1Acupotomy versus sodium hyaluronate for treatment of knee osteoarthritis in rabbits显示文摘OBJECTIVE: To investigate the possible advantages of acupotomy over sodium hyaluronate injection for the treatment of knee osteoarthritis(KOA).METHODS: Twenty rabbits were divided randomly into four groups(n = 5 in each): a control group,model group, acupotomy group, and sodium hyaluronate injection group. The model, acupotomy,and sodium hyaluronate groups underwent anterior cruciate ligament transection plus partial medial meniscectomy. Sodium hyaluronate injection and acupotomy were administered to the respective groups from weeks 5 to 8, and samples of the tibial plateau and medial condyle of the femur were collected in week 9. Vascular endothelial growth factor(VEGF) expression was assessed in cartilage and subchondral bone by immunohistochemical staining.RESULTS: Articular cartilage degeneration was less pronounced in the acupotomy compared with the model and sodium hyaluronate groups. VEGF expression levels in cartilage and subchondral bone were increased in the model group compared with the control group(P < 0.01), and acupotomy had a more pronounced therapeutic effect than sodium hyaluronate injection(P < 0.01).CONCLUSION: Acupotomy and sodium hyaluronate injection may both reduce degeneration in the cartilage and subchondral bone in KOA based on the results from a rabbit model, but acupotomy improved the histopathology and reduced the VEGF content more effectively than sodium hyaluronate injection, probably by reducing venous stasis and intraosseous pressure. 2The enhanced protective effects of salvianic acid A: A functionalized nanoparticles against ischemic stroke through increasing the permeability of the blood-brain barrier显示文摘Ischemic stroke is the leading cause of disability and death worldwide. Currently, the only proven treatment for ischemic stroke is restoring the cerebral blood supply. In addition, some of the tissue is damaged during the subsequent reperfusion because of the overproduction of reactive oxygen species (ROS). Furthermore, antioxidant therapies have shown promise in preclinical studies for the treatment of ischemia-reperfusion injury. However, their therapeutic efficacy has been limited because of their low bioavailability in brain. To resolve this issue, we synthesized ROS-responsive, fan-shaped dendrimer nanoparticles (NPs) and conjugated them with a blood-brain barrier (BBB)-targeting peptide, COG1410, and salvianic acid A (SA), which is an effective antioxidant in ischemic stroke. The BBB targeting peptide acts as a ligand of the nanocarrier system and penetrates the BBB through the endocytosis of the ligand receptor. The results showed that T-SA-NPs not only target and accumulate in the infarct area, they also reduce over 2 times of the infarct area and reverse the behavioral deficits in MCAO mice, which illustrates that these NPs have an effective therapeutic effect on the ischemic stroke. In addition, these NPs had no toxicity in any organs of the body. 318beta-glycyrrhetinic acid induces ROS-mediated apoptosis to ameliorate hepatic fibrosis by targeting PRDX1/2 in activated HSCs显示文摘Hepatic stellate cells(HSCs)are essential drivers of fibrogenesis.Inducing activated-HSC apoptosis is a promising strategy for treating hepatic fibrosis.18beta-glycyrrhetinic acid(18b-GA)is a natural compound that exists widely in herbal medicines,such as Glycyrrhiza uralensis Fisch,which is used for treating multiple liver diseases,especially in Asia.In the present study,we demonstrated that 18b-GA decreased hepatic fibrosis by inducing the apoptosis in activated HSCs.18b-GA inhibited the expression of a-smooth muscle actin and collagen type Ⅰ alpha-1.Using a chemoproteomic approach derived from activity-based protein profiling,together with cellular thermal shift assay and surface plasmon resonance,we found that 18b-GA covalently targeted peroxiredoxin 1(PRDX1)and peroxiredoxin 2(PRDX2)proteins via binding to active cysteine residues and thereby inhibited their enzymatic activities.18b-GA induced the elevation of reactive oxygen species(ROS),resulting in the apoptosis of activated HSCs.PRDX1 knockdown also led to ROS-mediated apoptosis in activated HSCs.Collectively,our findings revealed the target proteins and molecular mechanisms of 18b-GA in ameliorating hepatic fibrosis,highlighting the future development of 18b-GA as a novel therapeutic drug for hepatic fibrosis.Qian Zhang Piao Luo Liuhai Zheng Jiayun Chen Junzhe Zhang Huan Tang Dandan Liu Xueling He Qiaoli Shi Liwei Gu Jiahao Li Qiuyan Guo Chuanbin Yang Yin Kwan Wong Fei Xia Jigang Wang 2022Journal of Pharmaceutical Analysis2022,12,4:3

5Ion flux dropout observed near dipolarization front显示文摘An ion flux dropout near the dipolarization front(DF) at around XGSM=-11 REin the Earth's plasma sheet was observed by Time History of Events and Macroscale Interaction during substorms(THEMIS) on March31, 2009. The ion differential energy fluxes at energies from 450 e V to 150 ke V measured by the ESA and SST instruments from THC began to decrease about 2 s before the detection of the DF and reached a local minimum 6 s later. Then, the ion fluxes gradually increased to form a dropout around the DF. The spatial extent of the dropout was about 4,000 km. For energies above 20 ke V, the ion fluxes after the dropout are greater than those before it,contrary to the fluxes at energies below 20 ke V. The associated ion density variation indicates that the ion flux dropout coincides with the ion density dropout. Taking advantage of multipoint observations, THD, THC, and THE detected the same DF consecutively. Only THC detected an obvious ion flux dropout; THD observed an indistinct one about 2 s before THC; no high-energy(E [ 30 ke V) ion flux dropout was observed by THE. 7kdm4aa is required for reproduction and development of zebrafish显示文摘Lysine-specific demethylase 4A(KDM4A)catalyzes demethylation of histone lysine residues,which regulates chromatin state and transcription.In drosophila and mice,KDM4A plays an important role in multiple biological processes including development,aging,metabolism,and immunity,however the functions of KDM4A in fish are still unclear.There are two copies of the kdm4a gene in zebrafish,namely kdm4aa and kdm4ab,kdm4aa was edited using CRISPR/Cas9 technology in the present study,then homozygous kdm4aa mutants(kdm4aa^(-/-)��)were obtained,and loss of kdm4aa was confirmed by sequencing and increased H3K9me3.Whole-mount in situ hybridization showed that kdm4aa is widely expressed during the embryonic development of zebrafish.Compared with WT zebrafish,kdm4aa^(-/-)��zebrafish showed no significant difference in gamete formation and fertilization,but the survival rate of kdm4aa^(-/-)��embryos dramatically reduced to 21%at 26 hpf.Further observation showed that about 80%of survived kdm4aa^(-/-)��zebrafish experienced disruption in stripe formation,and 10%of survived kdm4aa^(-/-)��zebrafish underwent vertebral malformation.Alizarin red S staining demonstrated the abnormal spinal development in kdm4aa^(-/-)��zebrafish.These results indicated that kdm4aa is required for normal embryonic development of zebrafish,loss of kdm4aa function leads to decreased survival during the early stages of zebrafish development and morphological variation in adult zebrafish.Lijie Han Juntao Luo Songya Qu Xueling Shi Junfang Zhang Bingshe Han 2023Aquaculture and Fisheries2023,8,6:0

9Influence of inherent iron and oxygen concentrations on selenite sorption process using bentonite显示文摘The oxygen concentration and inherent Fe in bentonite have a significant influence on the Se(IV) sorption process.In this study,the sorption of selenite on natural bentonite was investigated using a batch experiment method,and the distribution coefficient(K_d)values were obtained in the pH range from 2.0 to 10.0 under oxic/anoxic conditions.The K_d values always reached a maximum value at a pH of 4 under oxic conditions and at a pH of 8 under anoxic conditions;meanwhile,the K_d value under anoxic conditions was larger than the value under oxic conditions,especially in regard to the maximum K_d values.The oxygen conditions have a significant influence on the ratio of redox-sensitive Fe^(2+)/Fe^(3+),which was closely related to the difference in the K_d values under both oxic/anoxic conditions.Ferric selenite and green rust could be responsible for the maximum K_d values under oxic/anoxic conditions.In the leaching experiments,we found that the Fe^(2+) in bentonite could replace Mg^(2+) and Al^(3+) in the octahedral sheet.Spectroscopy methods,such as Fourier transform infrared spectroscopy(FTIR),X-ray diffraction(XRD) and X-ray absorption spectroscopy(XAS) were used to characterize the surface properties of the samples after reaction.Overall,this study shows that the addition of Fe^(2+)-containing materials into backfill/buffer materials under anoxic condition could enhance the sorption of ^(79)Se(IV).Jiangang He Xueling Qiao Yanlin Shi Yao Li Xiaoyu Yang Wanqiang Zhou Chunli Liu 2017Science China Chemistry2017,60,9:0

11Self-assembly of insulated molecular wires of a water-soluble cationic PPV and anionic dendrons显示文摘Insulated molecular wires of poly(phenylenevinylene) (PPV) were prepared by wrapping the conjugated backbones with dendrons through a noncovalent approach. It was found that electrostatic interaction between the quaternary ammonium groups of PPV-1 and the carboxylate moieties in dendrons induced the packing of dendrons along PPV-1 conjugated backbones. Absorption and emission spectroscopic examinations in solution and solid film indicated that the PPV-1 backbones adopted a more planar and isolated conformation in the complexes.
