1Characterization of dissolved organic matter fractions from Lake Hongfeng, Southwestern China Plateau显示文摘With XAD-series and ion exchange resins, dissolved organic matter (DOM) from Lake Hongfeng in Southwestern China Plateau was isolated into 6 fractions, i.e., humic acid (HA), fulvic acid (FA), hydrophobic neutrals (HON), hydrophilic acids (HIA), hydrophilic bases (HIB) and hydrophilic neutrals (HIN). Those fractions were characterized by high performance size exclusion chromatography, fluorescence spectroscopy and UV absorbance. Among the 6 fractions, FA was predominant and accounted for 51% of the total DOM. The weight-average (Mw) and number-averaged (Mn) molecular weight of these fractions ranged from 1688 to 2355 Da and from 1338 to 1928 Da, respectively. A strong correlation was observed between specific UV absorbance at 280 nm, E2/E3 (absorbance at 250 nm to 365 nm), and the molecular weight for DOM fractions. UV-Vis fulvic-like fluorescence peaks were found in all fractions. Protein-like fluorescence peaks existed in HON may indicate that microbial activity was severely in Lake Hongfeng. There was a significant relationship between fluorescence intensities and specific UV absorbance at 254 nm for those DOM fractions, suggesting their similar luminescence characteristics. The values of fluorescence index (f450/500) indicated that hydrophobic fractions may derive from terrestrial sources, and the hydrophilic fractions from microbial and terrestrial origins. 3Binding characteristics of perylene, phenanthrene and anthracene to different DOM fractions from lake water显示文摘Six hydrophobic and hydrophilic fractions were isolated using XAD-8 and XAD-4 resins, and were extensively characterized. Partition coefficients of perylene, phenanthrene and anthracene binding to the six fractions were determined by fluorescence quenching titration. The Kdoc values obtained for the polycyclic aromatic hydrocarbons (PAHs) binding to the hydrophobic fractions were larger than those to the hydrophilic fractions. Nonlinear Stern-Volmer plots were observed when binding phenanthrene and anthracene to some hydrophilic fraction samples, suggesting saturation of polar interaction binding sites. A significant correlation of logKdoc values with molecular weights and molar absorptivities at 280 nm was observed, while atomic ratio of C/H was found to be a poor indicator for aromaticity. Other structural descriptors such as paraffinic carbon and polarity influenced the DOM-fraction ability to bind PAHs. 5Construction and Count of 1-Resilient Rotation Symmetric Boolean Functions on p^r Variables显示文摘Construction and count of 1-resilient Rotation symmetric Boolean functions(RSBFs) on p r variables are demonstrated. It is proved that constructions of1-resilient RSBFs on p r variables are equivalent to solving an equation system. An accurate enumeration formula of all 1-resilient RSBFs on p r variables is also proposed. 6Antiproliferative effects of cinobufacini on human hepatocellular carcinoma HepG2 cells detected by atomic force microscopy显示文摘AIM:To investigate the antiproliferative activity of cinobufacini on human hepatocellular carcinoma HepG2 cells and the possible mechanism of its action.METHODS:HepG2 cells were treated with different concentrations of cinobufacini.Cell viability was measured by methylthiazolyl tetrazolium(MTT) assay.Cell cycledistribution was analyzed by flow cytometry(FCM).Cytoskeletal and nuclear alterations were observed by fluorescein isothiocyanate-phalloidin and DAPI staining under a laser scanning confocal microscope.Changes in morphology and ultrastructure of cells were detected by atomic force microscopy(AFM) at the nanoscale level.RESULTS:MTT assay indicated that cinobufacini significantly inhibited the viability of HepG2 cells in a dosedependent manner.With the concentration of cinobufacini increasing from 0 to 0.10 mg/m L,the cell viability decreased from 74.9% ± 2.7% to 49.41% ± 2.2% and 39.24% ± 2.1%(P < 0.05).FCM analysis demonstrated cell cycle arrest at S phase induced by cinobufacini.The immunofluorescence studies of cytoskeletal and nuclear morphology showed that after cinobufacini treatment,the regular reorganization of actin filaments in HepG2 cells become chaotic,while the nuclei were not damaged seriously.Additionally,high-resolution AFM imaging revealed that cell morphology and ultrastructure changed a lot after treatment with cinobufacini.It appeared as significant shrinkage and deep pores in the cell membrane,with larger particles and a rougher cell surface.CONCLUSION:Cinobufacini inhibits the viability of HepG2 cells via cytoskeletal destruction and cell membrane toxicity.Qing Wu Wei-Dong Lin Guan-Qun Liao Li-Guo Zhang Shun-Qian Wen Jia-Ying Lin 2015World Journal of Gastroenterology2015,21,3:12
7Aspartic acid in the hippocampus:a biomarker for postoperative cognitive dysfunction显示文摘This study established an aged rat model of cognitive dysfunction using anesthesia with 2% isoflurane and 80% oxygen for 2 hours. Twenty-four hours later, Y-maze test results showed that isoflurane significantly impaired cognitive function in aged rats. Gas chromatography-mass spectrometry results showed that isoflurane also significantly increased the levels of N,N-diethylacetamide, n-ethylacetamide, aspartic acid, malic acid and arabinonic acid in the hippocampus of isoflurane-treated rats. Moreover, aspartic acid, N,N-diethylacetamide, n-ethylacetamide and malic acid concentration was positively correlated with the degree of cognitive dysfunction in the isoflurane-treated rats. It is evident that hippocampal metabolite changes are involved in the formation of cognitive dysfunction after isoflurane anesthesia. To further verify these results, this study cultured hippocampal neurons in vitro, which were then treated with aspartic acid(100 μmol/L). 8Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 12Laparoscopy-assisted total gastrectomy with trans-orally inserted anvil(OrVil^(TM)):A single institution experience显示文摘AIM:To investigate the feasibility of laparoscopyassisted total gastrectomy(LATG)using trans-orally inserted anvil(OrVilTM)in terms of operative characteristics and short term outcomes. RESULTS:Characteristics of 27 patients with gastric cancer who underwent LATG from October 2009 to October 2012 in the Foshan Affiliated Hospital of South Medical University were retrospectively reviewed. Among these patients,six were reconstructed by minilaparotomy and 21 by OrVilTM.The clinicopathological characteristics,total operation time,total blood loss, abdominal incision and complications of anastomosis including stenosis and leakage,were compared between the groups undergoing LATG with OrVilTM and the group undergoing minilaparotomy. RESULTS:The operations were successfully performed on all the patients without intraoperative complications or conversion to open surgery.Two(10%)patients received palliative procedure under laparoscope who were prepared for LATG preoperatively.One case had hepatic metastatic carcinoma and 1 case had tumor recurrence near the anastomosis 8 mo after surgery.The mean follow-up duration was 10 mo(range,2-24 mo). Operation time was significantly reduced by the use of OrVilTM(198.42±30.28 min vs 240.83±8.23 min). The postoperative course with regard to occurrence of stenosis and leakage was not different between the two groups.There were no significant differences in estimated blood loss.The upper abdominal incision was smaller in OrVilTM group than in minilaparotomy group (4.31±0.45 cm vs 6.43±0.38 cm). 13Effects of Water-Collecting and -Retaining Techniques on Photosynthetic Rates, Yield, and Water Use Efficiency of Millet Grown in a Semiarid Region显示文摘Field experiments were conducted in 2003 and 2004 to study the effects of plastic ridges and furrow film mulching (plastic film on sowing, as well as plastic film on flat soil and hole sowing) and chemicals (a drought resistant agent and a water- retaining agent) on growth, photosynthetic rate, yield, and water use efficiency (WUE) of spring millet (Setaria italica L.). The experimental results showed that water-collecting and -retaining techniques can effectively increase soil moisture content, the leaf photosynthetic rate and crop growth. Due to increased soil moisture under the plastic-covered ridge and furrow water-collecting in July and August, dry matter and plant height had a increase at the booting stage (late growth advantage). However, the plastic-covered flat soil and hole sowing reduced soil evaporation during early growth, the increase of dry matter and plant height appeared at the seedling stage (early growth advantage). Plastic-covered ridge and furrow sowing supplemented with chemical reagents had significant positive effects on water collection and soil moisture retention. Improvement of soil moisture resulted into the increase of the photosynthetic rate, dry matter accumulation yield and WUE. The water-collecting and -retaining techniques can improve WUE and enhance crop yield. Correlation analysis demonstrated that the photosynthetic rate under the water-collecting and -retaining techniques was significantly associated with the soil moisture, but had no significant relationship with leaf chlorophyll content. 14Defined tumor antigen-specific T cells potentiate personalized TCR-T cell therapy and prediction of immunotherapy response显示文摘Personalized immunotherapy targeting tumor-specific antigens(TSAs)could generate efficient and safe antitumor immune response without damaging normal tissues.Although neoantigen vaccines have shown therapeutic effect in clinic trials,precise prediction of neoantigens from tumor mutations is still challenging.The host antitumor immune response selects and activates T cells recognizing tumor antigens.Hence,T cells engineered with T-cell receptors(TCRs)from these naturally occurring tumor antigen-specific T(Tas)cells in a patient will target personal TSAs in his/her tumor.To establish such a personalized TCR-T cell therapy,we comprehensively characterized T cells in tumor and its adjacent tissues by single-cell mRNA sequencing(scRNA-seq),TCR sequencing(TCR-seq)and in vitro neoantigen stimulation.Compared to bystander T cells circulating among tissues,Tas cells were characterized by tumor enrichment,tumor-specific clonal expansion and neoantigen specificity.We found that CXCL13 is a unique marker for both CD4^(+)and CD8^(+)Tas cells.Importantly,TCR-T cells expressing TCRs from Tas cells showed significant therapeutic effects on autologous patient-derived xenograft(PDX)tumors.Intratumoral Tas cell levels measured by CXCL13 expression precisely predicted the response to immune checkpoint blockade,indicating a critical role of Tas cells in the antitumor immunity.We further identified CD200 and ENTPD1 as surface markers for CD4^(^(+))and CD8^(^(+))Tas cells respectively,which enabled the isolation of Tas cells from tumor by Fluorescence Activating Cell Sorter(FACS)sorting.Overall,our results suggest that TCR-T cells engineered with Tas TCRs are a promising agent for personalized immunotherapy,and intratumoral Tas cell levels determine the response to immunotherapy.Jingjing He Xinxin Xiong Han Yang Dandan Li Xuefei Liu Shuo Li Shuangye Liao Siyu Chen Xizhi Wen Kuai Yu Lingyi Fu Xingjun Dong Kaiyu Zhu Xiaojun Xia Tiebang Kang Chaochao Bian Xiang Li Haiping Liu Peirong Ding Xiaoshi Zhang Zhenjiang Liu Wende Li Zhixiang Zuo Penghui Zhou 2022Cell Research2022,32,6:10
15Intraperitoneal hemorrhage during and after percutaneous radiofrequency ablation of hepatic tumors: reasons and management显示文摘Background Introperitoneal hemorrhage is one of the mos t common complications of radiofrequency (RF) ablation of hepatic tumors. This study was designed to investigate the reason and management of intraperitoneal h emorrhage occurred during or after percutaneous RF ablation of hepatic tumors. Methods Three hundred and fifty-six patients with hepatic tumo rs have been treated at 592 procedures of ultrasound guided RF ablation. Intrap eritoneal hemorrhage occurred in 5 patients (0.8%). The reasons and management of intraperitoneal hemorrhage in these 5 cases were retrospectively analyzed. Results Two patients with liver metastasis and one hepatocellul ar carcinoma (HCC) patient suffered from hemorrhage during the RF treatment. Tw o patients with recurrent HCC after surgery developed hemorrhage 20 minutes or 4 hours after RF treatment. One case of hemorrhage was due to the inappropriate electrode positioning induced liver laceration while treating a 1 cm liver metas tasis near the liver capsule. One was due to the injury of a small vessel by th e RF needle in another liver metastasis patient. Three cases were due to tumor rupture with two cases induced by cough or position change after treating large protruding HCC lesions. Four (80%) of the 5 cases of hemorrhage were rapidly id e ntified by ultrasound. The causes and sites of bleeding during the RF treatment in three cases were confirmed through ultrasound, which were successfully treat ed using RF coagulation to achieve hemostasis of the bleeding site. Two patient s with post-ablation hemorrhage recovered in one hour and 24 hours, respectivel y after given blood transfusion and other conservative measures. No surgical in tervention was required. Two patients died of wide spread metastasis 23-36 mon ths afterwards and the other three patients have lived for 18-25 months to date . Conclusions It is important to perform close monitoring during and after RF ablation in order to identify intraperitoneal hemorrhage in time. RF ablation of the bleeding sites was a simple and effective management when the bleeding site could be confirmed by ultrasound. 18Risks faced by donors of right lobe for living donor liver transplantation显示文摘BACKGROUND:Because of the shortage of deceased donors with livers fit for transplantation,living donor liver transplantation(LDLT)is becoming an attractive alternative.Attention should be paid to the donors,especially to those of the right lobe.In this study,we evaluated the risks faced by donors of the right lobe for adult-to-adult LDLT.METHODS:The perioperative data from 105 consecutive living donors of the right lobe performed in West China Hospital from January 2002 to December 2007 were retrospectively studied.Preoperative evaluation included CT,MRCP,and intraoperative cholangiography,showing liver volume,hepatic vasculature and the biliary system.The standard liver volume(SLV)and the ratio of left lobe volume to SLV were calculated.The right lobe grafts were obtained by transecting the liver on the right side of the middle hepatic vein without inflow vascular occlusion,using an ultrasonic dissector.After operation the donors were monitored in the Intensive Care Unit for about three days.Each donor was followed up for at least 6 months.RESULTS:There was no donor mortality.Major complications occurred in 14 donors(13.3%),of whom 3 received conservative treatment,8 required invasive paracentesis,and 3 required further surgery.All donors were recovered well and resumed their previous occupations.CONCLUSIONS:Donors of the right lobe face low risks.The preoperative evaluation,especially evaluation of the volume of the remnant liver,should be exact.During the operation,the patency of the remnant hepatic vasculature and bile duct must be preserved,and the extent of injury to the remnant liver should be limited as much as possible.The detection and treatment of postoperative complications should be diligently performed.Ran, Shun Wen, Tian-Fu Yan, Lu-Nan Li, Bo Zeng, Yong Chen, Zhe-Yu Zhang, Yu Liao, Zhi-Xue Liang, Guan-Lin Li, Guo Zhang, Xian-Hua 2009Hepatobiliary & Pancreatic Diseases International2009,8,6:8
19Pulmonary embolism and deep vein thrombosis caused by nitrous oxide abuse: A case report显示文摘BACKGROUND Nitrous oxide(N2O)has gained increasing popularity as a recreational drug,causing hallucinations,excitation,and psychological dependence.However,side effects have been reported in recent years.Our case report proposes a correlation among N2O,pulmonary embolism(PE),and deep vein thrombosis(DVT)and emphasizes the role of homocysteine(Hcy)in thrombotic events.CASE SUMMARY A 29-year-old man with long-term N2O abuse sought evaluation after acute chest pain.A diagnostic workup revealed PE,DVT,and hyperhomocysteinemia.The patient was successfully treated with thrombolytic and anticoagulant therapy.Moreover,his Hcy level decreased and returned to normal after Hcy-lowering therapy.CONCLUSION Chronic N2O abuse might increase the risk of PE and DVT,although there have been few studies previously.Wen Sun Ji-Ping Liao Yan Hu Wei Zhang Jing Ma Guang-Fa Wang 2019World Journal of Clinical Cases2019,7,23:8
20THE VOID FRACTION DISTRIBUTION IN TWO-DIMENSIONAL GAS-LIQUID TWO PHASE FLOW USING IMAGE PROCESS METHOD显示文摘An image processing method for measuring the instantaneous two-dimensional (2D) local void fraction at high bubble number density was proposed. It was based on computing a correlation between the locally averaged brightness of the bubble plume images and the real void fraction. The method was tested using the 2D distribution of local void fraction from visualized image, where a correlation between the locally averaged brightness and the projection void fraction was derived by introducing an ideal of overlapping probability of the bubble-images under various conditions. This research was used to study the behavior of the bubbly flows with aspect ratio of 1.0 1.5, and 2.0. The present method was applied to the instantaneous measurement. The void fraction for bubble plumes of three different aspect ratios with three different bubble generating flow rates were measured for the clarification of the effect of the bubble on the behavior of the bubbly flows. Dependency of the void fraction on the gas flow rate was measured and compared to other results from the literature.
