Erianin,a novel dibenzyl compound in Dendrobium extract, inhibits lung cancer cell growth and migration via calcium/calmodulin-dependent ferroptosis显示文摘Ferroptosis,a novel form of programmed cell death,is characterized by iron-dependent lipid peroxidation and has been shown to be involved in multiple diseases, including cancer.Stimulating ferroptosis in cancer cells may be a potential strategy for cancer therapy.Therefore,ferroptosis-inducing drugs are attracting more attention for cancer treatment. Here,we showed that erianin,a natural product isolated from Dendrobium chrysotoxum Lindl, exerted its anticancer activity by inducing cell death and inhibiting cell migration in lung cancer cells.Subsequently,we demonstrated for the first time that erianin induced ferroptotic cell death in lung cancer cells, which was accompanied by ROS accumulation, lipid peroxidation, and GSH depletion. The ferroptosis inhibitors Fer-1 and Lip-1 but not Z-VAD-FMK,CQ,or necrostatin-1 rescued erianin-induced cell death, indicating that ferroptosis contributed to erianin-induced cell death.Furthermore,we demonstrated that Ca^(2+)/CaM signaling was a critical mediator of erianin-induced ferroptosis and that blockade of this signaling significantly rescued cell death induced by erianin treatment by suppressing ferroptosis.Taken together, our data suggest that the natural product erianin exerts its anticancer effects by inducing Ca^(2+)/CaMdependent ferroptosis and inhibiting cell migration, and erianin will hopefully serve as a prospective compound for lung cancer treatment. | Peng Chen Qibiao Wu Jiao Feng Lili Yan Yitian Sun Shuiping Liu Yu Xiang Mingming Zhang Ting Pan Xiaying Chen Ting Duan Lijuan Zhai Bingtao Zhai Wengang Wang Ruonan Zhang Bi Chen Xuemeng Han Yicong Li Liuxi Chen Ying Liu Xingxing Huang Ting Jin Wenzheng Zhang Hong Luo Xiaohui Chen Yongqiang Li Qiujie Li Guohua Li Qin Zhang Lvjia Zhuo Zuyi Yang Huifen Tang Tian Xie Xiaoping Ouyang Xinbing Sui | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 42
Nanocrystals:Solution-Based Synthesis and Applications as Nanocatalysts显示文摘Nanocrystals are emerging as key materials due to their novel shape-and size-dependent chemical and physical properties that differ drastically from their bulk counterparts. The main challenges in this field remain rationally controlled synthesis and large scale production. This article reviews recent progress in our laboratory related to solution-based synthesis of various nanostructures, including zero-dimensional(0-D)nanocrystals,1-D nanowires and nanorods, hollow structures, and superlattice materials. On the other hand, the essential goal for nanoresearchers is to achieve industrial applications of nanostructured materials. In the past decades, these fascinating materials have been widely used in many promising fields such as nanofabrication, nanodevices, nanobiology, and nanocatalysis.Herein,we focus on their applications as nanocatalysts and try to illustrate the main problems and future directions in this area based on our recent endeavors in catalytic applications of nanocrystals. | Dingsheng Wang Ting Xie Yadong Li | 2009 | Nano Research2009,2,1: | 28
Magma mixing of granites at Lianghe:In-situ zircon analysis for trace elements, U-Pb ages and Hf isotopes显示文摘In this paper, we report an integrated study of trace element, U-Pb age and Hf isotopic composition of zircons from alkali feldspar granites, granodiorites and diorite enclaves in a recently discovered ring complex at Lianghe in western Yunnan, Chi na. The granitoids showed identical U-Pb ages of 127, 115 and 122 Ma, from felsic to mafic, but had different zircon trace el ements and Hf isotopic compositions. Trace element content decreased with a gradual increase in εHf(t) values of ?9.1 to ?5.4, ?4.5 to 0, and 3.6 to 6.2, respectively. Results indicate that changes in zircon trace elements generally correlate with changes in Hf isotope signatures within single samples and among various granitoids. These relationships reflect the mixing of felsic and mafic magmas. Evidence indicates that depleted mantle-derived mafic magma underplating caused ancient crustal melting, and then formed large-scale granites in Lianghe during the Early Cretaceous. These granodiorites were formed mainly by the mix ing of mafic magma and granitic magma. | CONG Feng LIN ShiLiang ZOU GuangFu LI ZaiHui XIE Tao PENG ZhiMin LIANG Ting | 2011 | Science China Earth Sciences2011,54,9: | 26
Baicalin induces ferroptosis in bladder cancer cells by downregulating FTH1显示文摘Ferroptosis is a non-apoptotic regulated cell death caused by iron accumulation and subsequent lipid peroxidation.Currently, the therapeutic role of ferroptosis on cancer is gaining increasing interest.Baicalin an active component in Scutellaria baicalensis Georgi with anticancer potential various cancer types;however, the effects of baicalein on bladder cancer and the underlying molecular mechanisms remain largely unknown. In the study,we investigated the effect of baicalin on bladder cancer cells5637 and KU-19-19.As a result,we show baicalin exerted its anticancer activity by inducing apoptosis and cell death in bladder cancer cells.Subsequently,we for the first time demonstrate baicalin-induced ferroptotic cell death in vitro and in vivo, accompanied by reactive oxygen species(ROS) accumulation and intracellular chelate iron enrichment. The ferroptosis inhibitor deferoxamine but not necrostatin-1,chloroquine(CQ),N-acetyl-L-cysteine,L-glutathione reduced,or carbobenzoxy-valyl-alanyl-aspartyl-[O-methyl]-fluoromethylketone(Z-VAD-FMK) rescued baicalin-induced cell death, indicating ferroptosis contributed to baicalin-induced cell death.Mechanistically,we show that ferritin heavy chain1(FTH1) was a key determinant for baicalin-induced ferroptosis.Overexpression of FTH1 abrogated the anticancer effects of baicalin in both 5637 and KU19-19 cells.Taken together, our data for the first time suggest that the natural product baicalin exerts its anticancer activity by inducing FTH1-dependent ferroptosis, which will hopefully provide a prospective compound for bladder cancer treatment. | Na Kong Xiaying Chen Jiao Feng Ting Duan Shuiping Liu Xueni Sun Peng Chen Ting Pan Lili Yan Ting Jin Yu Xiang Quan Gao Chengyong Wen Weirui Ma Wencheng Liu Mingming Zhang Zuyi Yang Wengang Wang Ruonan Zhang Bi Chen Tian Xie Xinbing Sui Wei Tao | 2021 | Acta Pharmaceutica Sinica B2021,11,12: | 23
Kinetics and equilibrium of adsorption of dissolved organic matter fractions from secondary effluent by fly ash显示文摘Fly ash was used as a low-cost adsorbent for removing dissolved organic matter (DOM) in secondary effluent. Batch experiments were conducted under various adsorbent dosages, pH, contact time, temperatures and DOM fractional characteristics. Under the optimum conditions of fly ash dosage of 15 g/L, temperature of 303 K and contact time of 180 min, a removal of 22.5% of the dissolved organic carbon (DOC), 23.7% of UV-254, 25.9% of the trihalomethanes precursors in secondary effluent was obtained. The adsorption of DOM fractions onto fly ash all followed the pseudo second-order kinetic model, and the hydrophilic fraction adsorption by fly ash also fitted the intraparticle diffusion model quite well. Freundlich and Langmuir models were applicable to the fly ash adsorption and their constants were evaluated. The maximum adsorption capacities of the adsorptions revealed that fly ash was more effective in adsorbing hydrophilic fraction than the acidic fractions. Structure changes of the DOM fractions after fly ash adsorption were also characterized via spectrum analyzing. Those mechanisms presented critical step toward improved efficiencies of fly ash adsorption via further surface-modification. | Liangliang Wei Kun Wang Qingliang Zhao Chunmei Xie Wei Qiu Ting Jia | 2011 | Journal of Environmental Sciences2011,23,7: | 21
TRANSFORMING GROWTH FACTOR-β1 AND SMAD4 SIGNALING PATHWAY DOWN-REGULATES RENAL EXTRACELLULAR MATRIX DEGRADATION IN DIABETIC RATS显示文摘Objective To investigate the role of transforming growth factor-β1(TGF-β1)/Smad4 pathway in development of renal fibrosis in streptozotocin(STZ)-induced diabetic nephropathy(DN) rats and explore its possible mechanism. Methods Male Wistar rats weighing 180-220 g were divided into 5 groups:group A(normal control),group B [diabetes mellitus(DM) 2 weeks],group C(DM 4 weeks),group D(DM 8 weeks),and group E(DM 16 weeks).Except for the normal control group, other groups were induced DM by single injection of STZ(55 mg/kg) respectively.Blood glucose level, serum creatinine, and 24-hour urine protein were examined.Expressions of TGF-β1 and Smad4 protein and mRNA in kidney were detected using immunohistochemical technique,Western blot, and real-time PCR.mRNA expressions of stromelysin-1(MMP-3),tissue inhibitor of metalloproteinase-1(TIMP-1),and collagen Ⅲ in kidney were also detected by real-time PCR..Results The levels of blood glucose, serum creatinine, and 24-hour urine protein in rats of group B,C,D,and E were higher than those of the control group. With the progression of renal fibrosis, the expressions of TGF-β1 and Smad4 protein and mRNA in kidney of diabetic rats elevated. In addition, the renal MMP-3 mRNA expression diminished in diabetic rats, while TIMP-1 and collagen Ⅲ mRNA increased. Conclusions In STZ-induced diabetic rats, the TGF-β1/Smad4 appears to play an important role in renal fibrosis of DN.The increased expression of TGF-β1 and Smad4 might result in the transcriptional regulation of downstream target genes of TGF-β1/Smad4 pathway, which contributes to the progression of renal fibrosis in diabetic rats. | Qin Yang Ru-jia Xie Ting Yang Li Fang Bing Han Guo-zhong Zhang Ming-liang Cheng | 2007 | Chinese Medical Sciences Journal2007,22,4: | 19
Self-perceived burden and influencing factors in patients with cervical cancer administered with radiotherapy显示文摘BACKGROUND Cervical cancer is the fourth commonest malignancy in women around the world. It represents the second most commonly diagnosed cancer in South East Asian women, and an important cancer death cause in women of developing nations.Data collected in 2018 revealed 5690000 cervical cancer cases worldwide,85%of which occurred in developing countries.AIM To assess self-perceived burden(SPB)and related influencing factors in cervical cancer patients undergoing radiotherapy.METHODS Patients were prospectively included by convenient sampling at The Fifth Affiliated Hospital of Sun Yat-Sen University,China between March 2018 and March 2019.The survey was completed using a self-designed general information questionnaire, the SPB scale for cancer patients, and the self-care self-efficacy scale,Strategies Used by People to Promote Health, which were delivered to patients with cervical cancer undergoing radiotherapy.Measurement data are expressed as the mean±SD.Enumeration data are expressed as frequencies or percentages.Caregivers were the spouse, offspring, and other in 46.4,40.9,and 12.7%,respectively,and the majority were male(59.1%).As for pathological type,90 and 20 cases had squamous and adenocarcinoma/adenosquamous carcinomas, respectively.Stage IV disease was found in 12(10.9%)patients.RESULTS A total of 115 questionnaires were released, and five patients were excluded for too long evaluation time(n=2)and the inability to confirm the questionnaire contents(n=3).Finally,a total of 110 questionnaires were collected. They were aged 31-79 years, with the 40-59 age group being most represented(65.4%of all cases). Most patients were married(91.8%)and an overwhelming number had no religion(92.7%).Total SPB score was 43.13±16.65.SPB was associated with the place of residence, monthly family income, payment method, transfer status, the presence of radiotherapy complications, and the presence of pain(P<0.05).The SPB and self-care self-efficacy were negatively correlated(P<0.01).In multivariate analysis,self-care self-efficacy,place of residence, monthly family income, payment method, degree of radiation dermatitis, and radiation proctitis were influencing factors of SPB(P<0.05).CONCLUSION Patients with cervical cancer undergoing radiotherapy often have SPB.Self-care self-efficacy scale, place of residence, monthly family income, payment method, and radiation dermatitis and proctitis are factors independently influencing SPB. | Ting Luo Rong-Zhi Xie Yan-Xia Huang Xiao-Hua Gong Hui-Ying Qin Yi-Xiao Wu | 2021 | World Journal of Clinical Cases2021,9,17: | 19

An HBV-encoded miRNA activates innate immunity to restrict HBV replication显示文摘We previously identified that hepatitis B virus(HBV)encodes a microRNA(HBV-miR-3)that restrains HBV replication by targeting the HBV transcript. However, whether HBV-miR-3 affects host innate immunity to modulate HBV replication remains unclear. Here,we examined the vital functions of HBV-miR-3 in the innate immune response after HBV infection. We found that HBV-miR-3 expression gradually increased in a dose-and time-dependent manner in HBV-infected HepG2-NTCP cells.HBV-miR-3 activated the JAK/STAT signaling pathway by downregulating SOCS5 in hepatocytes, thereby enhancing the IFN-induced anti-HBV effect. In addition,HBVmiR-3 in exosomes facilitated the Ml polarization of macrophages.Furthermore, exosomes containing HBV-miR-3 enhanced the secretion of IL-6 via inhibiting the SOCS5-mediated ubiquitination of EGFR.In short, these results demonstrate that HBV-miR-3 activates the innate immune response to restrain HBV replication by multiple pathways, which may suppress HBV-induced acute liver cell injury and affect the progression of persistent HBV infection. | Xiaoqing Zhao Lu Sun Ting Mu Jianying Yi Chaoqun Ma Hong Xie Min Liu Hua Tang | 2020 | Journal of Molecular Cell Biology2020,12,4: | 17
De Novo Plant Genome Assembly Based on Chromatin Interactions: A Case Study of Arabidopsis thaliana显示文摘Dear Editor, Genome assembly is typically a two-stage process:paired sequencing reads are joined into contigs, and contigs are assembled into scaffolds.Over the past 10 years,next-generation sequencing (NGS) technologies have become inexpensive, routine, and widespread. As a result, most plant genomes that are sequenced by NGS produce 'drafts' that are always contig or scaffold maps remaining in a more or less advanced stage of completion (Claros et al.,2012).Slow advances in the technology for the de novo assembly of scaffolds into chromosome-scale scaffolds have become the main restriction to the completion of and high-quality genome sequence acquisition. | Ting Xie | 2015 | Molecular Plant2015,8,3: | 15
Integrated(de)multiplexer for orbital angular momentum fiber communication显示文摘The quickly increasing data transfer load requires an urgent revolution in current optical communication. Orbital angular momentum(OAM) multiplexing is a potential candidate with its ability to considerably enhance the capacity of communication. However, the lack of a compact, efficient, and integrated OAM(de)multiplexer prevents it from being widely applied. By attaching vortex gratings onto the facets of a few-mode fiber, we demonstrate an integrated fiber-based OAM(de)multiplexer. A vortex grating fabricated on the fiber facet enables the direct multiplexing of OAM states at one port and the demultiplexing of OAM states at the other port. The measured bit error rate of the carrier signal after propagating through a 5-km few-mode fiber confirms the validity and effectiveness of the proposed approach. The scheme offers advantages in future high-capacity OAM communication based on optical fiber. | ZHENWEI XIE SHECHENG GAO TING LEI SHENGFEI FENG YAN ZHANG FAN LI JIANBO ZHANG ZHAOHUI LI XIAOCONG YUAN | 2018 | Photonics Research2018,6,7: | 13
Role of pyroptosis in inflammation and cancer显示文摘Pyroptosis is a form of programmed cell death mediated by gasdermin and is a product of continuous cell expansion until the cytomembrane ruptures, resulting in the release of cellular contents that can activate strong inflammatory and immune responses.Pyroptosis,an innate immune response, can be triggered by the activation of inflammasomes by various influencing factors.Activation of these inflammasomes can induce the maturation of caspase-1 or caspase-4/5/11,both of which cleave gasdermin D to release its N-terminal domain, which can bind membrane lipids and perforate the cell membrane. Here,we review the latest advancements in research on the mechanisms of pyroptosis, newly discovered influencing factors, antitumoral properties, and applications in various diseases. Moreover, this review also provides updates on potential targeted therapies for inflammation and cancers, methods for clinical prevention, and finally challenges and future directions in the field. | Xiang Wei Feng Xie Xiaoxue Zhou Yuchen Wu Haiyan Yan Ting Liu Jun Huang Fangwei Wang Fangfang Zhou Long Zhang | 2022 | Cellular & Molecular Immunology2022,19,9: | 11
Mode-locked 2.8-μm fluoride fiber laser:from soliton to breathing pulse显示文摘The mode-locked fluoride fiber laser(MLFFL)is an exciting platform for directly generating ultrashort pulses in the mid-infrared(mid-IR).However,owing to difficulty in managing the dispersion in fluoride fiber lasers,MLFFLs are restricted to the soliton regime, hindering pulse-energy scaling. We overcame the problem of dispersion management by utilizing the huge normal dispersion generated near the absorption edge of an infrared-bandgap semiconductor and promoted MLFFL from soliton to breathing-pulse mode-locking.In the breathing-pulse regime, the accumulated nonlinear phase shift can be significantly reduced in the cavity, and the pulse-energy-limitation effect is mitigated. The breathing-pulse MLFFL directly produced a pulse energy of 9.3 nJ and pulse duration of 215 fs,with a record peak power of 43.3 kW at 2.8μm.Our work paves the way for the pulse-energy and peak-power scaling of mid-IR fluoride fiber lasers, enabling a wide range of applications. | Zhipeng Qin Guoqiang Xie Hongan Gu Ting Hai Peng Yuan Jingui Ma Liejia Qian | 2019 | Advanced Photonics2019,1,6: | 9

Curcumenol triggered ferroptosis in lung cancer cells via lncRNA H19/miR-19b-3p/FTH1 axis显示文摘Curcumenol,an effective ingredient of Wenyujin, has been reported that exerted its antitumor potential in a few cancer types. However, the effect and molecular mechanism of curcumenol in lung cancer are largely unknown. Here,we found that curcumenol induced cell death and suppressed cell proliferation in lung cancer cells.Next,we demonstrated that ferroptosis was the predominant method that contributed to curcumenol-induced cell death of lung cancer in vitro and vivo for the first time.Subsequently, using RNA sequencing,we found that the long non-coding RNA H19(lncRNA H19)was significantly downregulated in lung cancer cells treated with curcumenol, when compared to untreated controls.Overexpression of lncRNA H19 eliminated the anticancer effect of curcumenol, while lncRNA H19 knockdown promoted ferroptosis induced by curcumenol treatment.Mechanistically,we showed that lncRNA H19 functioned as a competing endogenous RNA to bind to miR-19b-3p,thereby enhanced the transcription activity of its endogenous target, ferritin heavy chain 1(FTH1),a marker of ferroptosis. In conclusion, our data show that the natural product curcumenol exerted its antitumor effects on lung cancer by triggering ferroptosis, and the lncRNA H19/miR-19b-3p/FTH1 axis plays an essential role in curcumenol-induced ferroptotic cell death. Therefore, our findings will hopefully provide a valuable drug for treating lung cancer patients. | Ruonan Zhang Ting Pan Yu Xiang Mingming Zhang Han Xie Zimao Liang Bi Chen Cong Xu Jing Wang Xingxing Huang Qianru Zhu Ziming Zhao Quan Gao Chengyong Wen Wencheng Liu Weirui Ma Jiao Feng Xueni Sun Ting Duan Elaine Lai-Han Leung Tian Xie Qibiao Wu Xinbing Sui | 2022 | Bioactive Materials2022,7,7: | 8
Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear. Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks. Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. Women with comorbidities were more likely to have severe disease, while men with comorbidities were more likely to have unfavorable treatment outcomes. | MA Yan ZHU Dong Shan CHEN Ren Bo SHI Nan Nan LIU Si Hong FAN Yi Pin WU Gui Hui YANG Pu Ye BAI Jiang Feng CHEN Hong CHEN Li Ying FENG Qiao GUO Tuan Mao HOU Yong HU Gui Fen HU Xiao Mei HU Yun Hong HUANG Jin HUANG Qiu Hua HUANG Shao Zhen JI Liang JIN Hai Hao LEI Xiao LI Chun Yan LI Min Qing LI Qun Tang LI Xian Yong LIU Hong De LIU Jin Ping LIU Zhang MA Yu Ting MAO Ya MO Liu Fen NA Hui WANG Jing Wei SONG Fang Li SUN Sheng WANG Dong Ting WANG Ming Xuan WANG Xiao Yan WANG Yin Zhen WANG Yu Dong WU Wei WU Lan Ping XIAO Yan Hua XIE Hai Jun XU Hong Ming XU Shou Fang XUE Rui Xia YANG Chun YANG Kai Jun YUAN Sheng Li ZHANG Gong Qi ZHANG Jin Bo ZHANG Lin Song ZHAO Shu Sen ZHAO Wan Ying ZHENG Kai ZHOU Ying Chun ZHU Jun Teng ZHU Tian Qing ZHANG Hua Min WANG Yan Ping WANG Yong Yan 无 | 2020 | Biomedical and Environmental Sciences2020,33,12: | 8
Combined prednisone and levothyroxine improve treatment of severe thrombocytopenia in hepatitis B with compensatory cirrhosis accompanied by subclinical and overt hypothyroidism显示文摘The aim of the present study was to investigate the relationship between hypothyroidism and thrombocytopenia in hepatitis Brelated compensatory liver cirrhosis and to determine whether treatment with levothyroxine and prednisone is superior in a multicenter, open-label, observational study in China. In total, 125 consecutive hepatitis B-related compensated liver cirrhosis patients with severe thrombocytopenia accompanied by hypothyroidism were included. The patients were divided into four groups according to treatment strategy: a control group(n=29), a prednisone group(n=25), a levothyroxine group(n=32) and a prednisone plus levothyroxine group(n=39). Severe thrombocytopenia was more prevalent in hepatitis B-associated compensatory liver cirrhosis patients with hypothyroidism than in euthyroid patients(29.6% vs. 14.7%, P<0.05). Combination treatment with prednisone and levothyroxine decreased the risk of bleeding and improved platelet recovery compared to control treatment and treatment with either prednisone or levothyroxine alone. The platelet count before therapy, serum thyroid stimulating hormone and combination treatment with prednisone and levothyroxine were associated with bleeding events. Therefore, the present study suggests that hypothyroidism is associated with severe thrombocytopenia in hepatitis B-associated compensatory liver cirrhosis. Treatment with prednisone plus levothyroxine may present a novel approach in these patients. | Jing Xue Ru Feng Haixia Fu Qian Jiang Hao Jiang Jing Lu Hui Liu Jingwen Wang Ting Niu Xin Wang YANDi Xie Hao Wang Lanping Xu Kaiyan Liu Xiaojun Huang Xiaohui Zhang | 2018 | Science China(Life Sciences)2018,61,8: | 8
Cholesterol transport through the peroxisome-ER membrane contacts tethered by PI(4,5)P2 and extended synaptotagmins显示文摘Most mammalian cells take up cholesterol from low-density lipoproteins(LDLs) via receptor-mediated endocytosis. After reaching lysosomes,LDL-derived cholesterol continues to transport to downstream organelles including the ER for specific structural and functional needs.Peroxisomes are recently found to receive cholesterol from lysosomes through lysosomeperoxisome membrane contacts. However, whether and how cholesterol is conveyed from peroxisomes to the ER remain unknown. Here,by combining high-resolution microscopic analyses and in vitro reconstitution of highly purified organelles or artificial liposomes,we demonstrate that peroxisomes form membrane contacts with the ER through the interaction between peroxisomal PI(4,5)P2 and ER-resident extended synaptotagmin-1,2 and 3(E-Syts).Depletion of peroxisomal PI(4,5)P2 or ESyts markedly decreases peroxisome-ER membrane contacts and induces cholesterol accumulation in lysosomes.Furthermore,we show that cholesterol is delivered from 3H-labeled peroxisomes or PI(4,5)P2-containing liposomes to the ER in vitro, and that the presence of peroxisomes augments cholesterol transfer from lysosomes to the ER.Together, our study reveals a new cholesterol transport pathway along the lysosome-peroxisome-ER membrane contacts in the cell. | Jian Xiao Jie Luo Ao Hu Ting Xiao Meixin Li Zekai Kong Luyi Jiang Zimu Zhou Yacheng Liao Chang Xie Beibei Chu Honghua Miao Boliang Li Xiongjie Shi Bao-Liang Song | 2019 | Science China(Life Sciences)2019,62,9: | 8

Historical trends of PCBs and PBDEs as reconstructed in a lake sediment from southern Tibetan Plateau显示文摘High-altitude lake sediment can be used as a natural archive to reconstruct the history of pollutants. In this work, the temporal distribution of polychlorinated biphenyls(PCBs) and polybrominated diphenyl ethers(PBDEs) were determined using a high-resolution gas chromatography coupled with high-resolution mass spectrometer(HRGC/HRMS) in an alpine lake sediment core collected from the southern Tibetan Plateau(TP) to examine whether the expected decreasing trends due to the implementation of the international Conventions could be observed. The concentrations of PCBs and PBDEs in the sediment core were in the range of 11.8–142 pg/g dw and ND-457 pg/g dw, and their fluxes were in the range of 2.51–31.7 ng/(m^2 ·yr) and ND-43.2 ng/(m^2 ·yr), respectively. The prevalence of low-chlorinated(tri-CB) PCBs and low-brominated(tri-to tetra-) PBDEs in most sections of the sediment profiles was observed, suggesting that the light molecular weight PCBs and PBDEs have most likely reached lake sediments by long-range atmospheric transport from distant sources. Despite the restrictions on their applications, the sediment records for the concentrations and fluxes showed no corresponding decreasing trend with restrictions for PCBs, which suggested that these POPs(e.g., PCBs) were still emitted to the environment owing to the influence of primary or secondary emissions. To our knowledge, this is the first report on input history of atmospheric PCBs and PBDEs recorded in TP Lake sediment. | Ruiqiang Yang Ting Xie Pu Wang Yingming Li Qinghua Zhang Guibin Jiang | 2020 | Journal of Environmental Sciences2020,32,12: | 7
