| 1 | Hymecromone:a clinical prescription hyaluronan inhibitor for efficiently blocking COVID-19 progression显示文摘Currently,there is no effective drugs for treating clinically COVID-19 except dexamethasone.We previously revealed that human identical sequences of SARS-CoV-2 promote the COVID-19 progression by upregulating hyaluronic acid(HA).As the inhibitor of HA synthesis,hymecromone is an approved prescription drug used for treating biliary spasm.Here,we aimed to investigate the relation between HA and COVID-19,and evaluate the therapeutic effects of hymecromone on COVID-19.Firstly,HA was closely relevant to clinical parameters,including lymphocytes(n=158;r=−0.50;P<0.0001),C-reactive protein(n=156;r=0.55;P<0.0001),D-dimer(n=154;r=0.38;P<0.0001),and fibrinogen(n=152;r=0.37;P<0.0001),as well as the mass(n=78;r=0.43;P<0.0001)and volume(n=78;r=0.41;P=0.0002)of ground-glass opacity,the mass(n=78;r=0.48;P<0.0001)and volume(n=78;r=0.47;P<0.0001)of consolidation in patient with low level of hyaluronan(HA<48.43 ng/mL).Furthermore,hyaluronan could directly cause mouse pulmonary lesions.Besides,hymecromone remarkably reduced HA via downregulating HAS2/HAS3 expression.Moreover,89%patients with hymecromone treatment had pulmonary lesion absorption while only 42%patients in control group had pulmonary lesion absorption(P<0.0001).In addition,lymphocytes recovered more quickly in hymecromone-treated patients(n=8)than control group(n=5)(P<0.05).These findings suggest that hymecromone is a promising drug for COVID-19 and deserves our further efforts to determine its effect in a larger cohort. | Shuai Yang Yun Ling Fang Zhao Wei Li Zhigang Song Lu Wang Qiuting Li Mengxing Liu Ying Tong Lu Chen Daoping Ru Tongsheng Zhang Kaicheng Zhou Baolong Zhang Peng Xu Zhicong Yang Wenxuan Li Yuanlin Song Jianqing Xu Tongyu Zhu Fei Shan Wenqiang Yu Hongzhou Lu | 2022 | Signal Transduction and Targeted Therapy2022,7,4: | 2 |

| 3 | Amniotic Membrane Transplantation for Conjunctival Tumor显示文摘Purpose: To evaluate the possibility of amniotic membrane transplantation (AMT) for the treatment of conjunctival tumor.Methods: Preserved AMT was performed in 26 patients (26 eyes) with conjunctival tumor, including 9 eyes (34.62%) with malignant tumor (conjunctival malignant melanoma,corneal and conjunctival squamous cell carcinoma, conjunctival lymphoma), 17 eyes(65.38%) with benign tumor(conjunctival papilloma, conjunctival dermoid tumor, conjunctival nevus, hemangioma etc.).Result: All the patients are followed up for 1 ~53 months. No acute rejection was observed after preserved AMT. Ideal healing was found in conjunctiva wound.Conclusion: Preserved AMT is a very effective method to repair wound after giant conjunctival tumor operation. | 5 | Features Associated with Recurrence of Idiopathic Orbital Inflammatory Pseudotumor显示文摘Purpose: To explore factors which lead to recurrence of idiopathic orbital inflam-matory pseudotumor (IOIP). Methods: Idiopathic orbital inflammatory pseudotumor in 209 cases between Jan 1, 1978 and Dec 31, 1999 in our hospital was evaluated retrospectively. The comparison of clinical and pathological parameters between patients with at least one episode of recurrence and those with no recurrence at all was performed and analyzed using logistic regression method. Results: Follow-up results (with a mean follow-up time of 3.4 years, ranging from 0.5 year to 21.0 years) showed that the recurrence rate of IOIP was 41%. Sex and proptosis were associated with the recurrence of IOIP. Male gender was more likely to relapse than female counterparts, with the male being 52% and female being 25%. The severer the proptosis is, the higher the rate of recurrence. Among the clinical subtypes of IOIP, the rate of recurrence (17%) in cases with dacryoadinitis was the lowest, followed by anterior local orbital mass (44%), posterior orbital mass (54%), myositis (75%) and diffuse subtype (100%). However, the clinical subtypes did not show significant relationship with the recurrence of IOIP. | 8 | Warming and elevated nitrogen deposition accelerate the invasion process of Solidago canadensis L.显示文摘Background:Invasive species can threaten native diversity and alter ecosystem processes while interacting with other components of global environmental change.Invasive plants are becoming increasingly problematic and this can be stimulated by changes in the environment.However,existing studies have primarily investigated the effects of environmental change on a specific stage of plant invasion rather than the continuous invasion process.Methods:A space-for-time substitution experiment was performed to investigate how warming and nitrogen deposition affects the invasion process of a plant.Specifically,different ratios of invasive Solidago canadensis L.to native Artemisia argyi Levl.et Van were employed as a proxy to represent successive levels of invasion.A total of seven treatments were applied in the experiment:ambient(CK),N addition(+5,+12 g m^(−2)year^(−1)),warming(+1.15,+1.86℃)and their interaction(5 g N m^(−2)year^(−1)+1.15℃,12 g N m^(−2)year^(−1)+1.86℃).The growth performance and competitiveness of S.canadensis were investigated.Results:The competitiveness of Solidago canadensis decreased linearly with its invasion degree(p<0.05).Non-linear regression showed that S.canadensis invasion levels of 53%,53%,68%,55%and 58%were the critical thresholds for shifting the direction or magnitude of chlorophyll,leaf nitrogen,leaf shape index,diameter,and root/shoot ratio,respectively.Compared with the ambient treatment(CK,no warming and no N addition),the diameter,height,bio-mass and relative competitiveness of S.canadensis were each limited by warming,to a certain extent,whereas these and the above parameters were significantly increased by nitrogen deposition.The interaction of increased temperature and nitrogen deposition led to significant increases in the growth and competitiveness of S.canadensis,and this effect was detected in every stage of the invasion,throughout the invasion process.Conclusions:Environmental change might have a continuous,progressive,and augmentative effect on the phenotypic traits of S.canadensis.This study provides fairly robust evidence that environmental change promotes the invasion process of S.canadensis in general,not simply in specific stages.In the future,rather than focusing on specific stages,experimental studies should consider examining invasion on a broader scale. | Guangqian Ren Bin Yang Miaomiao Cui Zhicong Dai Yan Xiang Haiyan Zhang Guanlin Li Jian Li Qaiser Javed Daolin Du | 2022 | Ecological Processes2022,11,1: | 0 |

| 10 | A unified global genotyping framework of dengue virus serotype-1 for a stratified coordinated surveillance strategy of dengue epidemics显示文摘Background: Dengue is the fastest spreading arboviral disease, posing great challenges on global public health. A reproduceable and comparable global genotyping framework for contextualizing spatiotemporal epidemiological data of dengue virus (DENV) is essential for research studies and collaborative surveillance.Methods: Targeting DENV-1 spreading prominently in recent decades, by reconciling all qualified complete E gene sequences of 5003 DENV-1 strains with epidemiological information from 78 epidemic countries/areas ranging from 1944 to 2018, we established and characterized a unified global high-resolution genotyping framework using phylogenetics, population genetics, phylogeography, and phylodynamics.Results: The defined framework was discriminated with three hierarchical layers of genotype, subgenotype and clade with respective mean pairwise distances 2-6%, 0.8-2%, and ≤ 0.8%. The global epidemic patterns of DENV-1 showed strong geographic constraints representing stratified spatial-genetic epidemic pairs of Continent-Genotype, Region-Subgenotype and Nation-Clade, thereby identifying 12 epidemic regions which prospectively facilitates the region-based coordination. The increasing cross-transmission trends were also demonstrated. The traditional endemic countries such as Thailand, Vietnam and Indonesia displayed as persisting dominant source centers, while the emerging epidemic countries such as China, Australia, and the USA, where dengue outbreaks were frequently triggered by importation, showed a growing trend of DENV-1 diffusion. The probably hidden epidemics were found especially in Africa and India. Then, our framework can be utilized in an accurate stratified coordinated surveillance based on the defined viral population compositions. Thereby it is prospectively valuable for further hampering the ongoing transition process of epidemic to endemic, addressing the issue of inadequate monitoring, and warning us to be concerned about the cross-national, cross-regional, and cross-continental diffusions of dengue, which can potentially trigger large epidemics.Conclusions: The framework and its utilization in quantitatively assessing DENV-1 epidemics has laid a foundation and re-unveiled the urgency for establishing a stratified coordinated surveillance platform for blocking global spreading of dengue. This framework is also expected to bridge classical DENV-1 genotyping with genomic epidemiology and risk modeling. | 15 | Involvement of the ammonium assimilation mediated by glutamate dehydrogenase in response to heat stress in the scleractinian coral Pocillopora damicornis显示文摘Glutamate dehydrogenase(GDH)plays an important role in the ammonium assimilation and nitrogen metabolism by catalyzing the reversible oxidative deamination of L-glutamate toα-ketoglutarate.In the present study,the potential functions of GDH in response to heat stre ss were explored in the scleractinian coral Pocillopora damicornis(designated as PdGDH).The cDNA of PdGDH contained an open reading frame of 1611 bp encoding a polypeptide of 536 amino acids,which exhibited the highest sequence identity to GDH of Stylophora pistillata(96%identity),and the deduced PdGDH protein was predicted to contain one GdhA domain(from Val95 to Tyr525).The recombinant protein of PdGDH(rPdGDH)was expressed in Escherichia coli BL21(DE3)-Transetta,and its catalytic activity was measured under different temperatures,pH conditions and epigallocatechin-3-gallate(EGCG,a GDH inhibitor)concentrations.The purified rPdGDH only used reduced coenzyme nicotinamide adenine dinucleotide(NADH)as coenzyme,and its highe st activity was observed at 35℃and pH 7.5,re spectively.The rPdGDH activity was negatively correlated with the concentration of EGCG,and was inhibited by more than half(65%,P<0.05)at 10mol/L EGCG.No significant alteration of PdGDH mRNA expression was detected at 12 h after exposure to heat and ammonium(P>0.05).Furthermore,the activities of NADH-GDH in the scleractinian coral P.damicornis increased significantly at 12 h after the heat and ammonium stress,and the NADH-GDH activity in the heat stress group(32.66 U/mg,P<0.05)was significantly higher than that in the heat and ammonium stress group(11.26 U/mg).These results collectively suggested that PdGDH,as a homologue of glutamate dehydrogenase in the scleractinian coral P.damicornis,could respond to heat stress at the protein level,which would have ability to further promote ammonium assimilation to increase the heat acclimatization of the coral-Symbiodiniaceae symbiotic association. | Jia TANG Wenqi CAI Zhicong YAN Zhongjie WU Qianxi YANG Zhi ZHOU | 2022 | Journal of Oceanology and Limnology2022,40,5: | 0 |
| 16 | mTORC1 induces plasma membrane depolarization and promotes preosteoblast senescence by regulating the sodium channel Scn1a显示文摘Senescence impairs preosteoblast expansion and differentiation into functional osteoblasts,blunts their responses to bone formation-stimulating factors and stimulates their secretion of osteoclast-activating factors.Due to these adverse effects,preosteoblast senescence is a crucial target for the treatment of age-related bone loss;however,the underlying mechanism remains unclear.We found that mTORC1 accelerated preosteoblast senescence in vitro and in a mouse model.Mechanistically,mTORC1 induced a change in the membrane potential from polarization to depolarization,thus promoting cell senescence by increasing Ca^(2+)influx and activating downstream NFAT/ATF3/p53 signaling.We further identified the sodium channel Scn1a as a mediator of membrane depolarization in senescent preosteoblasts.Scn1a expression was found to be positively regulated by mTORC1 upstream of C/EBPα,whereas its permeability to Na^(+)was found to be gated by protein kinase A(PKA)-induced phosphorylation.Prosenescent stresses increased the permeability of Scn1a to Na^(+)by suppressing PKA activity and induced depolarization in preosteoblasts.Together,our findings identify a novel pathway involving mTORC1,Scn1a expression and gating,plasma membrane depolarization,increased Ca^(2+)influx and NFAT/ATF3/p53 signaling in the regulation of preosteoblast senescence.Pharmaceutical studies of the related pathways and agents might lead to novel potential treatments for agerelated bone loss. | Ajuan Chen Jian Jin Shasha Cheng Zezheng Liu Cheng Yang Qingjing Chen Wenquan Liang Kai Li Dawei Kang Zhicong Ouyang Chenfeng Yao Xiaochun Bai Qingchu Li Dadi Jin Bin Huang | 2022 | Bone Research2022,10,2: | 0 |
| 17 | Electrophoretic Determination of Aqueous and Serum Neuron-specific Enolase in the Diagnosis of Retinoblastoma显示文摘Purpose: Neuron-specific enolase (NSE) of containing γ-enolase is considered valuable in the diagnosis of tumours of neuroectodermal origin.Method : We used rapid electrophoretic method on cellulose acetate plate to determine the pattern of enolase isoenzymes in 21 aqueous humor and 23 serum specimens from retinoblastoma (Rb) and 21 aqueous and 25 serum specimens from 25 control cases to evaluate NSE in the diagnosis of Rb.
