| 1 | Genetic analysis and fine mapping of an enclosed panicle mutant esp2 in rice (Oryza sativa L.)显示文摘The phenomenon of panicle enclosure in rice is mainly caused by the shortening of uppermost internode.Elucidating the molecular mechanism of panicle enclosure will be helpful for solving the problem of panicle enclosure in male sterile lines and creating new germplasms in rice.We acquired a monogenic recessive enclosed panicle mutant,named as esp2 (enclosed shorter panicle 2),from the tissue culture progeny of indica rice cultivar Minghui-86.In the mutant,panicles were entirely enclosed by flag leaf sheaths and the uppermost internode was almost completely degenerated,but the other internodes did not have obvious changes in length.Genetic analysis indicated that the mutant phenotype was controlled by a recessive gene,which could be steadily inherited and was not affected by genetic background.Apparently,ESP2 is a key gene for the development of uppermost internode in rice.Using an F 2 population of a cross between esp2 and a japonica rice cultivar Xiushui-13 as well as SSR and InDel markers,we fine mapped ESP2 to a 14-kb region on the end of the short arm of chromosome 1.According to the rice genome sequence annotation,only one intact gene exists in this region,namely,a putative phosphatidylserine synthase gene.Sequencing analysis on the mutant and the wild type indicated that this gene was inserted by a 5287-bp retrotransposon sequence.Hence,we took this gene as a candidate of ESP2.The results of this study will facilitate the cloning and functional analysis of ESP2 gene. | GUAN HuaZhong DUAN YuanLin LIU HuaQing CHEN ZhiWei ZHUO Ming ZHUANG LiJun QI WenMing PAN RunSen MAO DaMei ZHOU YuanChang WANG Feng WU WeiRen | 2011 | Chinese Science Bulletin2011,56,14: | 6 |
| 2 | Analysis of Review Helpfulness Based on Consumer Perspective显示文摘When consumers make purchase decisions, they generally refer to the reviews generated by other consumers who have already purchased similar products in order to get more information. Online transaction platforms provide a highly convenient channel for consumers to generate and retrieve product reviews. In addition,consumers can also vote reviews perceived to be helpful in making their decision. However, due to diverse characteristics, consumers can have different preferences on products and reviews. Their voting behavior can be influenced by reviews and existing review votes. To explore the influence mechanism of the reviewer, the review,and the existing votes on review helpfulness, we propose three hypotheses based on the consumer perspective and perform statistical tests to verify these hypotheses with real review data from Amazon. Our empirical study indicates that review helpfulness has significant correlation and trend with reviewers, review valance, and review votes. | 3 | Modeling study of source contributions and emergency control effects during a severe haze episode over the Beijing-Tianjin-Hebei area显示文摘In February 2014, the Beijing-Tianjin-Hebei(BTH) area experienced a weeklong episode of heavy haze pollution. Cities such as Beijing(BJ) and Shijiazhuang(SJZ) issued heavy pollution alerts for the first time and took emergency control measures. This study employed the Nested Air Quality Prediction Modeling System(NAQPMS) to simulate and analyze the three-dimensional structure of the source contributions of PM2.5 in the BTH area during this pollution episode and quantitatively assessed the effects of the emergency control measures. The results showed that during the polluted period(February 19–26), surface PM2.5 mainly originated from local sources(48%–72%). In the entire BTH area, southern Hebei(SHB) represented the largest internal contribution(33%), while the main external contributions came from Shandong(SD)(10%) and Henan(HN)(4%). Vertically, the local contribution was constrained below the near-ground layer, and rapidly decreased with altitude. The regional transport path from SHB and Shanxi(SX) to BJ appeared at 0.5–1.5 and 1.5–2.5 km, with contributions of 32%–42% and 13%–27%, respectively. The non-local source regions for the BTH area were SD below 1 km and mainly SX and HN above 1 km. Compared to the non-polluted period(February 27–28), the contribution from regional transport increased during the polluted period, indicating the key role of regional transport in the pollution formation. The emergency control measures had a relatively large effect on NOx and SO2 concentrations, but a limited effect on PM2.5. The stronger regional transport during the polluted period may have weakened the effects of the local emergency control measures. | 4 | Effect of rifampicin pre-and post-treatment on rotenone-induced dopaminergic neuronal apoptosis and alpha-synuclein expression显示文摘BACKGROUND: Rifampicin inhibits the formation of α-synuclein multimer and protects against 1-methyl-4-phenyl-1,2,3,6-tetrahydropyritine (MPTP)-induced PC12 cell apoptosis.OBJECTIVE: To compare the effect of rifampicin pre-and post-treatment on tyrosine hydroxylase and α-synuclein expression in substantia nigra pars compacta in a rat model of Parkinson’s disease.DESIGN,TIME AND SETTING: A randomized,controlled experiment was performed at the Experimental Animal Center of Sun Yat-sen University North Campus (China) from November 2006 to October 2008.MATERIALS: Rifampicin was purchased from MD,USA;rotenone was purchased from Sigma,USA;mouse anti-rat α-synuclein monoclonal antibody was purchased from B&D,USA;and rabbit anti-rat tyrosine hydroxylase monoclonal antibody was purchased from Chemicon,USA.METHODS: A total of 72 male,Sprague Dawley rats,aged 8 weeks,were randomly assigned to 5 groups: blank control (n = 12),rifampicin (n = 12),rotenone (n = 16),rifampicin pre-treatment (n = 16),and rifampicin post-treatment (n = 16).Parkinson’s disease model rats were established via a subcutaneous injection of rotenone (1.5 mg/kg per day) in the three treatment groups,once a day for 3 successive weeks.Rifampicin (30 mg/kg per day) was intragastrically administered in the rifampicin pre-treatment group 3 days prior to rotenone induction and in the rifampicin post-treatment group 7 days after rotenone induction.Rats were treated with a subcutaneous injection of 1 mL/kg per day sunflower oil in the blank control group and an intragastric injection of 30 mg/kg per day rifampicin in the rifampicin group,once a day for 3 successive weeks in total.MAIN OUTCOME MEASURES: Prior to treatment and in the end of the 3rd week after treatment,the rats were evaluated using the modified neurological severity score.The substantia nigra from the rats was extracted for hematoxylin-eosin staining.Western blot analysis was performed to determine tyrosine hydroxylase and α-synuclein expression.RESULTS: Hematoxylin-eosin staining revealed a significant reduction in the number of substantia nigral neurons in the rotenone group,in addition to neurodegradation,hypopigmentation,and pyknosis.In the rifampicin pre-treatment and post-treatment groups,the number of dopaminergic neurons was significantly increased compared with the rotenone group (P < 0.01),with slight neuronal damage.Compared with the rotenone group,substantia nigral tyrosine hydroxylase expres-sion was significantly increased in the rifampicin pre-treatment and post-treatment groups (P < 0.01),but α-synuclein expression and modified neurological severity scores were significantly decreased (P < 0.01).In addition,the effect of rifampicin in the pre-treatment group was superior to the post-treatment group.There was no significant difference in tyrosine hydroxylase and α-synuclein expression,or in the modified neurological severity scores,between the blank control and rifampicin groups (P > 0.05).CONCLUSION: Rifampicin significantly attenuated neuropathological and behavioral motor deficits induced by rotenone.Moreover,rifampicin enhanced tyrosine hydroxylase expression,but inhibited α-synuclein expression.The effect of rifampicin pre-treatment was superior to rifampicin post-treatment. | Yuanlin Sun Guohua Zhang Jie Xu Shiwen Chen Enxiang Tao Changqing Xu M. | 6 | Genome-wide identification and cold stress-induced expression analysis of the CBF gene family in Liriodendron chinense显示文摘Cold-resistance pathways that operate in model plants such as Arabidopsis thaliana and Oryza sativa have been studied extensively.It has been found that CBF genes play an important role in plant cold resistance.Liriodendron chinense,a tree known for its graceful tree shape and widely spread in south China,has weak cold tolerance.However,little is known about its response to cold.To further study the function of L.chinense CBF gene family,we started by characterizing all members of this gene family in the L.chinense genome and their expression profiling.Phylogenetic analysis found that 14 CBF genes in L.chinense are more closely related to their homologues in woody plants and A.thaliana than those in O.sativa.Cis-acting elements and GO analysis showed that some LcCBF genes participated in the biological process of cold stress response.The transcriptomic and RT-qPCR data showed that most of LcCBF genes displayed an initially increasing and subsequently decreasing trend during cold stress course and the expression profile of each member was different.Some LcCBF genes exhibited a different abundance in callus,root,stem and leaf tissues.The structure and expression characteristics of LcCBF genes imply that they may have similar and different functions in response to cold stress conditions.The identification and analysis of LcCBF gene family have laid the foundation for future studies into L.chinense cold stress mechanisms and for the cultivation of cold-resistance cultivars. | Yuanlin Guan Siqin Liu Weihuang Wu Kaiyue Hong Rongxue Li Liming Zhu Yang Liu Ye Lu Jinhui Chen Liming Yang Jisen Shi | 2021 | Journal of Forestry Research2021,32,6: | 3 |

| 10 | Hymecromone:a clinical prescription hyaluronan inhibitor for efficiently blocking COVID-19 progression显示文摘Currently,there is no effective drugs for treating clinically COVID-19 except dexamethasone.We previously revealed that human identical sequences of SARS-CoV-2 promote the COVID-19 progression by upregulating hyaluronic acid(HA).As the inhibitor of HA synthesis,hymecromone is an approved prescription drug used for treating biliary spasm.Here,we aimed to investigate the relation between HA and COVID-19,and evaluate the therapeutic effects of hymecromone on COVID-19.Firstly,HA was closely relevant to clinical parameters,including lymphocytes(n=158;r=−0.50;P<0.0001),C-reactive protein(n=156;r=0.55;P<0.0001),D-dimer(n=154;r=0.38;P<0.0001),and fibrinogen(n=152;r=0.37;P<0.0001),as well as the mass(n=78;r=0.43;P<0.0001)and volume(n=78;r=0.41;P=0.0002)of ground-glass opacity,the mass(n=78;r=0.48;P<0.0001)and volume(n=78;r=0.47;P<0.0001)of consolidation in patient with low level of hyaluronan(HA<48.43 ng/mL).Furthermore,hyaluronan could directly cause mouse pulmonary lesions.Besides,hymecromone remarkably reduced HA via downregulating HAS2/HAS3 expression.Moreover,89%patients with hymecromone treatment had pulmonary lesion absorption while only 42%patients in control group had pulmonary lesion absorption(P<0.0001).In addition,lymphocytes recovered more quickly in hymecromone-treated patients(n=8)than control group(n=5)(P<0.05).These findings suggest that hymecromone is a promising drug for COVID-19 and deserves our further efforts to determine its effect in a larger cohort. | Shuai Yang Yun Ling Fang Zhao Wei Li Zhigang Song Lu Wang Qiuting Li Mengxing Liu Ying Tong Lu Chen Daoping Ru Tongsheng Zhang Kaicheng Zhou Baolong Zhang Peng Xu Zhicong Yang Wenxuan Li Yuanlin Song Jianqing Xu Tongyu Zhu Fei Shan Wenqiang Yu Hongzhou Lu | 2022 | Signal Transduction and Targeted Therapy2022,7,4: | 2 |
| 11 | Rifampicin inhibits apoptosis in rotenone-induced differentiated PC12 cells by ameliorating mitochondrial oxidative stress显示文摘BACKGROUND:Previous studies have shown that rifampicin exhibits neuroprotective effects,but the precise mechanisms remain unclear.Rifampicin is thought to exert the neuroprotective effect as a hydroxyl free radical scavenger.OBJECTIVE:To investigate the protective effects of rifampicin pretreatment on rotenone-induced mitochondrial oxidative stress in differentiated PC12 cells.DESIGN,TIME AND SETTING:A repeated measure,cell-based study was performed at the Department of Neurology,Second Affiliated Hospital,Sun Yat-sen University,China between December 2007 and November 2008.MATERIALS:PC12 cells were a kind gift from the Physiology Laboratory of Zhongshan Medical School,Sun Yat-sen University,China.Rotenone and rifampicin were purchased from Sigma,USA.METHODS:PC12 cells were differentiated by culturing with 100 ng/mL 7S nerve growth factor for 9 days in Dulbecco's modified Eagle's medium/Nutrient Mix F12(DMEM/F12) supplemented with 10% fetal bovine serum.The cells were assigned to six groups according to various treatment conditions:control,cultured with normal media;rifampicin group,treated with 300 μmol/L rotenone for 26 hours;rotenone group,treated with 2.5 μmol/L rotenone for 24 hours;rifampicin pretreatment groups,pretreated with 100,200,and 300 μmol/L rifampicin for 2 hours,respectively,followed by 2.5 μmol/L rotenone for 24 hours.MAIN OUTCOME MEASURES:Mitochondrial membrane potential was measured by fluorescence microscopy and flow cytometry,respectively,using rhodamine123 staining.Intracellular reactive oxygen species formation was analyzed by flow cytometry using 2',7'-dichlorofluorescin-diacetate staining,and intracellular reduced glutathione was measured with a microplate reader.Cell viability was determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay.Cell apoptosis was detected by Hoechst 33342 staining and flow cytometry.RESULTS:Increased apoptosis in rotenone-induced,differentiated,PC12 cells was accompanied by the loss of mitochondrial transmembrane potential,the formation of reactive oxygen species,and reduced glutathione depletion(P<0.01).Rotenone-induced mitochondrial dysfunction was blocked in a dose-dependent manner by rifampicin(P<0.05 or P<0.01).CONCLUSION:Pretreatment of differentiated PC12 cells with rifampicin blocked rotenone-induced apoptosis by ameliorating mitochondrial dysfunction and oxidative stress. | Shiwen Chen Yuanlin Sun Zhifeng Zeng Enxiang Tao | 2010 | Neural Regeneration Research2010,5,4: | 2 |
