1Jixuecao(Herba Centellae Asiaticae) alleviates mesangial cell proliferation in IgA nephropathy by inducing mitofusin 2 expression显示文摘OBJECTIVE:To investigate the effect of mitofusin 2 (Mfn2) and its downstream signaling pathway on glomerular mesangial cells (GMCs) proliferation in IgA nephropathy (IgAN),as well as the mechanism of action of Jixuecao (Herba Centellae Asiaticae,HCA) in the treatment of IgAN.METHODS:Adenovirus-mediated Mfn2 gene transfection and Mfn2 expression were analyzed by real-time polymerase chain reaction (PCR) and Western blotting.IgA1 induced the proliferation of GMCs,which were then treated with HCA.Cell proliferation was detected with cell counting kit-8 (CCK-8),and Mfn2 expression was analyzed by real-time PCR and western blotting.An IgAN animal model was also established and treated with HCA.GMCs proliferation was detected by hematoxylin-eosin staining,mitochondrial structure was analyzed by electron microscopy,mitochondrial function was determined by the Clark oxygen electrode method,and the expression of Mfn2,Phospho-extracellular regulated protein kinases1/2 (P-ERK1/2),Cyclin-dependent kinase 2 (CDK2),Phospho-p27 (p-p27),and cyclin A was analyzed by Western blotting.RESULTS:In vitro,HCA inhibited GMCs in a concentration-dependent manner in association with the upregulation of Mfn2 expression.The overexpression of Mfn2 inhibited IgA1-induced GMCs proliferation and elevated the effect of HCA.In vivo,treatment with HCA could alleviate albuminuria and creatinine and GMCs proliferation.These effects were related to the upregulation of Mfn2,p-p27 and inhibition of p-ERK1/2,CDK2,and cyclinA.Mitochondrial swelling,vacuolar degeneration,and reduction of respiratory control rate were identified in IgAN,but HCA could improve the mitochondrial structure and function.CONCLUSION:HCA inhibited GMCs proliferation via the upregulation Mfn2 and the inhibition of Ras-Raf-ERK/MAPK.We revealed that changes of mitochondrial structure and function are associated with IgAN,but that HCA can improve these mitochondrial features.Chen Hongyu Du Yuanyuan Li Yayu Zeng Jiali Miao Jianxia Jiang Xue 2019Journal of Traditional Chinese Medicine2019,39,3:6
2Comparison of Clinical and Epidemiological Characteristics of Asymptomatic and Symptomatic SARS-CoV-2 Infection in Children显示文摘To understand the epidemiological and clinical features of the symptomatic and asymptomatic pediatric cases of COVID-19,we carried out a prospective study in Shanghai during the period of January 19 to April 30,2020.A total of 49 children(mean age 11.5±5.12 years)confirmed with SARS-CoV-2 infection were enrolled in the study,including 11(22.4%)domestic cases and 38(77.6%)imported cases.Nine(81.8%)local cases and 12(31.6%)imported cases had a definitive epidemiological exposure.Twenty-eight(57.1%)were symptomatic and 21(42.9%)were asymptomatic.Neither asymptomatic nor symptomatic cases progressed to severe diseases.The mean duration of viral shedding for SARS-CoV-2 in upper respiratory tract was 14.1±6.4 days in asymptomatic cases and 14.8±8.4 days in symptomatic cases(P>0.05).Forty-five(91.8%)cases had viral RNA detected in stool.The mean duration of viral shedding in stool was 28.1±13.3 days in asymptomatic cases and 30.8±18.6 days in symptomatic participants(P>0.05).Children<7 years shed viral RNA in stool for a longer duration than school-aged children(P<0.05).Forty-three(87.8%)cases had seropositivity for antibodies against SARS-CoV-2 within 1–3 weeks after confirmation with infection.In conclusion,asymptomatic SARS-CoV-2 infection may be common in children in the community during the COVID-19 pandemic wave.Asymptomatic cases shed viral RNA in a similar pattern as symptomatic cases do.It is of particular concern that asymptomatic individuals are potentially seed transmission of SARS-CoV-2 and pose a challenge to disease control.Jiehao Cai Xiangshi Wang Jun Zhao Yanling Ge Jin Xu He Tian Hailing Chang Aimei Xia Jiali Wang Jinqiang Zhang Zhongqiu Wei Jingjing Li Chuning Wang Jianshe Wang Qirong Zhu Xiaowen Zhai Mei Zeng 2020Virologica Sinica2020,35,6:4
3Electrodeposited NiSe2 on carbon fiber cloth as a flexible electrode for high-performance supercapacitors显示文摘A flexible electrode of nickel diselenide/carbon fiber cloth(NiSe_2/CFC) is fabricated at room temperature by a simple and efficient electrodeposition method. Owing to NiSe_2 character of nanostructure and high conductivity, the as-synthesized electrodes possess perfect pseudocapacitive property with high specific capacitance and excellent rate capability. In three-electrode system, the electrode specific capacitance of the NiSe_2/CFC electrode varies from 1058 F g^(-1) to 996.3 F g^(-1) at 2 A g^(-1) to 10 A g^(-1) respectively, which shows great rate capability. Moreover, the NiSe_2 electrode is assembled with an active carbon(AC) electrode to form an asymmetric supercapacitor with an extended potential window of 1.6 V. The asymmetric supercapacitor possesses an excellent energy density 32.7 Wh kg^(-1) with a power density 800 W kg^(-1) at the current density of 1 A g^(-1). 4Spirulina platensis aqueous extracts ameliorate colonic mucosal damage and modulate gut microbiota disorder in mice with ulcerative colitis by inhibiting inflammation and oxidative stress显示文摘Ulcerative colitis(UC)is a chronic and recurrent inflammatory bowel disease(IBD)that has become a major gastroenterologic problem during recent decades.Numerous complicating factors are involved in UC development such as oxidative stress,inflammation,and microbiota disorder.These factors exacerbate damage to the intestinal mucosal barrier.Spirulina platensis is a commercial alga with various biological activity that is widely used as a functional ingredient in food and beverage products.However,there have been few studies on the treatment of UC using S.platensis aqueous extracts(SP),and the underlying mechanism of action of SP against UC has not yet been elucidated.Herein,we aimed to investigate the modulatory effect of SP on microbiota disorders in UC mice and clarify the underlying mechanisms by which SP alleviates damage to the intestinal mucosal barrier.Dextran sulfate sodium(DSS)was used to establish a normal human colonic epithelial cell(NCM460)injury model and UC animal model.The mitochondrial membrane potential assay 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)and staining with Annexin V-fluorescein isothiocyanate(FITC)/propidium iodide(PI)and Hoechst 33258 were carried out to determine the effects of SP on the NCM460 cell injury model.Moreover,hematoxylin and eosin(H&E)staining,transmission electron microscopy(TEM),enzyme-linked immunosorbent assay(ELISA),quantitative real-time polymerase chain reaction(qPCR),western blot,and 16S ribosomal DNA(rDNA)sequencing were used to explore the effects and underlying mechanisms of action of SP on UC in C57BL/6 mice.In vitro studies showed that SP alleviated DSS-induced NCM460 cell injury.SP also significantly reduced the excessive generation of intracellular reactive oxygen species(ROS)and prevented mitochondrial membrane potential reduction after DSS challenge.In vivo studies indicated that SP administration could alleviate the severity of DSS-induced colonic mucosal damage compared with the control group.Inhibition of inflammation and oxidative stress was associated with increases in the activity of antioxidant enzymes and the expression of tight junction proteins(TJs)post-SP treatment.SP improved gut microbiota disorder mainly by increasing antioxidant enzyme activity and the expression of TJs in the colon.Our findings demonstrate that the protective effect of SP against UC is based on its inhibition of pro-inflammatory cytokine overproduction,inhibition of DSS-induced ROS production,and enhanced expression of antioxidant enzymes and TJs in the colonic mucosal barrier.Jian WANG Liqian SU Lun ZHANG Jiali ZENG Qingru CHEN Rui DENG Ziyan WANG Weidong KUANG Xiaobao JIN Shuiqing GUI Yinghua XU Xuemei LU 2022Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2022,23,6:3

7Enhanced ferroelectric polarization with less wake-up effect and improved endurance of Hf_(0.5)Zr_(0.5)O_(2)thin films by implementing W electrode显示文摘This paper reports the improvement of electrical,ferroelectric and endurance of Hf_(0.5)Zr_(0.5)O_(2)(HZO)thinfilm capacitors by implementing W electrode.The W/HZO/W capacitor shows excellent pristine 2 P_(r)values of 45.1 gC/cm^(2)at±6 V,which are much higher than those of TiN/HZO/W(34.4μC/cm^(2))and W/HZO/TiN(26.9μC/cm^(2))capacitors.Notably,the maximum initial 2 P_(r)value of W/HZO/W capacitor can reach as high as 57.9μC/cm^(2)at±7.5 V.These strong ferroelectric polarization effects are ascribed to the W electrode with a fairly low thermal expansion coefficient which provides a larger in-plane tensile strain compared with TiN electrode,allowing for enhancement of o-phase formation.Moreover,the W/HZO/W capacitor also exhibits higher endurance,smaller wake-up effect(10.1%)and superior fatigue properties up to 1.5×10^(10)cycles compared to the TiN/HZO/W and W/HZO/TiN capacitors.Such improvements of W/HZO/W capacitor are mainly due to the decreased leakage current by more than an order of magnitude compared to the W/HZO/TiN capacitor.These results demonstrate that capping electrode material plays an important role in the enhancement of o-phase formation,reduces oxygen vacancies,mitigates wake-up effect and improves reliability.Dao Wang Yan Zhang Jiali Wang Chunlai Luo Ming Li Wentao Shuai Ruiqiang Tao Zhen Fan Deyang Chen Min Zeng Jiyan Y.Dai Xubing B.Lu J-M.Liu 2022Journal of Materials Science & Technology2022,,9:1

11Picosecond electrical response in graphene/MoTe_(2) heterojunction with high responsivity in the near infrared region显示文摘Understanding the fundamental charge carrier dynamics is of great significance for photodetectors with both high speed and high responsivity.Devices based on two-dimensional(2D)transition metal dichalcogenides can exhibit picosecond photoresponse speed.However,2D materials naturally have low absorption,and when increasing thickness to gain higher responsivity,the response time usually slows to nanoseconds,limiting their photodetection performance.Here,by taking time-resolved photocurrent measurements,we demonstrated that graphene/MoTe_(2) van der Waals heterojunctions realize a fast 10 ps photoresponse time owing to the reduced average photocurrent drift time in the heterojunction,which is fundamentally distinct from traditional Dirac semimetal photodetectors such as graphene or Cd_(3)As_(2) and implies a photodetection bandwidth as wide as 100 GHz.Furthermore,we found that an additional charge carrier transport channel provided by graphene can ef-fectively decrease the photocurrent recombination loss to the entire device,preserving a high responsivity in the near-infrared region.Our study provides a deeper understanding of the ultrafast electrical response in van der Waals heterojunctions and offers a promising approach for the realization of photodetectors with both high responsivity and ultrafast electrical response.Zhouxiaosong Zeng Kai Braun Cuihuan Ge Martin Eberle Chenguang Zhu Xingxia Sun Xin Yang Jiali Yi Delang Liang Yufan Wang Lanyu Huang Ziyu Luo Dong Li Anlian Pan Xiao Wang 2022Fundamental Research2022,2,3:0
12Two New Species of Free-Living Marine Nematodes of the Desmodoridae from Mangrove Wetlands of Xiamen Bay, China显示文摘Mangroves are unique in their biodiversity,but studies on their meiobenthic biodiversity in China are scarce.Despite the importance of mangroves,little work has been done on the classification of nematodes in mangrove wetlands.Fujian Province is the most northern point of China’s natural mangrove distribution,and it is also one of the provinces with the earliest constructed mangrove forest.In this paper,two new free-living marine nematode species of Desmodoridae from the Xiamen mangrove wetlands in China are described.Metachromadora xiamenensis sp.nov.is characterized by a cylindrical body,and smooth head capsule set off from the rest of the body.The cuticle is finely annulated and thickened at the midbody.Lateral ridges run from the posterior end of the pharynx to the middle of the tail.The amphid foveae is loop shaped and opens at the top with a double contour amphidial,pharynx with bipartite cuticularized internal cavity.There are 18 tubular precloacal supplements and tail with three small protuberances.Molgolaimus euryformis sp.nov.is characterized by a relatively short and plump body with finely annulated cuticle,which is particularly obvious in the tail.The head is small and wide with intensive striates.The inner and outer labial sensilla are indistinct with short spicules and ventral apophysis,a gubernaculum with a block-shaped hook,a swollen conical-cylindrical tail and an absence of precloacal supplements.ZHOU Xiping ZENG Jiali CAI Lizhe FU Sujing TAN Wenjuan 2020Journal of Ocean University of China2020,19,1:0
13New insights into the carotenoid biosynthesis in Torreya grandis kernels显示文摘Torreya grandis is a characteristic rare economic tree species in subtropical mountainous areas. The nut has a variety of bioactive compounds, such as polyphenols and vitamins. However, the carotenoids compositions and its regulatory mechanisms underlying carotenoid biosynthesis in kernels remain unknown. Here, the major carotenoids in the kernel were analyzed. The result showed that lutein and β-carotene were the predominant carotenoids in the kernel, while low levels of a-carotenoid and zeaxanthin were detected. Lutein and β-carotene were decreased during the maturation of kernel. Expression analysis by RNA-seq and q RT-PCR indicated that the expressions of TgCYP97A3 and TgLCYB were also reduced during the maturation of kernel. The contents of lutein or β-carotene were obviously increased in tobacco transiently overexpressing TgCYP97A3 or TgLCYB. Moreover, DPPH radical scavenging activity and FRAP were also significantly enhanced. In addition, several MYB and WRKY transcription factors TgMYBS3, TgMYB48 and TgWRKY11 were identified to positively regulate the TgCYP97A3 expression, while TgMYB48, TgWRKY2 and TgWRKY11 could upregulate the TgLCYB expression. 14MCloudSim:modeling and simulating distributed clouds显示文摘Many distributed clouds which try to integrate the advantages of centralized clouds and distributed systems have been studied in recent years.Traditional cloud simulators focus on large scale data centers with virtualized servers,and cannot meet the needs of distributed cloud simulations.This paper introduces a generalized and extensible simulation framework,named MCloudSim,which is used for modeling and simulating distributed clouds.MCloudSim has the following characteristics;(1)the overlay protocol in MCloudSim can be freely replaced without affecting the function of other modules;(2) a Petri net workflow theory based task model is proposed to simulate distributed tasks;(3) a distributed cloud can be easily built by assembling and expanding the basic resource entities provided by MCloudSim.Finally,simulation results of scenarios with a 3-tier central-controlled distributed cloud and a P2 P based cloud prove that MCloudSim has high efficiency and satisfies performance in supporting large scale experiments and different distributed clouds.郑鹏飞 You Jiali Wang Jinlin Zeng Xuewen 2014High Technology Letters2014,20,2:0
15MapReduce based computation of the diffusion method in recommender systems显示文摘The performance of existing diffusion-based algorithms in recommender systems is still limited by the processing ability of a single computer. In order to conduct the diffusion computation on large data sets,a parallel implementation of the classic diffusion method on the MapReduce framework is proposed. At first,the diffusion computation is transformed from a summation format to a cascade matrix multiplication format,and then,a parallel matrix multiplication algorithm based on dynamic vector is proposed to reduce the CPU and I / O cost on the MapReduce framework,which can also be applied to other parallel matrix multiplication scenarios. 16Gut microbiota-derived short-chain fatty acids ameliorate methamphetamine-induced depression-and anxiety-like behaviors in a Sigmar-1 receptor-dependent manner显示文摘Methamphetamine(Meth)abuse can cause serious mental disorders,including anxiety and depression.The gut microbiota is a crucial contributor to maintaining host mental health.Here,we aim to investigate if microbiota participate in Meth-induced mental disorders,and the potential mechanisms involved.Here,15 mg/kg Meth resulted in anxiety-and depression-like behaviors of mice successfully and suppressed the Sigma-1 receptor(SIGMAR1)/BDNF/TRKB pathway in the hippocampus.Mean-while,Meth impaired gut homeostasis by arousing the Toll-like receptor 4(TLR4)-related colonic inflammation,disturbing the gut microbiome and reducing the microbiota-derived short-chain fatty acids(SCFAs).Moreover,fecal microbiota from Meth-administrated mice mediated the colonic inflam-mation and reproduced anxiety-and depression-like behaviors in recipients.Further,SCFAs supple-mentation optimized Meth-induced microbial dysbiosis,ameliorated colonic inflammation,and repressed anxiety-and depression-like behaviors.Finally,Sigmarl knockout(Sigmar1^(-/-))repressed the BDNF/TRKB pathway and produced similar behavioral phenotypes with Meth exposure,and elim-inated the anti-anxiety and-depression effects of SCFAs.The activation of SIGMAR1 with fluvoxamine attenuated Meth-induced anxiety-and depression-like behaviors.Our findings indicated that gut microbiota-derived SCFAs could optimize gut homeostasis,and ameliorate Meth-induced mental disorders in a SIGMAR1-dependent manner.This study confirms the crucial role of microbiota in Methrelated mental disorders and provides a potential preemptive therapy.Kaikai Zhang Lijian Chen Jianzheng Yang Jiali Liu Jiahao Li Yi Liu Xiuwen Li Long Chen Clare Hsu Jiahao Zeng Xiaoli Xie Qi Wang 2023Acta Pharmaceutica Sinica B2023,13,12:0
17The role of cAMP-dependent protein kinase A in the formation of long-term memory in Bactrocera dorsalis显示文摘The cAMP-dependent protein kinase A(PKA)signaling pathway has long been considered critical for long-term memory(LTM)formation.Previous studies have mostly focused on the role of PKA signaling in LTM induction by multiple spaced conditioning with less attention to LTM induction by a single conditioning.Here,we conducted behavioral-pharmacology,enzyme immunoassay and RNA interference experiments to study the role of the PKA signaling pathway in LTM formation in the agricultural pest Bactrocera dorsalis,which has a strong memory capacity allowing it to form a two-day memory even from a single conditioning trial.We found that either blocking or activating PKA prior to conditioning pretreatment affected multiple spaced LTM,and conversely,they did not affect LTM formed by single conditioning.This was further confirmed by enzyme-linked immunosorbent assay(ELISA)and silencing of the protein kinase regulatory subunit 2 and catalytic subunit 1.Taken together,these results suggest that activating PKA during memory acquisition helps to induce the LTM formed by multiple spaced conditioning but not by a single conditioning.Our findings challenge the conserved role of PKA signaling in LTM,which provides a basis for the greater diversity of molecular mechanisms underlying LTM formation across species,as well as possible functional and evolutionary implications.Jinxin Yu Yanmin Hui Jiayi He Yinghao Yu Zhengbing Wang Siquan Ling Wei Wang Xinnian Zeng Jiali Liu 2024Journal of Integrative Agriculture2024,23,2:0
18Information technology and artificial intelligence support in management experiences of the pediatric designated hospital during the COVID-19 epidemic in 2022 in Shanghai显示文摘Objective:To describe the information technology and artificial intelligence support in management experiences of the pediatric designated hospital in the wave of COVID-19 in Shanghai.Methods:We retrospectively concluded the management experiences at the largest pediatric designated hospital from March 1st to May 11th in 2022 in Shanghai.We summarized the application of Internet hospital,face recognition technology in outpatient department,critical illness warning system and remote consultation system in the ward and the structed electronic medical record in the inpatient system.We illustrated the role of the information system through the number and prognosis of patients treated.Results:The COVID-19 designated hospitals were built particularly for critical patients requiring high-level medical care,responded quickly and scientifically to prevent and control the epidemic situation.From March 1st to May 11th,2022,we received and treated 768 children confirmed by positive RT-PCR and treated at our center.In our management,we use Internet Information on the Internet Hospital,face recognition technology in outpatient department,critical illness warning system and remote consultation system in the ward,structed electronic medical record in the inpatient system.No deaths or nosocomial infections occurred.The number of offline outpatient visits dropped,from March to May 2022,146,106,48,379,57,686 respectively.But the outpatient volume on the internet hospital increased significantly(3,347 in March 2022 vs.372 in March 2021;4,465 in April 2022 vs.409 in April 2021;4,677 in May 2022 vs.538 in May 2021).Conclusions:Information technology and artificial intelligence has provided significant supports in the management.The system might optimize the admission screening process,increases the communication inside and outside the ward,achieves early detection and diagnosis,timely isolates patients,and timely treatment of various types of children.Yu Shi Jin Fu Mei Zeng Yanling Ge Xiangshi Wang Aimei Xia Weijie Shen Jiali Wang Weiming Chen Siyuan Jiang Xiaowen Zhai 2023Intelligent Medicine2023,3,1:0
19Oscillation-specific nodal alterations in early to middle stages Parkinson’s disease显示文摘Background:Different oscillations of brain networks could carry different dimensions of brain integration.We aimed to investigate oscillation-specific nodal alterations in patients with Parkinson’s disease(PD)across early stage to middle stage by using graph theory-based analysis.Methods:Eighty-eight PD patients including 39 PD patients in the early stage(EPD)and 49 patients in the middle stage(MPD)and 36 controls were recruited in the present study.Graph theory-based network analyses from three oscillation frequencies(slow-5:0.01–0.027 Hz;slow-4:0.027–0.073 Hz;slow-3:0.073–0.198 Hz)were analyzed.Nodal metrics(e.g.nodal degree centrality,betweenness centrality and nodal efficiency)were calculated.Results:Our results showed that(1)a divergent effect of oscillation frequencies on nodal metrics,especially on nodal degree centrality and nodal efficiency,that the anteroventral neocortex and subcortex had high nodal metrics within low oscillation frequencies while the posterolateral neocortex had high values within the relative high oscillation frequency was observed,which visually showed that network was perturbed in PD;(2)PD patients in early stage relatively preserved nodal properties while MPD patients showed widespread abnormalities,which was consistently detected within all three oscillation frequencies;(3)the involvement of basal ganglia could be specifically observed within slow-5 oscillation frequency in MPD patients;(4)logistic regression and receiver operating characteristic curve analyses demonstrated that some of those oscillation-specific nodal alterations had the ability to well discriminate PD patients from controls or MPD from EPD patients at the individual level;(5)occipital disruption within high frequency(slow-3)made a significant influence on motor impairment which was dominated by akinesia and rigidity.Conclusions:Coupling various oscillations could provide potentially useful information for large-scale network and progressive oscillation-specific nodal alterations were observed in PD patients across early to middle stages.Xiaojun Guan Tao Guo Qiaoling Zeng Jiaqiu Wang Cheng Zhou Chunlei Liu Hongjiang Wei Yuyao Zhang Min Xuan Quanquan Gu Xiaojun Xu Peiyu Huang Jiali Pu Baorong Zhang Min-Ming Zhang 2019Translational Neurodegeneration2019,8,1:0
20SETD2 mediates immunotherapy and radiotherapy efficacy via regulating DNA damage responses and genomic stability in lung adenocarcinoma显示文摘SETD2 is the main transferase for the trimethylation of histone H3 at lysine 36 (H3K36me3) in mammals. SETD2 plays important role in repairing DNA double strand breaks and maintaining chromatin integrity.1 In renal carcinoma, SETD2 deficiency caused DNA replication fork instability and DNA damage.2 The absence of SETD2 was also reported to have strong tumor-promoting effects in lung adenocarcinomas (LUAD).3 However, the other roles of SETD2 remain poorly understood in LUAD. In this study, we utilized comprehensive omics-data analysis to determine that SETD2 was associated with DNA damage and immune-related signals in LUAD. SETD2 knockdown induced DNA damage and cGAS activation in LUAD cells, and reduced the number of cells at the G1 phase. Moreover, SETD2 deficiency was conducive to mutation burden, immune cell infiltration, and immunotherapy responses. High SETD2 expression was associated with high radiocurability.
