1Relocation of the M8.0 Wenchuan earthquake and its aftershock sequence显示文摘We relocated M8.0 Wenchuan earthquake and 2706 aftershocks with M≥2.0 using double-difference algorithm and obtained relocations of 2553 events. To reduce the influence of lateral variation in crustal and upper mantle velocity structure, we used different velocity models for the east and west side of Longmenshan fault zone. In the relocation process, we added seismic data from portable seismic sta-tions close to the shocks to constrain focal depths. The precisions in E-W, N-S, and U-D directions after relocation are 0.6, 0.7, and 2.5 km respectively. The relocation results show that the aftershock epi-centers of Wenchuan earthquake were distributed in NE-SW direction, with a total length of about 330 km. The aftershocks were concentrated on the west side of the central fault of Longmenshan fault zone, excluding those on the north of Qingchuan, which obviously deviated from the surface fault and passed through Pingwu-Qingchuan fault in the north. The dominant focal depths of the aftershocks are between 5 and 20 km, the average depth is 13.3 km, and the depth of the relocated main shock is 16.0 km. The depth profile reveals that focal depth distribution in some of the areas is characterized by high-angle westward dipping. 3An MYB Transcription Factor from Malus xiaojinensis Has a Potential Role in Iron Nutrition显示文摘Regulation of iron uptake and use is critical for plant survival and growth. We isolated an MYB gene from Malus xiaojinensis named MxMYB1, which is induced under Fe-deficient conditions. Expression of MxMYB1 was upregulated by Fe starvation in the roots but not in leaves, suggesting that MxMYB1 might play a role in iron nutrition in roots. Transgenic Arabidopsis plants expressing MxMYB1 exhibited lower iron content as compared with wild type plants under both Fe-normal (40 μM) and Fe-deficient conditions (Fe omitted and Ferrozine 300 μM). However, the contents of Cu, Zn and Mn were not changed in these transgenic plants. Gene chip and real-time polymerase chain reaction analyses indicated that the expression of two Fe-related genes encoding an iron transporter AtIRT1 and an iron storage protein ferritin AtFER1 might be negatively regulated by MxMYB1 as the expression levels of these genes were lower in MxMYB1 expressing transgenic Arabidopsis plants as compared with wild type plants under both Fe-normal and Fe-deficient conditions. 4A novel miRNA negatively regulates resistance to Glomerella leaf spot by suppressing expression of an NBS gene in apple显示文摘Glomerella leaf spot(GLS)of apple(Malus×domestica Borkh.),caused by Glomerella cingulata,is an emerging fungal epidemic threatening the apple industry.Little is known about the molecular mechanism underlying resistance to this devastating fungus.In this study,high-throughput sequencing technology was used to identify microRNAs(miRNAs)involved in GLS resistance in apple.We focused on miRNAs that target genes related to disease and found that expression of a novel miRNA,Md-miRln20,was higher in susceptible apple varieties than in resistant ones.Furthermore,its target gene Md-TN1-GLS exhibited the opposite expression pattern,which suggested that the expression levels of Md-miRln20 and its target gene are closely related to apple resistance to GLS.Furthermore,downregulation of MdmiRln20 in susceptible apple leaves resulted in upregulation of Md-TN1-GLS and reduced the disease incidence.Conversely,overexpression of Md-miRln20 in resistant apple leaves suppressed Md-TN1-GLS expression,with increased disease incidence.We demonstrated that Md-miRln20 negatively regulates resistance to GLS by suppressing Md-TN1-GLS expression and showed,for the first time,a crucial role for miRNA in response to GLS in apple.Yi Zhang Qiulei Zhang Li Hao Shengnan Wang Shengyuan Wang Wenna Zhang Chaoran Xu Yunfei Yu Tianzhong Li 2019Horticulture Research2019,6,1:4
5A CRISPR/Cas12a-empowered surface plasmon resonance platform for rapid and specific diagnosis of the Omicron variant of SARS-CoV-2显示文摘The outbreak of the COVID-19 pandemic was partially due to the challenge of identifying asymptomatic and presymptomatic carriers of the virus,and thus highlights a strong motivation for diagnostics with high sensitivity that can be rapidly deployed.On the other hand,several concerning SARS-CoV-2 variants,including Omicron,are required to be identified as soon as the samples are identified as‘positive’.Unfortunately,a traditional PCR test does not allow their specific identification.Herein,for the first time,we have developed MOPCS(Methodologies of Photonic CRISPR Sensing),which combines an optical sensing technology-surface plasmon resonance(SPR)with the‘gene scissors’clustered regularly interspaced short palindromic repeat(CRISPR)technique to achieve both high sensitivity and specificity when it comes to measurement of viral variants.MOPCS is a low-cost,CRISPR/Cas12a-systemempowered SPR gene-detecting platform that can analyze viral RNA,without the need for amplification,within 38 min from sample input to results output,and achieve a limit of detection of 15 fM.MOPCS achieves a highly sensitive analysis of SARS-CoV-2,and mutations appear in variants B.1.617.2(Delta),B.1.1.529(Omicron)and BA.1(a subtype of Omicron).This platform was also used to analyze some recently collected patient samples from a local outbreak in China,identified by the Centers for Disease Control and Prevention.This innovative CRISPR-empowered SPR platform will further contribute to the fast,sensitive and accurate detection of target nucleic acid sequences with single-base mutations.Zhi Chen Jingfeng Li Tianzhong Li Taojian Fan Changle Meng Chaozhou Li Jianlong Kang Luxiao Chai Yabin Hao Yuxuan Tang Omar AAl-Hartomy Swelm Wageh Abdullah GAl-Sehemi Zhiguang Luo Jiangtian Yu Yonghong Shao Defa Li Shuai Feng William J.Liu Yaqing He Xiaopeng Ma Zhongjian Xie Han Zhang 2022National Science Review2022,9,8:3
6Two pathogenesis-related proteins interact with leucine-rich repeat proteins to promote Alternaria leaf spot resistance in apple显示文摘Alternaria leaf spot in apple(Malus x domestica),caused by the fungal pathogen Alternaria alternata f.sp.mali(also called A.mali),is a devastating disease resulting in substantial economic losses.We previously established that the resistance(R)protein MdRNL2,containing a coiled-coil,nucleotide-binding,and leucine-rich repeat(CC R-NB-LRR)domain,interacts with another CC R-NB-LRR protein,MdRNL6,to form a MdRNL2–MdRNL6 complex that confers resistance to A.mali.Here,to investigate the function of the MdRNL2–MdRNL6 complex,we identified two novel pathogenesis-related(PR)proteins,MdPR10-1 and MdPR10-2,that interact with MdRNL2.Yeast two-hybrid(Y2H)assays and bimolecular fluorescence complementation(BiFC)assays confirmed that MdPR10-1 and MdPR10-2 interact with MdRNL2 and MdRNL6 at the leucine-rich repeat domain.Transient expression assays demonstrated that accumulation of MdPR10-1 and MdPR10-2 enhanced the resistance of apple to four strains of A.mali that we tested:ALT1,GBYB2,BXSB5,and BXSB7.In vitro antifungal activity assays demonstrated that both the proteins contribute to Alternaria leaf spot resistance by inhibiting fungal growth.Our data provide evidence for a novel regulatory mechanism in which MdRNL2 and MdRNL6 interact with MdPR10-1 and MdPR10-2 to inhibit fungal growth,thereby contributing to Alternaria leaf spot resistance in apple.The identification of these two novel PR proteins will facilitate breeding for fungal disease resistance in apple.Qiulei Zhang Chaoran Xu Haiyang Wei Wenqi Fan Tianzhong Li 2021Horticulture Research2021,8,1:2

8Studies on the Attached Cultivation of Filamentous Oleaginous Microalga Tribonema minus显示文摘Attached cultivation is a promising method for microalgal biomass production. Filamentous oleaginous microalga Tribonema minus(hereafter T. minus) has shown a remarkable potential for biofuel production in terms of its high lipid content. However, the strain has only been cultivated in suspended cultivation systems including open pond and closed photobioreactors. Here, we attempted to study the attached cultivation of T. minus, which might be helpful for its scale-up cultivation and industrial applications. As the results, the optimal conditions for T. minus growth in the attached biofilm are 200 μmol photons m^-2 s^-1 of light intensity and 5% of CO2, and the maximum biomass density of 223 g m^-2 has been achieved under the light intensity. The non-woven fabric as substratum was found as the best substratum in thin layer attached bioreactor, on which the average biomass productivity of T. minus is about(9.73 ± 2.19) g m^-2)d^-1. Furthermore, two attached bioreactor systems, rotary drum and rotation disc, were designed following the light dilution strategy and introduced into T. minus cultivation. The highest footprint areal biomass productivity of these two systems is 33 and 47.1 g m^-2 d^-1, respectively, much higher than that in suspended cultivation system. 9Efficient Breeding and Cultivation of Type 2 Red-fleshed Apple Cultivars Using a Search System for Suitable Apple Cultivar Combination显示文摘As apple possesses self-incompatibility, knowledge of the S-genotypes is very important for apple breeding and cultivation. We investigated the S-RNase genotypes of 13 type 2 red-fleshed apple cultivars and 8 lineages. All of the investigated type 2 red-fleshed apples contained the S3-RNase allele closely linked to the red-fleshed trait. We made a search system for ovule parents to efficiently produce new type 2 red-fleshed cultivars based on the S-RNase allele information(http://www.agr.nagoya-u.ac.jp/~hort/apple/red/). More than 90% of progenies from crossing a selected white-fleshed ovule parent with a type 2 red-fleshed pollen parent are expected to show the red-fleshed phenotype. 10Comparison of growth,hydrocarbon accumulation and metabolites of Botryococcus braunii between attached cultivation and aqueous-suspension cultivation显示文摘Botryococcus braunii is a colonial green microalga which can produce extracellular hydrocarbons at a high rate,it is considered as one of the most promising feedstocks for sustainable biofuel production.However,B.braunii is generally recognized difficulties for cultivating and has limited amount of substantive scale-up and productivity assessments with conventional aqua-suspended cultivation systems(open pond and varieties of closed photobioreactors).This paper introduces a novel cultivation system based on biofilm technology,which is called“attached cultivation”.To investigate the potential of attached cultivation method,attached cultivation of B.braunii SAG 807-1 was compared with aqueous-suspension cultivation(flat plate reactors).The growth,hydrocarbon accumulation and metabolites were studied under identical conditions(e.g.temperature,light intensity,CO2 concentration).The main research results obtained are as follows:compared with conventional aqueous-suspension cultivation,the biomass productivity of B.braunii under biofilm attached cultivation was 4.78 g/(m^(2)·d),which was higher than of 4.43 g/(m^(2)·d)by aqueous-suspension,and hydrocarbon productivity of the two cultivation methods were 2.52 g/(m^(2)·d)and 2.37 g/(m^(2)·d),respectively.The contents of carbohydrate and protein were also similar.This attached cultivation method showed a new model of commercialization for the microalgae-derived biofuels.Cheng Pengfei Wang Yan Yang Qiyong Liu Tianzhong 2017International Journal of Agricultural and Biological Engineering2017,10,1:1

18Functional identification of lncRNAs in sweet cherry(Prunus avium)pollen tubes via transcriptome analysis using single-molecule long-read sequencing显示文摘Sweet cherry(Prunus avium)is a popular fruit with high nutritional value and excellent flavor.Although pollen plays an important role in the double fertilization and subsequent fruit production of this species,little is known about its pollen tube transcriptome.In this study,we identified 16,409 transcripts using single-molecule sequencing.After filtering 292 transposable elements,we conducted further analyses including mRNA classification,gene function prediction,alternative splicing(AS)analysis,and long noncoding RNA(lncRNA)identification to gain insight into the pollen transcriptome.The filtered transcripts could be matched with 3,438 coding region sequences from the sweet cherry genome.GO and KEGG analyses revealed complex biological processes during pollen tube elongation.A total of 2043 AS events were predicted,7 of which were identified in different organs,such as the leaf,pistil and pollen tube.Using BLASTnt and the Coding-Potential Assessment Tool(CPAT),we distinguished a total of 284 lncRNAs,among which 154 qualified as natural antisense transcripts(NATs).As the NATs could be the reverse complements of coding mRNA sequences,they might bind to coding sequences.Antisense transfection assays showed that the NATs could regulate the expression levels of their complementary sequences and even affect the growth conditions of pollen tubes.In summary,this research characterizes the transcripts of P.avium pollen and lays the foundation for elucidating the physiological and biochemical mechanisms underlying sexual reproduction in the male gametes of this species.Yang Li Chuanbao Wu Chunsheng Liu Jie Yu Xuwei Duan Wenqi Fan Jing Wang Xiaoming Zhang Guohua Yan Tianzhong Li Kaichun Zhang 2019Horticulture Research2019,6,1:1
