microarray data will be of great interest to the Tetrahymena research community. | 20 | Quantification of viable bacteria in wastewater treatment plants by using propidium monoazide combined with quantitative PCR(PMA-qPCR)显示文摘The detection of viable bacteria in wastewater treatment plants(WWTPs) is very important for public health, as WWTPs are a medium with a high potential for waterborne disease transmission. The aim of this study was to use propidium monoazide(PMA) combined with the quantitative polymerase chain reaction(PMA-qPCR) to selectively detect and quantify viable bacteria cells in full-scale WWTPs in China. PMA was added to the concentrated WWTP samples at a final concentration of 100 μmol/L and the samples were incubated in the dark for 5 min, and then lighted for 4 min prior to DNA extraction and qPCR with specific primers for Escherichia coli and Enterococci, respectively. The results showed that PMA treatment removed more than 99% of DNA from non-viable cells in all the WWTP samples, while matrices in sludge samples markedly reduced the effectiveness of PMA treatment. Compared to qPCR, PMA-qPCR results were similar and highly linearly correlated to those obtained by culture assay, indicating that DNA from non-viable cells present in WWTP samples can be eliminated by PMA treatment, and that PMA-qPCR is a reliable method for detection of viable bacteria in environmental samples. This study demonstrated that PMA-qPCR is a rapid and selective detection method for viable bacteria in WWTP samples, and that WWTPs have an obvious function in removing both viable and non-viable bacteria.
