The World Health Organization's (WHO) Global Tuberculosis Report 2018[1] has reported that in 2017, 10.0 million people across the world had developed TB diseases that resulted in an estimated 1.6 million deaths, and 889, 000 people developed TB in China that led to 39, 000 TB-related deaths. Therefore, rapid and accurate detection of Mycobacterium tuberculosis (MTB) is important for initiating early treatment and reducing mortality. | 2 | Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. | 3 | Prevalence and Risk Factors of Primary Drug-Resistant Tuberculosis in China显示文摘Objective To investigate the prevalence of primary drug-resistant tuberculosis(TB)and associated risk factors in China.We also explored factors contributing to the transmission of multidrug-resistant tuberculosis(MDR-TB).Methods A total of 2794 representative,Mycobacterium tuberculosis isolates from treatment-naive patients were subjected to drug susceptibility testing,and risk factors for drug-resistant TB were analyzed.We also analyzed MDR-TB strain sublineages,drug-resistance-conferring mutations,and risk factors associated with clustered primary MDR strains.Results Among 2794 Mycobacterium tuberculosis isolates from treatment-naive patients,the prevalence of any resistance to first-line drugs was 33.2%and the prevalence of MDR-TB was 5.7%.We did not find any risk factors significantly associated with resistance to first-line drugs.The 93 primary MDR-TB isolates were classified into six sublineages,of which,75(80.6%)isolates were the RD105-deleted Beijing lineage.The largest sublineage included 65(69.9%)isolates with concurrent deletions of RD105,RD207,and RD181.Twenty-nine(31.2%)primary MDR strains grouped in clusters;MDR isolates in clusters were more likely to have S531L rpoB mutation.Conclusion This study indicates that primary drug-resistant TB and MDR-TB strains are prevalent in China,and multiple measures should be taken to address drug-resistant TB. | WANG Sheng Fen ZHOU Yang PANG Yu ZHENG Hui Wen ZHAO Yan Lin | 2016 | Biomedical and Environmental Sciences2016,29,2: | 6 |
| 4 | The EGFR-P38 MAPK axis up-regulates PD-L1 through miR-675-5p and down-regulates HLA-ABC via hexokinase-2 in hepatocellular carcinoma cells显示文摘Background:Immunotherapy has been shown to be a promising strategy against human cancers.A better understanding of the immune regulation in hepatocellular carcinoma(HCC)could help the development of immunotherapy against HCC.The epidermal growth factor receptor(EGFR)signaling is frequently activated in HCC and plays important roles in tumorigenesis.However,its role in HCC immunity is still largely unknown.This study aimed to investigate the impact of EGFR signaling on programmed death-ligand 1(PD-L1)and human leukocyte antigen class-I(HLA-I)expression in HCC cells and its underlying mechanisms.Methods:The expression of phosphorylated EGFR(p-EGFR),PD-L1,and HLAI(HLA-ABC)in HCC specimens was detected by immunohistochemistry,and their correlations were analyzed.PD-L1 and HLA-ABC expression in EGFRactivated HCC cells were detected by quantitative real-time PCR,Western blotting,and flow cytometry,and T cell-mediated lysis was performed to test the immunosuppressive effects of PD-L1 and HLA-ABC alterations in HCC cells.Furthermore,the underlying mechanisms of EGFR activation-induced PD-L1 up-regulation and HLA-ABC down-regulation were explored by animal experiments,luciferase reporter assay,and gene gain-and loss-of-function studies.Results:p-EGFR was positively correlated with PD-L1 and negatively correlated with HLA-ABC expression in HCCs.EGFR activation by its ligand EGF up-regulated PD-L1 and down-regulated HLA-ABC in HCC cells,which was functionally important and could be abolished by the EGFR inhibitor,gefitinib,both in vitro and in vivo.Mechanistically,enhanced P38 mitogenactivated protein kinase(MAPK)activation down-regulated microRNA-675-5p(miR-675-5p)and up-regulated glycolysis-related enzyme hexokinase 2(HK2);miR-675-5p down-regulation enhanced the stability of PD-L1 mRNA probably via the 3’-untranslated region(3’-UTR)of PD-L1 and thereby caused PD-L1 accumulation,and HK2 up-regulation enhanced aerobic glycolysis and mediated a decrease in HLA-ABC.Conclusions:The EGFR-P38 MAPK axis could up-regulate PD-L1 through miR-675-5p and down-regulate HLA-ABC via HK2 in HCC cells.Our study reveals a novel signaling network that may cause immune suppression in HCC and suggests that EGFR signaling can be targeted for HCC immunotherapy. | Zongcai Liu Fen Ning Yanna Cai Huiying Sheng Ruidan Zheng Xi Yin Zhikun Lu Ling Su Xiaodan Chen Chunhua Zeng Haifang Wang Li Liu | 2021 | Cancer Communications2021,41,1: | 5 |
| 5 | The Changing Trends of HIV/AIDS in An Ethnic Minority Region of China: Modeling the Epidemic in Liangshan Prefecture, Sichuan Province显示文摘Objective This study was to investigate the HIV current situation in Liangshan prefecture, in order to predict prevalence and transmission trends. Methods Region-specific population, behavior, serosurveillence, and policy/program data (from 1995 to 2010) were gathered from various local and national organizations and applied to the Asian Epidemic Model (AEM) and used to derive estimates of future HIV prevalence, epidemic trends, and outcomes of intervention strategies. Results The AEM projections for 2020 included increased number of people living with HIV (PLHIV; to 136 617), increased HIV prevalence (2.51%), and 8037 deaths from acquired immunodeficiency syndrome (AIDS) in this region. However, the overall HIV incidence rate (per 10 000) was projected to decline from 27 in 2015 to 22 in 2020, largely due to a predicted decrease in HIV infection rate (per 10 000) from 658 in 2013 to 621 in 2020 among intravenous drug users. In contrast, the cases of HIV infection per 10 000 was projected to increase from 420 in 2010 to 503 in 2020 among men who have sex with men, and from 8 in 2010 to 15 in 2020 among the general population. The predominant risk factor for HIV transmission over the next decade in Liangshan was casual sex. Community-based outreach strategies to reduce injected drug use and casual sex, and to promote condom use, were predicted as effective interventions to decrease HIV transmission. | 6 | Fe_(2)TiO_(5)nanochains as anode for high-performance lithium-ion capacitor显示文摘The unique crystal structure and multiple redox couples of iron titanate(Fe_(2)TiO_(5)) provide it a high theoretical capacity and good cycling stability when used as an electrode. In this study, the electrospinning method is employed to synthesize one-dimensional(1 D) Fe_(2)TiO_(5) nanochains. The as-prepared Fe_(2)TiO_(5) nanochains exhibited superior specific capacity(500 mAh·g^(-1) at 0.10 A·g^(-1)),excellent rate performance(180 mAh·g^(-1) at 5.00 A·g^(-1)),and good cycling stability(retaining 100% of the initial specific capacity at a current density of 1.00 A·g^(-1) after1000 cycles). The as-assembled Fe_(2)TiO_(5)/SCCB lithiumion capacitor(LIC) also delivered a competitive energy density(137.8 Wh·kg^(-1))andpowerdensity(11,250 W·kg^(-1)). | 7 | Genetic Diversity and Drug Susceptibility of Mycobacterium tuberculosis Isolates in a Remote Mountain Area of China显示文摘Objective We determined the genetic diversity of Mycobacterium tuberculosis(MTB) in a remote mountainous area of southwest China and evaluated the resolving ability of single nucleotide polymorphism(SNP) genotyping combined with variable number tandem repeat(VNTR) genotyping for Beijing family strains in association with drug resistance status.Methods Three hundred thirty-one MTB strains were isolated from patients living in mountainous regions of southwest China,and 8-loci SNP,VNTR-15 genotyping assays,and drug susceptibility testing of 9 drugs were performed.Results A total of 183 [55.29%(183/331)] strains were classified into the Beijing family.Of the 183 strains,111(60.66%) were defined as modern Beijing strains.The most predominant modern Beijing sub-lineage and ancient Beijing sub-lineage were Bmyc10 [39.34%(72/183)] and Bmyc25 [20.77%(38/183)],respectively.Of the isolates,19.64%(65/331) were resistant to at least 1 of the 9 anti-TB drugs and 17 [4.98%(17/331)] MTB isolates were multi-drug resistant tuberculosis(MDR-TB).Two hundred sixty-one isolates showed a clustering rate of 14.18%(37/261) and a discriminatory index of 0.9990.The Beijing lineage exhibited a significantly higher prevalence of MDR-TB,as well as resistance to isoniazid(INH),rifampin(RIF),and para-aminosalicylic acid(PAS) when analyzed independently(P = 0.005,P = 0.017,P = 0.014,and P = 0.006 respectively).The Beijing lineage was not associated with genetic clustering or resistance to any drug.In addition,genetic clustering was not associated with drug resistance.Conclusion MTB strains demonstrate high genetic diversity in remote mountainous areas of southwest China.Beijing strains,especially modern Beijing strains,are predominant in remote mountainous area of China.The combination of 8-loci SNPs and VNTR-15 genotyping is a useful tool to study the molecular epidemiology of MTB strains in this area. | ma ai jing wang sheng fen fan jia le zhao bing he guang xue zhao yan lin | 2018 | Biomedical and Environmental Sciences2018,31,5: | 2 |
| 8 | A score model for predicting post-liver transplantation survival in HBV cirrhosis-related hepatocellular carcinoma recipients: a single center 5-year experience显示文摘BACKGROUND: The prognostic prediction of liver transplantation(LT) guides the donor organ allocation. However, there is currently no satisfactory model to predict the recipients' outcome, especially for the patients with HBV cirrhosis-related hepatocellular carcinoma(HCC). The present study was to develop a quantitative assessment model for predicting the post-LT survival in HBV-related HCC patients.METHODS: Two hundred and thirty-eight LT recipients at the Liver Transplant Center, First Affiliated Hospital, Zhejiang University School of Medicine between 2008 and 2013 were included in this study. Their post-LT prognosis was recorded and multiple risk factors were analyzed using univariate and multivariate analyses in Cox regression.RESULTS: The score model was as follows: 0.114×(Child-Pugh score)-0.002×(positive HBV DNA detection time)+0.647×(number of tumor nodules)+0.055×(max diameter of tumor nodules)+0.231×ln AFP+0.437×(tumor differentiation grade).The receiver operating characteristic curve analysis showed that the area under the curve of the scoring model for predicting the post-LT survival was 0.887.
