Nobiletin downregulates the SKP2-p21/p27-CDK2 axis to inhibit tumor progression and shows synergistic effects with palbociclib on renal cell carcinoma显示文摘Objective:Natural extracts, including nobiletin, have been reported to enhance the efficacy and sensitivity of chemotherapeutic drugs. However, whether and how nobiletin affects tumor growth and progression in renal cell carcinoma(RCC)are still unclear. Methods:Cell proliferation, cell cycle and apoptosis analyses,colony-formation assays, immunoblotting analysis, and q RT-PCR analysis were performed to investigate how nobiletin affected RCC cell proliferation in vitro. The nude mouse model was used to test the efficacy of nobiletin alone or in combination with palbociclib. Results:Nobiletin inhibited cell proliferation by inducing G1 cell cycle arrest and cell apoptosis in RCC cells.Mechanistically, nobiletin decreased SKP2 protein expression by reducing its transcriptional level. The downregulated SKP2 caused accumulation of its substrates,p27 and p21,which further inhibited the activity of the G1 phase-related protein,CDK2,leading to inhibition of cell proliferation and tumor formation.A higher SKP2 protein level indicated less sensitivity to the CDK4/6 inhibitor, palbociclib.A combination of nobiletin and palbociclib showed a synergistic tumor inhibition in vitro and in an in vivo model. Conclusions:Nobiletin downregulated the SKP2-p21/p27-CDK2 axis to inhibit tumor progression and showed synergistic tumor inhibition effects with the CDK4/6 inhibitor, palbociclib,on RCC,which indicates a potential new therapeutic strategy. | Tingting Chen Liu Liu Yonghong Zou Xiaoyan Hu Wenfeng Zhang Tao Zhou Xi Luo Weihua Fu Jie Xu | 2021 | Cancer Biology & Medicine2021,18,1: | 3
Dual Control of Depositional Facies on Uranium Mineralization in Coal-bearing Series: Examples from the Tuanyushan Area of the Northern Qaidam Basin, NW China显示文摘The uranium deposits in the Tuanyushan area of northern Qaidam Basin commonly occur in coal-bearing series. To decipher the U-enrichment mechanism and controlling factors in this area, a database of 72 drill cores, including 56 well-logs and 3 sampling wells, was examined for sedimentology and geochemistry in relation to uranium concentrations. The results show that coal-bearing series can influence uranium mineralization from two aspects, i.e., spatial distribution and dynamic control. Five types of uranium-bearing rocks are recognized, mainly occurring in the braided river and braided delta sedimentary facies, among which sandstones near the coals are the most important. The lithological associations of sandstone-type uranium deposits can be classified into three subtypes, termed as U-coal type, coal-U-coal type, and coal-U type, respectively. The coal and fine siliciclastic rocks in the coalbearing series confined the U-rich fluid flow and uranium accumulation in the sandstone near them. Thus, the coal-bearing series can provide good accommodations for uranium mineralization. Coals and organic matters in the coal-bearing series may have served as reducing agents and absorbing barriers. Methane is deemed to be the main acidolysis hydrocarbon in the U-bearing beds, which shows a positive correlation with U-content in the sandstones in the coal-bearing series. Additionally, the δ13 C in the carbonate cements of the U-bearing sandstones indicates that the organic matters, associated with the coal around the sandstones, were involved in the carbonation, one important component of alteration in the Tuanyushan area. Recognition of the dual control of coal-bearing series on the uranium mineralization is significant for the development of coal circular economy, environmental protection during coal utilization and the security of national rare metal resources. | GUO Biao SHAO Longyi WEN Huaijun HUANG Guangnan ZOU Mingjun LI Yonghong | 2018 | Acta Geologica Sinica(English Edition)2018,92,2: | 2

Effect Evaluation and Intelligent Prediction of Power Substation Project Considering New Energy显示文摘The evaluation of the implementation effect of the power substation project can find out the problems of the project more comprehensively, which has important practical significance for the further development of the power substation project. To ensure accuracy and real-time evaluation, this paper proposes a novel hybrid intelligent evaluation and prediction model based on improved TOPSIS and Long Short-Term Memory(LSTM)optimized by a Sperm Whale Algorithm(SWA).Firstly, under the background of considering the development of new energy, the influencing factors of power substation project implementation effect are analyzed from three aspects of technology, economy and society. Moreover,an evaluation model based on improved TOPSIS is constructed. Then,an intelligent prediction model based on SWA optimized LSTM is designed. Finally, the scientificity and accuracy of the proposed model are verified by empirical analysis, and the important factors affecting the implementation effect of power substation projects are pointed out. | Huiying Wu Meihua Zou Ye Ke Wenqi Ou Yonghong Li Minquan Ye | 2022 | Computer Modeling in Engineering & Sciences2022,,9: | 1
Wenshen Jianpi recipe(温肾健脾方) induced immune reconstruction and redistribution of natural killer cell subsets in immunological non-responders of human immunodeficiency virus/acquired immune deficiency syndrome:a randomized controlled trial显示文摘OBJECTIVE:To evaluate the effects of the Wenshen Jianpi recipe(温肾健脾方,WJR)on immune reconstruction and natural killer(NK)cells in immunological non-responders(INRs)of people living with human immunodeficiency virus(HIV)(PLWH)and propose new therapeutic strategies for HIV.METHODS:Based on Traditional Chinese Medicine treatment principle“invigorating Qi and warming Yang in the spleen and kidneys”,WJR combined with antiretroviral therapy(ART)therapy was performed in a randomized,double-blind,placebo-controlled study of 60 patients with non-responders.The randomized process was executed by the Clinical Evaluation Center of China Academy of Chinese Medical Sciences.Sixty patients who met the inclusion criteria obtained random numbers(that is the drug number)was randomly divided into a treatment group and a placebo control group according to a 1∶1 ratio.CD4+T cell counts and natural killer(NK)cells counts were evaluated at baseline and 12-week,24-week follow-ups.RESULTS:Four participants received random numbers and did not enter the group due to the patient’s own reasons.A total of 56 patients were enrolled, including 28 in the treatment group and 28 in the control group.CD4+T cell counts in the treatment group were significantly increased at week 24(P=0.01<0.05),which were significantly higher than those in the control group(P=0.01<0.05).Although no significant differences were observed between two groups, the CD56bri CD16-NK cell counts in the treatment group were significantly increased after duration.and CD56dim CD16+NK cell counts in the treatment group were significantly higher than those in the control group after 24 weeks of treatment(P=0.025<0.05).As compared with the control group, the treatment group had significantly lower CD56neg CD16+NK cell counts after 24 weeks of treatment(P=0.023<0.05).CONCLUSIONS:WJR promotes the immune reconstruction of INRs and redistribution of NK cell subsets, notably decreasing CD56neg CD16+NK cell counts in INRs.However, the redistribution of NK cell subsets is not beneficial for immune reconstruction in INRs.Further large-scale RCTs are required to evaluate the effect of WJR on immune recovery in INRs and decipher the underlying mechanism. | TAO Zhuang WANG Jian CHEN Xin LI Yonghong YAN Yuguang ZHANG Ao ZOU Wen LIU Ying | 2022 | Journal of Traditional Chinese Medicine2022,42,5: | 0
含异倍体的结直肠癌高表达CD133且预后不良(英文)显示文摘Objective:The aim of the study was to investigate the relationship of DNA ploidy status in colorectal cancers with patients' prognosis and also the relationship of DNA ploidy status with expression of the colorectal cancer stem cell marker CD133.Methods:The DNA ploidy status and CD133 expression in colorectal cancers were detected by flow cytometry. The clinicopathological characteristics and progression-free survival analysis of patients was evaluated based on the clinical data. Results:DNA ploidy pattern did not correlated with gender, age, lesion diameter, histological type, depth of tumor invasion, lymphatic invasion and Dukes stage.Only primary lesion cite showed significant correlation with DNA ploidy pattern, more aneuploids were observed in colonic cancer than rectal cancer,P < 0.05.The 2-year progression-free survival rate and total progression-free time in patients with aneuploids were lower than that with diploids,P < 0.05.Tumors contained aneuploids showed higher expression of CD133 than tumors of only diploids,P < 0.05.Conclusion:Tumor DNA ploidy status is a significant prognostic factor in patients with colorectal cancer and also associated with the existence of CD133 positive colorectal cancer stem cells. | Dongdong Yu Yonghong Zhang You Zou Ming Tian Deding Tao Junbo Hu Jianping Gong | 2010 | The Chinese-German Journal of Clinical Oncology2010,9,10: | 0

The unconventional prefoldin RPB5 interactor mediates the gravitropic response by modulating cytoskeleton organization and auxin transport in Arabidopsis显示文摘Gravity-induced root curvature involves the asymmetric distribution of the phytohormone auxin. This response depends on the concerted activities of the auxin transporters such as PIN-FORMED(PIN)proteins for auxin efflux and AUXIN RESISTANT 1(AUX1)for auxin influx. However, how the auxin gradient is established remains elusive. Here we identified a new mutant with a short root, strong auxin distribution in the lateral root cap and an impaired gravitropic response. The causal gene encoded an Arabidopsis homolog of the human unconventional prefoldin RPB5 interactor(URI).At URI interacted with prefoldin 2(PFD2)and PFD6,twoβ-type PFD members that modulate actin and tubulin patterning in roots. The auxin reporter DR5_(rev):GFP showed that asymmetric auxin redistribution after gravistimulation is disordered in aturi-1 root tips.Treatment with the endomembrane protein trafficking inhibitor brefeldin A indicated that recycling of the auxin transporter PIN2 is disrupted in aturi-1 roots as well as in pfd mutants. We propose that At URI cooperates with PFDs to recycle PIN2and modulate auxin distribution. | Yi Yang Fang Liu Le Liu Mingyue Zhu Jinfeng Yuan Yan‐Xia Mai Jun‐Jie Zou Jie Le Yonghong Wang Klaus Palme Xugang Li Yong Wang Long Wang | 2022 | Journal of Integrative Plant Biology2022,64,10: | 0
