ZBP1(DAI/DLM-1) promotes osteogenic differentiation while inhibiting adipogenic differentiation in mesenchymal stem cells through a positive feedback loop of Wnt/β-catenin signaling显示文摘The lineage specification of mesenchymal stem/stromal cells(MSCs) is tightly regulated by a wide range of factors. Recently, the versatile functions of ZBP1(also known as DAI or DLM-1) have been reported in the blood circulation and immune systems. However, the biological function of ZBP1 during the lineage specification of MSCs is still unknown. In the present study, we found that ZBP1 was upregulated during osteogenesis but downregulated during adipogenesis in mouse bone marrow-derived MSCs(m BMSCs). ZBP1 was highly expressed in osteoblasts but expressed at a relatively low level in marrow adipocytes. Knockdown of ZBP1 inhibited alkaline phosphataseactivity, extracellular matrix mineralization, and osteogenesis-related gene expression in vitro and reduced ectopic bone formation in vivo. Knockdown of ZBP1 also promoted adipogenesis in MSCs in vitro. Conversely, the overexpression of ZBP1 increased the osteogenesis but suppressed the adipogenesis of MSCs. When the expression of ZBP1 was rescued, the osteogenic capacity of ZBP1-depleted m BMSCs was restored at both the molecular and phenotypic levels.Furthermore, we demonstrated that ZBP1, a newly identified target of Wnt/β-catenin signaling, was required for β-catenin translocation into nuclei. Collectively, our results indicate that ZBP1 is a novel regulator of bone and fat transdifferentiation via Wnt/β-catenin signaling. | Xuefeng Zhao Liang Xie Zhiyong Wang Jiongke Wang Hao Xu Xianglong Han Ding Bai Peng Deng | 2020 | Bone Research2020,8,2: | 8
Rapid in-situ reaction synthesis of novel TiC and carbon nanotubes reinforced titanium matrix composites显示文摘In-situ TiC and remained multi-walled carbon nanotubes(MWCNTs) reinforced Ti composites were synthesized using vacuum hot-press sintering and hot rolling. The effect of weight fraction of MWCNTs on microstructural evolution and mechanical properties of the Ti composites was investigated. The results indicated that both proportion and particle size of TiC increased in proportion to MWCNTs content, which resulted in different matrix microstructure, and the grains were obviously refined after rolling deformation. The hardness tests indicated that MWCNTs addition could make the composites harden, and 18.4%improvement in hardness was obtained after hot rolling. The significant improvement in both strength and hardness could be attributed to grain refinement, solid solution strengthening of carbon and dispersion strengthening of TiC particles and remained MWCNTs. A good combination of strength and ductility were achieved in Ti–1 wt% MWCNTs composites, which were in accordance with the uniform distribution of smaller-sized TiC particles in Ti matrix. | Xianglong Sun Yuanfei Han Sanchen Cao Peikun Qiu Weijie Lu | 2017 | Journal of Materials Science & Technology2017,33,10: | 6
Tephra records from abyssal sediments off western Sumatra in recent 135 ka: evidence from Core IR-GC1显示文摘Three volcanic ash layers were identified in a deep-sea Core IR-GC1 from the north-eastern Indian Ocean, adjacent to western Indonesian arc. They were dominated by glass shards with minor mineral crystals, such as plagioclase, biotite, and hornblende. According to the morphology and major element compositions of the representative glass shards, combined with the δ18O-based age, it is suggested that ash Layer A is correlated to the youngest Toba tuff(YTT), Layer B is supposed to be associated with a new eruption of Toba caldera in an age of 98 to 100 ka. Ash Layer C is different the geochemistry characteristics than those of Layer A and Layer B, suggesting that Layer C was not originated from Toba but registered another volcanic eruption event. | QIU Zhongyan HAN Xiqiu JIN Xianglong WANG Yejian ZHU Jihao | 2014 | Acta Oceanologica Sinica2014,33,12: | 1

Three-dimensional FEM analysis of stress distribution in dynamic maxillary canine movement显示文摘An alteration in stress/strain distribution within periodontal ligament (PDL) occurs when the tooth is mechanical stimulated in orthodontic treatment. The majority of previous finite element studies were confined to a static evaluation on initial loading status, and a dynamic long-term analysis was rarely performed in orthodontics. The present study sought to analyze the stress distribution patterns within tooth root, PDL, and alveolar bone during dynamic canine tipping and bodily movement with three-dimensional finite element method in four consecutive therapeutic weeks. We observed that the highest stress was on root surface, secondly alveolar bone, and finally, PDL for either tipping or bodily movement. Throughout the PDL and root surface, the highest stress was always around the cervical area in both patterns of movement through four phases. Moreover, a lower and more uniform stress distribution was produced during bodily movement compared with tipping movement. These results indicated that canine translation might be healthier for periodontal tissue than tipping movement; the cervical region bore a greater load and might also be a susceptible area for tissue damage (except for the apex) regardless of movement pattern. | JING Yan HAN XiangLong CHENG BiHuan BAI Ding | 2013 | Chinese Science Bulletin2013,58,20: | 1

Risk SNP-induced lncRNA-SLCCl drives colorectal cancer through activating glycolysis signaling显示文摘Long non-coding RNAs(lncRNAs)play key roles in colorectal carcinogenesis. Here,we aimed to identify the risk SNP-induced lncRNAs and to investigate their roles in colorectal carcinogenesis.First,we identified rs6695584 as the causative SNP in 1 q41 locus. The A>G mutation of rs6695584 created a protein-binding motif of BATF,altered the enhancer activity, and subsequently activated IncSLCCl expression.Further validation in two independent CRC cohorts confirmed the upregulation of IncSLCCl in CRC tissues, and revealed that increased IncSLCCl expression was associated with poor survival in CRC patients.Mechanistically,lncRNA-SLCCl interacted with AHR and transcriptionally activated HK2 expression, the crucial enzyme in glucose metabolism, thereby driving the glycolysis pathway and accelerating CRC tumor growth. The functional assays revealed that IncSLCCl induced glycolysis activation and tumor growth in CRC mediated by HK2.In addition,HK2 was upregulated in colorectal cancer tissues and positively correlated with IncSLCCl expression and patient survival.Taken together, our findings reveal a risk SNP-mediated oncogene lncRNA-SLCCl promotes CRC through activating the glycolysis pathway. | Tingting Yan Chaoqin Shen Penglei Jiang Chenyang Yu Fangfang Guo Xianglong Tian Xiaoqiang Zhu Shiyuan Lu Bingshe Han Ming Zhong Jinxian Chen Qiang Liu Yingxuan Chen Junfang Zhang Jie Hong Haoyan Chen Jing-Yuan Fang | 2021 | Signal Transduction and Targeted Therapy2021,6,3: | 1
Engineering of a NIR-activable hydrogel-coated mesoporous bioactive glass scaffold with dual-mode parathyroid hormone derivative release property for angiogenesis and bone regeneration显示文摘Osteogenesis, osteoclastogenesis, and angiogenesis play crucial roles in bone regeneration.Parathyroid hormone(PTH),an FDA-approved drug with pro-osteogenic,pro-osteoclastogenic and proangiogenic capabilities, has been employed for clinical osteoporosis treatment through systemic intermittent administration. However, the successful application of PTH for local bone defect repair generally requires the incorporation and delivery by appropriate carriers.Though several scaffolds have been developed to deliver PTH,they suffer from the weaknesses such as uncontrollable PTH release, insufficient porous structure and low mechanical strength.Herein,a novel kind of NIR-activable scaffold(CBP/MBGS/PTHrP-2)with dual-mode PTHrP-2(a PTH derivative)release capability is developed to synergistically promote osteogenesis and angiogenesis for high-efficacy bone regeneration, which is fabricated by integrating the PTHrP-2-loaded hierarchically mesoporous bioactive glass(MBG)into the N-hydroxymethylacrylamide-modified,photothermal agent-doped,poly(N-isopropylacrylamide)-based thermosensitive hydrogels through assembly process.Upon on/off NIR irradiation, the thermoresponsive hydrogel gating undergoes a reversible phase transition to allow the precise control of on-demand pulsatile and long-term slow release of PTHrP-2 from MBG mesopores.Such NIR-activated dual-mode delivery of PTHrP-2 by this scaffold enables a well-maintained PTHrP-2 concentration at the bone defect sites to continually stimulate vascularization and promote osteoblasts to facilitate and accelerate bone remodeling. In vivo experiments confirm the significant improvement of bone reparative effect on critical-size femoral defects of rats. This work paves an avenue for the development of novel dual-mode delivery systems for effective bone regeneration. | Shi Liu Zhengzhe Han Ji-Na Hao Dapeng Zhang Xianglong Li Yuanyuan Cao Jinghuan Huang Yongsheng Li | 2023 | Bioactive Materials2023,,8: | 1

Simulation Study of the Control Strategy of a DC Inverter Heat Pump Using a DC Distribution Network显示文摘Photovoltaics, energy storage, direct current and flexibility(PEDF)are important pillars of achievement on the path to manufacturing nearly zero energy buildings(NZEBs).HVAC systems, which are an important part of public buildings, play a key role in adapting to PDEF systems. This research studied the basic principles and operational control strategies of a DC inverter heat pump using a DC distribution network with the aim of contributing to the development and application of small DC distribution systems.Along with the characteristics of a DC distribution network and different operating conditions,a DC inverter heat pump has the ability to adapt to changes in the DC bus voltage and adds flexibility to the system.Theoretical models of the DC inverter heat pump integrated with an ice storage unit were developed. The control strategies of the DC inverter heat pump system considered the influence of both room temperature and varied bus voltage.A simulation study was conducted using MATLAB&Simulink software with simulation results validated by experimental data. The results showed that:(1)The bus fluctuation under the rated working voltage had little effect on the operation of the unit;(2)When the bus voltage was fluctuating from 80%-90%or 105%-107%,the heat pump could still operate normally by reducing the frequency;(3)When the bus voltage was less than 80%or more than 107%,the unit needed to be shut down for the sake of equipment safety,so that the energy storage device could adjust to the sharp decrease or rise of voltage. | Siwei Han Xianglong Li Wei Zhao Linyu Wang Anqi Liang Shuang Zeng | 2023 | Energy Engineering2023,120,6: | 0
Rapid fabrication of zwitterionic sulfobetaine vinylimidazole-based monoliths via photoinitiated copolymerization for hydrophilic interaction chromatography显示文摘Zwitterionic sulfobetaine-based monolithic stationary phases have attracted increasing attention for their use in hydrophilic interaction chromatography. In this study,a novel hydrophilic polymeric monolith was fabricated through photo-initiated copolymerization of 3-(3-vinyl-1-imidazolio)-1-propanesulfonate(SBVI)with pentaerythritol triacrylate using methanol and tetrahydrofuran as the porogenic system.Notably, the duration for the preparation of this novel monolith was as little as 5 min, which was significantly shorter than that required for previously reported sulfobetaine-based monoliths prepared via conventional thermally initiated copolymerization. Moreover, these monoliths showed good morphology,permeability,porosity(62.4%),mechanical strength(over 15 MPa),column efficiency(51,230 plates/m),and reproducibility(relative standard deviations for all analytes were lower than 4.6%).Mechanistic studies indicated that strong hydrophilic and negative electrostatic interactions might be responsible for the retention of polar analytes on the zwitterionic SBVI-based monolith. In particular, the resulting monolith exhibited good anti-protein adhesion ability and low nonspecific protein adsorption. These excellent features seem to favor its application in bioanalysis. Therefore, the novel zwitterionic sulfobetaine-based monolith was successfully employed for the highly selective separation of small bioactive compounds and the efficient enrichment of N-glycopeptides from complex samples. In this study,we prepared a novel zwitterionic sulfobetaine-based monolith with good performance and developed a simpler and faster method for preparation of zwitterionic monoliths. | Qiqin Wang Lingjue Sun Huihui Wu Ning Deng Xianglong Zhao Jingwei Zhou Tingting Zhang Hai Han Zhengjin Jiang | 2022 | Journal of Pharmaceutical Analysis2022,12,5: | 0
Clean-DM1, a Korean Polyherbal Formula, Improves High Fat Diet-Induced Diabetic Symptoms in Mice by Regulating IRS/PI3K/AKT and AMPK Expressionsin Pancreas and Liver Tissues显示文摘Objective:To investigate the effects of Clean-DM1(C-DM1),a polyherbal formulation of Radix Scrophulariae,Radix Astragali,Rhizoma Atractylodis, and Radix Salviae Miltiorrhizae,on high-fat diet(HFD)-induced diabetes mice. Methods:The information about active components of C-DM1 extract and molecular mechanism was obtained from network pharmacology analysis.Main compounds of C-DM1 extract by high performance liquid chromatography-mass spectrometry(HPLC-MS)analysis were conducted for quality control. For in vivo study, mice were induced diabetes by HFD for 12 weeks. The mice in the normal group(Nor)were maintained with a regular diet and treated with saline by gavage. The HFD model mice were randomly divided into 3 groups, including a HFD diabetic model group,a C-DM1 extract-administered group(C-DM1,500 mg/kg),and metformin-administered groups(Met,500 mg/kg),8 mice in each group.Food intake, body weight(BW),and fasting blood glucose(FBG)levels were recorded weekly for 4 weeks. After 4 weeks of treatment, alanine aminotransferase(ALT),aspartate aminotransferase(AST),blood glucose,low-density lipoprotein cholesterol(LDL-C)were determined using an automated clinical chemistry analyzer, and homeostatic model for assessing insulin resistance(HOMA-IR)levels and oral glucose tolerance test(OGTT)were detected. The histopathological changes of liver and pancreatic tissues were observed by hematoxylin-eosin staining.Insulin receptor substrate(IRS)/phosphatidylinositol 3 kinase(PI3K)/protein kinase B(AKT)and adenosine 5'-monophosphate-activated protein kinase(AMPK)expressions in liver and pancreas tissues were detected by Western blot analysis.Results:HPLC-MS identified dihydroisotanshinone, dihydroisotanshinone I,cryptotanshinone, harpagoside, and atractyloside A in C-DM1 extract. The administration of C-DM1 extract significantly decreased body weight, calorie intake, and the levels of blood glucose and insulin in the diabetic mice(P<0.05 or P<0.01).The C-DM1 extract administration improved the impaired glucose tolerance and insulin resistance in the diabetic mice and significantly decreased the levels of LDL-C,ALT and AST(P<0.01).The C-DM1 extract inhibited the histopathological changes of fatty liver and hyperplasia of pancreatic islets in the diabetic mice. The C-DM1 extract significantly increased the phosphorylation of IRS,AKT,and AMPK and the expression of PI3K in pancreas and liver tissues(P<0.05 or P<0.01),which was consistent with the analysis results of network pharmacology.Conclusion:C-DM1 extract improved diabetes symptoms in longterm HFD-induced mice by regulation of IRS/PI3K/AKT and AMPK expressions in pancreas and liver tissues, suggesting that C-DM1 formulation may help prevent the progression of T2DM. | Piao Wang Yi Liu Seok Yong Kang Chenzi Lyu Xiang Han Tianjun Ho Kyung Jae Lee Xianglong Meng Yong-Ki Park Hyo Won Jung | 2024 | Chinese Journal of Integrative Medicine2024,30,2: | 0
An analysis of correctness for API recommendation:are the unmatched results useless?显示文摘API recommendation is a promising approach which is widely used during software development. However, the evaluation of API recommendation is not explored with sufficient rigor. The current evaluation of API recommendation mainly focuses on correctness, the measurement is conducted by matching recommended results with ground-truth results. In most cases, there is only one set of ground-truth APIs for each recommendation attempt, but the object code can be implemented in dozens of ways. The neglect of code diversity results in a possible defect in the evaluation. To address the problem,we invite15 developers to analyze the unmatched results in a user study. The online evaluation confirms that some unmatched APIs can also benefit to programming due to the functional correlation with ground-truth APIs.Then we measure the API functional correlation based on the relationships extracted from API knowledge graph,API method name, and API documentation.Furthermore,we propose an approach to improve the measurement of correctness based on API functional correlation. Our measurement is evaluated on a dataset of 6141 requirements and historical code fragments from related commits. The results show that 28.2%of unmatched APIs can contribute to correctness in our experiments. | Xianglong KONG Weina HAN Li LIAO Bixin LI | 2020 | Science China(Information Sciences)2020,63,9: | 0
Hydrothermal synthesized delafossite CuGaO_(2) as an electrocatalyst for water oxidation显示文摘Hydrogen production from water splitting provides an effective method to alleviate the ever-growing global energy crisis. In this work, delafossite CuGaO_(2)(CGO)crystal was synthesized through hydrothermal routes with Cu(NO_(3))_(2)·3H_(2)O and Ga(NO_(3))_(3)·xH_(2)O used as reactants. The addition of cetyltrimethylammonium bromide(CTAB)was found to play an important role in modifying the morphology of CuGaO_(2)(CGO-CTAB).With the addition of CTAB,the morphology of CGO-CTAB samples changed from irregular flake to typical hexagonal sheet microstructure, with an average size of 1-2μm and a thick-ness of around 100 nm.Furthermore, the electrocatalytic activity of CGO-CTAB crystals for oxygen evolution reaction(OER)was also studied and compared with that of CGO crystals.CGO-CTAB samples exhibited better activity than CGO.An overpotential of 391.5 mV was shown to be able to generate a current density of 10 mA/cm^(2).The as-prepared samples also demonstrate good stability for water oxidation and relatively fast OER kinetics with a Tafel slope of 56.4 mV/dec.This work highlights the significant role of modification of CTAB surfactants in preparing CGO related crystals, and the introduc-tion of CTAB was found to help to improve their electrocatalytic activity for OER. | Han Gao Miao Yang Xing Liu Xianglong Dai Xiao-Qing Bao Dehua Xiong | 2022 | Frontiers of Optoelectronics2022,15,1: | 0
