| 1 | Cryopreservation of Gametophytes of Laminaria japonica (Phaeophyta) Using Encapsulation-Dehydration with Two-Step Cooling Method显示文摘Gametophytes of Laminaria japonica were cryopreserved in liquid nitrogen using encapsulation-dehydration with two-step cooling method. Gametophytes cultured at 10℃ and under continuous irradiance of 30 μmol m-2 s-1 for 3 weeks were en- capsulated in calcium alginate beads. The beads were dehydrated in 0.4 molL-1 sucrose prepared with seawater for 6 h, desiccated in an incubator set at 10℃ and 70% relative humidity for 4 h, pre-frozen at either ?40℃ or ?60℃ for 30 min, and stored in liquid nitrogen for >24 h. As high as 43% of survival rate was observed when gametophytes were thawed by placing the beads in 40℃ seawater and re-hydrated in 0.05 molL-1 citrate sodium prepared using 30‰ NaCl 7 d later. More cells of male gametophytes sur- vived the whole procedure in comparison with female gametophytes. | 2 | Influence of Temperature and Salinity on Germination of Eelgrass (Zostera marina L.) Seeds显示文摘Seagrass restoration as part of ocean ecosystem protection has been launched for many years all over the world, but intensive research on this subject in China has just begun in recent years. Seed broadcasting has been widely accepted as the most potentially useful method for seagrass restoration over large areas. We examined the influence of key environmental factors on seed germination to help promote eelgrass bed restoration. Under anoxic conditions, the influence of temperature and salinity on the germination rate of eelgrass (Zostera marina L.) seeds was examined at different combinations of four temperatures (4, 9, 14, and 24℃) and nine salinities (5 to 45, increment of 5). The effect of significant interaction of temperature and salinity on germination rate was observed (ANOVA) (P<0.001). The highest germination rate (83.3 ± 3.5)% was reached in 8 weeks at 14℃ and salinity 5. Higher temperature significantly increased the germination rate at salinity 5 (P<0.001) during the whole observation period except for 24℃, while lower salinity significantly increased the germination rate at 14℃ (P<0.001). Although significant interaction was found between temperature and salinity (P<0.001), the influence of salinity was stronger than that of temperature for the germination of eelgrass seeds. | 4 | Aberrant translation regulated by METTL1/WDR4-mediated tRNA N7-methylguanosine modification drives head and neck squamous cell carcinoma progression显示文摘Background:Cancer cells selectively promote the translation of oncogenic tran-scripts to stimulate cancer progression.Although growing evidence has revealed that tRNA modifications and related genes participate in this process,their roles in head and neck squamous cell carcinoma(HNSCC)remain largely unchar-acterized.Here,we sought to investigate the function and mechanisms of the transfer RNA(tRNA)N7-methylguanosine(m'G)modification in regulating the occurrence and development of HNSCC.Methods:Cell lost of-function and gain-of function assays,xenograft models,conditional knockout and knockin mouse models were used to study the physi-ological functions of tRNA m'G modification in HNSCC tumorigenesis.tRNA modification and expression profiling,mRNA translation profiling and res-cue assays were performed to uncover the underlying molecular mechanisms.Single-cell RNA sequencing(scRNA seq)was conducted to explore the tumor microenvironment changes.Results:The tRNA.m7G methyltransferase complex components Methyltransferase-like 1(METTL1)/WD repeat domain 4(WDR4)were upregulated in HNSCC and associated with a poor prognosis.Functionally,METTL1/WDR4 promoted HNSCC progression and metastasis in cell-based and transgenic mouse models.Mechanistically,ablation of METTL1 reduced the m'G levels of 16 tRNAS,inhibiting the translation of a subset of oncogenic transcripts,including genes related to the phosphatidylinositol-3-kinase/protein kinase B/mammalian target of rapamycin(PI3K/AKT/mTOR)signaling pathway.In addition,chemical modulators of the PI3K/Akt/mTOR signaling pathway reversed the effects of Mettll in mouse HNSCC.Furthermore,scRNA-seq results revealed that Mettll knockout in mouse tumor cells altered the immune landscape and cell-cell interaction between the tumor and stromal compartment.Conclusions:The tRNA m?G methyltransferase METTLI was found to promote the development and malignancy of HNSCC through regulating global mRNA translation,including the PI3K/AKT/mTOR signaling pathway,and found to alter immune landscape.METTLI could be a promising treatment target for HNSCC patients. | Jie Chen Kang Li Jianwen Chen Xiaochen Wang Rongsong Ling Maosheng Cheng Zhi Chen Fangfang Chen Qianting He Shuai Li Caihua Zhang Yizhou Jiang Qianming Chen Anxun Wang Demeng Chen | 2022 | Cancer Communications2022,42,3: | 6 |
| 5 | Dendritic Growth Pattern and Dendritic Network Distortion in the Platform of a Ni-based Superalloy显示文摘The pattern of dendritic growth and distortion of dendritic network in the platform have been investigated by one mold casting with different platform length during directional solidification. As the platform length elongates, the symmetry of dendritic growth along left and right edges gradually worsens in platform base. While the platform length reaches 14 mm, the distortion of dendritic network is first observed in outward platform. It is found that the distortion of dendritic network along platform inside is more serious than that along platform edges. Both[001] deviation and accumulated misorientation along platform inside, up to 9 and 16.3, respectively, are far greater than those along lefte outwarde right edges. The alloy exhibits excellent comprehensive mechanical performance during tension at temperature range of 600–700℃. The microstructure reveals the intersection of twintwin is the prevailing deformation mechanism and the twins play a dual role in strengthening and toughening the alloy in the thermomechanical process. The deformation twin variants I and were formed by the edge dislocation 112 and the mixed dislocation 211 on the {111} crystal planes, respectively. Besides, the dislocation jogs and kinks caused by twin intersection on the slip planes can strengthen the alloy, which may contribute to the high strength(the tensile strengths at the 600°and 700°tensile tests are respectively780 MPa and 630 MPa.). | 8 | Microstructure of Carbides at Grain Boundaries in Nickel Based Superalloys显示文摘It is well known that carbides at grain boundaries play an important role in affecting mechanical properties of nickel based superalloys. In order to deeply understand the relationship between grain boundary structures and properties, in this work, we have investigated the microstructure of grain boundaries with different misorientation angles in bicrystals of nickel based superalloys. It is found that the bicrystals with smaller misorientation angles contain denser M 23 C 6 but sparse MC particles at grain boundaries, and this kind of bicrystals presents longer stress rupture lives. It was observed that MC carbides were decorated by M 23 C 6 and M 6 C particles at grain boundaries. The formation of these carbide particles, therefore, is likely due to the local fluctuation of chemical composition around MC carbides. | 18 | Microstructural evolution of a Ni-Co based superalloy during hot compression atγ'sub-/super-solvus temperatures显示文摘The effects of strain rate on the microstructural evolution and deformation mechanism of a Ni-Co based superalloy were investigated by isothermal compression tests performed atγ'sub-solvus(1090℃)andγ'super-solvus temperatures(1150℃)with a wide strain rate range from 0.001 to 10 s^(-1)under a true strain of 0.693.Electron backscatter diffraction(EBSD),electron channeling contrast imaging(ECCI)and transmission electron microscope(TEM)techniques were used to characterize the microstructures.The results revealed that the dynamic recrystallization(DRX)volume fraction increased and stored energy of theγ'matrix grains decreased with increasing the strain rate duringγ'sub-solvus temperature deformation,while the opposite phenomena were observed duringγ'super-solvus temperature deformation.The comprehensive effect of initial grain size,primaryγ'phase,twins and adiabatic temperature rise led to these results.The primaryγ'particles undergone the deformation behavior within itself and obviously accelerated the DRX of the matrix.The microstructural evolution proved that discontinuous dynamic recrystallization(DDRX)was the dominant mechanism during the hot deformation carried out at bothγ'sub-solvus andγ'super-solvus temperatures.Primaryγ'particles obviously accelerated the nucleation step and retarded the growth step of DDRX duringγ'sub-solvus temperature deformation.Besides,the acceleration effect of primaryγ'particles on DDRX increased with the increase of strain rate.Continuous dynamic recrystallization(CDRX)was confirmed to be an assistant mechanism duringγ'super-solvus temperature deformation and was promoted with the increase of strain rate. | Peng Liu Rui Zhang Yong Yuan Chuanyong Cui Faguang Liang Xi Liu Yuefeng Gu Yizhou Zhou Xiaofeng Sun | 2021 | Journal of Materials Science & Technology2021,,18: | 2 |

During heat treatment. Reretards γ phases coarsening and equalizes the distribution of γ' phases. As Re content increases, fatigue life increases and slip bands distribute more inhomogeneously. Moreover, adding Re not only reduces stacking fault energy of the matrix, but also promotes the element segregation to increase the lattice misfit. However, the larger lattice misfit does not lead to the formation of dislocation networks, but which activates dislocation movement and promotes dislocations cross-slip and climbing movement under high temperature and applied stress.
