| 2 | Embryonic endothelial evolution towards first hematopoietic stem cells revealed by single-cell transcriptomic and functional analyses显示文摘Hematopoietic stem cells(HSCs)in adults are believed to be born from hemogenic endothelial cells(HECs)in mid-gestational embryos.Due to the rare and transient nature,the HSC-competent HECs have never been stringently identified and accurately captured,let alone their genuine vascular precursors.Here,we first used high-precision single-cell transcriptomics to unbiasedly examine the relevant EC populations at continuous developmental stages with intervals of 0.5 days from embryonic day(E)9.5 to E11.0.As a consequence,we transcriptomically identified two molecularly different arterial EC populations and putative HSCprimed HECs,whose number peaked at El 0.0 and sharply decreased thereafter,in the dorsal aorta of the aorta-gonadmesonephros(AGM)region.Combining computational prediction and in vivo functional validation,we precisely captured HSCcompetent HECs by the newly constructed Neurl3-EGFP reporter mouse model,and realized the enrichment further by a combination of surface markers(Procr+Kit+CD44+,PK44).Surprisingly,the endothelial-hematopoietic dual potential was rarely but reliably witnessed in the cultures of single HECs.Noteworthy,primitive vascular ECs from E8.0 experienced two-step fate choices to become HSC-primed HECs,namely an Initial arterial fate choice followed by a hemogenic fate conversion.This finding resolves several previously observed contradictions.Taken together,comprehensive understanding of endothelial evolutions and molecular programs underlying HSC-primed HEC specification in vivo will facilitate future investigations directing HSC production in vitro. | Siyuan Hou Zongcheng Li Xiaona Zheng Yun Gao Ji Dong Yanli Ni Xiaobo Wang Yunqiao Li Xiaochen Ding Zhilin Chang Shuaili Li Yuqiong Hu Xiaoying Fan Yu Hou Lu Wen Bing Liu Fuchou Tang Yu Lan | 2020 | Cell Research2020,30,5: | 8 |
| 3 | DNA degradation within mouse brain and dental pulp cells 72 hours postmortem显示文摘In this study, we sought to elucidate the process of DNA degradation in brain and dental pulp cells of mice, within postmortem 0-72 hours, by using the single cell gel electrophoresis assay and professional comet image analysis and processing techniques. The frequency of comet-like cells, the percentage of tail DNA, tail length, tail moment, Olive moment and tail area increased in tandem with increasing postmortem interval. In contrast, the head radius, the percentage of head DNA and head area showed a decreasing trend. Linear regression analysis revealed a high correlation between these parameters and the postmortem interval. | 15 | Cultivars and oil extraction techniques affect Cd/Pb contents and health risks in oil of rapeseed grown on Cd/Pb-contaminated farmland显示文摘Substitute planting with rapeseed offers promise for safely using large areas of Cd/Pb-contaminated farmland.Cd/Pb distributions during rapeseed oil production were investigated and health risks posed by the oil were assessed.Tests were performed using three cultivars(Brassica rapa SYH and ZS100 and Brassica napus QY-1)and four oil extraction techniques(mechanical and low-temperature pressing and n-hexane and subcritical low-temperature butane extraction).The amounts of Cd and Pb in oil were 0.73%-8.44%and 3.14%-11.76%,respectively,of the amounts in rapeseed and were strongly affected by the cultivar and oil extraction technique.The heavy metal(HM)concentrations were lower in solvent-extracted oil(particularly subcritical low-temperature butane extracted oil,in which HMs were not detected)than mechanically pressed oil.The Cd and Pb transfer indices were lower(meaning larger proportions of HMs were retained by the rapeseed meal)for B.rapa than B.napus.This was attributed to a high HM binding protein content of B.rapa seed.Health risks to humans were assessed using a probabilistic risk assessment model.The carcinogenic risk was mainly(97.1%-99.9%)caused by Cd and poses more concern than non-carcinogenic risk.Stronger health risks are posed by mechanically pressed than solvent-extracted oil,and higher carcinogenic risks are posed to people living in rural areas than urban areas.Substitute planting with B.rapa and extracting oil with organic solvent(preferably subcritical low-temperature butane)are optimal for safely utilizing Cd/Pb-contaminated soil.Attention should be paid to the health risks posed by Cd in oil to rural populations. | Junmei Guo Yuexing Wei Junxing Yang Tongbin Chen Guodi Zheng Tianwei Qian Xiaona Liu Xiaofei Meng Mengke He | 2023 | Frontiers of Environmental Science & Engineering2023,17,7: | 0 |
