| 2 | Genome-wide CRISPR screen identifies synthetic lethality between DOCK1 inhibition and metformin in liver cancer显示文摘Metformin is currently a strong candidate anti-tumor agent in multiple cancers.However,its anti-tumor effectiveness varies among different cancers or sub-populations,potentially due to tumor heterogeneity.It thus remains unclear which hepatocellular carcinoma(HCC)patient subpopulation(s)can benefit from met-formin treatment.Here,through a genome-wide CRISPR-Cas9-based knockout screen,we find that DOCK1 levels determine the anti-tumor effects of met-formin and that DOCK1 is a synthetic lethal target of metformin in HCC.Mechanistically,metformin promotes DOCK1 phosphorylation,which activates RAC1 to facilitate cell survival,leading to metformin resistance.The DOCK1-selective inhibitor,TBOPP,potentiates anti-tumor activity by metformin in vitro in liver cancer cell lines and patient-derived HCC organoids,and in vivo in xenografted liver cancer cells and immunocompetent mouse liver cancer models.Notably,metformin improves overall survival of HCC patients with low DOCK1 levels but not among patients with high DOCK1 expression.This study shows that metformin effective-ness depends on DOCK1 levels and that combining metformin with DOCK1 inhibition may provide a promising personalized therapeutic strategy for met-formin-resistant HCC patients. | Junru Feng Hui Lu Wenhao Ma Wenjing Tian Zhuan Lu Hongying Yang Yongping Cai Pengfei Cai Yuchen Sun Zilong Zhou Jiaqian Feng Jiazhong Deng Ying Shu Kun Qu Weidong Jia Ping Gao Huafeng Zhang | 2022 | Protein & Cell2022,13,11: | 5 |

| 7 | Structural mechanism of cooperative activation of the human calcium-sensing receptor by Ca^(2+) ions and L-tryptophan显示文摘The human calcium-sensing receptor(CaSR)is a class C G protein-coupled receptor(GPCR)responsible for maintaining Ca^(2+)homeostasis in the blood.The general consensus is that extracellular Ca^(2+)is the principal agonist of CaSR.Aliphatic and aromatic L-amino acids,such as L-Phe and L-Trp,increase the sensitivity of CaSR towards Ca^(2+)and are considered allosteric activators.Crystal structures of the extracellular domain(ECD)of CaSR dimer have demonstrated Ca^(2+)and L-Trp binding sites and conformational changes of the ECD upon Ca^(2+)/L-Trp binding.However,it remains to be understood at the structural level how Ca^(2+)/L-Trp binding to the ECD leads to conformational changes in transmembrane domains(TMDs)and consequent CaSR activation.Here,we determined the structures of full-length human CaSR in the inactive state,Ca^(2+)-or L-Trp-bound states,and Ca^(2+)/L-Trp-bound active state using single-particle cryo-electron microscopy.Structural studies demonstrate that L-Trp binding induces the closure of the Venus flytrap(VFT)domain of CaSR,bringing the receptor into an intermediate active state.Ca^(2+)binding relays the conformational changes from the VFT domains to the TMDs,consequently inducing close contact between the two TMDs of dimeric CaSR,activating the receptor.Importantly,our structural and functional studies reveal that Ca^(2+)ions and L-Trp activate CaSR cooperatively.Amino acids are not able to activate CaSR alone,but can promote the receptor activation in the presence of Ca^(2+).Our data provide complementary insights into the activation of class C GPCRs and may aid in the development of novel drugs targeting CaSR. | Shenglong Ling Pan Shi Sanling Liu Xianyu Meng Yingxin Zhou Wenjing Sun Shenghai Chang Xing Zhang Longhua Zhang Chaowei Shi Demeng Sun Lei Liu Changlin Tian | 2021 | Cell Research2021,31,4: | 3 |

| 11 | The ER membrane protein complex subunit Emc3 controls angiogenesis via the FZD4/WNT signaling axis显示文摘The endoplasmic reticulum(ER) membrane protein complex(EMC) regulates the synthesis and quality control of membrane proteins with multiple transmembrane domains. One of the membrane spanning subunits, EMC3, is a core member of the EMC complex that provides essential hydrophilic vestibule for substrate insertion. Here, we show that the EMC subunit Emc3 plays critical roles in the retinal vascular angiogenesis by regulating Norrin/Wnt signaling. Postnatal endothelial cell(EC)-specific deletion of Emc3 led to retarded retinal vascular development with a hyperpruned vascular network, the appearance of bluntended, aneurysm-like tip endothelial cells(ECs) with reduced numbers of filopodia and leakage of erythrocytes at the vascular front. Diminished tube formation and cell proliferation were also observed in EMC3 depleted human retinal endothelial cells(HRECs). We then discovered a critical role for EMC3 in expression of FZD4 receptor of β-catenin signaling using RNA sequencing, real-time quantitative PCR(RT-q PCR) and luciferase reporter assay. Moreover, augmentation of Wnt activity via lithium chloride(Li Cl) treatment remarkably enhanced β-catenin signaling and cell proliferation of HRECs. Additionally, Li Cl partially reversed the angiogenesis defects in Emc3-c KO mice. | 14 | Hyperactive PI3Kδ predisposes naive T cells to activation via aerobic glycolysis programs显示文摘Activated phosphoinositide 3-kinaseδsyndrome(APDS)is an autosomal-dominant combined immunodeficiency disorder resulting from pathogenic gain-of-function(GOF)mutations in the PIK3CD gene.Patients with APDS display abnormal T cell homeostasis.However,the mechanisms by which PIK3CD GOF contributes to this feature remain unknown.Here,with a cohort of children with PIK3CD GOF mutations from multiple regions of China and a corresponding CRISPR/Cas9 gene-edited mouse model,we reported that hyperactive PI3Kδdisrupted TNaive cell homeostasis in the periphery by intrinsically promoting the growth,proliferation,and activation of TNaive cells.Our results showed that PIK3CD GOF resulted in loss of the quiescence-associated gene expression profile in naive T cells and promoted naive T cells to overgrow,hyperproliferate and acquire an activated functional status.Naive PIK3CD GOF T cells exhibited an enhanced glycolytic capacity and reduced mitochondrial respiration in the resting or activated state.Blocking glycolysis abrogated the abnormal splenic T cell pool and reversed the overactivated phenotype induced by PIK3CD GOF in vivo and in vitro.These results suggest that enhanced aerobic glycolysis is required for PIK3CD GOF-induced overactivation of naive T cells and provide a potential therapeutic approach for targeting glycolysis to treat patients with APDS as well as other immune disorders. | Yanjun Jia Qiuyun Yang Yanping Wang Wenyan Li Xuemei Chen Tao Xu Zhirui Tian Minxuan Feng Liang Zhang Wenjing Tang Na Tian Lina Zhou Wenxia Song Xiaodong Zhao | 2021 | Cellular & Molecular Immunology2021,18,7: | 2 |

| 18 | Electronic structures and optical properties of two anthracene derivatives显示文摘The electronic structures and the optical properties of two anthracene derivatives, DBMA and DAA, are investigated by both experimental tech- niques and quantum chemical calculations. The cy- clic voltammetry and differential pulse polarograph measurement revealed that the introduction of ben- zol-imidazol and pyrrolo-pyridine group on the an- thracene block can affect the electrochemical be- havior of DBMA and DAA. Both UV/visible absorption and emission spectra of DBMA and DAA are red-shifted in contrast to the unsubstituted anthra- cene, so that the anthracene derivatives emit at blue-green region and the luminescence yields are remarkably elevated (over 90%). The B3LYP/6-31G theoretical calculations explored that the electronic structures of the anthracene derivatives are per- turbed by the side substitutes on the anthracene block, and the slight variation of the electronic struc- tures results in the enhanced electron accepting abil- ity and the decrease of the HOMO-LUMO energy gap, which is the origin of the emission to be shifted to blue-green region.
