1BING: Binarized normed gradients for objectness estimation at 300fps显示文摘Training a generic objectness measure to produce object proposals has recently become of significant interest. We observe that generic objects with well-defined closed boundaries can be detected by looking at the norm of gradients, with a suitable resizing of their corresponding image windows to a small fixed size. Based on this observation and computational reasons, we propose to resize the window to 8 × 8 and use the norm of the gradients as a simple 64 D feature to describe it, for explicitly training a generic objectness measure. We further show how the binarized version of this feature, namely binarized normed gradients(BING), can be used for efficient objectness estimation, which requires only a few atomic operations(e.g., add, bitwise shift, etc.). To improve localization quality of the proposals while maintaining efficiency, we propose a novel fast segmentation method and demonstrate its effectiveness for improving BING's localization performance, when used in multithresholding straddling expansion(MTSE) postprocessing. 2Effect of Combined Chilling and GA_3 Treatment on Bud Abortion in Forced ‘Luoyanghong' Tree Peony(Paeonia suffruticosa Andr.)显示文摘This research investigated the effect of combined chilling and GA_3 treatment on bud abortion of tree peony under greenhouse conditions to determine the dynamics of exogenous hormones and chilling effects on bud abortion. Research of such nature will increase the understanding of the effects of GA_3 and chilling level on flower development thereby, promoting off-season cultivation techniques in tree peonies to increase production that meets market demand. Hormone and sugar levels in the ‘Luoyanghong' tree peony were respectively determined using High-Performance Liquid Chromatography(HPLC) and spectrophotometer. Non-potted chilling with 200 mg·L^(-1) GA_3 treatment reduced bud abortion rate. Flowering rate in non-potted chilling with 200 mg·L^(-1) GA_3 plants was 58%, as against 21% in potted chilling plants with 200 mg·L^(-1) GA_3 treatment. Sugar accumulation in retained or aborted flower buds was conversely related to ABA level. Low ABA and high sugar levels were noted in retained buds whereas the reverse induced bud abortion. ABA level apparently inhibited sucrose and reducing sugar uptake in aborted buds whereas GA_3 enhanced sucrose uptake in retained buds. GA_3 and IAA levels were higher in retained than in aborted buds, which made plants with retained bud more competitive in nutrient uptake(by root)for growth and development. 3Local neuronal circuits that may shape the discharge patterns of inferior collicular neurons显示文摘The discharge patterns of neurons in auditory centers encode information about sounds.However,few studies have focused on the synaptic mechanisms underlying the shaping of discharge patterns using intracellular recording techniques.Here,we investigated the discharge patterns of inferior collicular(IC)neurons using intracellular recordings to further elucidate the mechanisms underlying the shaping of discharge patterns.Under in vivo intracellular recording conditions,recordings were obtained from 66 IC neurons in 18 healthy adult mice(Mus musculus,Km)under free field-stimulation.Fiftyeight of these neurons fired bursts of action potentials(APs)to auditory stimuli and the remaining eight just generated local responses such as excitatory(n=4)or inhibitory(n=4)postsynaptic potentials.Based on the APs and subthreshold responses,the discharge patterns were classified into seven types:phasic(24/58,41.4%),phasic burst(8/58,13.8%),pauser(4/58,6.9%),phasic-pauser(1/58,1.7%),chopper(2/58,3.4%),primary-like tonic(14/58,24.1%)and sound-induced inhibitory(5/58,8.6%).We concluded that(1)IC neurons exhibit at least seven distinct discharge patterns;(2)inhibition participates in shaping the discharge pattern of most IC neurons and plays a role in sculpting the pattern,except for the primary-like tonic pattern which was not shapedby inhibition;and(3)local neural circuits are the likely structural basis that shapes the discharge patterns of IC neurons and can be formed either in the IC or in lower-level auditory structures.Zi-Ying Fu Hui-Xian Mei Liang Cheng Jing Bai Jia Tang Philip Hung-Sun Jen Qi-Cai Chen 2013Neuroscience Bulletin2013,29,5:5
4Cell cycle-related kinase reprograms the liver immune microenvironment to promote cancer metastasis显示文摘The liver is an immunologically tolerant organ and a common metastatic site of multiple cancer types.Although a role for cancer cell invasion programs has been well characterized,whether and how liver-intrinsic factors drive metastatic spread is incompletely understood.Here,we show that aberrantly activated hepatocyte-intrinsic cell cycle-related kinase(CCRK)signaling in chronic liver diseases is critical for cancer metastasis by reprogramming an immunosuppressive microenvironment.Using an inducible liverspecific transgenic model,we found that CCRK overexpression dramatically increased both B16F10 melanoma and MC38 colorectal cancer(CRC)metastasis to the liver,which was highly infiltrated by polymorphonuclear-myeloid-derived suppressor cells(PMNMDSCs)and lacking natural killer T(NKT)cells.Depletion of PMN-MDSCs in CCRK transgenic mice restored NKT cell levels and their interferon gamma production and reduced liver metastasis to 2.7% and 0.7%(metastatic tumor weights)in the melanoma and CRC models,respectively.Mechanistically,CCRK activated nuclear factor-kappa B(NF-κB)signaling to increase the PMN-MDSC trafficking chemokine C-X-C motif ligand 1(CXCL1),which was positively correlated with liver-infiltrating PMN-MDSC levels in CCRK transgenic mice.Accordingly,CRC liver metastasis patients exhibited hyperaaivation of hepatic CCRK/NF-κB/CXCL1 signaling,which was associated with accumulation of PMN-MDSCs and paucity of NKT cells compared to healthy liver transplantation donors.In summary,this study demonstrates that immunosuppressive reprogramming by hepatic CCRK signaling undermines antimetastatic immunosurveillance.Our findings offer new mechanistic insights and therapeutic targets for liver metastasis intervention.Xuezhen Zeng Jingying Zhou Zhewen Xiong Hanyong Sun Weiqin Yang Myth T.S.Mok Jing Wang Jingqing Li Man Liu Wenshu Tang Yu Feng Hector Kwong-Sang W ang Shun-Wa Tsang King-Lau Chow Philip Chun Yeung John Wong Paul Bo-San Lai Anthony Wing-Hung Chan Ka Fai To Stephen Lam Chan Qiang Xia Jing Xue Xiao Chen Jun Yu Sui Peng Joseph Jao-Yiu Sung Ming Kuang Alfred Sze-Lok Cheng 2021Cellular & Molecular Immunology2021,18,4:5
5Randomised controlled trial comparing modified Sano's and narrow band imaging international colorectal endoscopic classifications for colorectal lesions显示文摘AIM To assess the utility of modified Sano′s(MS) vs thenarrow band imaging international colorectal endoscopic(NICE) classification in differentiating colorectal polyps.METHODS Patients undergoing colonoscopy between 2013 and 2015 were enrolled in this trial.Based on the MS or the NICE classifications,patients were randomised for real-time endoscopic diagnosis.This was followed by biopsies,endoscopic or surgical resection.The endoscopic diagnosis was then compared to the final(blinded) histopathology.The primary endpoint was the sensitivity(Sn),specificity(Sp),positive predictive value(PPV) and negative predictive value(NPV) of differentiating neoplastic and non-neoplastic polyps(MSⅡ/Ⅱo/Ⅲa/Ⅲb vs I or NICE 1 vs 2/3).The secondary endpoints were 'endoscopic resectability'(MSⅡ/Ⅱo/Ⅲa vs Ⅰ/Ⅲb or NICE 2 vs 1/3),NPV for diminutive distal adenomas and prediction of post-polypectomy surveillance intervals.RESULTS A total of 348 patients were evaluated.The Sn,Sp,PPV and NPV in differentiating neoplastic polyps from non-neoplastic polyps were,98.9%,85.7%,98.2% and 90.9% for MS;and 99.1%,57.7%,95.4% and 88.2% for NICE,respectively.The area under the receiver operating characteristic curve(AUC) for MS was 0.92(95%CI:0.86-0.98);and AUC for NICE was 0.78(95%CI:0.69,0.88).The Sn,Sp,PPV and NPV in predicting 'endoscopic resectability' were 98.9%,86.1%,97.8% and 92.5% for MS;and 98.6%,66.7%,94.7% and 88.9% for NICE,respectively.The AUC for MS was 0.92(95%CI:0.87-0.98);and the AUC for NICE was 0.83(95%CI:0.75-0.90).The AUC values were statistically different for both comparisons(P = 0.0165 and P = 0.0420,respectively).The accuracy for diagnosis of sessile serrated adenoma/polyp(SSA/P) with high confidence utilizing MS classification was 93.2%.The differentiation of SSA/P from other lesions achieved Sp,Sn,PPV and NPV of 87.2%,91.5%,89.6% and 98.6%,respectively.The NPV for predicting adenomas in diminutive rectosigmoid polyps(n = 150) was 96.6% and 95% with MS and NICE respectively.The calculated accuracy of post-polypectomy surveillance for MS group was 98.2%(167 out of 170) and for NICE group was 92.1%(139 out of 151).CONCLUSION The MS classification outperformed the NICE classification in differentiating neoplastic polyps and predicting endoscopic resectability.Both classifications met ASGE PIVI thresholds.Leonardo Zorrón Cheng Tao Pu Kuan Loong Cheong Doreen Siew Ching Koay Sze Pheh Yeap Amanda Ovenden Mahima Raju Andrew Ruszkiewicz Philip W Chiu James Y Lau Rajvinder Singh 2018World Journal of Gastrointestinal Endoscopy2018,10,9:4
