1Comparing the Diagnostic Criteria for Gestational Diabetes Mellitus of World Health Organization 2013 with 1999 in Chinese Population显示文摘INTRODUCTION Gestational diabetes mellitus (GDM) was earlier defined as 'hyperglycemia first recognized during pregnancy' and has more recently been described by American Diabetes Association (ADA) (2012) as diabetes diagnosed during pregnancy that is not clearly overt diabetes.[1] The hyperglycemia and adverse pregnancy outcomes study (HAPO) demonstrated that the risk of adverse maternal,fetal,and neonatal outcomes continuously increase as a function of maternal glycemia at 24-28 weeks,even within ranges previously considered normal for pregnancy.[2] After reviewing the results of the HAPO study,many international diabetes study groups,including the International Association of Diabetes and Pregnancy Study Groups and ADA have adopted the 75 g oral glucose tolerance test (OGTT) at 24-28 weeks as a screening and diagnostic test and defined new cut-off values for GDM diagnosis.The Ministry of Health of China published the criteria for GDM,that is the same as the ADA criteria on July 1,2011.World Health Organization (WHO) has recommended the 75 g OGTT with different cut-offvalues as a diagnostic test since 1999,changed its recommendation on 2013.In Hongkong and Yunnan province in China,most hospital adopted the WHO 1999 criteria,nevertheless,there is no evidence of multicenter and large sample to show that the GDM population within the same ethnic group diagnosed by the new WHO criteria matches those diagnosed by the old one and consistency between the two criteria has not been reported.Thus,we conducted this study to analyze the two criteria in Chinese pregnant women.Weiwei Zhu Huixia Yang Yumei Wei Zilian Wang Xuelan Li Hairong Wu Nan Li Meihua Zhang Xinghui Liu Hua Zhang Yunhui Wang Jianmin Niu Yujie Gan Liruo Zhong Yunfeng Wang Anil Kapur 2015Chinese Medical Journal2015,,1:42
2Discrimination of the rare medicinal plant Dendrobium officinale based on naringenin, bibenzyl, and polysaccharides显示文摘The aim of this study was to establish a method for discriminating Dendrobium officinale from four of its close relatives Dendrobium chrysanthum, Dendrobium crystallinum, Dendrobium aphyllum and Dendrobium devonianum based on chemical composition analysis. We analyzed 62 samples of 24 Dendrobium species. High performance liquid chromatography analysis confirmed that the four low molecular weight compounds 4′,5,7-trihydroxyflavanone (naringenin), 3,4-dihydroxy-4′,5-dime- thoxybibenzyl (DDB-2), 3′,4-dihydroxy-3,5′-dimethoxybibenzyl (gigantol), and 4,4′-dihydroxy-3,3′,5-trimethoxybibenzyl (moscatilin), were common in the genus. The phenol-sulfuric acid method was used to quantify polysaccharides, and the monosaccharide composition of the polysaccharides was determined by gas chromatography. Stepwise discriminant analysis was used to differentiate among the five closely related species based on the chemical composition analysis. This proved to be a simple and accurate approach for discriminating among these species. The results also showed that the polysaccharide content, the amounts of the four low molecular weight compounds, and the mannose to glucose ratio, were important factors for species discriminant. 3Global Water Vapor Content Decreases from 2003 to 2012: An Analysis Based on MODIS Data显示文摘Water vapor in the earth′s upper atmosphere plays a crucial role in the radiative balance, hydrological process, and climate change. Based on the latest moderate-resolution imaging spectroradiometer(MODIS) data, this study probes the spatio-temporal variations of global water vapor content in the past decade. It is found that overall the global water vapor content declined from 2003 to 2012(slope b = –0.0149, R = 0.893, P = 0.0005). The decreasing trend over the ocean surface(b = –0.0170, R = 0.908, P = 0.0003) is more explicit than that over terrestrial surface(b = –0.0100, R = 0.782, P = 0.0070), more significant over the Northern Hemisphere(b = –0.0175, R = 0.923, P = 0.0001) than that over the Southern Hemisphere(b = –0.0123, R = 0.826, P = 0.0030). In addition, the analytical results indicate that water vapor content are decreasing obviously between latitude of 36°N and 36°S(b = 0.0224, R = 0.892, P = 0.0005), especially between latitude of 0°N and 36°N(b = 0.0263, R = 0.931, P = 0.0001), while the water vapor concentrations are increasing slightly in the Arctic regions(b = 0.0028, R = 0.612, P = 0.0590). The decreasing and spatial variation of water vapor content regulates the effects of carbon dioxide which is the main reason of the trend in global surface temperatures becoming nearly flat since the late 1990 s. The spatio-temporal variations of water vapor content also affect the growth and spatial distribution of global vegetation which also regulates the global surface temperature change, and the climate change is mainly caused by the earth's orbit position in the solar and galaxy system. A big data model based on gravitational-magmatic change with the solar or the galactic system is proposed to be built for analyzing how the earth's orbit position in the solar and galaxy system affects spatio-temporal variations of global water vapor content, vegetation and temperature at large spatio-temporal scale. 4Pollen-mediated gene flow in Chinese commercial fields of glufosinate-resistant canola (Brassica napus)显示文摘One of the most pressing issues surrounding transgenic oilseed rape cultivation is the potential impact of GM genes entering neighboring non-GM oilseed rape plants, related weeds or closely related wild relatives. Following the principle of a case-by-case for transgenic crop safety assessment, we designed experiments to study pollen-mediated gene flow from herbicide-resistant transgenic oilseed rape to Chinese commercial varieties. The pollen dispersal was detected as the progeny of recipient plants that were identified as glufosinate resistant. The results indicated that pollen dispersal occurred up to 2000 m and its rate sharply decreased as the distance from the pollen source increased. However, the rate of long-distance pollen dispersal from 33 to 2000 m was less than 0.015% and did not present a gradual decrease, indicating the randomization of dispersal and pollination. Most pollen dispersed within a 4.5 m area around the transgenic pollen source plot, with a maximum of 1.19% at the sampling site of 1.4 m. Wind direction significantly affected the direction and the distance of pollen dispersal, resulting in the more and farther dispersal in southwest direction. However, the number of potentially pollinating bees was not correlated with direction and distance from the pollen source plot, suggesting that bee density and distribution would not contribute to the differences in pollen dispersal among different directions. This paper is the first report on research in a large-scale experiment of pollen-mediated gene flow un-der the China’s environmental conditions and provided scientific evidence for the effective commer-cialization of transgenic oilseed rape and its safe management. 5Trehalose phosphate synthase 5-dependent trehalose metabolism modulates basal defense responses in Arabidopsis thaliana显示文摘Despite the recent discovery that trehalose synthesis is important for plant development and abiotic stress tolerance, the effects of trehalose on biotic stress responses remain relatively unknown. In this study, we demonstrate that TREHALOSE PHOSPHATE SYNTHASE 5(TPS5)-dependent trehalose metabolism regulates Arabidopsis thaliana defenses against pathogens(necrotrophic Botrytis cinerea and biotrophic Pseudomonas syringae).Pathogen infection increased trehalose levels and upregulated TPS5 expression. Application of exogenous trehalose significantly improved plant defenses against B. cinerea, but increased the susceptibility of plants to P. syringae. We demonstrate that elevated trehalose biosynthesis, in transgenic plants over-expressing TPS5, also increased the susceptibility to P. syringae, but decreased the disease elcit r symptoms caused by B. cinerea. The knockout of TPS5 prevented the accumulation of trehalose and enhanced defense responses against P. syringae. Additionally, we observed that a TPS5-interacting protein(multiprotein bridging factor 1c) was required for induced expression of TPS5 during pathogen infections. Furthermore, we show that trehalose promotes P. syringae growth and disease development, via a mechanism involving suppression of the plant defense gene, Pathogenesis-Related Protein 1. 6First-principles investigation of the effects of B impurities on the mechanical properties of NiAl intermetallics显示文摘We have investigated the effects of B impurities on the structure and mechanical properties of NiAl intermetallics by using a first-principles pseudopotential total-energy method, based on the density functional theory with a generalized gradient approximation. We found that the impurity B atoms can either replace Ni atoms or Al atoms or both, depending on the surrounding environment. We demonstrated that the presence of B will cause an increase in brittleness and a decrease in the ductility of NiAl for the Al-substitutional case, while causing an increase in the ductility of NiAl for the Ni-subtitutional case, based on the calculated elastic constants and the empirical criterions. 7Effect of the population heterogeneity on growth behavior and its estimation显示文摘Different types of the Logistic model are constructed based on a simple assumption that the microbial populations are all composed of homogeneous members and consequently, the condition of design for the initial value of these models has to be rather limited in the case of N(t_0)=N_0. Therefore, these models cannot distinguish the dynamic behavior of the populations possessing the same N0 from heteroge-neous phases. In fact, only a certain ratio of the cells in a population is dividing at any moment during growth progress, termed as θ, and thus, ddNt not only depends on N, but also on θ. So θ is a necessary element for the condition design of the initial value. Unfortunately, this idea has long been neglected in widely used growth models. However, combining together the two factors (N0 and θ ) into the initial value often leads to the complexity in the mathematical solution. This difficulty can be overcome by using instantaneous rates (Vinst) to express growth progress. Previous studies in our laboratory sug-gested that the Vinst curve of the bacterial populations all showed a Guassian function shape and thus, the different growth phases can be reasonably distinguished. In the present study, the Gaussian dis-tribution function was transformed approximately into an analytical form (0.5x ibxYi αe=20) that can be conveniently used to evaluate the growth parameters and in this way the intrinsic growth behavior of a bacterial species growing in heterogeneous phases can be estimated. In addition, a new method has been proposed, in this case, the lag period and the double time for a bacterial population can also be reasonably evaluated. This approach proposed could thus be expected to reveal important insight of bacterial population growth. 8Acute toxicity study of Aspidopterys obcordata aqueous extract in Sprague-Dawley rats显示文摘OBJECTIVE: To examine the acute toxicity of an aqueous extract of Aspidopterys obcordata(A. obcordata) in Sprague Dawley rats.METHODS: The rats were orally administered a dose of 5000 mg/kg body weight and observed continuously for 6 h and then daily for 14 days. Control rats were administered distilled water. The effect of the extract on general behavior, body weight, and food and water intake were measured.After 14 days, the rats were sacrificed and their organs(liver, heart, spleen, lungs, kidney, adrenal glands, ovaries, and testes) were removed for macroscopic examination. The body and organ weights in addition to hematology(e.g., hemoglobin and white blood cell counts) and clinical blood biochemistry(e.g., albumin and bilirubin) were also examined.RESULTS: There were no deaths recorded, and the rats treated with A. obcordata showed no signs of toxicity. All measured parameters in rats treated with A. obcordata were unaffected when compared with those in control rats. 10Effects of Heat Shock Factor AtHsfA1a on Caspase-3 Activity in Arabidopsis thaliana under High Temperature Stress显示文摘[Objective] This study aimed to investigate the effects of heat shock factor AtHsf A1 a on Caspase-3 activity in Arabidopsis thaliana under high temperature stress,thus revealing the relationship between heat shock factor At Hsf A1 a and programmed cell death in A. thaliana. [Method] Different genotypes of A. thaliana( AtHsf A1a-silenced transgenic and wild-type) seedlings were treated at 42 ℃. According to the fragmentation level of fluorogenic substrate Ac-DEVDp NA,Caspase-3 activity was determined by spectrophotometry. [Result]After high temperature treatment,Caspase-3 activity in A. thaliana was enhanced significantly. Caspase-3 activity in At Hsf A1a-silenced transgenic A. thaliana was higher than that in wild-type A. thaliana,which indicated that At Hsf A1 a could inhibit Caspase-3 activity in A. thaliana under high temperature stress. [Conclusion] Under high temperature stress,heat shock factor At Hsf A1 a might exert inhibitory effects on programmed cell death by reducing Caspase-3 activity. 12Global vegetation change analysis based on MODIS data in recent twelve years显示文摘Vegetation cover change is critical for understanding impacts and responses of vegetation to climate change. A study found that vegetation in the regions between 45°N-70°N was increasing using normalized difference vegetation index( NDVI) from 1981 to 1991 ten years ago. The global vegetation growth has changed because of climate change in recent twelve years( 2001- 2012). After thorough analysis based on satellite data,it is found that it is evident that the global vegetation changed( NDVI) little,and it is increasing slightly in Northern hemisphere while it is decreasing slightly in Southern Hemisphere. For different latitudes,vegetation is increasing 0.17% every year from 60°N to 70 °N( R^2= 0.47,P > 0.013),while the vegetation is decreasing 0.11% every year from 10°N to 10° S( R^2= 0.54,P > 0.004). For different continents,the vegetation in South America is decreasing 0.16% every year( R^2= 0.78,P > 0.0001) and it is increasing 0.05% every year in Asia( R^2= 0.28,P > 0.072) and 0.25% every year in Oceania( R^2= 0.24,P > 0.1). The analysis of global vegetation in different seasons indicates that spatial distribution of global temperature and water vapor will affect the spatial distribution of vegetation,in turn,the spatial distribution of vegetation will also regulate the global temperature and water vapor spatial distribution at large scale. The growth and distribution of vegetation are mainly caused by the orbit of the celestial bodies,and a big data model based on gravitational-magmatic change with the solar or the galactic system as its center is proposed to be built for analyzing how the earth's orbit position in the solar and galaxy system affects spatial-temporal variations of global vegetation and temperature at large scale. 13Epigenetic drug library screening identified an LSD1 inhibitor to target UTX-deficient cells for differentiation therapy显示文摘UTX(also known as KDM6A),a histone 3 lysine 27 demethylase,is among the most frequently mutated epigenetic regulators in myelodysplastic syndrome(MDS)and acute myeloid leukemia(AML).Recent studies have suggested that UTX mutations promote MDS and AML by blocking the differentiation of hematopoietic stem and progenitor cells(HSPCs).Here,we performed an epigenetic drug library screening for small molecules able to release the differentiation block on HSPCs induced by UTX deficiency.We found that SP2509,a selective inhibitor of LSD1,specifically promoted the differentiation of Utx-null HSPCs while sparing wild-type HSPCs.Transcriptome profiling showed that Utx loss reduced the expression of differentiation-related and tumor suppressor genes,correlating with their potential roles in HSPC self-renewal and leukemogenesis.In contrast,SP2509 treatment reversed these changes in gene expression in Utx-null HSPCs.Accordingly,Utx loss decreased H3K4 methylation level probably through the COMPASS-like complex,while LSD1 inhibition by SP2509 partially reversed the reduction of H3K4 methylation in Utx-deficient HSPCs.Further,SP2509 promoted the differentiation of Utx-null AML cells in vitro and in vivo and,therefore,extended the survival of these leukemic mice.Thus,our study identified a novel strategy to specifically target both premalignant and malignant cells with Utx deficiency for differentiation therapy and provided insights into the molecular mechanisms underlying the role of Utx in regulating HSPCs and related diseases.Baohong Wu Xiangyu Pan Xuelan Chen Mei Chen Kaidou Shi Jing Xu Jianan Zheng Ting Niu Chong Chen Xiao Shuai Yu Liu 2019Signal Transduction and Targeted Therapy2019,4,1:2

17Performance of iteratively decoded bit-interleaved coded modulation with chaos based interleaving strategies显示文摘Bit-interleaved coded modulation with iterative decoding (BICM-ID) is a bandwidth efficient trans- mission scheme which is well suitable for next-generation mobile communications. The interleaver design is critical to the high performance of BICM-ID. In this paper we investigate the application of the chaos theory to the interleaver design to further improve the performance of BICM-ID. A new interleaving scheme based on the chaos theory is proposed. Compared with the random interleaver, the proposed approach has the ad- vantages including lower system delay and higher transmission efficiency. 18Landscape dynamic network biomarker analysis reveals the tipping point of transcriptome reprogramming to prevent skin photodamage显示文摘Skin,as the outmost layer of human body,is frequently exposed to environmental stressors including pollutants and ultraviolet(UV),which could lead to skin disorders.Generally,skin response process to ultraviolet B(UVB)irradiation is a nonlinear dynamic process,with unknown underlying molecular mechanism of critical transition.Here,the landscape dynamic network biomarker(lDNB)analysis of time series transcriptome data on 3D skin model was conducted to reveal the complicated process of skin response to UV irradiation at both molecular and network levels.The advanced l-DNB analysis approach showed that:(i)there was a tipping point before critical transition state during pigmentation process,validated by 3D skin model;(ii)13 core DNB genes were identified to detect the tipping point as a network biomarker,supported by computational assessment;(iii)core DNB genes such as COL7A1 and CTNNB1 can effectively predict skin lightening,validated by independent human skin data.Overall,this study provides new insights for skin response to repetitive UVB irradiation,including dynamic pathway pattern,biphasic response,and DNBs for skin lightening change,and enables us to further understand the skin resilience process after external stress.Chengming Zhang Hong Zhang Jing Ge Tingyan Mi Xiao Cui Fengjuan Tu Xuelan Gu Tao Zeng Luonan Chen 2021Journal of Molecular Cell Biology2021,13,11:1
